| 2 | Clinical curative effect of electric acupuncture on acute cerebral infarction:a randomized controlled multicenter trial显示文摘OBJECTIVE: To examine whether electric acupuncture can improve the daily life of patients with ischemic cerebral apoplexy at acute stage.METHODS: A stratified-block randomized controlled multicenter trial was designed for this study.Totally 340 patients with acute ischemic cerebral apoplexy were randomly divided into an electric acupuncture group and a control group. In the electric acupuncture group, 170 patients were treated with electric acupuncture and routine therapy, and170 patients in the control group with routine therapy alone. Major indexes for judging curative effect were Barthel index at 3- and 6- months follow-up visits and number of re-hospitalized patients. Mi-nor indexes for judging curative effect were change in the score for nervous dysfunction at 4and 12 weeks follow-up visits and number of patients persisting in rehabilitation treatment with acupuncture during follow-up visit.RESULTS: Baseline data at the time of case selection between the two groups were similar. The odds ratio(OR) was 0.92, and the 95% confidence interval(CI) was 0.49-1.73 in disabled rate and 0.73 and 0.51-1.05 in the number of re-hospitalized patients in the electric acupuncture group at 6-month follow up visit compared with the control group.There was no difference in the score for nervous dysfunction at the end of 12-week follow-up visit between the two groups. The score for nervous dysfunction at the end of 4-week treatment in the electric acupuncture group was significantly higher than that in the control group(P<0.05). The number of patients discharged from hospital who persisted in rehabilitation treatment with acupuncture in the acupuncture group was significantly higher than that in the control group.CONCLUSION: Using electric acupuncture to treat patients with acute ischemic cerebral apoplexy can effectively improve the nervous dysfunction scores after 4-week treatment and their ability to deal with daily life after 6-month follow-up visit. | 6 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. | 7 | Effect of Cooling Rate and Vanadium Content on the Microstructure and Hardness of Medium Carbon Forging Steel显示文摘This paper reports the effect of cooling rate on the microstructure and hardness of a kind of medium carbon steel microalloyed with two levels of V content(0.15%and 0.28%)after hot deformation by using single compression tests on a Gleeble-3800 thermal simulator.The results show that cooling rate has a significant effect on the microstructure and hardness of the tested steels.Both the fraction of pearlite and hardness increase with increasing cooling rate,whereas a further increase of the cooling rate above a critical value promotes the formation of acicular ferrite(AF),and thus leads to a decrease of hardness mainly owing to the decrease of pearlite fraction and replacing it by AF and the less effective precipitation strengthening.Increasing V content results in a significant increase of hardness,and this tendency enhances with increasing cooling rate until the formation of AF.Furthermore,increasing V content also significantly enhances the formation of AF structure at a lower cooling rate.The results also suggest that by controlling microstructure,especially the precipitation of fine V(C,N)particles through adjusting postforging cooling,the strengthening and gradient function in one hot-forging part could be obtained. | Weijun Hui Yongjian Zhang Chengwei Shao Silian Chen Xiaoli Zhao Han Dong | 2016 | Journal of Materials Science & Technology2016,32,6: | 7 |

Compared with traditional methods, picosecond laser-induced surface modification provides a fast and facile method for surface modification without chemical damage and environmental pollution. As a result of treatment by 100 ps laser pulses, microstructures are fabricated on the stainless steel sample surface, contributing to the increase of the contact angle from 88° to 105°, which realizes a transformation from hydrophilicity to hydrophobicity. | 12 | Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. | 13 | Amending Research on the Expression of the Contact Force of the Spindle Barrel Finishing Based on EDEM Simulation显示文摘The spindle barrel finishing is commonly used to improve the surface integrity of the important parts of the high-end equipment while it is difficult to provide enough test artifacts for the traditional trial and error experiment to obtain the desirable processing technology.The EDEM simulation of the spindle barrel finishing can provide effective help for the process design,however,the difference between the simulation and experiment is closely related to the selection of the contact model during simulation.In this paper,simulations and experiments are conducted based on the identical apparatus and conditions to facilitate the comparison and validation between each other.Based on the Hertz contact theory,the effect of the material properties of contact objects and the relative position of the workpiece on the contact force is qualified.The expression of the correlation coefficient of the contact model is deduced.Then the formula for calculating the contact force between the barrel finishing abrasive and the workpiece that includes influence coefficient of the material properties and the relative positions is established.Finally,the contact force calculation formula is verified by changing the rotating speed.The result shows that the material correction coefficient ranges from 1.41 to 2.38,which is inversely related to the equivalent modulus E.The position correction coefficient ranges from 2.0 to 2.3.The relative error value between the calculation result and the experimental test result is from 0.58%to 14.07%.This research lay a theoretical foundation for the correction theory of the core elements of the spindle barrel finishing process. | Na Wang Shengqiang Yang Tingting Zhao Bo Cao Chengwei Wang | 2020 | Chinese Journal of Mechanical Engineering2020,33,6: | 1 |
| 14 | Highly efficient CRISPR-SaKKH tools for plant multiplex cytosine base editing显示文摘Base editing, as an expanded clustered regularly interspaced short palindromic repeats(CRISPR)-Cas genome editing strategy, permits precise and irreversible nucleotide conversion. SaKKH, an efficient variant of a Cas9 ortholog from Staphylococcus aureus(SaCas9), is important in genome editing because it can edit sites with HHHAAT protospacer adjacent motif(PAM) that the canonical Streptococcus pyogenes Cas9(SpCas9) or its variants(e.g. xCas9, Cas9-NG) cannot. However, several technical parameters of SaKKH involved base editors have not been well defined and this uncertainty limits their application. We developed an effective multiplex cytosine base editor(SaKKHn-pBE) and showed that it recognized NNARRT, NNCRRT, NNGRGT, and NNTRGT PAMs. Based on 27 targets tested, we defined technical parameters of SaKKHn-pBE including the editing window, the preferred sequence context, and the mutation type. The editing efficiency was further improved by modification of the SaKKH sgRNA. | 17 | Nano silica aerogel-induced formation of an organic/alloy biphasic interfacial layer enables construction of stable high-energy lithium metal batteries显示文摘Lithium metal batteries represent promising candidates for high-energy-density batteries, however, many challenges must still be overcome,e.g., interface instability and dendrite growth. In this work, nano silica aerogel was employed to generate a hybrid film with high lithium ion conductivity(0.6 mS cm^(-1)at room temperature) via an in situ crosslinking reaction. TOF-SIMS profile analysis has revealed conversion mechanism of hybrid film to Li–Si alloy/Li F biphasic interface layer, suggesting that the Li–Si alloy and Li F-rich interface layer promoted rapid Li+transport and shielded the Li anodes from corrosive reactions with electrolyte-derived products. When coupled with nickel-cobalt-manganese-based cathodes, the batteries achieve outstanding capacity retention over 1000 cycles at 1 C. Additionally the developed film coated on Li enabled high coulombic efficiency(99.5%) after long-term cycling when coupled with S cathodes.
