1Therapeutic effect of acupuncture and massage for shoulder-hand syndrome in hemiplegia patients:a clinical two-center randomized controlled trial显示文摘OBJECTIVE:To evaluate the therapeutic effects of acupuncture and massage for shoulder-hand syndrome in hemiplegia patients.METHODS:One hundred and twenty hemiplegia patients with stage I shoulder-hand syndrome were randomly divided into a group treated with standardized electric acupuncture and massage,and a group treated with rehabilitation therapy for 6 weeks.The primary indices evaluated were pain on passive movement of the shoulder using the numeric pain rating scale(NPRS),and the number of patients with shoulder-hand syndrome at Steinbrocker stage II or III after treatment.The secondary indices were Fugl-Meyer evaluation of functional movement of the upper limb and hand using the modified rankin scale(MRS).RESULTS:At post-treatment evaluation and a 12-week follow-up visit,NPRS score,number of patients with stage II or III shoulder-hand syndrome,and MRS score were all improved in the acupuncture-massage group compared with the rehabilitation group(P<0.05).On Fugl-Meyer evaluation,functional movement of the upper limb was also improved in the acupuncture-massage group compared with the rehabilitation group(P<0.05).CONCLUSION:Standardized acupuncture-massage therapy may have curative effects on shoulder-hand syndrome in hemiplegia patients.Ning Li Fengwei Tian Chengwei Wang Pengming Yu Xi Zhou Qian Wen Xiulan Qiao Lu Huang 2012Journal of Traditional Chinese Medicine2012,32,3:25

4Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 5Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. 13A robust method for identification of plant SUMOylation substrates in a library-based reconstitution system显示文摘Dear Editor,SUMOylation,which transfers a small ubiquitin-like protein to the lysine residues of target proteins,is an important type of posttranslational modification in eukaryotic cells.This modification plays a critical role in plant development and stress responses(Miura and Hasegawa,2010);thus,the efficient identification of SUMOylation substrates in plant cells is a fundamental issue in the field.In Arabidopsis,hundreds of SUMOylated proteins have been identified by a mass spectrometry(MS)approach in which modified targets in transgenic plants harboring an H89R variant of SUMO1 are enriched via affinity chromatography and a four-residue footprint is left on the substrates after trypsin digestion for MS analysis(Miller et al.,2010).Given that antibodies generated against SUMO molecules are not very specific,it is impractical to catch endogenous SUMO moieties via antibody-based one-step purification.Thus,it is difficult to obtain SUMOylation substrates via existing approaches in plant species that cannot be efficiently transformed.Given that SUMOylation is highly reversible(Yates et al.,2016),another problem is that the modification may be removed quickly during sample preparation.In addition,based on the efficiency and sensitivity of affinity enrichment and MS,substrates with low protein levels may be missed in the analyses.Ruiqiang Lai Wenliang Li Zewei Xu Wen Liu Qianrui Zeng Wenxiong Lin Jieming Jiang Jianbin Lai Chengwei Yang 2023Plant Communications2023,4,4:0
14Starved lubrication analysis of angular contact ball bearing based on a multi-degree-of-freedom tribo-dynamic model显示文摘When the oil supply is not adequate to maintain the ideal lubrication,angular contact ball bearing will enter into the starved lubrication regime resulting in the potential performance degradation and consequently the severe failures.To study the effects of starved lubrication on the performance of angular contact ball bearing,this paper first proposes a multi-degree-of-freedom(DOF)tribo-dynamic model by introducing five-DOF inner ring,six-DOF balls,and six-DOF cage.The model considers the starved lubrication in the ball-raceway contact and the full multi-body interactions between the bearing components.With different ball-raceway starvation degrees being analyzed,the effects of starved lubrication on the bearing tribo-dynamic performance are first revealed.By comparison,it is found that the oil film thickness,the skidding performance,and the traction forces in the ball-raceway contact are significantly influenced by the starvation degrees.It is also found that the starvation-induced change of the ball-pocket contact force is dramatical under combined loads,and the maximum contact force under this load condition increases with the increasing starvation degrees.Chengwei WEN Xianghui MENG Jiaming GU Lin XIAO Shuang JIANG Hanbing BI 2023Friction2023,11,8:0
15Electroactive and antibacterial wound dressings based on Ti_(3)C_(2)T_(x)MXene/poly(ε-caprolactone)/gelatin coaxial electrospun nanofibrous membranes显示文摘Endogenous electric fields(EFs)are capable of regulating the behaviors of skin cells in wound healing.However,majority of current dressings are primarily engaged in the passive repair of defective tissue,as they lack the ability to actively respond to physiological electrical signals.In this work,a series of nanofibrous membranes(NFMs)were fabricated by coaxial electrospinning,combining the good mechanical properties of poly(ε-caprolactone)(PCL),the bioactivity of gelatin and the electroactivity of Ti_(3)C_(2)T_(x)MXene,as electroactive and antibacterial dressings for cutaneous wound healing.The obtained NFMs exhibited suitable mechanical properties and hydrophilicity,excellent electroactivity,antibacterial activity,and biocompatibility.Especially,Ti_(3)C_(2)T_(x)MXene/PCL/gelatin-6(MPG-6,6 wt.%of Ti_(3)C_(2)T_(x)MXene in sheath spinning liquids)showed the optimal conductivity and antibacterial activity.Excitingly,this scaffold significantly promoted the adhesion,proliferation,and migration of NIH 3T3 cells under the electrical stimulation(ES).The in vivo evaluation in a full-thickness wounds defect model demonstrated that the MPG-6 films significantly accelerated wound closure,increased granulation tissue formation,increased collagen deposition,and promoted wound vascularization.In summary,the versatile scaffold is expected to be an ideal candidate as wound dressings due to its ability to promote the transmission of physiological electrical signals and thus improved the therapeutic outcomes of wound regeneration.Shixin Xu Chen Du Miaomiao Zhang Ruoying Wang Wei Feng Chengwei Wang Qinsong Liu Wen Zhao 2023Nano Research2023,16,7:0
