1Generation of transgenic wheat resistant to wheat yellow mosaic virus and identification of gene silence induced by virus infection显示文摘The plasmid containing the promoter Actl, the coat protein (cp) gene of wheat yellow mosaic virus (WYMV) and the selectable bar gene, was delivered via particle bombardment, directly into immature embryos of a wheat culti-vars. PCR and PCR-RFLP were employed to screen the existence of the cp gene in T0 and T1 generations. Seeds from the positive T1 plants were sowed in fields heavily contaminated with WYMV to detect their resistance. In field trial of virus infection, one of the transgenic wheat lines, P8-T2, exhibited highly disease-resistance. Western blot and RT-PCR analysis showed that the expression level of cp gene in the resistant transgenic line was reduced greatly compared to those susceptible to WYMV infection. 3Exploration on Separation of Periplaneta Americana Small Peptides with Tricine-SDS-PAGE System显示文摘[Objective] This study aimed to explore a Tricine-SDS-PAGE method for separation and purification of Periplaneta Americana small peptides. [Method] By comparing the separation effects of three types of gels with different ratios on P. Americana small peptides, an appropriate Tricine-SDS-PAGE method for separation of small peptides was established. [Result] Standard low molecular weight Marker was preliminarily separated to a certain extent with polyacrylamide gel electrophoresis, while the sample mixture was not separated due to the different levels of aggregation in separating gel, spacer gel and stacking gel. 5Genetic investigation of haemoglobinopathies in a large cohort of asymptomatic individuals reveals a higher carrier rate for b-thalassaemia in Sichuan Province (Southwestern China)显示文摘The incidence of haemoglobinopathy is high in China,especially south of the Yangtze River.However,the exact status of haemoglobinopathy in Sichuan is unknown.To carry out a detailed research of haemoglobinopathy in individuals living in Sichuan,13,298 subjects without clinical symptoms who were living in Sichuan Province,with an age distribution of 5e73 years,were included in this study.Between March 2014 and July 2017,these subjects received examinations at the Medical Lab of Chengdu Women’s&Children’s Central Hospital.Mean corpuscular volume(MCV)<82 fL or mean corpuscular haemoglobin(MCH)<27 pg was used to indicate haemoglobinopathy carriers.Abnormal haemoglobin was screened by electrophoresis,and genes were sequenced to identify genotypes.Genotype diagnosis of alpha-and beta-thalassaemia was carried out by using PCR and shunt hybridization.There were 638 suspected haemoglobinopathy carriers(4.80%,638/13,298).DNA sequencing identified 6 subjects with abnormal haemoglobin genotypes and 15 subjects with Hb E.The frequency of heterozygosity for thalassaemia was 4.12%(1.48%for α-thalassaemia and 2.61%for β-thalassaemia)in Sichuan Province.The mutation spectrum of α-thalassaemia consisted of the five most common mutations:–^(SEA),-α^(3.7),-α^(4.2),α^(CS),and α^(QS).Seven types of β-thalassaemia mutation were found in this study:CD41-42(-TTCT)was the most frequent(28.47%),followed by 17(A>T),28(A>G),and IVS-Ⅱ-654(C>T).The main abnormal haemoglobin genotype(HbE)and thalassaemia genotype(–^(SEA),CD41-42(-TTCT))were consistent with those in other regions of China,but the carrier rate of β-thalassaemia in Sichuan was higher than that of α-thalassaemia.Xia Yu Min Lin Chenggui Liu Zhiyong Liao Yongqiong Wei Rui Liu Jing Zhu 2021Genes & Diseases2021,8,2:1

7Functional analysis of beet necrotic yellow vein virus (BNYVV) RNA4 in fungal transmission显示文摘Full-length RNA4 of beet necrotic yellow vein virus (BNYVV) and its mutants, including a frame-shift and deletions within the coding region, were inserted into a transcription plasmid. Transcripts derived from these plasmids and RNAs extracted from a BNYVV isolate containing only RNAs 1, 2 and 3 were coinoculated to Tetragona expansa. Then these recombinant isolates were inoculated to sugar beet plants grown in a sand culture system containing Po-lymyxa betae that had been freed of virus particles to test efficiency of BNYVV transmission. The result showed that the efficiency of BNYVV transmission among the plants by Polymyxa betae was highly decreased by different deletions in the coding region of RNA4, but not affected by the frame-shift mutant with four nucleotides insertion. 8A Comparative Study on Four Staining Methods for Antitumor Active Fraction of Periplaneta americana显示文摘[Objective] This study aimed to compare four staining methods for proteins of SDS-polyacrylamide gel electrophoresis(SDS-PAGE) and explore a suitable staining method for the antitumor active fraction of P.americana after polyacrylamide gel electrophoresis.[Method] BSA was used as the standard for the comparison of Coomassie brilliant blue staining method,potassium staining method,calcium staining method and silver staining method,on the basis,antitumor active fraction samples of P.americana were used for SDS-PAGE electrophoresis and staining.[Result] The results showed that silver staining method could be accurately,quickly and easily used for SDS-PAGE staining of the antitumor active fraction of P.americana.[Conclusion] This study laid the foundation for exploring the medicinal value of P.americana.Shaohui WU Chenggui ZHANG Xiumei WU Guangming LIU 2012Agricultural Biotechnology2012,1,1:0

10Caspase-1 and Gasdermin D Afford the Optimal Targets with Distinct Switching Strategies in NLRP1b Inflammasome-Induced Cell Death显示文摘Inflammasomes are essential complexes of innate immune system, which form the first line of host defense against pathogens. Mounting evidence accumulates that inflammasome signaling is highly correlated with coronavirus disease 2019 (COVID-19). However, there remains a significant gap in our understanding of the regulatory mechanism of inflammasome signaling. Combining mathematical modeling with experimental analysis of NLRP1b inflammasome signaling, we found that only the expression levels of caspase-1 and GSDMD have the potential to individually switch cell death modes. Reduction of caspase-1 or GSDMD switches cell death from pyroptosis to apoptosis. Caspase-1 and GSDMD present different thresholds and exert distinct pathway choices in switching death modes. Pyroptosis switches to apoptosis with an extremely low threshold level of caspase-1, but with a high threshold of GSDMD. Caspase-1-impaired cells employ ASC-caspase-8-dependent pathway for apoptosis, while GSDMD-impaired cells primarily utilize caspase-1-dependent pathway. Additionally, caspase-1 and GSDMD can severally ignite the cooccurrence of pyroptosis and apoptosis. Landscape topography unravels that the cooccurrence is dramatically different in caspase-1- and GSDMD-impaired cells. Besides pyroptosis state and apoptosis state, a potential new “coexisting” state in single cells is proposed when GSDMD acts as the driving force of the landscape. The “seesaw model” is therefore proposed, which can well describe the death states that are controlled by caspase-1 or GSDMD in single cells. 11Effect of the RNA5 component on pathogenicity of beet necrotic yellow vein virus显示文摘The transcripts of beet necrotic yellow virus RNA5 were prepared from the cloned cDNA in a bacterio-phage T7 in vitro run-off transcription system. The RNA5 transcripts and RNAs extracted from two BNYVV mutants, Hu0 containing RNA1, 2 and Hu3 containing RNA1, 2, 3 respectively, were co-inoculated to Tetragonia expansa and sugar beet seedlings. The results indicated that the virus infections and accumulation in the hosts could be improved by presence of RNA5. Cooperating with RNA3, the RNA5 seems to cause more severe symptom and yield losses in sugar beet.
