6Responses of ground-dwelling spiders to four hedgerow species on sloped agricultural fields in Southwest China显示文摘Spiders are important predators in agroecosystems and contribute to the control of agricultural pest populations. Habitat manage- ment such as the creation of new semi-natural habitats around or within fields enhances spider abundance and species diversity. Using pitfall traps, we investigated the effects of four hedgerow plant species, which serve as undisturbed and permanent semi-natural habitats, on ground-dwelling spider activity density (a parameter of population density and relative activity) and species richness. Samples were collected over two winter wheat and two summer maize growing seasons during 2005-2007 in trial field 1 (slope gradient of 20%) and field 2 (slope gradient of 12%) at the Ziyang Experimental Site in Sichuan Province, China. The hedgerow species evaluated were Amorpha fruticosa (field 1), Vetiveria zizanioides (field 1), Eulaliopsis binata (field 2), and Medicago sativa (field 2). Compared to control plots, hedgerow plots had significantly higher activity density and species richness of ground-dwelling spiders within strips in both fields 1 and 2 during the wheat growing season. The presence of hedgerow strips did not augment the activity density and species richness of ground-dwelling spiders within the crop fields during the wheat or maize growing seasons. The ground-dwelling spider activity density within hedgerow strips was significantly higher in the Vetiveria than in the Amorpha plots and in the Medicago than in the Eulaliopsis plots, and the species richness was significantly higher in the Vetiveria than in the Amorpha plots during the wheat season. Our results suggest that hedgerows may serve as important overwintering sites for ground-dwelling spiders during the wheat growing season. In addition, the diversification of agroecosystems by using hedgerow strips may be a viable strategy for maintaining ground-dwelling spider populations in agricultural areas. 12Diagnostic performance of different specimens in detecting enterovirus A71 in children with hand,foot and mouth disease显示文摘Hand,foot and mouth disease(HFMD)is a major public health problem among children in the Asia-Pacific region.The optimal specimen for HFMD virological diagnosis remains unclear.Enterovirus A71(EV-A71)neutralizing antibody titres detected in paired sera were considered the reference standard for calculating the sensitivity,specificity,positive and negative predictive value of throat swabs,rectal swabs,stool,blood samples and cerebrospinal fluid(CSF)by RT-PCR or ELISA assay.In this study,clinical samples from 276 HFMD patients were collected for analysing the sensitivity of different kind of specimens.Our results showed that stool had the highest sensitivity(88%,95%CI:74%–96%)and agreement with the reference standard(91%).The order of diagnostic yield for EV-A71 infection was stool samplerectal swab>throat swab>blood sample>CSF sample,and using a combination of clinical samples improved sensitivity for enterovirus detection.The sensitivity of ELISA for IgM antibody detection in sterile-site specimens was significantly higher than that of RT-PCR(serum/plasma:62%vs.2%,CSF:47%vs.0%)(P<0.002).In conclusion,our results suggest that stool has the highest diagnostic yield for EV-A71-infected HFMD.If stool is unavailable,rectal swabs can be collected to achieve a similar diagnostic yield.Otherwise,throat swabs may be useful in detecting positive samples.Although IgM in blood or CSF is diagnostically accurate,it lacks sensitivity,missing 40%–50%of cases.The higher proportion of severe cases and shorter interval between onset and sampling contributed to the increase in congruency between clinical testing and the serological reference standard.Yonghong Zhou Chongchen Zhou Kai Wang Qi Qiu Yibing Cheng Yu Li Peng Cui Lu Liang Peng Li Xiaowei Deng Lili Wang Wen Zheng Hui Gong Fang Wang Meng Xu Justin Jang Hann Chu Lance Turtle Hongjie Yu 2023Virologica Sinica2023,38,2:1

18A non-ACE2 competing human single-domain antibody confers broad neutralization against SARS-CoV-2 and circulating variants显示文摘The current COVID-19 pandemic has heavily burdened the global public health system and may keep simmering for years.The frequent emergence of immune escape variants have spurred the search for prophylactic vaccines and therapeutic antibodies that confer broad protection against SARS-CoV-2 variants.Here we show that the bivalency of an affinity maturated fully human singledomain antibody(n3113.1-Fc)exhibits exquisite neutralizing potency against SARS-CoV-2 pseudovirus,and confers effective prophylactic and therapeutic protection against authentic SARS-CoV-2 in the host cell receptor angiotensin-converting enzyme 2(ACE2)humanized mice.The crystal structure of n3113 in complex with the receptor-binding domain(RBD)of SARS-CoV-2,combined with the cryo-EM structures of n3113 and spike ecto-domain,reveals that n3113 binds to the side surface of up-state RBD with no competition with ACE2.The binding of n3113 to this novel epitope stabilizes spike in up-state conformations but inhibits SARS-CoV-2 S mediated membrane fusion,expanding our recognition of neutralization by antibodies against SARS-CoV-2.Binding assay and pseudovirus neutralization assay show no evasion of recently prevalent SARS-CoV-2 lineages,including Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2)for n3113.1-Fc with Y58L mutation,demonstrating the potential of n3113.1-Fc(Y58L)as a promising candidate for clinical development to treat COVID-19.Zhenlin Yang Yulu Wang Yujia Jin Yuanfei Zhu Yanling Wu Cheng Li Yu Kong Wenping Song Xiaolong Tian Wuqiang Zhan Ailing Huang Shanshan Zhou Shuai Xia Xiaoxu Tian Chao Peng Cuicui Chen Yibing Shi Gaowei Hu Shujuan Du Yuyan Wang Youhua Xie Shibo Jiang Lu Lu Lei Sun Yuanlin Song Tianlei Ying 2021Signal Transduction and Targeted Therapy2021,6,12:1
