| 1 | Polycyclic aromatic hydrocarbon in urban soil from Beijing, China显示文摘Polycyclic aromatic hydrocarbons (EPA-PAHs) in the urban surface soils from Beijing were determined using gas chromatography and mass spectrometry (GC-MS). It is significantly complementary for understanding the PAHs pollution in soil of integrated Beijing city on the basis of the information known in the outskirts. The total concentration of 16 EPA-PAH was from 0.467 to 5.470 ?滋g/g and was described by the contour map. Compound profiles presented that the 4-, 5- and 6-ring PAHs were major compositions. The correlation analysis showed that PAHs have the similar source in the most sampling sites and BaP might be considered as the indicator of PAHs. Characteristic ratios of anthracene (An)/(An+ phenanthrene (Phe)), fluoranthene (Flu)/(Flu+ pyrene (Pyr)) and benzo[a]pyrene (BaP)/benzo[g,h,i]perylene (BghiP) indicated that the PAHs pollutants probably mainly originated from the coal combustion and it was not negligible from vehicular emission. | 4 | Dietary resveratrol improves antioxidant status of sows and piglets and regulates antioxidant gene expression in placenta by Keap1-Nrf2 pathway and Sirt1显示文摘Background: Resveratrol, a plant phenol, affords protection against inflammation and oxidative stress. The objective of this study was to investigate the effects of dietary resveratrol supplementation during pregnancy and lactation on the antioxidant status of sows and piglets and on antioxidant gene expression and pathway in placenta.Methods: Forty sows were allotted to 2 dietary treatments 20 d after breeding. Sows were fed a control diet and a control diet with 300 mg/kg resveratrol. Oxidative stress biomarkers and antioxidant enzymes were measured in the placenta, milk, and plasma of sows and piglets. Antioxidant gene expression and protein expression of Kelch-like ECH-associated protein 1-Nuclear factor E2-related factor 2(Keap1-Nrf2), nuclear factor kappa B-p65(NFκB-p65) and sirtuin1(Sirt1) were quantified in the placenta.Results: Dietary resveratrol increased the litter and piglets weaning weights. Antioxidant status in the milk, placenta and plasma of sows and piglets was partially improved by dietary resveratrol. In placenta, Nrf2 protein expression was increased and Keap1 protein expression was decreased by dietary resveratrol. The m RNA expression of antioxidant genes including catalase(CAT), glutathione peroxidase 1(GPX1), GPX4, superoxide dismutase 1(SOD1)and heme oxygenase 1(HO1), and phase 2 detoxification genes, including glutamate-cysteine ligase modifier(GCLM), microsomal glutathione S-transferase 1(MGST1) and UDP glucuronosyltransferase family 1 member A1(UGT1 A1), was increased by dietary resveratrol. Dietary resveratrol also increased Sirt1 and phosphorylated NFκB-p65 protein expression in the placenta. We failed to observe any influences of dietary resveratrol on pro-inflammatory cytokine levels, including those of interleukin 1β(IL-1β), IL-6, IL-8 and tumor necrosis factor α(TNF-α). However, we observed that the m RNA expression of IL-8 in placenta was reduced by maternal resveratrol. In addition, dietary resveratrol showed interactive effects with day of lactation on activities of SOD and CAT and levels of malonaldehyde(MDA) and hydrogen peroxide(H2 O2) in milk.Conclusions: Dietary resveratrol supplementation during pregnancy and lactation improves the antioxidant status of sows and piglets, which is beneficial to the reproductive performance of sows. | 7 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |

| 10 | Survival of transplanted neurotrophin-3 expressing human neural stem cells and motor function in a rat model of spinal cord injury显示文摘<正>BACKGROUND:Many methods have been attempted to repair nerves following spinal cord injury, including peripheral nerve transplantation,Schwann cell transplantation,olfactory ensheathing cell transplantation,and embryonic neural tissue transplantation.However,there is a need for improved outcomes. OBJECTIVE:To investigate the repair feasibility for rat spinal cord injury using human neural stem cells(hNSCs) genetically modified by lentivirus to express neurotrophin-3. DESIGN,TIME AND SETTING:In vitro cell biological experiment and in vivo randomized,controlled, genetic engineering experiment were performed at the Third Military Medical University of Chinese PLA and First People's Hospital of Yibin,China from March 2006 to December 2007. MATERIALS:A total of 64 adult,female,Wistar rats were used for the in vivo study.Of them,48 rats were used to establish models of spinal cord hemisection,and were subsequently equally and randomly assigned to model,genetically modified hNSC,and normal hNSC groups.The remaining 16 rats served as normal controls. METHODS:hNSCs were in vitro genetically modified by lentivirus to secrete both green fluorescence protein and neurotrophin-3.Neurotrophin-3 expression was measured by Western blot. Genetically modified hNSC or normal hNSC suspension(5×10~5) was injected into the rat spinal cord following T_(10) spinal cord hemisection.A total of 5μL Dulbecco's-modified Eagle's medium was infused into the rat spinal cord in the model group.Transgene expression and survival of transplanted hNSCs were determined by immunohistochemistry.Motor function was evaluated using the Basso,Beattie,and Bresnahan(BBB) scale. MAIN OUTCOME MEASURES:The following parameters were measured:expression of neurotrophin-3 produced by genetically modified hNSCs,transgene expression and survival of hNSCs in rats,motor function in rats. RESULTS:hNSCs were successfully genetically modified by lentivirus to stably express neurotrophin-3.The transplanted hNSCs primarily gathered at,or around,the injection site two weeks following transplantation,and gradually migrated towards the surrounding tissue. Transplanted hNSCs were observed 7.0-8.0 mm away from the injection site.In addition,hNSCs were observed 10 weeks after transplantation.At week 4,BBB locomotor scores were significantly greater in the genetically modified hNSC and normal hNSC groups,compared with the model group (P<0.05),and scores were significantly greater in the genetically modified hNSC group compared with the normal hNSC group(P<0.05). | 11 | Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. | 12 | Responses of soil nitrogen, phosphorous and organic matter to vegetation succession on the Loess Plateau of China显示文摘Revegetation is a traditional practice widely used for soil protection. We evaluated the effect of natural revegetation succession on soil chemical properties and carbon fractions(particulate organic carbon(POC), humus carbon(HS-C), humic acid carbon(HA-C) and fulvic acid carbon(FA-C)) on the Loess Plateau of China. The vegetation types, in order from the shortest to the longest enclosure duration, were:(a) abandoned overgrazed grassland(Ab G3; 3 years);(b) Hierochloe odorata Beauv.(Hi O7; 7 years);(c) Thymus mongolicus Ronnm(Th M15; 15 years);(d) Artemisia sacrorum Ledeb(At S25; 25 years);(e) Stipa bungeana Trin Ledeb(St B36; 36 years) and(f) Stipa grandis P. Smirn(St G56; 56 years). The results showed that the concentrations of soil organic carbon, total nitrogen and available phosphorus increased with the increase of restoration time except for Th M15. The concentration of NH4-N increased in the medium stage(for Th M15 and At S25) and decreased in the later stage(for St B36 and St G56) of vegetation restoration. However, NO3-N concentration significantly increased in the later stage(for St B36 and St G56). Carbon fractions had a similar increasing trend during natural vegetation restoration. The concentrations of POC, HS-C, FA-C and HA-C accounted for 24.5%–49.1%, 10.6%–15.2%, 5.8%–9.1% and 4.6%–6.1% of total carbon, respectively. For Ab G3, the relative changes of POC, HS-C and FA-C were significantly higher than that of total carbon during the process of revegetation restoration. The higher relative increases in POC, HS-C and FA-C confirmed that soil carbon induced by vegetation restoration was sequestrated by higher physical and chemical protection. Methods:Diabetic rat models were established by inducing diabetes via intra-peritoneal injection of STZ.Rats were randomly divided into normal(control),diabetic(DM),and DM+ puerarin groups.After intra-gastric administration of puerarin(500 mg/kg/day for 4 weeks),levels of SOD and malondialdehyde(MDA) were determined in serum and retina.mRNA and protein expression levels of RAGE and VEGF in retinas were determined by real-lime polymerase chain reaction(RT-PCR)(mRNA) and Western blot analysis(protein levels).Results:There was significantly lower SOD activity and significantly higher MDA in serum and retinas of the DM group compared with the two other groups(P<0.05).After treatment with puerarin,SOD activity increased and MDA content decreased in this group(P<0.05).mRNA and protein expression levels of RACE and VECF in the DM group were significantly higher than those of the other groups (P<0.05),and decreased after puerarin treatment(P<0.05).Conclusions:Puerarin is able to enhance SOD activity,and inhibit RAGE and VEGF expressions in retinas of STZ-induced early diabetic ruts. | Fang Chen Hong-Quan Zhang Jun Zhu Kai-Yang Liu Hong Cheng Guo-Li Li Shan Xu Wei-Hong Lv Zheng-Gao Xie | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,11: | 16 |
| 14 | Double-balloon enteroscopy for obscure gastrointestinal bleeding: A single center experience in China显示文摘AIM: To evaluate the diagnostic value of double-balloon enteroscopy (DBE) for obscure gastrointestinal bleeding (OGIB). METHODS: The data about 75 OGIB patients who underwent DBE in January 2007-June 2009 in our hospital were retrospectively analyzed. RESULTS: DBE was successfully performed in all 75 patients without complication. Of the 75 patients, 44 (58.7%) had positive DBE findings, 22 had negative DBE findings but had potential bleeding at surgery and capsule endoscopy, etc . These 66 patients were finally diagnosed as OGIB which was most commonly caused by small bowel tumor (28.0%), angiodysplasia (18.7%) and Crohn’s disease (10.7%). Lesions occurred more frequently in proximal small bowel than in distal small bowel (49.3% vs 33.3%, P = 0.047).
