| 1 | Effect of phosphorylation of MAPK and Stat3 and expression of c-fos and c-jun proteins on hepatocarcinogenesis and their clinical significance显示文摘AIM To study the effect of phosphorylation ofMAPK and Stat3 and the expression of c-fos andc-jun proteins on hepatocellular carcinogenesisand their clinical significance.METHODS SP immunohistochemistry was usedto detect the expression of p42/44MAPK, p-Stat3,c-fos and c-jun proteins in 55 hepatocellularcarcinomas (HCC) and their surrounding livertissues.RESULTS The positive rates and expressionlevels of p42/44MAPK, p-Stat3, c-fos and c-junproteins in HCCs were significantly higher thanthose in pericarcinomatous liver tissues (PCLT).A positive correlation was observed between theexpression of p42/44MAPK and c-fos proteins, andbetween p-Stat3 and c-jun, but there was nosignificant correlation between p42/44MAPK and p-Stat3 in HCCs and their surrounding livertissues.CONCLUSION The abnormalities of Ras/Rat/MAPK and JAKs/ Stat3 cascade reaction maycontribute to malignant transformation ofhepatocytes. | 3 | Cidea controls lipid droplet fusion and lipid storage in brown and white adipose tissue显示文摘Excess lipid storage in adipose tissue results in the development of obesity and other metabolic disorders including diabetes,fatty liver and cardiovascular diseases.The lipid droplet(LD)is an important subcellular organelle responsible for lipid storage.We previously observed that Fsp27,a member of the CIDE family proteins,is localized to LD-contact sites and promotes atypical LD fusion and growth.Cidea,a close homolog of Fsp27,is expressed at high levels in brown adipose tissue.However,the exact role of Cidea in promoting LD fusion and lipid storage in adipose tissue remains unknown.Here,we expressed Cidea in Fsp27-knockdown adipocytes and observed that Cidea has similar activity to Fsp27 in promoting lipid storage and LD fusion and growth.Next,we generated Cidea and Fsp27 double-deficient mice and observed that these animals had drastically reduced adipose tissue mass and a strong lean phenotype.In addition,Cidea/Fsp27 double-deficient mice had improved insulin sensitivity and were intolerant to cold.Furthermore,we observed that the brown and white adipose tissues of Cidea/Fsp27double-deficient mice had significantly reduced lipid storage and contained smaller LDs compared to those of Cidea or Fsp27single deficient mice.Overall,these data reveal an important role of Cidea in controlling lipid droplet fusion,lipid storage in brown and white adipose tissue,and the development of obesity. | WU LiZhen ZHOU LinKang CHEN Cheng GONG JingYi XU Li YE Jing LI De LI Peng | 2014 | Science China(Life Sciences)2014,57,1: | 11 |

The growth kinetics of β-NaYF4 NPs includes several stages: nucleation, growth of aNaYF4, Ostwald ripening, size shrinkage and growth. The results prove that the temperatures are preferred to the phase transformation compared with the NaF content when other experimental conditions are unchanged. | 12 | Endoscopic ultrasound-guided vs endoscopic retrograde cholangiopancreatography biliary drainage for obstructed distal malignant biliary strictures: A systematic review and meta-analysis显示文摘BACKGROUND For palliation of malignant biliary obstruction(MBO), the gold-standard method of biliary drainage is endoscopic retrograde cholangiopancreatography(ERCP)with the placement of metallic stents. Endoscopic ultrasound(EUS)-guided drainage is an alternative that is typically reserved for cases of ERCP failure.Recently, however, there have been robust randomized clinical trials(RCTs)comparing EUS-guided drainage and ERCP as primary approaches to MBO.AIM To compare EUS guidance and ERCP in terms of their effectiveness and safety in palliative biliary drainage for MBO.METHODS This was a systematic review and meta-analysis, in which we searched the MEDLINE, Excerpta Medica, and Cochrane Central Register of Controlled Trials databases. Only RCTs comparing EUS and ERCP for primary drainage of MBO were eligible. All of the studies selected provided data regarding the rates of technical and clinical success, as well as the duration of the procedure, adverse events, and stent patency. We assessed the risk of biases using the Jadad score and the quality of evidence using the Grading of Recommendations Assessment,Development and Evaluation criteria.RESULTS The database searches yielded 5920 records, from which we selected 3 RCTs involving a total of 222 patients(112 submitted to EUS and 110 submitted to ERCP). In the EUS and ERCP groups, the rate of technical success was 91.96%n and 91.81%, respectively, with a risk difference(RD) of 0.00%(95%CI:-0.07, 0.07;P = 0.97; I^2 = 0%). The clinical success was 84.81% and 85.53% in the EUS and ERCP groups, respectively, with an RD of-0.01%(95%CI:-0.12, 0.10; P = 0.90; I^2 =0%). The mean difference(MD) for the duration of the procedure was-0.12%(95%CI:-8.20, 7.97; P = 0.98; I^2 = 84%). In the EUS and ERCP groups, there were14 and 25 adverse events, respectively, with an RD of-0.06%(95%CI:-0.23, 0.12; P= 0.54; I^2 = 77%). The MD for stent patency was 9.32%(95%CI:-4.53, 23.18; P =0.19; I^2 = 44%). The stent dysfunction rate was significantly lower in the EUS t group(MD =-0.22%; 95 CI:-0.35,-0.08; P = 0.001; I^2 = 0%).CONCLUSION EUS represents an interesting alternative to ERCP for MBO drainage,demonstrating lower stent dysfunction rates compared with ERCP. | 17 | Exhumation history of the Huangshan granite pluton,southern Anhui Province:New insights from fission-track analysis显示文摘The dating of the uplift onset of the Huangshan pluton in the southern part of Anhui Province associated with the collision between paleo-Pacific Block and the Eurasian Block is a fundamental issue to better understand the uplift mechanisms and the regional tectonic evolution.χ2 values of seven zircon fission track(ZFT) samples collected from the south part of the Huangshan pluton were all<5%.Based on the grain ages of four typical ZFT samples,three thermal overprints ranging in 113-123,72-95 and 49-66 Ma were distinguished respectively using the binomial peak-fitting method in accordance with the tectonic thermal events after south China shifted into circum-Pacific tectonic region.Apatite fission track(AFT) study of the Huangshan pluton shows ages lie between 15±3 and 56±6 Ma with allχ2 values>5%,and all are significantly younger than their host rock formation ages,indicating that the samples have experienced post-formation thermal history.Based on the AFT results and topography characteristics in the Huangshan pluton,three zones with different denudation intensities caused by the differential uplift of the fault blocks were recognized in this paper,indicating that denudation was driven by the increase in elevation significantly.Modeling of the AFT data shows that the earlier cooling phase took place in the early Cenozoic and involved 3.3,3.8 and 6.0°C/Ma of cooling rates equivalent to the exhumation rates of 90,104 and 167 m/Ma in different fault blocks.The last phase of cooling took place from~10 Ma with average cooling rate of~5.6°C/Ma,equivalent to average denudation rates of~156 m/Ma.The fission track results imply that the regional compression field plays an important role for the differential exhumation between individual fault blocks of the Huangshan pluton. | ZHENG Yong YU XinQi YUAN WanMing WANG De'En JIANG GongBing ZHU DeLai WANG Cheng JIA YingYing | 2011 | Science China Earth Sciences2011,54,4: | 3 |

| 20 | Seismic stability of loess tunnels under the effects of rain seepage and a train load显示文摘Loess tunnels are widely used in transportation engineering and are irreplaceable parts of transportation infrastructure. In this paper, a dynamic finite element method is used to analyze the coupled effects of a train vibration load and rainfall seepage. By calculating the variation in the safety factor of a loess tunnel because of the effects of various factors, such as different rainfall intensities and soil thicknesses, the dynamic stability of the loess tunnel is studied under the condition of a near-field pulse-like earthquake. The results show that the security and stability of the tunnel decrease gradually with decreasing burial depth. In addition, the plastic strain of the tunnel is mainly distributed on both sides of the vault and the feet, and the maximum value of the critical strain occurs on both sides of the arch feet. Because of the effects of the train vibration load and rainfall seepage, the safety factor of the loess tunnel structure decreases to a certain degree.
