| 1 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. | 2 | Mycoplasma infections and different human carcinomas显示文摘AIM To explore relationships between human carcinomas and mycoplasma infection.METHODS Monoclonal antibody PD4, which specifically recognizes a distinct protein from mycoplasma hyorhinis, was used to detect mycoplasma infection in different paraffinembedded carcinoma tissues with immunohistochemistry. PCR was applied to amplify the mycoplasma DNA from the positive samples for confirming immunohistochemistry.RESULTS Fifty of 90 cases (56%) of gastric carcinoma were positive for mycoplasma hyorhinis. In other gastric diseases, the mycoplasma infection ratio was 28% (18/49) in chronic superficial gastritis, 30% (14/ 46) in gastric ulcer and 37% (18/ 49) in intestinal metaplasia. The difference is significant with gastric cancer (X2=12.06, P<0.05). In colon carcinoma, the mycoplasma infection ratio was 55.1% (32/58), but it was 20.9% (10/49) in adenomarous polyp (X2=13.46, P<0.005).Gastric and colon cancers with high differentiation had a higher mycoplasma infection ratio than those with low differentiation (P< 0.05). Mycoplasma infection in esophageal cancer, lung cancer, breast cancer and glioma was 50.9% (27/53), 52.6% (31/ 59), 39.7%(25/63) and 41% (38/91), respectively. The mycoplasma DNA was successfully amplified with the DNA extracted from the cancer tissues that were positive for mycoplasma infection (detected with antibody PD4).CONCLUSION There was high correlation between mycoplasma infection and different cancers, which suggests the possibility of an association between the two. | 3 | Establishment of an artificial β-cell line expressing insulin under the control of doxycycline显示文摘AIM: Artificial β-cell lines may offer an abundant source of calls for the treatment of type Ⅰ diabetes, but insulin secretion in β-cells is tightly regulwted in physiological conditions The Tet-On system is a 'gene switch' system,which can induce gene expression by administration of tetracycline (Tet) derivatives such as doxcycline (D ox).Using this system, we established 293 cells to an artificial cell line secreting insulin in response to stimulation by Dox.METHODS: The mutated proinsulin cDNA was obtained fromplasmid pcDNA3.1/C-mlNS by the polymerase chain reaction(PCR), and was inserted downstream from the promoter onthe expression vector pTRE2, to construct a recombinedexpression vector pTRE2mlNS. The promoter on pTRE2consists of the tetracycline-response element and the CMVminimal promoter and is thus activated by the reversetetracycline-controlled transactivator (rtTA) when Dox isadministrated. pTRE2mlNS and plasmid pTK-Hyg encodinghygromycin were co-transfected in the tet293 cells, whichexpress rtTA stably. Following hygromycin screening, thesurvived cells expressing insulin were selected andenriched. Dox was used to control the expression of insulinin these cells. At the levels of mRNA and protein, theregulating effect of Dox in culture medium on the expressionof proinsulin gene was estimated respectively with Northernblot, RT-PCR, and radioimmunoassay.RESULTS: From the 28 hygromycin-resistant cell strains, weselected one cell strain (tet293/Ins6) secreting insulin notonly automatically, but in response to stimulation by Dox.The amount on insulin secretion was dependent on the Doxdose (0,10,100,200,400,800 and 1000 μg@ L-1 ), the level ofinsulin secreted by the cells treated with Dox ( 1000μg. L-1 )wes 241.0 pU@d1 @cell-1 , which was 25-fold that of 9.7 pU@d1@ cell-1 without Dox treatment. Northern blot analyses andRT-PCR further confinned that the transcription of insulingene had already been up-regulated after exposing tet293/Ins6 cells to Dox for 15 minutes, and was also induced in adose-depeodent manner. | 4 | Restudies on Body Surface of Dung Beetle and Application of Its Bionics Flexible Technique显示文摘A scanning electron microscope was used to observe the structures of the setae on the surface of a dung beetle Copris ochus, Motschulsky. There are lots of setae on the body surface, especially on the ventral part surface and lateral to the legs which are different in size, arrangement and shape. These setae have different lengths and many thorns on the whole seta. The top ends of these setae stand up without furcations which direct uprightly towards the surface of the touched soil. By the method of removing these setae, getting the insect weight before and after digging into the dung we affirm farther that the setae on the beetle body surface form the anti-stick and non-adherent gentle interface. | 5 | Syntheses of amphiphilic biodegradable copolymers of poly(ethyl ethylene phosphate) and poly(3-hydroxybutyrate) for drug delivery显示文摘Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxybutyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a coreshell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. | 7 | Structure comparisons of Aedes albopictus densovirus with other parvoviruses显示文摘Parvoviridae is a family of the smallest viruses known with a wide variety of hosts. The capsid structure of the Aedes albopictus C6/36 cell densovirus (C6/36 DNV) at 1.2-nm resolution was obtained by elec-tron cryomicroscopy (cryoEM) and three-dimensional (3D) image reconstruction. Structure compari-sons between the C6/36 DNV and other parvoviruses reveal that the degree of structural similarity be-tween C6/36 DNV and the human parvovirus B19 is higher than that between C6/36 DNV and other in-sect parvoviruses. The amino acid sequence comparisons of structural and non-structural proteins also reveal higher levels of similarity between C6/36 DNV and parvovirus B19 than those between C6/36 DNV and other parvoviruses. It is divided into three types by eruptive locations and occurrences of eruptive rocks, including the pattern of eruption along fault and occurrence (PEAFO), the pattern of eruption on footwall of a fault and occurrence (PEOFO) and the pattern of eruption on hangingwall of fault and occurrence (PEOHO) in Xujiaweizi fault depression of Songliao basin, Northeast of China. Those basin filling patterns of volcanic eruptive rocks are of specific geometric characteristics controlling or affecting sedimentation, geometry of sedimentary body and sequence architecture during a sequence evolution.
