2PpINH1,an invertase inhibitor,interacts with vacuolar invertase PpVIN2 in regulating the chilling tolerance of peach fruit显示文摘Sucrose metabolism,particularly the decomposition of sucrose by invertase,plays a central role in plant responses to cold stress.Invertase inhibitors(INHs)evolved in higher plants as essential regulators of sucrose metabolism.By limiting invertase activity,INHs keep cellular sugar levels elevated,which provides enhanced protection to plants under stress.Our results showed that the expression of PpVIN2,the only vacuolar invertase(VIN)gene in peach fruit sensitive to chilling temperatures,increases significantly during cold storage,while VIN enzyme activity increases more modestly.We also found that peach fruit transiently overexpressing PpINH1 had decreased VIN activity.Interactions of PpINH1 and PpVIN2 with recombinant proteins were shown by yeast two-hybrid assays and bimolecular fluorescence complementation assays,as well as in vitro.During cold storage,trehalose-treated peach fruit had significantly increased PpINH1 expression,decreased VIN enzyme activity,and significantly higher sucrose content than did untreated fruit.As a result,the treated fruit had enhanced resistance to chilling injury.Collectively,our data show that the post-translational repression of VIN enzyme activity by PpINH1 helps maintain sucrose levels in peach fruit during cold storage,thereby improving resistance to chilling injury.Xingxing Wang Yi Chen Shu Jiang Feng Xu Hongfei Wang Yingying Wei Xingfeng Shao 2020Horticulture Research2020,7,1:1
3Evaluation of strategy for analyzing mouse liver plasma membrane proteome显示文摘Plasma membrane (PM) proteome is one of the major subproteomes present in the cell,and is very important in liver function. In the present work, C57 mouse liver PM was purified by density-gradient centrifugation. The purified PM was verified by electron microscope analysis and Western blotting. The results showed that the PM was enriched by more than 20-fold and the contamination of mitochondria was reduced by 2-fold compared with the homogenization fraction. Proteins were separated by 2DE and 1DE, trypsin-digested and submitted to ESI-Q-TOF and MALDI-TOF-TOF mass spectrometry or directly digested in solution and analyzed by LC-ESI ion trap mass spectrometry. In all, 547 non-redundant mouse liver PM proteins were identified, of which 34% contributed to plasma membrane or plasma membrane-related proteins. 5Initial research on the polarized reflectance of typical urban surface types measured by the Directional Polarimetric Camera显示文摘This paper retrieves the bidirectional polarization distribution functions (BPDFs) of the distinct urban cover in the Pearl River Delta (PRD) region employing the high spatial resolution of multi-angle polarized measurements made by the Directional Polarimetric Camera (DPC).The BPDF operational products of MICROPOL and POLDER measurements were used to validate the BPDF products of the DPC,with the results demonstrating that the BPDF product of the DPC measurements accurately expresses the surface polarized reflectance.The polarized reflectance of distinct surface types in the PRD region was studied using the DPC measurements.The results demonstrate that the polarized reflectances of different surface types differ and decrease as the normalized difference vegetation index increases.The polarized reflectance of a distinct surface type in the PRD region decreases with increasing scattering angle.The basic theory of investigating surface properties using multi-angle polarized measurements is proposed.XIE DongHai GU XingFa CHENG TianHai YU Tao LI ZhengQiang CHEN XingFeng CHEN Hao GUO Jing 2011Science China Earth Sciences2011,54,8:1

7Spatiotemporal distribution of seasonal bird assemblages on land-bridge islands: linking dynamic and static views of metacommunities显示文摘Background: Although assessing temporal dynamics of populations is crucial for understanding metacommunities, empirical studies have primarily analyzed only static snapshots of communities. Here, we present a holistic view of how species traits and habitat characteristics relate to metacommunity dynamics and use it to test for differences in the spatiotemporal distribution of seasonal bird assemblages. Methods: We surveyed forest birds in breeding and winter seasons within 36 islands for 9 years. We then grouped birds into four landbird assemblages, selected on the basis of published differences in biology or ecology: winter residents, migratory winter visitors, breeding summer residents, and migratory summer visitors. We estimated dynamic species colonization and extirpation through the 9-year period, and evaluated the associations among island attributes, species attributes and community composition. Results: Overall, winter and summer residents showed strong associations between composition and habitat structure of the islands. In addition, winter and summer residents on large islands had lower extirpation and turnover than winter and summer visitors. Visitor assemblages showed no significant habitat associations, and in winter had high extirpation rates and small body sizes. By contrast, local extirpation of summer visitors was correlated with local species richness, indicating a likely effect of competition on extirpation. Conclusions: Our results demonstrated repeated patterns among species composition, bird traits, habitat/island characteristics and observed metacommunity dynamics. Winter and summer residents best matched species sorting and patch dynamics, respectively, due to differences in resource availability and requirements of overwinter survival versus breeding. Summer visitors were consistent with species sorting and winter visitors were randomly distributed, likely because of interactions with resident competitors.
