| 1 | Cloned goats (Gapra hircus) from foetal fibroblast cell lines显示文摘Mammalian cloning has been one of the most active research topics in the world. Cioning with in vitro culured foetal fibroblast cells, in comparison with embryonic cells, can be used not only to theoretically study the embryonic or cellular development and differentiation in mammals, but also to utilize the unlimited fibroblast cells to produce large numbers of clonings. The preliminary results are as follows: (i) The division and development of the cloned embryos with embryonic donor cells and goat foetal fibroblast donor cells were 55%, 77% and 35%, 31%, respectively. There is no significant statistical difference between them. (? These studies result in the birth of two cloned goats derived from two 30-day foetal fibroblast celi lines, which are the first cloned mammals from somatic cells in China. | 3 | Morphological MRI and T2 mapping of cartilage repair tissue after mosaicplasty with tissue-engineered cartilage in a pig model显示文摘The aim of this study was to evaluate the efficacy of mosaicplasty with tissue-engineered cartilage for the treatment of osteochondral defects in a pig model with advanced MR technique.Eight adolescent miniature pigs were used.The right knee underwent mosaicplasty with tissue-engineered cartilage for treatment of focal osteochondral defects,while the left knee was repaired via single mosaicplasty as controls.At 6,12,18 and 26 weeks after surgery,repair tissue was evaluated by magnetic resonance imaging(MSI)with the cartilage repair tissue(MOCART)scoring system and T2mapping.Then,the results of MRI for 26 weeks were compared with findings of macroscopic and histologic studies.The MOCART scores showed that the repaired tissue of the tissue-engineered cartilage group was statistically better than mat of controls(P<0.001).A significant correlation was found between macroscopic and MOCART scores(P<0.001).Comparable mean T2 values were found between adjacent cartilage and repair tissue in the experimental group(P>0.05).For zonal T2 value evaluation,there were no significant zonal T2 differences for repair tissue in controls(P>0.05).For the experimental group,zonal T2 variation was found in repair tissue(P<0.05).MRI,macroscopy and histology showed better repair results and bony incorporation in mosaicplasty with the tissue-engineered cartilage group than those of the single mosaicplasty group.Mosaicplasty with the tissue-engineered cartilage is a promising approach to repair osteochodndral defects.Morphological MRI and T2 mapping provide a non-invasive method for monitoring the maturation and integration of cartilage repair tissue in vivo. | Qichun Chen Qiang Zuo Qianqian Hu Yang Feng Weiding Cui Weimin Fan Yuefen Zou | 2014 | The Journal of Biomedical Research2014,28,4: | 2 |

| 10 | TGF-β1-induced RAP2 regulates invasion in pancreatic cancer显示文摘Pancreatic cancer is highly lethal due to its aggressive invasive properties and capacity for metastatic dissemination.Additional therapeutic targets and effective treatment options for patients with tumours of high invasive capacity are required.Ras-related protein-2a(RAP2)is a member of the GTP-binding proteins.RAP2 has been reported to be widely upregulated in many types of cancers via regulating cytoskeleton reorganization,cell proliferation,migration,and adhesion,as well as inflammation.As a member of the RAS oncogene family,which has been demonstrated to drive pancreatic cancer oncogenesis and many other malignancies,the physiological roles of RAP2 in pancreatic cancer have seldom been discussed.In the present study,we explored the correlation between RAP2 expression and the prediction of overall survival of pancreatic cancer patients.Mechanistic studies were carried out to shed light on the role of RAP2 in pancreatic cancer invasion and how RAP2 is regulated in the invasive process.Our results demonstrated that patients with higher RAP2 expression showed unfavourable prognoses.In vitro studies demonstrated that silencing of RAP2 inhibited the invasion of pancreatic cancer cells.Moreover,our results demonstrated that transforming growth factor-β1(TGF-β1),an inducer of the metastatic potential of pancreatic cancer cells,regulates the expression of RAP2 via the transcription factor c-Myc.In conclusion,the present study uncovered RAP2 as a novel predictive marker and therapeutic target for pancreatic cancer. | Kaizhou Jin Chen Liu He Cheng Qinglin Fei Qiuyi Huang Zhiwen Xiao Xianjun Yu Weiding Wu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,3: | 0 |
