| 2 | Leishmania donovani whole cell antigen delivered with adjuvants protects against visceral leishmaniasis in vervet monkeys (Chlorocebus aethiops)显示文摘In a previous immunogenicity and efficacy study in mice,montanide ISA 720(MISA)was indicated to be a better adjuvant than bacillus calmette guerin vaccine(BCG)for a Leishmania vaccine.In the present study,we report the safety,immunogenicity and efficacy of Leishmania donovani(L.donovani)sonicated antigen delivered with alum-BCG(AlBCG),MISA or monophosphoryl lipid A(MPLA)in vervet monkeys following intradermal inoculums.Vaccinated and control animals were challenged with virulent L.donovani parasites and the parasitic burden was determined.Only animals vaccinated with alum-BCG adversely reacted to the inoculum by producing ulcerative erythematous skin indurations.Non-parametric ANOVA followed by a post test showed significantly higher IgG antibodies,and revealed the presence of lymphoproliferative and interferon gamma responses in both AlBCG+Ag and MISA+Ag as compared to the MPLA+Ag or other groups(P < 0.001).We conclude that L.donovani sonicated antigen containing MISA is safe and is associated with protective immune response against Leishmania donovani infection in the vervet monkey model. | Joshua Muli Mutiso John Chege Macharia Evans Taracha Michael Muita Gicheru | 2012 | The Journal of Biomedical Research2012,26,1: | 2 |
| 3 | Development ofLeishmania vaccines:predicting the future from past and present experience显示文摘Leishmaniasis is a disease that ranges in severity from skin lesions to serious disfigurement and fatal systemic infection. Resistance to infection is associated with a T-helper-1 immune response that activates macrophages to kill the intracellular parasite in a nitric oxide-dependent manner. Conversely, disease progression is generally associated with a T-helper-2 response that activates humoral immunity. Current control is based on chemotherapeutic treatments which are expensive, toxic and associated with high relapse and resistance rates. Vaccination remains the best hope for control of all forms of the disease, and the development of a safe, effective and affordable antileishmanial vaccine is a critical global public-health priority. Extensive evidence from studies in animal models indicates that solid protection can be achieved by immunization with defined subunit vaccines or live-attenuated strains of Leishmania. However, to date, no vaccine is available despite substantial efforts by many labo-ratories. Major impediments in Leishmania vaccine development include: lack of adequate funding from national and international agencies, problems related to the translation of data from animal models to human disease, and the transition from the laboratory to the field. Furthermore, a thorough understanding of protective immune responses and generation and maintenance of the immunological memory, an important but least-studied aspect of antiparasitic vaccine development, during Leishmania infection is needed. Moench]显示文摘Many attempts on optimization of sorghum[Sorghum bicolor(L.)Moench]tissue culture induction media have been made,but the culture system remains with some bottlenecks compared to that of other crops.This study aimed at assessing the suitability of various induction media to produce embryogenic callus(yellow and friable)with high induction rates and reduced phenolic exudation.The six culture medium modifications:3 based on Murashige and Skoog(MS)medium and one each based on Chu N6,Gamborg B5 and 190-2 media respectively were applied in the culture of mature embryos from 10 sorghum genotypes.Although there was a genotype influence on the attainment of a yellow callus,friability of the callus was determined to be dependent on the culture medium and not the genotype.Half strength MS medium with 0.2 mg/l 2,4-D with 2.8 g/l Gelrite®as the gelling agent modified with 1.0 g/l KH_(2)PO_(4),1.0 g/l L-proline,1.0 g/l L-asparagine and 0.16 mg/l CuSO_(4)·5H_(2)O(type E)was found to be the most effective resulting in about 60%yellow coloured callus induction with 25%friability.Addition of CuSO_(4)·5H_(2)O,KH_(2)PO_(4),L-proline and L-asparagine significantly reduced the phenolic production.Half strength MS medium was observed to contribute to quality callus production when compared to full strength MS media modified with the compounds.The half strength MS medium was also observed to suppress phenolic production.Medium 190-2 produced the highest regeneration frequency(40%)among the 3-regeneration media tested.The results provide information on a suitable sorghum callus induction medium necessary for embryogenesis. | Paul Chege Andrea Palagyi Csaba Lantos Erzsebet Kiss Janos Pauk | 2020 | Phyton-International Journal of Experimental Botany2020,89,1: | 0 |
| 20 | Opportunistic infection of Aspergillus and bacteria in captive Cape vultures (Gyps coprotheres)显示文摘Objective:To describe clinical signs,pathology,diagnosis and treatment of Cape vultures in which Aspergillus fumigatus(A.fumigatus)and mixed species of bacteria were isolated.Methods:Six Cape vultures sourced from South Africa for exhibition at Al Ain Zoo developed illness manifesting as anorexia,dyspnea,polyuria and lethargy.Three vultures died manifesting'pneumonia-like syndrome'.These three vultures were necropsied and gross lesions recorded,while organ tissues were collected for histopathology.Internal organs were swabbed for bacteriology and mycology.From live vultures,blood was collected for hematology and biochemistry,oropharyngeal and cloacal swabs were collected for mycology and bacteriology.Results:A.fumigatus was isolated from the three dead vultures and two live ones that eventually survived.One of the dead vulture and two live vultures were co-infected with A.fumigatus and mixed species of bacteria that included Clostridium perfringens,Pseudomonas,Staphylococcus,Escherichia,Proteus,Enterococcus and Enterbacter.One of the Cape vulture and a Lappetfaced vulture,however,were free of Aspergillus or bacterial infections.At necropsy,intestinal hemorrhages were observed and the lungs were overtly congested with granulomas present on caudal air sac.Histopathologtcal examinations demonstrated granulomatous lesions that were infiltrated by mononuclear cells and giant cells.Conclusions:Aspergillosis is a persistent threat to captive birds and we recommend routine health assessments so that early diagnosis may prompt early treatment.It is likely that prompt prophylaxis by broad spectrum antibiotics and antifungals medication contributed to the survival of some of the vultures. | Stephen Chege Judith Howlett Majid Al Qassimi Arshad Toosy Joerg Kinne Vincent Obanda | 2013 | Asian Pacific Journal of Tropical Biomedicine2013,3,5: | 0 |
