| 1 | Fermented soybean meal improves the growth performance, nutrient digestibility, and microbial flora in piglets显示文摘In order to increase nutritive values of soybean meal(SBM), 3 species of microbes were used to ferment SBM. Through a 3 x 3 orthogonal design and parameter measurements of soybean peptide and antinutritional factor contents in the fermented soybean meal(FSBM), it was estimated that the best microbial proportion of Bacillus subtilis, Hansenula anomala and Lactobacillus casei was 2:1:2 for SBM fermentation(P < 0.05). The further piglet feeding experiment showed that 10% FSBM substitute for SBM had no significant effect on growth performance of suckling piglets(d 7-28)(P> 0.05). However, newlyweaned piglets(d 28-38) fed 10% FSBM and different levels of plasma protein obtained higher average daily gain(ADG) and feed conversion ratio(FCR), compared with those without FSBM but with 6% plasma protein(P < 0.05). Piglets(d 38-68) fed diets supplemented with FSBM and soybean protein concentrate(SBPC) at 3.75% and 7.5% respectively increased nutrient digestibility,fecal enzyme activity and lactic acid bacteria counts, and decreased fecal Escherichia coli counts(P < 0.05), compared with the control. | 6 | Crystalline Silica Promotes Rat Fibrocyte Differentiation in Vitro,and Fibrocytes Participate in Silicosis in Vivo显示文摘Objective The aim of this study was to investigate the effects of Si O2 on fibrocytes and whether fibrocytes participate in silicosis in vivo. Methods A macrophagocyte(AM)/fibrocyte coculture system was established, and AMs were treated with 100 μg/mL SiO 2. Flow cytometry was used to detect the number of fibrocytes. Real‐time PCR was performed to measure the expression of collagen I, collagen III, and α‐SMA mR NA. The levels of collagen I, collagen III, and TGF‐β1 protein were determined by ELISA. Immunohistochemical staining was performed to measure α‐SMA protein expression. A rat silicosis model was induced by intratracheal instillation of SiO 2. Lung histopathological evaluation was conducted using HE and Masson's trichrome staining after 1 and 9 weeks. The number of fibrocytes in peripheral blood or lung tissue of rat was detected by flow cytometry. Double‐color immunofluorescence was applied to identify fibrocytes in the lung tissue. Results Peripheral blood monocytes were found to differentiate into fibrocytes in vitro in a time‐dependent manner, and exposure to crystalline silica might potentiate fibrocyte differentiation. In addition, fibrocytes were able to migrate from peripheral blood to the lung tissue, and the number of fibrocytes was increased after SiO 2 exposure. | 7 | Mg-Zn-Y-Nd coated with citric acid and dopamine by layer-by-layer self-assembly to improve surface biocompatibility显示文摘Magnesium alloy has been generally accepted as an important biodegradable material on cardiovascular stent development for a long time. However, its limited biocompatibility, especially delayed endothelialization process restricts its further application. In this contribution, we modified the Mg-Zn-Y-Nd alloy surface with citric acid and dopamine via a layer-by-layer self-assembly assay, aiming at improving the biocompatibility of the magnesium alloy. The citric acid/dopamine(CA/PDA) layer exhibited a remarkable suppression of platelet activation/aggregation and thrombosis under 15 dyn/cm2 blood flowing. Inhibition on vascular smooth muscle cells growth and macrophages attachment/activation were also observed on this layer. In particular, the CA/PDA layer presented a promoted property for the vascular endothelial cells growth and spreading compared with the bare magnesium alloy, suggesting the pro-endotelialized function. | 9 | L-theanine inhibits nicotine-induced dependence via regulation of the nicotine acetylcholine receptor-dopamine reward pathway显示文摘In this study, the inhibitory effect of L-theanine, an amino acid derivative of tea, on the rewarding effects of nicotine and its underlying mechanisms of action were studied. We found that L-theanine inhibited the rewarding effects of nicotine in a con- ditioned place preference (CPP) model of the mouse and reduced the excitatory status induced by nicotine in SH-SY5Y cells to the same extent as the nicotine receptor inhibitor dihydro-beta-erythroidine (DH E). Further studies using high performance liquid chromatography, western blotting and immunofluorescence staining analyses showed that L-theanine significantly inhibited nicotine-induced tyrosine hydroxylase (TH) expression and dopamine production in the midbrain of mice. L-theanine treatment also reduced the upregulation of the 4, 2 and 7 nicotine acetylcholine receptor (nAChR) subunits induced by nico- tine in mouse brain regions that related to the dopamine reward pathway, thus decreasing the number of cells that could react to nicotine. In addition, L-theanine treatment inhibited nicotine-induced c-Fos expression in the reward circuit related areas of the mouse brain. Knockdown of c-Fos by siRNA inhibited the excitatory status of cells but not the upregulation of TH induced by nicotine in SH-SY5Y cells. Overall, the present study showed that L-theanine reduced the nicotine-induced reward effects via inhibition of the nAChR-dopamine reward pathway. | 11 | CHIP regulates bone mass by targeting multiple TRAF family members in bone marrow stromal cells显示文摘Carboxyl terminus of Hsp70-interacting protein(CHIP or STUB1) is an E3 ligase and regulates the stability of several proteins which are involved in different cellular functions. Our previous studies demonstrated that Chip deficient mice display bone loss phenotype due to increased osteoclast formation through enhancing TRAF6 activity in osteoclasts. In this study we provide novel evidence about the function of CHIP. We found that osteoblast differentiation and bone formation were also decreased in Chip KO mice. In bone marrow stromal(BMS) cells derived from Chip^(-/-) mice, expression of a panel of osteoblast marker genes was significantly decreased. ALP activity and mineralized bone matrix formation were also reduced in Chip-deficient BMS cells. We also found that in addition to the regulation of TRAF6, CHIP also inhibits TNFα-induced NF-κB signaling through promoting TRAF2 and TRAF5 degradation. Specific deletion of Chip in BMS cells downregulated expression of osteoblast marker genes which could be reversed by the addition of NF-κB inhibitor. These results demonstrate that the osteopenic phenotype observed in Chip^(-/-) mice was due to the combination of increased osteoclast formation and decreased osteoblast differentiation. | 18 | Clinical features and predictors of remission in children under the age of 7 years with Graves’disease显示文摘Importance:Graves’disease(GD)is rare in children under the age of 7 years.Children with this disease exhibit greater thyrotoxicity at diagnosis and require a longer course of medical therapy,compared with pubertal and postpubertal children and adults.Objective:To investigate the clinical features and identify predictors of remission in children under the age of 7 years with GD.Methods:This retrospective study included 77 children who were diagnosed with GD under the age of 7 years and were treated in the Department of Endocrinology,Beijing Children’s Hospital from 2010 to 2018.Clinical manifestations,laboratory data,and follow-up records were collected for all patients.Children who achieved remission of treatment with methimazole were compared with those who had persistent disease to identify which variables were associated with remission;multiple logistic regression and Cox regression analyses were used to evaluate interactions among predictive variables.Results:Sixty-three boys and 14 girls were included;the median age at diagnosis was 4.2 years(interquartile range:3.2-5.3 years).Forty-six(56.7%)patients had no family history of thyroid disease,17 patients had family history of thyroid disease and 14 patients with unknown family history.Of the 77 patients,18(23.4%)patients achieved remission of treatment with methimazole and 59 patients did not;moreover,51(66.2%)had Graves’ophthalmopathy.Univariate analyses revealed no significant differences between the remission group and non-remission group in terms of age at diagnosis,sex,initial goiter size,or initial thyroid hormone concentration.However,there were a trend of correlation between the initial level of thyroid peroxidase antibody(TPOAb)and remission status(univariate analysis OR 1.002,P=0.038;multivariate analysis OR 1.004,P=0.019).Similar results were observed in univariate analysis of the initial thyrotropin receptor antibody(TRAb)level,but this association was not significant in multivariate analysis.Cox regression analyses revealed that children with high TRAb level required longer duration of remission,compared with low TRAb level(OR 0.950,95%CI 0.904-0.997,P=0.037).Interpretation:Initial TRAb level was an independent predictor of remission outcome in young children under the age of 7 years with GD.Initial TRAb level may predict the likelihood of remission in patients with young-age-of-onset GD. | Yi Gu Xuejun Liang Ming Liu Di Wu Wenjing Li Bingyan Cao Yuchuan Li Chang Su Jiajia Chen Chunxiu Gong | 2020 | Pediatric Investigation2020,4,3: | 3 |

| 20 | Protective effect of paeoniflorin on H_(2)O_(2) induced Schwann cells injury based on network pharmacology and experimental validation显示文摘This study was to investigate the protective effect of paeoniflorin(PF)on hydrogen peroxide-induced injury.Firstly,“SMILES”of PF was searched in Pubchem and further was used for reverse molecular docking in Swiss Target Prediction database to obtain potential targets.Injury-related molecules were obtained from GeenCards database,and the predicted targets of PF for injury treatment were selected by Wayne diagram.For mechanism analysis,the protein-protein interactions were constructed by String,and the KEGG analysis was conducted in Webgestalt.Then,cell viability and cytotoxicity assay were established by CCK8 assay.Also,the experimental cells were allocated to control,model(200μmol·L^(−1) H_(2)O_(2)),SB20358010μmol·L^(−1)(200μmol·L^(−1) H_(2)O_(2)+SB20358010μmol·L^(−1)),PF 50μmol·L^(−1)(200μmol·L^(−1) H_(2)O_(2)+PF 50μmol·L^(−1)),and PF 100μmol·L^(−1)(200μmol·L^(−1) H_(2)O_(2)+PF 100μmol·L^(−1))groups.We measured the intracellular ROS,Hoechst 33258 staining,cell apoptosis,the levels of Bcl-xl,Bcl-2,Caspase-3,Cleaved-caspase3,Cleaved-caspase7,TRPA1,TRPV1,and the phosphorylation expression of p38MAPK.There are 96 potential targets that may be associated with PF for injury treatment.Then,we chose the“Inflammatory mediator regulation of TRP channels”pathway for the experimental verification from the first 10 KEGG pathway.In experimental verification,H_(2)O_(2) decreased the cell viability moderately(P<0.05),and 100μmol·L^(−1) PF increased the cell viability significantly(P<0.05).Depending on the difference of intracellular ROS fluorescence intensity,PF inhibited H_(2)O_(2)-induced reactive oxygen species production in Schwann cells.In Hoechst 33258 staining,PF reversed the condensed chromatin and apoptotic nuclei following H_(2)O_(2) treatment.Moreover,Flow cytometry results showed that PF could substantially inhibit H_(2)O_(2) induced apoptosis(P<0.05).Pretreatment with PF obviously reduced the levels of Caspase3,Cleaved-caspase3,Cleaved-caspase7,TRPA1,TRPV1,and the phosphorylation expression of p38MAPK after H_(2)O_(2) treatment(P<0.05),increased the levels of Bcl-2,and Bcl-xl(P<0.05).PF inhibited Schwann cell injury and apoptosis induced by hydrogen peroxide,which mechanism was linked to the inhibition of phosphorylation of p38MAPK. | ZHANG Di YANG Bing CHANG Shi-Quan MA Sheng-Suo SUN Jian-Xin YI Lin LI Xing SHI Hui-Mei JING Bei ZHENG Ya-Chun ZHANG Chun-Lan CHEN Feng-Guo ZHAO Guo-Ping | 2021 | Chinese Journal of Natural Medicines2021,19,2: | 3 |
