| 2 | Astrocyte reactivity in related brain regions in a mouse model of MPTP-induced Parkinson's disease显示文摘BACKGROUND:Severe injury to dopaminergic neuronal cell bodies and their axon terminals in the substantia nigra pars compacta(SNC) has been observed in both Parkinson's disease(PD) patients or in 1-methy-4-phenyl-1,2,3,6-tetrahydropyrindine(MPTP)-induced PD animal models, but only slight injury occurs in the adjacent ventral tegmental area(VTA).The mechanisms underlying this selective injury remain poorly understood. OBJECTIVE:To comparatively observe astrocyte reactivity in the SNC,caudate putamen(CPu), VTA,and frontal association cortex(FrA). DESIGN,TIME AND SETTING:A cellular and molecular biology,randomized,controlled experiment was performed at the Institute of Neurobiology,Department of Human Anatomy, Medical School of Nantong University,between December 2006 and September 2008. MATERIALS:A total of 80 healthy adult male C57BL/6 mice were included in this study.MPTP was purchased from Sigma,USA. METHODS:Mice were randomly divided into a model group(n=64) and a sham-operated group (n=16).PD was induced in the mice from the model group by intraperitoneal injection of 20 mg/kg MPTP,once every three hours,for a total of 4 times. MAIN OUTCOME MEASURES:Tyrosine hydroxylase(TH)-immunoreactive neurons and glial fibrillary acidic protein(GFAP)-immunoreactive astrocytes were examined by dual immunofluorescence labeling.GFAP-immunoreactive astrocytes in the CPu and FrA were determined by immunofluorescent staining.GFAP mRNA expression in the SNC,CPu,VTA,and FrA was detected using real-time polymerase chain reaction.TH protein levels in the TH-immunoreactive axon terminals of the CPu and FrA were detected by Western blotting. RESULTS:Numbers of TH-immunoreactive neurons in the SNC,and TH protein level in the CPu, markedly decreased(by approximately 68%) 1 day after MPTP injection,and gradually increased at 3 days.Simultaneously,astrocyte reactivity was strengthened,in particular at 7 days.However, after MPTP injection,decreases in the numbers of TH-immunoreactive neurons in the VTA,and TH protein levels in the FrA,were less apparent(approximately 15%).Also,no obvious astrocyte reactivity was observed. | 13 | VEGFR2 inhibition hampers breast cancer cell proliferation via enhanced mitochondrial biogenesis显示文摘Objective:Vascular endothelial growth factor(VEGF),apart from its predominant roles in angiogenesis,can enhance cancer cell proliferation,but its mechanisms remain elusive.The purpose of the present study was therefore to identify how VEGF regulates cancer cell proliferation.Methods:VEGF effects on cancer cell proliferation were investigated with the VEGF receptor 2 inhibitor,Ki8751,and the breast cancer cell lines,MCF-7 and MDA-MB-231,using flow cytometry,mass spectrometry,immunoblotting,and confocal microscopy.Data were analyzed using one-way analysis of variance followed by Tukey’s multiple comparison test.Results:VEGF blockade by Ki8751 significantly reduced cancer cell proliferation,and enhanced breast cancer cell apoptosis.Mass spectrometric analyses revealed that Ki8751 treatment significantly upregulated the expression of mitochondrial proteins,suggesting the involvement of mitochondrial biogenesis.Confocal microscopy and flow cytometric analyses showed that Ki8751 treatment robustly increased the mitochondrial masses of both cancer cells,induced endomitosis,and arrested cancer cells in the high aneuploid phase.VEGFR2 knockdown by sh RNAs showed similar effects to those of Ki8751,confirming the specificity of Ki8751 treatment.Enhanced mitochondrial biogenesis increased mitochondrial oxidative phosphorylation and stimulated reactive oxygen species(ROS)production,which induced cancer cell apoptosis.Furthermore,Ki8751 treatment downregulated the phosphorylation of Akt and PGC1α,and translocated PGC1αinto the nucleus.The PGC1αalterations increased mitochondrial transcription factor A(TFAM)expression and subsequently increased mitochondrial biogenesis.Conclusions:VEGF enhances cancer cell proliferation by decreasing Akt-PGC1α-TFAM signaling-mediated mitochondrial biogenesis,ROS production,and cell apoptosis.These findings suggested the anticancer potential of Ki8751 via increased mitochondrial biogenesis and ROS production. | Hao Ni Min Guo Xuepei Zhang Lei Jiang Shuai Tan Juan Yuan Huanhuan L Cui Yanan Min Junhao Zhang Susanne Schlisio Chunhong Ma Wangjun Liao Monica Nister Chunlin Chen Shuijie Li Nailin Li | 2021 | Cancer Biology & Medicine2021,18,1: | 0 |
| 14 | Spatial separation of phosphatase and kinase activity within the Bub complex is required for proper mitosis显示文摘The Bub1 and BubR1 kinetochore proteins support proper chromosome segregation and mitotic checkpoint activity. Bub1 and BubR1 are paralogs with Bub1 being a kinase, while BubR1 localizes the PP2A-B56 protein phosphatase to kinetochores in humans. Whether this spatial separation of kinase and phosphatase activity is important is unclear as some organisms integrate both activities into one Bub protein. Here, we engineer human Bub1 and BubR1 proteins integrating kinase and phosphatase activities into one protein and show that these do not support normal mitotic progression. A Bub1–PP2A-B56 complex can support chromosome alignment but results in impairment of the checkpoint due to dephosphorylation of the Mad1 binding site in Bub1. Furthermore, a chimeric BubR1 protein containing the Bub1 kinase domain induces delocalized H2ApT120 phosphorylation, resulting in the reduction of centromeric hSgo2 and chromosome segregation errors.
