1Review of a challenging clinical issue:Intrahepatic cholestasis of pregnancy显示文摘Intrahepatic cholestasis of pregnancy(ICP) is a reversible pregnancy-specific cholestatic condition characterized by pruritus, elevated liver enzymes, and increased serum bile acids. It commences usually in the late second or third trimester, and quickly resolves after delivery. The incidence is higher in South American and Scandinavian countries(9.2%-15.6% and 1.5%, respectively) than in Europe(0.1%-0.2%). The etiology is multifactorial where genetic, endocrine, and environmental factors interact. Maternal outcome is usually benign, whereas fetal complications such as preterm labor, meconium staining, fetal distress, and sudden intrauterine fetal demise not infrequently lead to considerable perinatal morbidity and mortality. Ursodeoxycholic acid is shown to be the most efficient therapeutic agent with proven safety and efficacy. Management of ICP consists of careful monitoring of maternal hepatic function tests and serum bile acid levels in addition to the assessment of fetal well-being and timely delivery after completion of fetal pulmonary maturity. 2A deep learning-enabled portable imaging flow cytometer for cost-effective, highthroughput, and label-free analysis of natural water samples显示文摘We report a deep learning-enabled field-portable and cost-effective imaging flow cytometer that automatically captures phase-contrast color images of the contents of a continuously flowing water sample at a throughput of 100 mL/h.The device is based on partially coherent lens-free holographic microscopy and acquires the diffraction patterns of flowing micro-objects inside a microfluidic channel.These holographic diffraction patterns are reconstructed in real time using a deep learning-based phase-recovery and image-reconstruction method to produce a color image of each micro-object without the use of external labeling.Motion blur is eliminated by simultaneously illuminating the sample with red,green,and blue light-emitting diodes that are pulsed.Operated by a laptop computer,this portable device measures 15.5 cm×15 cm×12.5 cm,weighs 1 kg,and compared to standard imaging flow cytometers,it provides extreme reductions of cost,size and weight while also providing a high volumetric throughput over a large object size range.We demonstrated the capabilities of this device by measuring ocean samples at the Los Angeles coastline and obtaining images of its micro-and nanoplankton composition.Furthermore,we measured the concentration of a potentially toxic alga(Pseudo-nitzschia)in six public beaches in Los Angeles and achieved good agreement with measurements conducted by the California Department of Public Health.The cost-effectiveness,compactness,and simplicity of this computational platform might lead to the creation of a network of imaging flow cytometers for largescale and continuous monitoring of the ocean microbiome,including its plankton composition.Zoltán Gӧrӧcs Miu Tamamitsu Vittorio Bianco Patrick Wolf Shounak Roy Koyoshi Shindo Kyrollos Yanny Yichen Wu Hatice Ceylan Koydemir Yair Rivenson Aydogan Ozcan 2018Light(Science & Applications)2018,7,1:14
3Effect of Intercritical Annealing Parameters on Dual Phase Behavior of Commercial Low-Alloyed Steels显示文摘The effects of some intercritical annealing parameters including heating rate,soaking temperature,soaking time,and quench media on the microstructure and mechanical properties of cold-rolled dual phase steel were studied.The microstructure of specimens quenched after each annealing stage was analyzed using optical microscope.The tensile properties,determined for specimens submitted to complete annealing cycles,were influenced by the volume fractions of multiphases(originated from martensite,bainite,and retained austenite),which depend on the annealing process parameters.The results obtained showed that the yield strength and the ultimate tensile strength increase with increasing the intercritical temperature and cooling rate.This can be explained by higher martensite volume ratio with the increased volume fraction of austenite formed at the higher temperatures and cooling rates.Aydin Hüseyin Kazdal Zeytin Havva Kubilay Ceylan 2010Journal of Iron and Steel Research(International)2010,17,4:10
4Motility-based label-free detection of parasites in bodily fluids using holographic speckle analysis and deep learning显示文摘Parasitic infections constitute a major global public health issue.Existing screening methods that are based on manual microscopic examination often struggle to provide sufficient volumetric throughput and sensitivity to facilitate early diagnosis.Here,we demonstrate a motility-based label-free computational imaging platform to rapidly detect motile parasites in optically dense bodily fluids by utilizing the locomotion of the parasites as a specific biomarker and endogenous contrast mechanism.Based on this principle,a cost-effective and mobile instrument,which rapidly screens~3.2 mL of fluid sample in three dimensions,was built to automatically detect and count motile microorganisms using their holographic time-lapse speckle patterns.We demonstrate the capabilities of our platform by detecting trypanosomes,which are motile protozoan parasites,with various species that cause deadly diseases affecting millions of people worldwide.Using a holographic speckle analysis algorithm combined with deep learningbased classification,we demonstrate sensitive and label-free detection of trypanosomes within spiked whole blood and artificial cerebrospinal fluid(CSF)samples,achieving a limit of detection of ten trypanosomes per mL of whole blood(~five-fold better than the current state-of-the-art parasitological method)and three trypanosomes per mL of CSF.We further demonstrate that this platform can be applied to detect other motile parasites by imaging Trichomonas vaginalis,the causative agent of trichomoniasis,which affects 275 million people worldwide.With its costeffective,portable design and rapid screening time,this unique platform has the potential to be applied for sensitive and timely diagnosis of neglected tropical diseases caused by motile parasites and other parasitic infections in resource-limited regions.Yibo Zhang Hatice Ceylan Koydemir Michelle M.Shimogawa Sener Yalcin Alexander Guziak Tairan Liu Ilker Oguz Yujia Huang Bijie Bai Yilin Luo Yi Luo Zhensong Wei Hongda Wang Vittorio Bianco Bohan Zhang Rohan Nadkarni Kent Hill Aydogan Ozcan 2018Light(Science & Applications)2018,7,1:6
5Comparison of visual and topographic outcomes of deep-anterior lamellar keratoplasty and penetrating keratoplasty in keratoconus显示文摘AIM:To compare visual,surgical and topographic outcomes of deep anterior lamellar keratoplasty(DALK)and penetrating keratoplasty(PK)for keratoconus(KC).METHODS:In this multicenter,prospective,randomized clinical trial 76 eyes of 71 KC patients operated between January 2011 and July 2014 in 2 tertiary referral hospitals were included. Consecutive patients were alternately selected to receive one of the two surgical methods. Thirty eight eyes underwent DALK with the big-bubble technique and 38 eyes underwent PK.RESULTS:Mean best spectacle corrected visual acuity(BSCVA)at the first postoperative week(P=0.012)and the first postoperative month(P<0.001)was statistically significantly higher in DALK group. The mean BSCVA at12 mo was not significantly different for DALK(0.30±1.99 log MAR)versus PK(0.40±0.33 log MAR)(P=0.104). The76.3% of the eyes had a BSCVA over 0.5 in DALK and 47.4%in PK group(P=0.009). The 7.9% of the eyes had a BSCVA of 1.0 in DALK and 5.3% in PK group(P=0.644). Mean spherical equivalent was-2.94 D in DALK and-3.09 D in PK group.Mean topographic astigmatism was 4.62 D and 4.18 D respectively. Regular topographic patterns were observed in 31(81.6%)of DALK and 29(76.3%)of PK(P=0.574). The most frequent topographic pattern was oblate asymmetric bow tie,seen in 39.5% in DALK and 23.7% in PK. CONCLUSION:Big bubble DALK provides an earlier visual improvement compare to PK. However,visual and topographic outcomes are similar to those in PK at 1y. Postoperative complications including rejection and intraocular pressure elevation are more frequent in PK. DALK is a safer alternative to PK for KC. 6Pancreatic hyperechogenicity on endoscopic ultrasound examination显示文摘There is an ongoing discussion on how to diagnose a hyperechogenic pancreas and what is the clinical significance of diffusely hyperechogenic pancreas. Computerized tomography and magnetic resonance imaging are the more appropriate methods to diagnose pancreatic hyperechogenicity when compared with transcutaneous or endoscopic ultrasound examination. More importantly, pancreatic hyperechogenicity may not be a certain indicator of pancreatic fat infiltration. Even if it is true, we do not know the clinical significances of pancreatic fat accumulation. Some suggested that excess fat in the pancreas is associated with chronic pancreatitis. However, several histological studies on human alcoholic chronic pancreatitis did not prove the presence of fatty pancreas in such cases. 7Therapeutic effect of caffeic acid phenethyl ester on cerulein-induced acute pancreatitis显示文摘AIM: To evaluate the therapeutic role of caffeic acid phenethyl ester (CAPE) in a rat model of ceruleaninduced acute pancreatitis (AP).METHODS: Seventy male Wistar albino rats were divided into seven groups. Acute edematous pancreatitis was induced by subcutaneous cerulein injection (20 μg/kg) four times at 1-h intervals. CAPE (30 mg/kg) was given by subcutaneous injection at the beginning (CAPE 1 group) and 12 h after the last cerulein injection (CAPE 2 group). Serum amylase, lipase, white blood cell count, and tumor necrosis factor (TNF)-α levels were measured, and pancreatic histopathology was assessed. RESULTS: In the AP group, amylase and lipase levels were found to be elevated and the histopathological evaluation showed massive edema and inflammation of the pancreas, with less fatty necrosis when compared with sham and control groups. Amylase and lipase levels and edema formation decreased signif icantly in the CAPE therapy groups (P < 0001); especially in the CAPE 2 group, edema was improved nearly completely (P = 0001). Inflammation and fatty necrosis were partially recovered by CAPE treatment. The pathologicalresults and amylase level in the placebo groups were similar to those in the AP group. 9Parkin deficiency accentuates chronic alcohol intake-induced tissue injury and autophagy defects in brain,liver and skeletal muscle显示文摘Alcoholism leads to organ injury including mitochondrial defect and apoptosis with evidence favoring a role for autophagy dysregulation in alcoholic damage.Parkin represents an autosomal recessive in herited gene for Parkins on's disease and an important member of selective autophagy for mitoch on dria.The associati on between Parkins on's disease and alcoholic injury remains elusive.This study aimed to examine the effect of parki n deficiency on chr onic alcohol in takeinduced organ injury in brain,liver and skeletal muscle(rectus femoris muscle).Adult parkinknockout(PRK-/-)and wild-type mice were placed on Liber-De Carli alcohol liquid diet(4%)for 12 weeks prior to assessment of liver enzymes,intraperitoneal glucose toleranee,protein carbonyl content apoptosis,hematoxylin and eosin morphological staining,and mitochondrial respiration(cytochrome c oxidase,NADH:cytochrome c reductase and succinate:cytochrome c reductase).Autophagy protein markers were monitored by western blot analysis.Our data revealed that chronic alcohol intake imposed liver injury as evidenced by elevated aspartate aminotransferase and alanine transaminase,glucose intoleranee,elevated protein carbonyl formation,apoptosis,focal inflammation,necrosis,microvesiculation,autophagy/mitophagy failure and dampened mitochondrial respiration(complexⅣ,complexesⅠandⅢ,and complexesⅡandⅢ)in the brain,liver and rectus femoris skeletal muscle.Although parkin ablation itself did not generate any notable effects on liver enzymes,insulin sensitivity,tissue carbonyl damage,apoptosis,tissue morphology,autophagy or mitochondrial respiration,it accentuated alcohol intake-induced tissue damage,apoptosis,morphological change,autophagy/mitophagy failure and mitochondrial injury without affecting insulin sensitivity.These data suggest that parkin plays an integral role in the preservation against alcohol-induced organ injury,apoptosis and mitochondrial damage.Hu Peng Xing Qin Sainan Chen Asli E Ceylan Maolong Dong Zhaofen Lin Jun Ren 2020Acta Biochimica et Biophysica Sinica2020,52,6:3
10Effect of Microstructure on Exhaust Manifold Cracks Produced From SiMo Ductile Iron显示文摘Ductile cast irons are used as high temperature materials in internal combustion engines,because they are microstructurally stable at high operating temperatures. SiMo granular graphite cast irons contain Fe2MoC and M6C carbide precipitates due to their higher concentration of both silicon and molybdenum. The microstructure of these cast irons consists of carbides dispersed within the ferrite matrix. The microstructural change and the crack formation mechanism in manifolds produced from SiMo ductile iron are studied. The liver keeps a delicate balance between hepatic screening of pathogenic antigens and immune tolerance to self-antigens. Herbal treatments with immunological effects have potential to alter this hepatic immune balance towards either therapeutic side or diseases side by inducing liver injury via hepatotoxicity or initiation of autoimmune diseases. 12Effects of moisture damage on asphalt mixtures显示文摘The reduction in the ability of bitumen to bond with the aggregate surface due to the infiltration of moisture has been recognised for years, and this deterioration phenomenon is called moisture damage. In general, the loss of bonding between bitumen and aggregate shortens the service life of the top layer of the pavement. Many investigations have been conducted to understand the mechanisms of moisture damage due to the loss of bonding strength between bitumen and aggregate and to find ways to improve and strengthen the bond to mitigate the effect of moisture. This paper reviews the extensive literature on the loss of bitumen-aggregate bonding strength due to moisture damage in asphalt mixtures.The general description of the theories and mechanisms that explain the effect of the thermodynamic, chemical, physical and mechanical characteristics of the bitumen and aggregate on the bonding phenomenon are discussed in this paper. In addition, the causes of and contributing factors to moisture damage and methods to improve the bond between bitumen and aggregates are also discussed. Moreover, a description of the test methods that can be used to evaluate moisture damage in poorly bonded and compacted mixtures are also presented. Special attention is given to a well-known method, known as the pulloff test, which has been successfully used to evaluate aggregate-binder bond strength, both for laboratory and in-situ tests. This includes the test methods, the factors that affect the bonding strength results and their correlation with other test method. 13Storage in ice incorporated antimelanotic agent and its effects on melanosis and quality of giant red shrimp(Aristaeomorpha foliacea)显示文摘Since melanosis starts very quickly after catching in shrimps,it is immediately treated with an antimelanotic agent.Shrimps are transported to land with ice.In this study,it was aimed to combine these two applications and to produce antimelanotic added ice and to preserve the shrimps in this ice.In ice production,4-hexylresorcinol and sodium metabisulphite were used at ratios 0.0025%and 0.625%,respectively.Shrimp(Aristaeomorpha foliacea)dipped in 4-hexylresorcinol and sodium metabisulphite solutions at ratios 0.005%and 1.25%,respectively,and shrimp kept in normal ice were considered as controls.The melanosis scores of the shrimp stored in ice containing antimelanotics were found to be lower than those of the shrimp dipped into antimelanotics.This situation showed that storage in ice containing antimelanotic agent delayed the formation of melanosis in shrimps.The highest TVB-N and TMA-N values were determined in the control group that did not contain antimelanotic agents.Shrimp stored in ice containing 4-hexylresorcinol had lower TVB-N and TMA-N values compared to sulphide added ice.The highest pH values were determined in shrimps dipped in 4-hexylresorcinol solution and in the control group.The pH values of shrimps treated with 4-hexylresorcinol were higher than those of shrimps treated with sulphite.The incorporation of antimelanotics to ice prevented the softening of shrimps and preserved the texture.Sodium metabisulphite and 4-hexylresorcinol showed a microbial inhibition effect in shrimp,and this effect was more clearly observed by storage in ice containing antimelanotic agent.Nalan Gokoglu Bahar Gumus Afsin Ceylan Mehmet Gokoglu 2022Food Bioscience2022,46,2:3
