| 1 | New Insights into Plant Isoprenoid Metabolism显示文摘Isoprenoids are a hugely diverse family of compounds derived from the C5 precursors isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP).Although all free-living organisms synthesize isoprenoids,they are particularly abundant and diverse in plants,with tens of thousands structures known to date.The highest variety of plant isoprenoids are specialized (secondary) metabolites that participate in the interaction of plants with their environment.These include pigments,volatiles,and defense compounds,some of which have applications in industry and agriculture.For example,isoprenoid drugs are used against cancer (taxol) or malaria (artemisin).But plants also synthesize isoprenoids with essential (primary) functions in respiration (ubiquinone),photosynthesis (carotenoids,chlorophylls,tocopherols,phylIoquinones,plastoquinone),membrane architecture (sterols),and growth regulation (brassinosteroids,cytokinins,gibberellins,abscisic acid,strigolactones).Despite their economic importance and biological relevance,our knowledge of the core pathways for the production of the universal isoprenoid precursors IPP and DMAPP in plant cells remained incomplete until the mid-1990s.Impressive progress in the last decade has resulted in the complete elucidation of several isoprenoid pathways,the identification of regulatory mechanisms,the discovery of new functions and properties of specific isoprenoids,and the successful manipulation of isoprenoid biosynthesis in a number of metabolic engineering approaches.In this update article,we will discuss some of the most recent advances in the plant isoprenoid field,focusing on the pathways supplying the C5 precursors in plant cells and the novel insights into regulatory matters. | Pablo Pulido Catalina Perello Manuel Rodriguez-Concepcion | 2012 | Molecular Plant2012,5,5: | 22 |
| 2 | Anti-oxidation actions of curcumin in two forms of bed rest:oxidative stress serum and salivary markers显示文摘Objective:To determine the preventive effects of curcumin on peroxidative damage under two bed rest conditions.Methods:20 healthy male(10 with curcumin and 10 without curcumin ) volunteers were selected.They were studied before,during,and just on bed rest conditions at -6°head-down-tilt(HDT) bed rest and bed rest position(BD) for 10 days.We measured the salivary and serum oxidative markers such as Malonaldehyde,8-hydroxydeoxyguanosine, vitamin C and E just before HDT & BD.during HDT & BD experiment,and in course time of recovery with curcumin and without curcumin groups.Results:The values of serum and salivary vitamin C & E showed statistically significant decrease in both bed rest conditions as compared to those of the conditions before and during the recovery stage.However,these levels were not significantly lowered in curcumin groups in contrast to the groups without curcumin (P>0.05).MDA and 8-OHdG levels showed significant increase in simulating microgravity and zero gravity conditions as compared to those before and in the recovery stage.However,these levels were lower in curcumin groups in contrast to the groups without curcumin(P<0.05).Serum and salivary correlation analysis revealed a strong and highly significant correlation for MDA. vitamin C & E and 8 dihydro-2 deoxyguanosine(8-OHdG) in the conditions before,during and in the recovery periods in both bed rest conditions.Since saliva collection is easy and noninvasive, measurements of salivary marker levels may prove to be useful in the space research. | 3 | Serum 25-hydroxyvitamin D concentration and inflammatory bowel disease characteristics in Romania显示文摘AIM:To describe the relationship between vitamin D levels and inflammatory bowel disease(IBD)characteristics in northeastern Romanian patients.METHODS:This was a prospective study of 47 consecutive IBD patients admitted to The Institute of Gastroenterology and Hepatology in Iasi,Romania between March 2011 and June 2012.The diagnosis of IBD was established based on endoscopic,histologic and radiologic findings.Demographic data,disease characteristics,ongoing treatments and biological parameters of patients(including markers of inflammation:C-reactive protein level,fibrinogen level,and erythrocyte sedimentation rate)were recorded.Serum vitamin D levels were measured and compared with age-and sexmatched healthy volunteers from the same geographic area.Vitamin D levels were defined as sufficient(>30ng/mL),insufficient(20-30 ng/mL),or severely defi-cient(<20 ng/mL).RESULTS:Thirty-three of the IBD patients included in this study had ulcerative colitis(UC)and 14 had Crohn’s disease(CD).Only 24%of the UC patients and 21%of the CD patients had sufficient vitamin D levels.The vitamin D levels were significantly lower in the CD patients with moderate to severe disease activity compared to the CD patients in remission or with mild disease activity(16±6 ng/mL vs 26±7 ng/mL;16±6 ng/mL vs 31±9 ng/mL,respectively,P<0.05).Vitamin D levels in the UC patients were not influenced by disease activity and no correlation was observed with the inflammation markers tested(C-reactive protein,fibrinogen,and erythrocyte sedimentation rate).No association was observed between vitamin D levels and smoking status or ongoing medication(5ASA,steroids,and anti-TNFα).Newly diagnosed IBD patients had lower vitamin D levels than patients with established cases,though these differences were not significant(UC:22±9 ng/mL vs 26±12 ng/mL;CD:18±6 ng/mL vs 27±11 ng/mL,respectively).Although no association was found between the season during which the visit was scheduled and vitamin D levels,the UC patients assessed during the winter tended to have lower levels than those assessed during the summer(22±9 ng/mL vs 28±13 ng/mL,respectively).CONCLUSION:Vitamin D levels are significantly reduced in IBD patients in northeastern Romania,with the lowest levels occurring in CD patients with moderate to severe disease activity. | Gabriela Dumitrescu Catalina Mihai Mihaela Dranga Cristina Cijevschi Prelipcean | 2014 | World Journal of Gastroenterology2014,20,9: | 8 |

| 5 | Portal hypertensive colopathy is associated with portal hypertension severity in cirrhotic patients显示文摘AIM:To assess the prevalence of portal hypertension (PH) related colorectal lesions in liver transplant candidates, and to evaluate its association with the severity of PH.METHODS: Between October 2004 and December 2005, colonoscopy was performed in 92 cirrhotic liver transplant candidates. We described the lesions resulting from colorectal PH and their association with the grade of PH in 77 patients who underwent measurement of hepatic venous pressure gradient (HVPG).RESULTS: Mean age was 55 years and 80.7% of patients were men. The main etiology of cirrhosis was alcoholism (45.5%). Portal hypertensive colopathy (PHC) was found in 23.9%, colonic varices in 7.6% and polyps in 38% of patients (adenomatous type 65.2%). One asymptomatic patient had a well-differentiated adenocarcinoma. The manifestations of colorectal PH were not associated with the etiology of liver disease or with the Child-Pugh grade. Ninety percent of patients with colopathy presented with gastroesopha-geal varices (GEV), and 27.5% of patients with GEV presented with colopathy (P=0.12). A relationship between higher values of HVPG and presence of col-opathy was observed (19.9±6.2 mmHg vs 16.8±5.4 mmHg, P=0.045), but not with the grade of col-opathy (P=0.13). Preneoplastic polyps and neoplasm (P=0.02) and spontaneous bacterial peritonitis (P=0.006) were more prevalent in patients with colopathy. It is expected that WSNs will be integrated into the Internet of Things(IoT)and participate in various tasks. WSNs play an important role monitoring and reporting environment information and collecting surrounding context. In this paper we consider a WSN deployed for an application such as environment monitoring, and a mobile sink which acts as the gateway between the Internet and the WSN. Data gathering is a challenging problem in WSNs and in the IoT because the information has to be available quickly and effectively without delays and redundancies. In this paper we propose several distributed algorithms for composite event detection and reporting to a mobile sink. Once data is collected by the sink, it can be shared using the IoT infrastructure. We analyze the performance of our algorithms using WSNet simulator, which is specially designed for event-based WSNs. | 10 | Red pigment production by Penicillium purpurogenum GH2 is influenced by pH and temperature显示文摘The combined effects of pH and temperature on red pigment production and fungal morphology were evaluated in a submerged culture of Penicillium purpurogenum GH2, using Czapek-Dox media with D-xylose as a carbon source. An experimental design with a factorial fix was used: three pH values (5, 7, and 9) and two temperature levels (24 and 34 °C) were evaluated. The highest production of red pigment (2.46 g/L) was reached with a pH value of 5 and a temperature of 24 °C. Biomass and red pigment production were not directly associated. This study demon- strates that P. purpurogenum GH2 produces a pigment of potential interest to the food industry. It also shows the feasibility of producing and obtaining natural water-soluble pigments for potential use in food industries. | 13 | Immunotherapy and immunoescape in colorectal cancer显示文摘Immunotherapy encompasses a variety of interventions and techniques with the common goal of eliciting tumor cell destructive immune responses. Colorectal carcinoma often presents as metastatic disease that impedes curative surgery. Novel strategies such as active immunization with dendritic cells (DCs), gene transfer of cytokines into tumor cells or administration of immunostimulatory monoclonal antibodies (such as anti-CD137 or anti-CTLA-4) have been assessed in preclinical studies and are at an early clinical development stage. Importantly, there is accumulating evidence that chemotherapy and immunotherapy can be combined in the treatment of some cases with colorectal cancer, with synergistic potentiation as a result of antigens cross-presented by dendritic cells and/or elimination of competitor or suppressive T lymphocyte populations (regulatory T-cells). However, genetic and epigenetic unstable carcinoma cells frequently evolve mechanisms of immunoevasion that are the result of either loss of antigen presentation, or an active expression of immunosuppressive substances. Some of these actively immunosuppressive mechanisms are inducible by cytokines that signify the arrival of an effector immune response. For example, induction of 2, 3 indoleamine dioxygenase (IDO) by IFNγ in colorectal carcinoma cells. | 18 | Management of Helicobacter pylori infection in Latin America: A Delphi technique-based consensus显示文摘AIM: To optimize diagnosis and treatment guidelines for this geographic region, a panel of gastroenterologists, epidemiologists, and basic scientists carried out a structured evaluation of available literature.METHODS: Relevant questions were distributed among the experts, who generated draft statements for consideration by the entire panel. A modified three-round Delphi technique method was used to reach consensus. Critical input was also obtained from representatives of the concerned medical community. The quality of the evidence and level of recommendation supporting each statement was graded according to United States Preventive Services Task Force criteria.RESULTS: A group of ten experts was established. The survey included 15 open-ended questions that were distributed among the experts, who assessed the articles associated with each question. The levels of agreement achieved by the panel were 50% in the first round, 73.3% in the second round and 100% in the third round. | 20 | The genus Thelonectria(Nectriaceae,Hypocreales,Ascomycota)and closely related species with cylindrocarpon-like asexual states显示文摘The genus Thelonectria and closely related species with cylindrocarpon-like asexual states are a group of perithecial ascomycetes in the family Nectriaceae that occur as saprobes and in few cases as pathogens of hardwood trees,shrubs or other plants.Although a key component of forest ecosystems around the world,species relationships and distributions of these fungi are largely unknown.The objectives of this study were to:1)infer species rank phylogenetic relationships of the genus Thelonectria and closely related species with cylindrocarpon-like asexual states and test the monophyly of each of the groups studied;2)delimit taxa establishing taxon circumscriptions;3)resolve nomenclatural issues by identifying redundantly used names and synonyms;and 4)provide an updated outline to the genus,geographical distributions data and identification tools,specifically diagnostic keys and molecular data that can be used as molecular barcodes.The recovered consensus phylogeny resulted in a narrow circumscription of the genus Thelonectria,based on the type T.discophora,excluding one of the common species T.jungneri.According to the phylogenetic analyses,T.jungneri belongs in a segregate clade that should be recognized as a different genus.In the genus Thelonectria,a total of four new species and three new combinations are recognized.Additionally,three new genera,closely related to Thelonectria,are described to accommodate species displaying a morphological resemblance to those of Thelonectria:Cinnamomeonectria gen.nov.with C.cinnamomea as type species,Macronectria gen.nov.with M.jungneri as type species and including four additional newly described species,and Tumenectria gen.nov.with T.laetidisca as type species. | Catalina Salgado-Salazar Amy YRossman Priscila Chaverri | 2016 | Fungal Diversity2016,,5: | 2 |
