| 3 | A complete compilation of matrix metalloproteinase expression in human malignant gliomas显示文摘Glioblastomas are characterized by an aggressive local growth pattern, a marked degree of invasiveness and poor prognosis. Tumor invasiveness is facilitated by the increased activity of proteolytic enzymes which are involved in destruction of the extracellular matrix of the surrounding healthy brain tissue. Elevated levels of matrix metalloproteinases(MMPs) were found in glioblastoma(GBM) cell-lines, as well as in GBM biopsies as compared with low-grade astrocytoma(LGA) and normal brain samples, indicating a role in malignant progression. A careful review of the available literature revealed that both the expression and role of several of the 23 human MMP proteins is controversely discussed and for some there are no data available at all. We therefore screened a panel of 15 LGA and 15 GBM biopsy samples for those MMPs for which there is either no, very limited or even contradictory dataavailable. Hence, this is the first complete compilation of the expression pattern of all 23 human MMPs in astrocytic tumors. | 4 | Polo-like kinase 1 expression is a prognostic factor in human colon cancer显示文摘AIM: To clarify the expression patterns and prognostic implications of the mitotic regulator Polo-like kinase 1(PLK1) in colon cancer.METHODS: Expression of PLK1 was investigated by immunohistochemistry (158 cases) and immunoblotting in tissue of colon adenomas and adenocarcinomas. PLK1expression patterns were correlated with clinicopathological parameters and patient prognosis. In addition, expression of PLK1 was evaluated by immunoblot and PCR in colon carcinoma cell lines, and coexpression of PLK1 with the proliferation marker Ki-67 was investigated.RESULTS: Weak PLK1 expression was observed in normal colon mucosa and adenomas. In contrast, 66.7% of carcinomas showed strong expression of PLK1.Overexpression of PLK1 correlated positively with Dukes stage (P<0.001), tumor stage (P = 0.001) and nodal status (P<0.05). Additionally, PLK1 expression was a prognostic marker in univariate survival analysis (P<0.01) and had independent prognostic significance (RR = 3.3, P = 0.02)in patients with locoregional disease. Expression of PLK1 mRNA and protein was detected in all cell lines investigated. Coexpression of PLK1 and Ki-67 was observed in the majority of colon cancer cells, but a considerable proportion of cells showed PLK1 positivity without Ki-67expression.CONCLUSION: PLK1 is a new prognostic marker for colon carcinoma patients and may be involved in tumorigenesis and progression of colon cancer. | 11 | Vessel density in OCT angiography permits differentiation between normal and glaucomatous optic nerve heads显示文摘AIM:To evaluate whether optical coherence tomography angiography(OCTA) can detect altered vessel density(VD) at the optic nerve head(ONH) in glaucoma patients.Special attention is paid to the accuracy of the OCTA technique for distinguishing healthy from glaucomatous eyes.METHODS:A total of 171 eyes were examined by the OCTA system Angio Vue^(TM)(Optovue):97 eyes diagnosed with glaucoma and 74 healthy control eyes.The papillary and peripapillary VD was measured.Furthermore,the VD was correlated with different structural and functional measurements.In order to test the accuracy of differentiation between eyes with and without glaucoma,we calculated the receiver operating characteristic curve(ROC) and the area under the curve(AUC).RESULTS:The papillary and peripapillary VD in glaucomatous eyes was significantly lower than in healthy eyes(P<0.05).The VD of the nasal peripapillary sector was significantly lower than in the other sectors.The further the disease had progressed [measured by determining the thickness of the ganglion cell complex(GCC) and the retinal nerve fiber layer(RNFL)] the greater the VD reduction.The AUC discriminated well between glaucomatous and normal eyes(consensus classifier 94.2%).CONCLUSION:OCTA allows non-invasive quantification of the peripapillary and papillary VD,which is significantly reduced in glaucomatous eyes and accurately distinguishes between healthy and diseased eyes.OCTA expands the spectrum of procedures for detecting and monitoring glaucoma. | Claudia Lommatzsch Kai Rothaus Joerg Michael Koch Carsten Heinz Swaantje Grisanti | 2018 | International Journal of Ophthalmology(English edition)2018,11,5: | 9 |
| 12 | 铅冷能源系统中液态金属与铁基合金相容性的研究进展显示文摘核电是一种可以有效解决能源与环境问题的清洁能源,受到国际上的广泛重视。目前的商用堆以第二代及第三代热中子反应堆为主,存在铀资源利用率低、放射性废物不断积累和潜在的核安全等问题,故具有更高安全性和经济性的第四代核能系统的研究成为当前的热点。四代堆中采用液态铅或铅基合金作为主冷却剂的反应堆称为铅冷快堆,由于液态铅及其合金具有优异的热工性能及核物理性能,铅冷快堆被认为是最有希望率先得到应用的第四代反应堆之一,液态铅也是加速器驱动次临界系统重要的液态靶材料兼冷却剂。此外,液态铅也被认为是太阳能热电系统最有前景的能量交换介质之一。铅冷堆具有可在较高温度条件下运行、发电效率高等明显优势,但也由于堆内的服役温度和中子辐照强度较高,对关键结构材料的服役性能提出很高要求。尤其是大多数合金在液态铅中都会由于合金元素的选择性溶解带来明显的腐蚀问题,结构材料与液态铅的相容性问题是铅冷能源系统工程应用的重要瓶颈。堆内的腐蚀情况包括材料的溶解和氧化、固液两相的输运以及腐蚀产物和杂质之间的反应等,是一个复杂的过程。腐蚀行为的影响因素包括材料自身特点及外界因素,例如材料类型、微观结构、化学成分和表面状态以及冷却剂类型、温度、氧浓度、流速和腐蚀时间等都会对腐蚀产生重要的影响。本文针对制约铅冷能源系统用结构材料发展的基础核心问题,把握材料成分和显微组织特点与其在液态铅中的溶解和氧化问题之间的关系这一主线,就国内外关于液态铅对合金的溶解腐蚀基础问题,金属及非金属腐蚀抑制剂的发展,不锈钢、氧化物弥散强化(ODS)钢、含Al奥氏体耐热钢(AFA)及FeCrAl合金在液态铅基中的腐蚀行为等研究内容进行总结和分析,对影响腐蚀的因素进行归纳,列举材料的腐蚀过程和机制以及相应的腐蚀产物和结构,分析各元素在腐蚀过程中对氧化层的作用及扩散迁移模式,并对存在的问题进行总结和展望,为满足铅冷能源系统关键结构材料的发展提供依据和参考. | 陈灵芝 周张健 Carsten Schroer | 2020 | 材料导报2020,34,5: | 9 |

| 16 | Oxidative stress: the lowest common denominator of multiple diseases显示文摘Oxygen is essential to the human life and life of all aerobic organisms. The complete oxidation of nutrients for the biological energy supply is one of the most important prerequisites for the formation of higher life forms. However, cells that benefit from oxidative respiration also suffer from reactive oxygen species because they adapted to oxygen as an energy source. Healthy cells balance the formation and elimination of reactive oxygen species thereby creating and keeping reactive oxygen species-homeostasis. When the concentration of free radicals exceeds a critical level and homeostasis is disturbed, oxidative stress occurs leading to damage of multiple cellular molecules and compartments. Therefore, oxidative stress plays an important role in the physiology and pathology of various diseases. Often, the antioxidant protection system becomes pathologically unbalanced in the genesis of several diseases, leading to functional losses of the organism, as in the case of amyotrophic lateral sclerosis, or cells develop metabolic mechanisms to use this system as protection against external influences, such as in the case of glioblastoma cells. Either way, understanding the underlying deregulated mechanisms of the oxidative protection system would allow the development of novel treatment strategies for various diseases. | 19 | 雌激素受体相关受体亚型在卵巢癌中的表达及其与预后的关系显示文摘目的探讨雌激素受体相关受体(ERRs)亚型α、β、γ在卵巢癌中的表达及其与预后的关系。方法激光共聚焦显微镜观察ERRα蛋白在卵巢癌细胞株SKOV3和OVCAR3细胞内的定位。RT-PCR技术检测33份卵巢癌组织以及12份正常卵巢组织中3种ERRs亚型的mRNA表达,并结合临床病理指标分析其预后情况。结果ERRα蛋白主要分布于SKOV3和OVCAR3细胞的细胞核内。卵巢癌组织中ERRαmRNA的阳性表达率(58%)和ERRγmRNA的阳性表达率(48%)显著高于正常卵巢组织(分别为17%和33%,P<0.05);ERRβmRNA的阳性表达率在卵巢癌组织和正常卵巢组织中都很低(分别为9%和0),两者比较,差异无统计学意义(P=0.795)。ERRαmRNA的阳性表达与卵巢癌的手术病理分期(r=0.639,P=0.017)、病理分级(r=0.520,P=0.022)呈正相关。ERRαmRNA阳性表达的卵巢癌患者,其中位数整体生存时间(19.0个月)明显短于ERRαmRNA阴性表达患者(31.5个月,P=0.015),但两者间的无瘤生存时间(分别为12.6和14.5个月)比较,差异无统计学意义(P=0.820);ERRγmRNA阳性表达患者的中位数无瘤生存时间(18.0个月)显著长于ERRγmRNA阴性表达患者(13.5个月,P=0.020),但两者间中位数整体生存时间(分别为23.4和19.6个月)比较,差异无统计学意义(P=0.093)。结论ERRα蛋白主要表达于卵巢癌细胞核内。ERRαmRNA高表达与卵巢癌患者的预后差有关,而ERRγmRNA高表达可能提示患者预后较好。 | 孙蓬明 魏丽惠 Jalid Sehouli Carsten Denkert 赵丹 高敏 孙秀丽 Werner Litchtenegger | 2005 | 中华妇产科杂志2005,40,8: | 6 |
| 20 | The IL-1R/TLR signaling pathway is essential for efficient CD8+ T-cell responses against hepatitis B virus in the hydrodynamic injection mouse model显示文摘The outcome of hepatitis B viral(HBV)infection is determined by the complex interactions between replicating HBV and the immune system.While the role of the adaptive immune system in the resolution of HBV infection has been studied extensively,the contribution of innate immune mechanisms remains to be defined.Here we examined the role of the interleukin-1 receptor/Toll-like receptor(IL-1R/TLR)signaling pathway in adaptive immune responses and viral clearance by exploring the HBV mouse model.Hydrodynamic injection with a replication-competent HBV genome was performed in wild-type mice(WT)and a panel of mouse strains lacking specific innate immunity component expression.We found higher levels of HBV protein production and replication in Tlr2^(−/−),Tlr23479^(−/−),3d/Tlr24^(−/−),Myd88/Trif^(−/−)and Irak4^(−/−)mice,which was associated with reduced HBV-specific CD8+T-cell responses in these mice.Importantly,HBV clearance was delayed for more than 2 weeks in 3d/Tlr24^(−/−),Myd88/Trif^(−/−)and Irak4^(−/−)mice compared to WT mice.HBV-specific CD8+T-cell responses were functionally impaired for producing the cytokines IFN-γ,TNF-αand IL-2 in TLR signaling-deficient mice compared to WT mice.In conclusion,the IL-1R/TLR signaling pathway might contribute to controlling HBV infection by augmenting HBV-specific CD8+T-cell responses. | Zhiyong Ma Jia Liu Weimin Wu Ejuan Zhang Xiaoyong Zhang Qian Li Gennadiy Zelinskyy Jan Buer Ulf Dittmer Carsten J Kirschning Mengji Lu | 2017 | Cellular & Molecular Immunology2017,14,12: | 6 |
