| 1 | Chronic intestinal pseudo-obstruction显示文摘Chronic intestinal pseudo-obstruction (CIPO) is a se- vere digestive syndrome characterized by derangement of gut propulsive motility which resembles mechanical obstruction, in the absence of any obstructive process. Although uncommon in clinical practice, this syndrome represents one of the main causes of intestinal failure and is characterized by high morbidity and mortality. It may be idiopathic or secondary to a variety of diseases. Most cases are sporadic, even though familial forms with either dominant or recessive autosomal inheritance have been described. Based on histological features in- testinal pseudo-obstruction can be classified into three main categories:neuropathies, mesenchymopathies, and myopathies, according on the predominant involvement of enteric neurones, interstitial cells of Cajal or smooth muscle cells, respectively. | 2 | Pioneering function of Isl1 in the epigenetic control of cardiomyocyte cell fate显示文摘Generation of widely differing and specialized cell types from a single totipotent zygote involves large-scale transcriptional changes and chromatin reorganization. Pioneer transcription factors play key roles in programming the epigenome and facilitating recruitment of additional regulatory factors during successive cell lineage specification and differentiation steps. Here we show that Isl1 acts as a pioneer factor driving cardiomyocyte lineage commitment by shaping the chromatin landscape of cardiac progenitor cells. Using an Isl1 hypomorphic mouse line which shows congenital heart defects, genome-wide profiling of Isl1 binding together with RNA- and ATAC-sequencing of cardiac progenitor cells and their derivatives, we uncover a regulatory network downstream of Isl1 that orchestrates cardiogenesis. Mechanistically, we show that Isl1 binds to compacted chromatin and works in concert with the Brg1-Baf60c-based SWI/SNF complex to promote permissive cardiac lineage-specific alterations in the chromatin landscape not only of genes with critical functions in cardiac progenitor cells, but also of cardiomyocyte structural genes that are highly expressed when Isl1 itself is no longer present. | 3 | Integrating beneficiaries into assessment of ecosystem services from managed forests at the Hubbard Brook Experimental Forest,USA显示文摘Background:Forests contribute to human wellbeing through the provision of important ecosystem services.Methods:In this study,we investigated how the perceived importance of ecosystem services may impact the overall benefit provided by managed watersheds at the Hubbard Brook Experimental Forest over a 45-year period,using standardized measures of service capacity weighted by service importance weights derived from a survey of beneficiaries.Results:The capacity of watersheds to regulate water flow and quality was high in all watersheds throughout the study period,whereas cultural services such as scenic beauty declined after harvest.Impacts on greenhouse gas regulation depended on the efficiency with which harvested biomass was used.Surveys revealed that stakeholders placed high value on all ecosystem services,with regulating and cultural services seen as more important than provisioning services.When service metrics were weighted by survey responses and aggregated into a single measure,total service provision followed the same overall trend as greenhouse gas regulation.Where biomass use was less efficient in terms of greenhouse gas emissions,harvesting resulted in an overall 'ecosystem service debt';where use was more efficient,this 'ecosystem service debt' was reduced.Beneficiaries' educational backgrounds significantly affected overall assessment of service provision.Beneficiaries with college or university degrees incurred smaller 'ecosystem service debts' and were less negatively affected by harvesting overall.Conclusions:This study highlights the importance of including empirical measures of beneficiary preference when attempting to quantify overall provision of ecosystem services to human beneficiaries over time. | Jesse Caputo Colin M.Beier Valerie A.Luzadis Peter M.Groffman | 2016 | Forest Ecosystems2016，3，4： | 2 |

| 6 | Peripheral leukocyte and endometrium molecular biomarkers of inflammation and oxidative stress are altered in peripartal dairy cows supplemented with Zn,Mn,and Cu from amino acid complexes and Co from Co glucoheptonate显示文摘Background: Immune dysfunction and a higher risk of uterine infections are characteristics of the transition into lactation in dairy cows. The supply of complexed trace minerals, which are more bioavailable, could help overcome the greater needs of these nutrients in tissues around parturition and early lactation.Results: Twenty Holstein cows received an oral bolus with a mix of inorganic trace minerals(INO) or complexed trace minerals(AAC) to achieve 75, 65, 11, and 1 ppm supplemental Zn, Mn, Cu, and Co, respectively, in the total diet dry matter from -30 d through +30 d relative to parturition. Blood for polymorphonuclear leukocyte(PMNL) isolation was collected at-30,-15, +10, and + 30 d relative to parturition, whereas endometrium biopsies were performed at +14 and +30 d. Feeding AAC led to greater PMNL expression of genes related with inflammation response(DDX58), oxidative stress response(MPO), eicosanoid metabolism(PLA2G4A and ALOX5AP), transcription regulation(PPARG), and cellular adhesion(TLN1). The upregulation by AAC in endometrium of genes related with inflammation response( TLR2, TLR4, NFKB1, TNF, IL6, IL1 B, IL10, IL8), prostaglandin synthesis(PTGS2, PTGES), and antioxidant responses(NFE2 L2, SOD1) indicated a faster remodeling of uterine tissue and potentially greater capacity to control a local bacterial invasion.Conclusions: Data indicate that trace mineral supplementation from amino acid complexes improves PMNL activity and allows the prompt recovery of uterine tissue during early lactation.
