1Sequencing a Juglans regia×J.microcarpa hybrid yields high-quality genome assemblies of parental species显示文摘Members of the genus Juglans are monecious wind-pollinated trees in the family Juglandaceae with highly heterozygous genomes,which greatly complicates genome sequence assembly.The genomes of interspecific hybrids are usually comprised of haploid genomes of parental species.We exploited this attribute of interspecific hybrids to avoid heterozygosity and sequenced an interspecific hybrid Juglans microcarpa×J.regia using a novel combination of single-molecule sequencing and optical genome mapping technologies.The resulting assemblies of both genomes were remarkably complete including chromosome termini and centromere regions.Chromosome termini consisted of arrays of telomeric repeats about 8 kb long and heterochromatic subtelomeric regions about 10 kb long.The centromeres consisted of arrays of a centromere-specific Gypsy retrotransposon and most contained genes,many of them transcribed.Juglans genomes evolved by a whole-genome-duplication dating back to the Cretaceous-Paleogene boundary and consist of two subgenomes,which were fractionated by numerous short gene deletions evenly distributed along the length of the chromosomes.Fractionation was shown to be asymmetric with one subgenome exhibiting greater gene loss than the other.The asymmetry of the process is ongoing and mirrors an asymmetry in gene expression between the subgenomes.Given the importance of J.microcarpa×J.regia hybrids as potential walnut rootstocks,we catalogued disease resistance genes in the parental genomes and studied their chromosomal distribution.We also estimated the molecular clock rates for woody perennials and deployed them in estimating divergence times of Juglans genomes and those of other woody perennials.Tingting Zhu Le Wang Frank M.You Juan C.Rodriguez Karin R.Deal Limin Chen Jie Li Sandeep Chakraborty Bipin Balan Cai-Zhong Jiang Patrick J.Brown Charles ALeslie Mallikarjuna KAradhya Abhaya M.Dandekar Patrick E.McGuire Daniel Kluepfel Jan Dvorak Ming-Cheng Luo 2019Horticulture Research2019,6,1:7
2The cellular metabolic landscape in the tumor milieu regulates the activity of myeloid infiltrates显示文摘Malignant cells upregulate distinct energy metabolism programs that support their proliferation,migration,and adaptation to the stressful tumor microenvironment(TME).Additionally,this exaggerated metabolic activity allows cancer cells to hijack essential nutrients and outcompete neighboring infiltrating immune cells,thereby impairing antitumor immunity.During recent years,there has been great interest in the field to understand the tumor-induced energy metabolism signals that regulate the function of immune cells in individuals with cancer.Accordingly,it is now well accepted that uncovering the mechanisms that instruct the metabolic behavior of cancer cells and tumor-associated immune cells is an indispensable strategy for the development of new approaches to overcome immune suppression in tumors.Thus,in this minireview,we briefly discuss the interaction between particular metabolic signaling pathways and immunosuppressive activity in different subsets of myeloid cells within the TME.Additionally,we illustrate potential central mechanisms controlling the metabolic reprogramming of myeloid cells in response to tumor-derived factors.Eslam Mohamed Amir A.Al-Khami Paulo C.Rodriguez 2018Cellular & Molecular Immunology2018,15,5:2

5Antibacterial activity of Leptogium cochleatum and Leptogium moluccanum显示文摘Currently there is an increase in the emergence and re-emergence of bacterial infections,and lichens are potential sources of pharmaceutically important secondary metabolites to halt this problem.Lichens are unique plant-like organisms which are relatively less explored in the Philippines.Here,lichen extracts of Leptogium cochleatum and Leptogium moluccanum were tested against Escherichia coli,Enterobacter agglomerans,Klebsiella pneumoniae,Pseudomonas aeruginosa,and Staphylococcus aureus using paper disk diffusion assay.Hexane extract of L.moluccanum led to a 17.14 mm zone of inhibition(ZOI)against K.pneumoniae,while hexane extract of L.cochleatum led to a 15.28 mm ZOI against E.agglomerans.Dichloromethane extracts of L.cochleatum exhibited a ZOI of 15.86 mm against E.agglomerans and 14.84 mm ZOI against E.coli.The bioactivities exhibited by the lichen extracts may be due to the secondary metabolites detected using thin layer chromatography.Indeed,lichen-derived bioactive compounds hold great promise for pharmaceutical applications for the benefit of human life.Andrea Pauline B.Manlapaz Mitzi I.Mariano Odessa Rona M.Reyes Laureena C.Rodriguez Jaycee Augusto G.Paguirigan 2022Studies in Fungi2022,7,1:0
6Work-related quality of life and performance appraisal among nurses at a tertiary hospital in Philippines显示文摘Objective:The objective of the study is to describe the work-related quality of life(WRQOL)among nurses and explore its association with performance evaluation ratings.Methods:A cross-sectional design was utilized measuring the WRQOL scale and the previous performance appraisal rating.Multistage sampling approach was utilized with nurses stratified based on their position and then systematically random sampled based on their unit assignment.Results:One hundred and eighty-two nurses were included.About half reported a high quality of work life(101;55.5%).Low scores were noted on the subscale working conditions(100;54.9%),low to average responses for home-work interface(109;59.9%),control at work(100;54.9%),and stress at work(90;49.5%).A higher proportion of nurses reported positive responses toward the areas of general well-being(113;62.1%)and job-career satisfaction(112;61.5%),than in the other subscales.There were notable differences between WRQOL ratings:(1)Head nurses had the highest perceived quality of work life,followed by nurse supervisors and charge nurses(F=6.1,P<0.01)and(2)Nurses in the pay-patient services reported lower quality of working life,while those in office and outpatient services had more positive scores(F=4.6,P<0.01).Conclusion:Only more than half of the nurses reported a high quality of work life,some of its dimensions,particularly job and career satisfaction and working conditions,appeared to vary in the perceived degree across years in service,work hours,and position.The assessment of the quality of work life may serve as an important tool to address staff burnout,absenteeism and other issues that affect job performance among health-care professionals.Alvin Duke R.SY Ma.Krisstella D.GONZALES Rachel Camille C.RODRIGUEZ 2023Journal of Integrative Nursing2023,5,3:0
