| 1 | Identification and classification of all potential hemolysin encoding genes and their products from Leptospira interrogans serogroup Icterohaemorrhagiae serovar Lai显示文摘Aim: To identify and classify all potential hemolysin candidates of Leptospira interrogans serogroup Icterohaemorrhagiae serovar Lai. Methods: All of the potential hemolysin encoding genes were characterized in silico. These genes were cloned and expressed in Escherichia coli. The hemolytic activities of the expressed proteins were assayed observing the hemolysis on sheep blood agar plates. Sphingomyelinase activities of the hemolysin candidates were measured by thin-layer chromatography (TLC) and HPLC for sphingomyelin-hydrolysis.Expression and secretion of the hemolysins in L interrogans were studied by reverse transcription polymerase chain reaction, Western blot, and enzyme-linked immunosorbent assays. Results and Conclusion: The hemolytic activities of hemolysin candidates (LA0327, LA0378, LA1027, LA1029, LA1650, LA3050, LA3937, LA4004) from L interrogans strain Lai were confirmed. They were further divided into two groups, sphingomyelinase hemolysins and non-sphingomyelinase hemolysins, based on their ability to hydrolyze sphingomyelin. Most of these hemolysins were actually expressed in living L interrogans and some of them were secreted into the environment. | 2 | Characterization of a novel toxin-antitoxin module,VapBC,encoded by Leptospira interrogans chromosome显示文摘Comparative genomic analysis of the coding sequences (CDSs) of Leptospira interrogans revealed a pair of closely linked genes homologous to the vapBC loci of many other bacteria with respect to both deduced amino acid sequences and operon organizations. Expression of single vapC gene in Escherichia coli resulted in inhibition of bacterial growth,whereas co-expression of vapBC restored the growth effectively. This phenotype is typical for three other characterized toxin-antitoxin systems of bacteria, i.e., mazEF[1], relBE[2] and chpIK[3]. The VapC proteins of bacteria and a thermophilic archeae, Solfolobus tokodaii, form a structurally distinguished group of toxin different from the other known toxins of bacteria. | 4 | E24 profile slope stability analysis in Haizhou Opencast Coal Mine of Fuxin显示文摘The E24 profile slope analyzed belongs to a series of excavated slopes of the Haizhou Opencast Coal Mine. It seems to be divided into Downslope Part and Upslope Part. Its profile comprises two noticeable coal seams, called the 8# and 9# weak layers, considered as the potential failure surfaces. In consideration of the actual configuration as in the perspective of any modification, assessing the stability of this slope with various profile forms under given conditions, and assessing the risk of instability and quantifying the influence of earthworks or other modifications to the stability of this slope, have constituted the primordial objectives carried out. From assumed potential failure surfaces, any specific profiles and specified slip surfaces are defined. A factor of safety (FoS) is computed for each specified slip surface; the smallest FoS found corresponds to the least favorable slip surface. The safety factor values obtained are compared to the suggested safety factor. Limit equilibrium methods of vertical slices implemented in Slope/W, computer program for slope stability analyses, have been adopted to perform the E24 slope stability analysis. The safety factor values computed with 9# weak layer are lower than for 8#; the factors of safety obtained with Sarma's method are the smallest; more, without groundwater (long term) overall values are greater than those determined under groundwater condition (short term). The lowest safety factor value is found for a profile depending on an adopted earthwork sequence. The E24 profile slope stability analysis shows the instability risk for the deepest weak layer, and also shows the short and long term stability of this slope for the envisaged earth movements.
