2Current progress and prospects of induced pluripotent stem cells显示文摘Induced pluripotent stem(iPS) cells are derived from somatic cells by ectopic expression of few transcription factors.Like embryonic stem(ES) cells,iPS cells are able to self-renew indefinitely and to differentiate into all types of cells in the body.iPS cells hold great promise for regenerative medicine,because iPS cells circumvent not only immunological rejection but also ethical issues.Since the first report on the derivation of iPS cells in 2006,many laboratories all over the world started research on iPS cells and have made significant progress.This paper reviews recent progress in iPS cell research,including the methods to generate iPS cells,the molecular mechanism of reprogramming in the formation of iPS cells,and the potential applications of iPS cells in cell replacement therapy.Current problems that need to be addressed and the prospects for iPS research are also discussed.CHEN LingYi & Liu Lin Key Laboratory of Bioactive Materials of Ministry of Education,College of Life Sciences,Nankai University,Tianjin 300071,China 2009Science China(Life Sciences)2009,52,7:11
3Inhibition of Emodin on LPS-induced Nitric Oxide Generation by Suppressing PLC-γ Phosphorylation in Rat Peritoneal Macrophages显示文摘Objective To investigate the inhibitory mechanism of emodin on lipopolysaccharide (LPS)-induced nitric oxide (NO) generation in rat peritoneal macrophages. Methods NO production and iNOS expression were measured through nitrite assay and Western blotting assay, respectively. NF-κB activity and nuclei P65 expression were estimated by dual-luciferase and Western blotting assay, respectively. Intracellular free Ca2+ ([Ca2+]i) was detected using the ratiometric fluorescent calcium indicator dye, Fura-2, and a microspectrofluorometer. PLC-γ phosporylation was analyzed by Western blotting assay. Results First, emodin was found playing active roles in suppressing LPS-induced NF-κB activation in rat peritoneal macrophages. Second, emodin down-regulated transient [Ca2+]i and could increase in NF-κB upstream signal. Finally, emodin suppressed phosphorylation of PLC-γ by LPS stimulation in the upstream of [Ca2+]i. 4Core-shell fibrous vascular grafts with the nitric oxide releasing property显示文摘Small-diameter vascular grafts with the nitric oxide(NO) releasing property were designed and prepared.Diazeniumdiolated N,N'-dibutyl-1,6-hexanediamine(DBHD/N2O2) was first synthesized as the NO donor and doped into the biocompatible polymer poly(ε-caprolactone)(PCL).The fibrous vascular grafts were fabricated by electrospinning.Despite the reduced platelet adhesion observed on the NO releasing grafts,the cytotoxicity and burst release were apparent,especially at a higher loading level of the NO donor.In order to solve this problem,core-shell structured nanofibers,in which PCL or gelatin is the shell and PCL doped with the NO donor is the core,were fabricated via coaxial electrospinning.The core-shell structure made the NO release more steady and controllable,and the cell compatibility was improved.ZHANG Lei,WANG Kai,ZHAO Qiang,ZHENG WenTing,WANG ZhiHong,WANG ShuFang & KONG DeLing Key Laboratory of Bioactive Materials,Ministry of Education College of Life Science,Nankai University,Tianjin 300071,China 2010Science China Chemistry2010,53,3:6
5High performance liquid chromatographic separation of thirteen drugs collected in Chinese Pharmacopoeia 2010(Ch.P2010) on cellulose ramification chiral stationary phase显示文摘The enantiomers separation of thirteen drugs collected in Ch.P2010 was performed on chiral stationary phase of cellulose ramification (chiralpak OD and chiralpak OJ) by high performance liquid chromatographic (HPLC) methods, which included ibuprofen (C1), ketoprofen (C2), nitrendipine (C3), nimodipine (C4), felodipine (C5), omeprazole (C6), praziquantel (C7), propranolol hydrochloride (C8), atenolol (C9), sulpiride (C10), clenbuterol hydrochloride (C11), verapamil hydrochloride (C12), and chlorphenamine maleate (C13). The mobile phase consisted of isopropanol and n-hexane. The detection wavelength was set at 254 nm and the fiow rate was 0.7 mL/min. The enantiomers separation of these thirteen racemates on chiralpak OD column and chiralpak OJ column was studied, while the effects of proportion of organic additives, alcohol displacer and temperature on the separation were studied. And the mechanism of some of racemates was discussed. The results indicated that thirteen chiral drugs could be separated onchiral stationary phase of cellulose ramification in normal phase chromatographic system. The chromatographic retention and resolution of enantiomers could be adjusted by factors including column temperature and the concentration of alcohol displacer and organic alkaline modifier in mobile phase. It was shown that the resolution was improved with reducing concentration of alcohol displacer. When concentration of organic alkaline modifier was 0.2% (v/v), the resolution and the peak shape were fairly good. Most racemates mentioned above had better resolution at column temperature of 25 1C. 7Computer Aided Study of Mechanism of Immunoadsorbent for Myasthenia Gravis显示文摘Myasthenia Gravis (MG) is an organ specific autoimmune disease mediated by autoantibodies (AChR Ab) against the acetylcholine receptor (AChR). Literature reported that the AChR Ab can be removed by absorbent linked with tryptophan. It was studied in detail in our lab. With the aid of computer, we docked some ligands into AChR Ab, and the results from scores of docking under different generations showed that there was no specific binding between tryptophan and scFv fragment just as the binding between antigen-antibody. 15A New Caffeate from the Aerial Parts of Anabasis aphylla显示文摘Objective To study the chemical constituents from the aerial parts of Anabasis aphylla. Methods The chemical constituents were isolated and purified by silica gel column and Sephadex LH-20 column chromatography. Spectroscopic methods such as MS and NMR spectra were used for the structural identification. Results A new caffeate ester, named eicosyl-(Z)-caffeate (1), along with fourteen known compounds was isolated from the EtOAc part. 17STRUCTURAL AND ADSORPTION RELATIONSHIP OF MACROPOROUS R ESIN IN HEMOPERFUSION显示文摘In order to achieve high adsorption capacity of adsorbents in hemoperfusion,macroporous resins were tailor made with appropriate pore sizes and specific area. This allows the toxin molecules to diffuse freely within the channels ofthe porous resins and rinally reach the adsorption sites.Macroporous resin NK - 107 having small pore size and large Surface area(1086cm2) were synthesized which showed a high adsorption capacity of 82 - 94%for small molecules such as hypnotic and sedative drugs. For the adsorption of middle molecules i. e. Vit B12 or cytochrome C adsorbents having large pore sizes weretailor made. In the case of the adsorption of unconjugated bilirubin which is a verybig molecule, the pore size of the adsorbent should be at least>160A. NK - 110 istailor made with a very big pore size,which have a high adsorption capacity of 87%for bilirubin. DNA immobilized on carbonized resins,which have a porous structurewith a large surface area of 1000m2/g,can adsorb DNA antibody effectively. 18Synthesis and Characterization of a Zwitterionic Copolymer Poly (ε-caprolactone)-b-poly(carboxybetaine methacrylate) with Good Cell Affinity显示文摘Design ofpolymeric scaffolds with specific physical and biological properties is a key objective of tissue engineering research. In this work, a novel copolymer poly (ε- caprolactone)- b- poly(carboxybetaine methacrylate) (PCL- PCBMA) containing zwitterion and polyester segments, was synthesized through ATRP technique. The chemical structure, composition,and molecular weight of the synthesized copolymer were characterized by 1HNMR and GPC. The polymer was further electrospun into fibrous film which has been characterized by SEM, XPS, water uptake test, and MTT cell culture assay. 20SYNTHESIS AND FLUORESCENCE STUDY OF SELF-AGGREGATION PROCESS IN AQUEOUS SOLUTION OF HYDROPHOBILIZED POLYSACCHARIDE显示文摘Cholesterol modified dextran (CHD) having self-aggregation or self-assembly property was synthesized from cholesterol and 1,6-hexyldiisocyanate. The degree of substitution of cholesteryl moiety in dextran main line is 3-5 cholesterols the 100 glucose units. We have prepared water solution of CHD using probe type sonifier and N-Phenyl-α-naphthylamine (PNA) as a fluorescent probe to study CHD self-aggregate process .For each solution of two samples, we found that the maximum emission of PNA in CHD solution was shifted to the lower wavelength and the intensity drastically increased above a critical CHD concentration.
