1Molecular mechanism about lymphogenous metastasis of hepatocarcinoma cells in mice显示文摘AIM To investigate the correlation between lymphogenous metastasis and matrix metalloproteinases (MMPs) activity and the expression of Fas Iigand of tumor cells in lymph nodes.METHODES Fifty-six inbred 615-mice were equally divided into 2 groups and inoculated with Hca-F and Hca-P cells. Their lymph node metastatic rates were examined.Growth fraction of lymphocytes in host lymph nodes was detected by flow cytometry. The Hca-F and Hca-P cells were cultured with extract of lymph node, liver or spleen.The quantity of MMPs in these supernatants was examined by zymographic analysis. The expression of Fas ligand,PCNA, Bcl-2 protein of Hca-F and Hca-P cells in the mice were examined by immunohistochemistry. The apoptosis signals of macrophages in lymph nodes were observed with in situ DNA fragmentation.RESULTS On the 28th day post-inoculation, the lymph node metastatic rate of Hca-F was 80% (16/20), whereas that of Hca-P was 25% (5/20). The growth fraction of lymphocytes was as follows: in the Hca-F cells, the proliferating peak of lymphocytes appeared on the 14th day post-inoculation and then decreased rapidly, while in HcaP cells, the peak appeared on the 7th day post-inoculation and then kept at a high level. With the extract of lymph node, the quantity of the MMP-9 activity increased (P<0.01) and active MMP-9 and MMP-2 were produced by both Hca-F and Hca-P tumor cells, which did not produce MMPs without the extract of lymph node or with the extracts of the liver and spleen. The expression of Fas Ligand of Hca-F cells was stronger than that of Hca-P cells (P<0.01). The expressions of PCNA and Bcl-2 protein of Hca-F cells in the tumors of inoculated area were the same as that of Hca-P cells. In situ DNA fragmentation showed that the positive signals of macrophages were around HcaF cells.CONCLUSION Secretion of MMPs which was associated with metastatic ability of Hca-F and Hca-P tumor cells depends on the environment of lymph nodes. 4Approximate Solution of Oil Film Load-carrying Capacity of Turbulent Journal Bearing with Couple Stress Flow显示文摘The instability of the rotor dynamic system supported by oil journal bearing is encountered frequently, such as the half-speed whirl of the rotor, which is caused by oil film lubricant with nonlinearity. Currently, more attention is paid to the physical characteristics of oil film due to an oil-lubricated journal bearing being the important supporting component of the bearing-rotor systems and its nonlinear nature. In order to analyze the lubrication characteristics of journal bearings efficiently and save computational efforts, an approximate solution of nonlinear oil film forces of a finite length turbulent journal bearing with couple stress flow is proposed based on Sommerfeld and Ocvirk numbers. Reynolds equation in lubrication of a finite length turbulent journal bearing is solved based on multi-parametric principle. Load-carrying capacity of nonlinear oil film is obtained, and the results obtained by different methods are compared. The validation of the proposed method is verified, meanwhile, the relationships of load-carrying capacity versus eccentricity ratio and width-to-diameter ratio under turbulent and couple stress working conditions are analyzed. The numerical results show that both couple stress flow and eccentricity ratio have obvious influence on oil film pressure distribution, and the proposed method approximates the load-carrying capacity of turbulent journal bearings efficiently with various width-to-diameter ratios. 6Land cover mapping using time series HJ-1 / CCD data显示文摘It is very difficult to have remote sensing data with both high spatial resolution and high temporal frequency; thus, two categories of land-use mapping methodology have been developed separately for coarser resolution and finer resolution data. The first category uses time series of data to retrieve the variation of land surface for classification, which are usually used for coarser resolution data with high temporal frequency. The second category uses fine spatial resolution data to classify different land surface. With the launch of Chinese satellite constellation HJ-1in 2008, four 30 m spatial resolution CCDs with about 360 km coverage for each one onboard two satellites made a revisit period of two days, which brought a new type of data with both high spatial resolution and high temporal frequency. Therefore, by taking the spatiotemporal advantage of HJ-1/CCD data we propose a new method for finer resolution land cover mapping using the time series HJ-1/CCD data, which can greatly improve the land cover mapping accuracy. 7Effects of bile acids on proliferation and ultrastructural alteration of pancreatic cancer cell lines显示文摘AIM: Pancreatic cancer in the head is frequently accompanied by jaundice and high bile acid level in serum. This study focused on the direct effects of bile acids on proliferation and ultrastructural alteration of pancreatic cancer.METHODS: Pancreatic cancer cell lines PANC-1, MIA PaCa2 and PGHAM-1 were explored in this study. The cell lines were cultured in media supplemented with certain bile acids,CA, DCA, LCA, TCDC, TDCA and GCA. Their influence on cell growth was measured with MTT assay after 72 h of incubation. Cell cycles of PANC-1 cells in 40 μM of bile acids media were analyzed by flow cytometry. Ultrastructural alteration of PANC-1 cells induced by DCA was observed using scanning and transmission electron microscope (SEM and TEM).RESULTS: At various concentrations of bile acids and incubation time, no enhanced effects of bile acids on cell proliferation were observed. Significant inhibitory effects were obtained in almost all media with bile acids. DCA and CA increased the percentage of G0+G1 phase cells, while GCA and TDCA elevated the S phase cell number. After 48 h of incubation in DCA medium, PANC-1 cells showed some structural damages such as loss of their microvilli and vacuolization of organelles in cytoplasm.CONCLUSION: Bile acids can reduce proliferation of pancreatic cancer cells due to their direct cytotoxicity. 8Modification of Inclusions in Liquid Iron by Mg Treatment显示文摘Effect of Mg addition on the compositions of inclusions were studied. The results show that Mg can minimize the inclusions of steel obviously. Under the present condition, Mg deoxidation products of low-S content experimental steel would be changed in the order of Al2O3→MgAl2O4→Mg–Al–O–S→Mg–O–S. Mg deoxidation products of high-S content experimental steel generate Mg–S(–O)+MnS type inclusions, except for usual oxysulfide. And it is consistent with the results of thermodynamics calculation. Mg is preferred to react with oxide, compared with sulfide. The reaction reaches the equilibrium after 1 min or 5 min. 9Preparation of the UPOV Guidelines for the Conduct of Tests for Distinctness, Uniformity and Stability–Tea Plant [Camellia sinensis (L.) O. Kuntze]显示文摘Distinctness, Uniformity and Stability (DUS) testing is the technical base of Plant Variety Protection (PVP) and the scientific basis for the approval of Plant Breeder’s Rights (PBR). DUS Test Guidelines are not only the technical manuals for the DUS testing authorities to conduct the testing, but also the technical standards for the competent authorities to examine the DUS of new varieties of plants. Tea plant, originated from Yunnan Province, China, is a very important woody cash species in the world. The International Union for the Protection of New Varieties of Plants (UPOV) DUS Test Guidelines for tea plant is the first Test Guidelines prepared by China for the UPOV. In this article, the subject, selection, and determination of characteristics, states of expression of characteristics and the selection of example varieties, assessment of the UPOV DUS Test Guidelines for tea plant were elucidated in detail. Finally, the proposal of PVP for tea plant in China was also proposed. 13A Diagnostic Analysis on the Effect of the Residual Layer in Convective Boundary Layer Development near Mongolia Using 20th Century Reanalysis Data显示文摘Although the residual layer has already been noted in the classical diurnal cycle of the atmospheric boundary layer,its effect on the development of the convective boundary layer has not been well studied. In this study, based on 3-hourly20 th century reanalysis data, the residual layer is considered as a common layer capping the convective boundary layer. It is identified daily by investigating the development of the convective boundary layer. The region of interest is bounded by(30°–60° N, 80°–120° E), where a residual layer deeper than 2000 m has been reported using radiosondes. The lapse rate and wind shear within the residual layer are compared with the surface sensible heat flux by investigating their climatological means, interannual variations and daily variations. The lapse rate of the residual layer and the convective boundary layer depth correspond well in their seasonal variations and climatological mean patterns. On the interannual scale, the correlation coefficient between their regional averaged(40°–50°N, 90°–110° E) variations is higher than that between the surface sensible heat flux and convective boundary layer depth. On the daily scale, the correlation between the lapse rate and the convective boundary layer depth in most months is still statistically significant during 1970–2012. 14Hybrid uncertainties-based analysis and optimization design of powertrain mounting systems显示文摘In this study,a hybrid uncertainties-based analysis and optimization method is presented for the designs of the powertrain mounting system(PMS)involving mixed uncertainties.In the presented method,the PMS parameters with sufficient data are treated as random variables,while those with limited information are defined as interval variables.Then,an uncertainty-based analysis method called as hybrid interval-random perturbation-central difference method(HIRP-CDM),is proposed to compute the hybrid interval-random outputs of the inherent characteristics of the PMS in concerned directions.In addition,the hybrid interval-random-Monte Carlo method(HIR-MCM)is developed to verify the computational accuracy of HIRP-CDM.Next,an optimization model mixed uncertainties is built up for the PMS design based on HIRP-CDM,in which the hybrid intervalrandom outputs of the concerned inherent characteristics are adopted to construct the design objective and constrains.The complex optimization problem can be effectively settled by means of HIRP-CDM.The effectiveness of the presented method is verified by a numerical example.CAI BoHao SHANGGUAN Wen-Bin LÜHui BO Tao 2020Science China(Technological Sciences)2020,63,5:4

17Shikonin attenuates hyperhomocysteinemia-induced CD4E^+T cell inflammatory activation and atherosclerosis in ApoE^−/− mice by metabolic suppression显示文摘T cell metabolic activation plays a crucial role in inflammation of atherosclerosis.Shikonin(SKN),a natural naphthoquinone with anti-inflammatory activity,has shown to exert cardioprotective effects,but the effect of SKN on atherosclerosis is unclear.In addition,SKN was found to inhibit glycolysis via targeting pyruvate kinase muscle isozyme 2(PKM2).In the present study,we investigated the effects of SKN on hyperhomocysteinemia(HHcy)-accelerated atherosclerosis and T cell inflammatory activation in ApoE^−/− mice and the metabolic mechanisms in this process.Drinking water supplemented with Hcy(1.8 g/L)was administered to ApoE^−/− mice for 2 weeks and the mice were injected with SKN(1.2 mg/kg,i.p.)or vehicle every 3 days.We showed that SKN treatment markedly attenuated HHcy-accelerated atherosclerosis in ApoE^−/− mice and significantly decreased inflammatory activated CD4^+T cells and proinflammatory macrophages in plaques.In splenic CD4^+T cells isolated from HHcy-ApoE^−/− mice,SKN treatment significantly inhibited HHcy-stimulated PKM2 activity,interferon-γsecretion and the capacity of these T cells to promote macrophage proinflammatory polarization.SKN treatment significantly inhibited HHcy-stimulated CD4^+T cell glycolysis and oxidative phosphorylation.Metabolic profiling analysis of CD4^+T cells revealed that Hcy administration significantly increased various glucose metabolites as well as lipids and acetyl-CoA carboxylase 1,which were reversed by SKN treatment.In conclusion,our results suggest that SKN is effective to ameliorate atherosclerosis in HHcy-ApoE^−/− mice and this is at least partly associated with the inhibition of SKN on CD4^+T cell inflammatory activation via PKM2-dependent metabolic suppression.Si-lin Lü Guo-hui Dang Jia-cheng Deng Hui-ying Liu Bo Liu Juan Yang Xiao-long Ma Yu-tong Miao Chang-tao Jiang Qing-bo Xu Xian Wang Juan Feng 2020Acta Pharmacologica Sinica2020,41,1:4
18Optimization of corn-stalk skin flake-wood shaving composite technology显示文摘We focused on the optimization of corn-stalk skin flake-wood shaving composite technology. We studied the effects of material-mixture ratio, glue content, hotpressing temperature, and hot-pressing time on the appearance, physical, and mechanical properties of the composite by the orthogonal experiment method. Our findings yielded highly significant results in all three cases: the effects of the material-mixture ratio on 2 h of thickness swelling(2h TS)and the modulus of rupture(MOR); the effects of glue content on 2h TS, internal bond strength, and modulus of elasticity(MOE); and the effects of hot-pressing temperature on MOR and MOE. 19Neonatal vitamin A injection promotes cattle muscle growth and increases oxidative muscle fibers显示文摘Background: Vitamin A and its metabolite,retinoic acid(RA),are important regulators of cell differentiation and organ morphogenesis.Its impact on beef cattle muscle growth remains undefined.Method: Angus steer calves were administrated with 0(control) or 150,000 IU vitamin A(retinyl palmitate in glycerol,i.m.) per calf at birth and 1 month of age.At 2 months of age,a biopsy of the Biceps femoris muscle was obtained to analyze the immediate effects of vitamin A injection on myogenic capacity of muscle cells.The resulting steers were harvested at 14 months of age.Results: Vitamin A administration increased cattle growth at 2 months.At 2 months of age,Vitamin A increased PAX7 positive satellite cells and the expression of myogenic marker genes including PAX7,MYF5,MYOD and MYOG.Muscle derived mononuclear cells were further isolated and induced myogenesis in vitro.More myotubes and a higher degree of myogenesis was observed in vitamin A groups.Consistently,vitamin A increased Latissimus dorsi(LD) muscle fiber size at harvest.In addition,vitamin A increased the ratio of oxidative type I and type IIA fibers and reduced the glycolic type IIX fibers.Furthermore,we found that RA,a key bioactive metabolite of vitamin A,activated PPARGC1 A promoter,which explains the upregulated expression of PPARGC1 A in skeletal muscle.Conclusion: Vitamin A administration to neonatal calves enhanced postnatal muscle growth by promoting myogenesis and increasing satellite cell density,accompanied with a shift to oxidative muscle fibers.Bo Wang Wei Nie Xing Fu Jeanene M de Avila Yannan Ma Mei-Jun Zhu Martin Maquivar Steven M Parish Jan R Busboom Mark L Nelson Min Du 2019Journal of Animal Science and Biotechnology2019,10,1:3
20Isolation and Expression Analysis of Two Genes Encoding Cinnamate 4-Hydroxylase from Cotton(Gossypium hirsutum)显示文摘Two genes(GhC4H1 and GhC4H2) that encode putative cotton cinnamate 4-hydroxylases that catalyze the second step in the phenylpropanoid pathway were isolated from developing cotton fibers. GhC4H1 and GhC4H2 each contain open reading frames of 1 518 base pairs(bp) in length and both encode proteins consisting of 505 amino acid residues. They are 90.89% identical to each other at the amino acid sequence level and belong to class I of plant C4 Hs. GhC4H1 and GhC4H2 genomic DNA are 2 247 and 2 161 bp long, respectively, and contain two introns located at conserved positions relative to the coding sequence. GhC4H1 and GhC4H2 promoters were isolated and found to contain many cis-elements(boxes P, L and AC-I element) previously identified in the promoters of other phenylpropanoid pathway genes. Histochemical staining showed GUS expression driven by the GhC4H1 and GhC4H2 promoters in ovules and fibers tissues. GhC4H1 and GhC4H2 were also widely expressed in other cotton tissues. GhC4H2 expression reached its highest level during the elongation stage of fiber development, whereas GhC4H1 expression increased during the secondary wall development period in cotton fibers.
