1PacBio Sequencing and Its Applications显示文摘Single-molecule, real-time sequencing developed by Pacific Bio Sciences offers longer read lengths than the second-generation sequencing(SGS) technologies, making it well-suited for unsolved problems in genome, transcriptome, and epigenetics research. The highly-contiguous de novo assemblies using Pac Bio sequencing can close gaps in current reference assemblies and characterize structural variation(SV) in personal genomes. With longer reads, we can sequence through extended repetitive regions and detect mutations, many of which are associated with diseases. Moreover, Pac Bio transcriptome sequencing is advantageous for the identification of gene isoforms and facilitates reliable discoveries of novel genes and novel isoforms of annotated genes,due to its ability to sequence full-length transcripts or fragments with significant lengths. Additionally, Pac Bio's sequencing technique provides information that is useful for the direct detection of base modifications, such as methylation. In addition to using Pac Bio sequencing alone, many hybrid sequencing strategies have been developed to make use of more accurate short reads in conjunction with Pac Bio long reads. In general, hybrid sequencing strategies are more affordable and scalable especially for small-size laboratories than using Pac Bio Sequencing alone. 4Laparoscopic natural orifice specimen extraction-colectomy: A systematic review显示文摘Over the last 20 years, laparoscopic colorectal surgery has shown equal efficacy for benign and malignant colorectal diseases when compared to open surgery. However, a laparoscopic approach reduces postoperative morbidity and shortens hospital stay. In the quest to optimize outcomes after laparoscopic colorectal surgery, reduction of access trauma could be a way to improve recovery. To date, one method to reduce access trauma is natural orifice specimen extraction(NOSE). NOSE aims to reduce access trauma in laparoscopic colorectal surgery. The specimen is delivered via a natural orifice and the anastomosis is created intracorporeally. Different methods are used to extract the specimen and to create a bowel anastomosis. Currently, specimens are delivered transcolonically, transrectally, transanally, or transvaginally. Each of these NOSEprocedures raises specific issues with regard to operative technique and application. The presumed benefits of NOSE-procedures are less pain, lower analgesia requirements, faster recovery, shorter hospital stay, better cosmetic results, and lower incisional hernia rates. Avoidance of extraction site laparotomy is the most important characteristic of NOSE. Concerns associated with the NOSE-technique include bacterial contamination of the peritoneal cavity, inflammatory response, and postoperative outcomes, including postoperative pain and the functional and oncologic outcomes. These issues need to be studied in prospective randomized controlled trials. 8Management of hepatocellular carcinoma with portal vein tumor thrombosis: Review and update at 2016显示文摘Portal vein tumor thrombosis(PVTT) is a common phenomenon in hepatocellular carcinoma(HCC). Compared to HCC without PVTT, HCC with PVTT is characterized by an aggressive disease course, worse hepatic function, a higher chance of complications related to portal hypertension and poorer tolerance to treatment. Conventionally, HCC with PVTT is grouped together with metastatic HCC during the planning of its management, and most patients are offered palliative treatment with sorafenib or other systemic agents. As a result, most data on the management of HCC with PVTT comes from subgroup analyses or retrospective series. In the past few years, there have been several updates on management of HCC with PVTT. First, it is evident that HCC with PVTT consists of heterogeneous subgroups with different prognoses. Different classifications have been proposed to stage the degree of portal vein invasion/thrombosis, suggesting that different treatment modalities may be individualized to patients with different risks. Second, more studies indicate that more aggressive treatment, including surgical resection or locoregional treatment, may benefit select HCC patients with PVTT. 12Standards of Ecological Compensation for Traditional Ecoagriculture:Taking Rice-Fish System in Hani Terrace as an Example显示文摘Traditional eco-agriculture provides many ecosystem services which are important for the local environment, especially in mountain areas. In order to encourage the farmers to engage in eco-agriculture,it is necessary to establish an eco-compensation mechanism for them. As the compensation standard is one of key issues in establishing eco-compensation mechanism, this paper calculated the standard based on the farmers' willingness to accept(WTA) and the input-output analysis of eco-compensation. We took the traditional rice-fish eco-agriculture as the research object, conducting field surveys in Hani Terrace of Honghe County. Through the questionnaires in 2011, we obtained the farmers' willingness to accept government compensation and market compensation. Then, the research evaluated the output of eco-compensation, the economic value of rice-fish paddy ecosystem services. Finally, under different market compensation standards, we compared the input and output of government compensation. The results show that, in 2011 the government should to pay farmers 7462 yuan·ha–1·a-1to meet their willingness, but the output(ecological benefit) was only 7393 yuan·ha–1·a-1. 13Multivariate adaptive regression splines and neural network models for prediction of pile drivability显示文摘Piles are long, slender structural elements used to transfer the loads from the superstructure through weak strata onto stiffer soils or rocks. For driven piles, the impact of the piling hammer induces compression and tension stresses in the piles. Hence, an important design consideration is to check that the strength of the pile is sufficient to resist the stresses caused by the impact of the pile hammer. Due to its complexity, pile drivability lacks a precise analytical solution with regard to the phenomena involved.In situations where measured data or numerical hypothetical results are available, neural networks stand out in mapping the nonlinear interactions and relationships between the system's predictors and dependent responses. In addition, unlike most computational tools, no mathematical relationship assumption between the dependent and independent variables has to be made. Nevertheless, neural networks have been criticized for their long trial-and-error training process since the optimal configuration is not known a priori. This paper investigates the use of a fairly simple nonparametric regression algorithm known as multivariate adaptive regression splines(MARS), as an alternative to neural networks, to approximate the relationship between the inputs and dependent response, and to mathematically interpret the relationship between the various parameters. 14古村落景区游客拥挤感知维度与游憩情绪影响机制研究--以西递、宏村为例显示文摘拥挤感知被认为是影响游客的游憩体验和满意度的重要指标,通过构建拥挤感知的多维度变化来探讨游客情绪的变化对满意度的影响研究仍不成熟。本文以西递、宏村为研究案例地,运用结构方程模型分析方法,构建空间拥挤、人潮拥挤和体验拥挤维度对游客游憩过程的影响模型,探讨了游客拥挤三个维度的影响因素。研究表明:(1)人潮拥挤对情感依附型游客积极情绪呈正向影响;(2)空间拥挤增加消极情绪,与游客满意度呈负相关关系;(3)体验拥挤对游客积极情绪呈显著负向影响;(4)人潮拥挤—积极情绪—满意度(0.359)之间的相互关系强于人潮拥挤—消极情绪—满意度(0.128);空间拥挤—积极情绪—游客满意度(-0.241)的关联度强于空间拥挤—消极情绪—游客满意度(-0.142);体验拥挤—积极情绪—游客满意度(-0.167)之间的影响关系弱于体验拥挤—消极情绪—游客满意度(-0.213);(5)人口特征对拥挤感知的影响差异显著。张圆刚 余向洋 Wong IpKin Anthony 程静静 尹寿兵 2018人文地理2018,33,2:34
