1Peripheral nerve regeneration and intraneural revascularization显示文摘Peripheral nerves are particularly vulnerable to injuries and are involved in numerous pathologies for which specific treatments are lacking. This review summarizes the pathophysiological features of the most common traumatic nerve injury in humans and the different animal models used in nerve regeneration studies. The current knowledge concerning Wallerian degeneration and nerve regrowth is then described. Finally, the involvement of intraneural vascularization in these processes is addressed. As intraneural vascularization has been poorly studied, histological experiments were carried out from rat sciatic nerves damaged by a glycerol injection. 2土源性线虫感染对儿童生长发育影响及相关因素分析显示文摘目的了解中国西南贫困地区儿童土源性线虫感染现状,分析其对儿童生长发育的影响。方法在四川省和贵州省随机选取3个国家级贫困县,采用改良加藤厚涂片法对受检儿童粪便进行病原学检查并进行问卷调查。采用普通最小二乘法(OLS)分析土源性线虫感染情况、个体特征、饮食卫生习惯和家庭特征等相关因素对儿童身体质量指数(BMI)、身高和体重的影响。结果共粪检1 707名儿童,土源性线虫感染率为22.2%。OLS分析显示感染土源性线虫、性别、年龄、母亲受教育程度、饮用生水、饲养畜禽等6个因素差异均有统计学意义,并与儿童身高和体重存在负相关关系。结论中国西南农村贫困地区儿童感染土源性线虫与生长发育存在负相关关系。土源性线虫感染与母亲的文化程度密切相关。应加大力度控制儿童土源性线虫的感染。王晓兵 王国飞 张林秀 罗仁福 王聚君 Alexis Medina Karen Eggleston Scott Rozelle Scott Smith 2013中国血吸虫病防治杂志2013,25,3:18
3On-the-fly active learning of interpretable Bayesian force fields for atomistic rare events显示文摘Machine learned force fields typically require manual construction of training sets consisting of thousands of first principles calculations,which can result in low training efficiency and unpredictable errors when applied to structures not represented in the training set of the model.This severely limits the practical application of these models in systems with dynamics governed by important rare events,such as chemical reactions and diffusion.We present an adaptive Bayesian inference method for automating the training of interpretable,low-dimensional,and multi-element interatomic force fields using structures drawn on the fly from molecular dynamics simulations.Within an active learning framework,the internal uncertainty of a Gaussian process regression model is used to decide whether to accept the model prediction or to perform a first principles calculation to augment the training set of the model.The method is applied to a range of single-and multi-element systems and shown to achieve a favorable balance of accuracy and computational efficiency,while requiring a minimal amount of ab initio training data.We provide a fully opensource implementation of our method,as well as a procedure to map trained models to computationally efficient tabulated force fields.Jonathan Vandermause Steven B.Torrisi Simon Batzner Yu Xie Lixin Sun Alexie M.Kolpak Boris Kozinsky 2020npj Computational Materials2020,,1:17
4TREM-1 multimerization is essential for its activation on monocytes and neutrophils显示文摘The triggering receptor expressed on myeloid cells-1(TREM-1)is a receptor expressed on innate immune cells.By promoting the amplification of inflammatory signals that are initially triggered by Toll-like receptors(TLRs),TREM-1 has been characterized as a major player in the pathophysiology of acute and chronic inflammatory diseases,such as septic shock,myocardial infarction,atherosclerosis,and inflammatory bowel diseases.However,the molecular events leading to the activation of TREM-1 in innate immune cells remain unknown.Here,we show that TREM-1 is activated by multimerization and that the levels of intracellular Ca 2+release,reactive oxygen species,and cytokine production correlate with the degree of TREM-1 aggregation.TREM-1 activation on primary human monocytes by LPS required a two-step process consisting of upregulation followed by clustering of TREM-1 at the cell surface,in contrast to primary human neutrophils,where LPS induced a rapid cell membrane reorganization of TREM-1,which confirmed that TREM-1 is regulated differently in primary human neutrophils and monocytes.In addition,we show that the ectodomain of TREM-1 is able to homooligomerize in a concentration-dependent manner,which suggests that the clustering of TREM-1 on the membrane promotes its oligomerization.We further show that the adapter protein DAP12 stabilizes TREM-1 surface expression and multimerization.TREM-1 multimerization at the cell surface is also mediated by its endogenous ligand,a conclusion supported by the ability of the TREM-1 inhibitor LR12 to limit TREM-1 multimerization.These results provide evidence for ligand-induced,receptor-mediated dimerization of TREM-1.Collectively,our findings uncover the mechanisms necessary for TREM-1 activation in monocytes and neutrophils.Kevin Carrasco Amir Boufenzer Lucie Jolly Helene Le Cordier Guanbo Wang Albert JR Heck Adelheid Cerwenka Emilie Vinolo Alexis Nazabal Alexandre Kriznik Pierre Launay Sebastien Gibot Marc Derive 2019Cellular & Molecular Immunology2019,16,5:13
5Application of MODIS satellite products to the air pollution research in Beijing显示文摘The direct correlation between NASA MODIS aerosol optical depth (AOD) products and the air pollution index (API) in Beijing was found relatively low based on the long-term comparison analysis. The correlation improved to some extent after taking account of the seasonal variation of scale height and the vertical distribution of aerosols. The correlation coefficient further improved significantly after considering the influencing factor of Relative Humidity (RH). This study concluded that satellite remote-sensing could serve as an efficient tool for monitoring the spatial distribution of particulate pollutants on the ground-level, as long as corrections have been made in the two aforementioned processes. Taking advantage of the MODIS information, we analyzed a pollution episode occurring in October 2004 in Beijing. It indicated that satellite remote-sensing could describe the formation process of the ground-level pollution episode in detail, and showed that regional transport and the topography were crucial factors to air quality in Beijing. The annual averaged distribution in the urban area of Beijing and its surroundings could be also obtained from the high-resolution retrieval results, implicating that high-resolution satellite remote-sensing might be potential in monitoring the source distribution of particulate pollutants.Alexis K.H.Lau 2005Science China Earth Sciences2005,48,z2:12

8中国农村医生医疗服务质量的测量——基于标准化病人法的实验研究显示文摘过去十年中,中国政府在农村地区进行了一系列的医疗改革,这些改革极大地提高了医疗服务的可及性,但却很少关注医疗服务的质量。本文采用标准化病人法,测量和评估中国农村基层医疗服务的质量。我们招募样本地区的农民作为'标准化病人',培训他们稳定地展示小儿痢疾和心绞痛这两种常见疾病的症状。基于标准化病人在村卫生室的就诊案例,我们发现作为基层医疗服务第一线的村医可能无法给患者提供高质量的医疗服务。村医在诊断过程中仅完成了18%的建议问诊条目,诊断正确率仅为26%;在所有开具处方的病例中,有64%的药物是不必要的,甚至是有害的。进一步分析表明,医疗服务质量与医生的受教育水平和是否获得医疗资格证书密切相关。此外,我们发现农民患者并不能很好地识别出哪些医生是相对好的医生,这样的医患信息不对称的情况可能使村医更加缺乏提高医疗服务质量的激励。研究结果建议政府在投资硬件改善基层医疗服务设施的同时,应更加注重提高农村医生的诊断和治疗水平。史耀疆 薛浩 王欢 Sean Sylvia Alexis Medina Scott Rozelle 2016劳动经济研究2016,4,2:10
9Osteocyte dysfunction promotes osteoarthritis through MMP13-dependent suppression of subchondral bone homeostasis显示文摘Osteoarthritis(OA),long considered a primary disorder of articular cartilage,is commonly associated with subchondral bone sclerosis.However,the cellular mechanisms responsible for changes to subchondral bone in OA,and the extent to which these changes are drivers of or a secondary reaction to cartilage degeneration,remain unclear.In knee joints from human patients with end-stage OA,we found evidence of profound defects in osteocyte function.Suppression of osteocyte perilacunar/canalicular remodeling(PLR)was most severe in the medial compartment of OA subchondral bone,with lower protease expression,diminished canalicular networks,and disorganized and hypermineralized extracellular matrix.As a step toward evaluating the causality of PLR suppression in OA,we ablated the PLR enzyme MMP13 in osteocytes while leaving chondrocytic MMP13 intact,using Cre recombinase driven by the 9.6-kb DMP1 promoter.Not only did osteocytic MMP13 deficiency suppress PLR in cortical and subchondral bone,but it also compromised cartilage.Even in the absence of injury,osteocytic MMP13 deficiency was sufficient to reduce cartilage proteoglycan content,change chondrocyte production of collagen II,aggrecan,and MMP13,and increase the incidence of cartilage lesions,consistent with early OA.Thus,in humans and mice,defects in PLR coincide with cartilage defects.Osteocyte-derived MMP13 emerges as a critical regulator of cartilage homeostasis,likely via its effects on PLR.Together,these findings implicate osteocytes in bone-cartilage crosstalk in the joint and suggest a causal role for suppressed perilacunar/canalicular remodeling in osteoarthritis.Courtney MMazur Jonathon JWoo Cristal SYee Aaron JFields Claire Acevedo Karsyn NBailey Serra Kaya Tristan WFowler Jeffrey CLotz Alexis Dang Alfred CKuo Thomas PVail Tamara Alliston 2019Bone Research2019,7,4:10

11中国西南地区儿童土源性线虫感染及影响因素调查显示文摘目的了解中国西南贫困地区儿童土源性线虫感染情况和主要影响因素,为制定土源性线虫病防治措施提供依据。方法采用改良加藤厚涂片法对受检儿童粪便进行病原学检查,其中部分儿童进行透明胶纸肛拭法检查蛲虫卵。采用问卷调查和Probit估计多元统计方法,分析儿童服药驱虫、个体特征、饮食卫生习惯和家庭特征等16个相关因素对土源性线虫感染的影响。结果共粪检1 707名儿童,土源性线虫感染率为22.2%,其中蛔虫、钩虫、鞭虫感染率分别为16.9%、3.8%和6.6%,且以蛔虫感染率和感染度为最高;蛲虫检查495名,感染率为5.1%。Probit估计多元统计显示,兄弟姐妹数量、母亲受教育程度、饮用生水、饲养畜禽等4个因素差异均有统计学意义(P均<0.05)。其中,母亲受教育程度能降低土源性线虫感染率,边际效应为-0.074;兄弟姐妹数量、饮用生水和饲养畜禽均能提高土源性线虫感染率,边际效应分别为0.028、-0.112和0.080。结论我国西南贫困地区儿童土源性线虫感染率仍然较高,且以蛔虫感染为主;改变儿童饮用生水的不良卫生习惯、家庭饲养畜禽习惯和针对其母亲开展寄生虫病防治知识的健康教育,对儿童土源性线虫病的预防和控制具有重要意义。王晓兵 王国飞 张林秀 罗仁福 田洪春 唐丽娜 王聚君 Alexis Medina Paul Wise Scott Rozelle 2012中国血吸虫病防治杂志2012,24,3:10
12Axonal regeneration after spinal cord injury in zebrafish and mammals: differences, similarities, translation显示文摘Spinal cord injury (SCI) in mammals results in functional deficits that are mostly permanent due in part to the inability of severed axons to regenerate. Several types of growth-inhibitory molecules expressed at the injury site contribute to this regeneration failure. The responses of axons to these inhibitors vary greatly within and between organisms, reflecting axons' characteristic intrinsic propensity for regeneration. In the zebrafish (Danio rerio) many but not all axons exhibit successful regeneration after SCi. This review presents and compares the intrinsic and extrinsic determinants of axonal regeneration in the injured spinal cord in mammals and zebrafish. 13Does urodynamics predict voiding after benign prostatic hyperplasia surgery in patients with detrusor underactivity?显示文摘Objective:We sought to determine if urodynamic study(UDS)predicted voiding outcomes in men with detrusor underactivity(DU)and benign prostatic enlargement(BPE)who underwent photovaporization of the prostate(PVP).Methods:Between September 2010 and July 2015,106 male patients with BPE and DU were identified.All patients underwent PVP.Urinary retention was noted by the preoperative necessity for an indwelling or intermittent catheter.Data collection included comorbidities,quality of life(QoL)scores,prostate volume,prostate-specific antigen(PSA),UDS and perioperative outcomes.UDS parameters included volume at first desire to void,volume at first urge to void,volume of severe urge,volume at capacity,compliance,detrusor contractions,maximum urinary flow rate(Qmax),and postvoid residual(PVR).Results:A total of 106 men were included in this analysis,who had urinary retention with a Foley catheter or clean intermittent catheterization(CIC)at the time of surgery.At baseline we found patients who voided had a detrusor pressure at Qmax(Pdet@Qmax)of 10.05±6.45 cmH2O compared to 16.78±12.17 cmH2O in those who did not void(p=0.071).Postoperatively,96(90.6%,mean age 76.9±26.2 years)of patients voided successfully while 10(9.4%,mean age 80.52±9.61 years)of patients remained in urinary retention.Mean baseline Qmax was 4.895±5.452 mL/s and 2.900±3.356 mL/s(p=0.087)in those who voided and did not respectively.PVR was 319.23±330.62 mL in those who voided and 276.88263.27 mL(p=0.344)in those who did not void.No UDS parameter predicted who would void postoperatively or improvements in QoL.Conclusions:The patients with DU and BPE might be able to successfully void after undergoing PVP regardless of UDS findings.All men who voided had improved international prostate symptom score and QoL scores compared to baseline and these parameters were durable up to 12 months.Dominique Thomas Kevin C.Zorn Nadir Zaidi Stephanie Ashley Chen Yiye Zhang Alexis Te Bilal Chughtai 2019Asian Journal of Urology2019,6,3:8

15西南两省贫困地区儿童土源性线虫感染状况及影响因素分析显示文摘目的:了解四川和贵州省贫困地区儿童土源性线虫感染情况及其主要影响因素,为制定土源性线虫病防治策略提供依据。方法采用分层随机抽样,选择贵州和四川两省6个贫困县的95个行政村及46所小学为调查点,每个样本村各选择11名3~5岁学龄前儿童及8~10岁就读于样本学校的儿童作为调查对象,收集其粪便,采用改良加藤厚涂片法进行检查。同时对各村村医和村干部进行问卷调查,了解该村2009年儿童驱虫情况及社会经济和卫生状况;采用Tobit估计多元统计方法分析儿童土源性线虫感染的影响因素。结果两省共粪检1707名儿童,贵州省样本村儿童土源性线虫感染率为46.1%,蛔虫、钩虫、鞭虫感染率分别为31.1%、4.1%、10.8%;四川省样本村儿童土源性线虫感染率为9.8%,上述3种寄生虫感染率分别为3.6%、3.5%、2.7%。两省共粪检890名学龄儿童,贵州省样本学校儿童土源性线虫和蛔虫、钩虫、鞭虫感染率分别为53.8%、32.7%、6.6%、14.4%,四川省分别为7.3%、2.2%、2.9%、2.2%。Tobit估计分析显示,儿童驱虫治疗和村水田比例是儿童土源性线虫感染的影响因素(P均<0.05)。结论我国西南贫困地区儿童土源性线虫感染率仍然较高。为有效降低贫困地区儿童的土源性线虫感染率,除构建长效驱虫工作机制和采取改水、改厕等措施外,还应加强健康教育,并将驱虫防治纳入新型农村合作医疗体系。王晓兵 张林秀 王国飞 罗仁福 Alexis Medina Scott Rozelle 2014中国血吸虫病防治杂志2014,26,3:7

18The role of photovaporization of the prostate in small volume benign prostatic hyperplasia and review of the literature显示文摘Objective:Our objective was to characterize the safety and efficacy of the 180 W XPS-GreenLight laser in men with lower urinary tract symptoms secondary to a small volume benign prostatic hyperplasia(BPH).Methods:A retrospective analysis was performed for all patients who underwent 180 W XPSlaser photoselective vaporization of the prostate(PVP)vaporization of the prostate between 2012 and 2016 at two-tertiary medical centers.Data collection included baseline comorbidities,disease-specific quality of life scores,maximum urinary flow rate(Qmax),postvoid residual(PVR),complications,prostate volume and prostate-specific antigen(PSA).The secondary endpoints were the incidence of intraoperative and postoperative adverse events.Complications were stratified using the Clavien-Dindo grading system up to 90 days after surgery.Results:Mean age of men was 67.8 years old,with a mean body mass index of 29.7 kg/m2.Mean prostate volume as measured by transrectal ultrasound was 29 mL.Anticoagulation use was 47%and urinary retention with catheter at time of surgery was 17%.Mean hospital stay and catheter time were 0.5 days.Median follow-up time was 6 months with the longest duration of follow-up being 22.5 months(interquartile range,3-22.5 months).The International Prostate Symptom Score improved from 22.8±7.0 at baseline to 10.7±7.4(p<0.01)and 6.3±4.4(p<0.01)at 1 and 6 months,respectively.The Qmax improved from 7.70±4.46 mL/s at baseline to 17.25±9.30 mL/s(p<0.01)and 19.14±7.19 mL/s(p<0.001)at 1 and 6 months,respectively,while the PVR improved from 216.0±271.0 mL preoperatively to 32.8±45.3 mL(p<0.01)and 26.2±46.0 mL(p<0.01)at 1 and 6 months,respectively.The PSA dropped from 1.97±1.76 ng/mL preoperatively to 0.71±0.61 ng/mL(p<0.01)and 0.74±0.63 ng/mL at 1 and 6 months,respectively.No patient had a bladder neck contracture postoperatively and no capsular perforations were noted intraoperatively.Conclusion:The 180 W GreenLight XPS system is safe and effective for men with small volume BPH.PVP produced improvements in symptomatic and clinical parameters without any safety concern.It represents a safe surgical option in this under studied population.Dominique Thomas Kevin C.Zorn Malek Meskawi Ramy Goueli Pierre-Alain Hueber Lesa Deonarine Vincent Misrai Alexis Te Bilal Chughtai 2019Asian Journal of Urology2019,6,4:6

20Ketamine anesthesia reduces intestinal ischemia/reperfusion injury in rats显示文摘AIM: To investigate the effects of ketamine anesthesia on the motility alterations and tissue injury caused by ischemia/reperfusion in rats. METHODS: Thirty male Wistar rats weighing 200-250 g were used. Ischemia was induced by ob-structing blood flow in 25% of the total small intesti-nal length (ileum) with a vascular clamp for 45 min, after which either 60 min or 24 h of reperfusion was allowed. Rats were either anesthetized with pento-barbital sodium (50 mg/kg) or ketamine (100 mg/kg). Control groups received sham surgery. After 60 min of reperfusion, the intestine was examined for mor-phological alterations, and after 24 h intestinal basic electrical rhythm (BER) frequency was calculated, and intestinal transit determined in all groups. RESULTS: The intestinal mucosa in rats that were anesthetized with ketamine showed moderate altera-tions such as epithelial lifting, while ulceration and hemorrhage was observed in rats that received pento-barbital sodium after 60 min of reperfusion. Quantita-tive analysis of structural damage using the Chiu scaleshowed significantly less injury in rats that received ketamine than in rats that did not (2.35 ± 1.14 vs 4.58 ± 0.50, P < 0.0001). The distance traveled by a mark-er, expressed as percentage of total intestinal length, in rats that received pentobarbital sodium was 20% ± 2% in comparison with 25.9% ± 1.64% in rats that re-ceived ketamine (P = 0.017). BER was not statistically different between groups.
