3Evaluation of SNPs in miR-196-a2, miR-27a and miR-146a as risk factors of colorectal cancer显示文摘AIM: To investigate whether selected single nucleotide polymorphisms (SNPs) in miR-196a2, miR-27a and miR146a genes are associated with sporadic colorectal cancer (CRC).METHODS: In order to investigate the effect of these SNPs in CRC, we performed a case-control study of 197 cases of sporadic CRC and 212 cancer-free controls originating from the Central-European Caucasian population using TaqMan Real-Time polymerase chain reaction and allelic discrimination analysis. RESULTS: The genotype and allele frequencies of SNPs were compared between the cases and the controls. None of the performed analysis showed any statistically significant results. 5Enforced PGC-1α expression promotes CD8 T cell fitness, memory formation and antitumor immunity显示文摘Memory CD8 T cells can provide long-term protection against tumors,which depends on their enhanced proliferative capacity,self-renewal and unique metabolic rewiring to sustain cellular fitness.Specifically,memory CD8 T cells engage oxidative phosphorylation and fatty acid oxidation to fulfill their metabolic demands.In contrast,tumor-infiltrating lymphocytes(TILs)display severe metabolic defects,which may underlie their functional decline.Here,we show that overexpression of proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),the master regulator of mitochondrial biogenesis(MB),favors CD8 T cell central memory formation rather than resident memory generation.PGC-1α-overexpressing CD8 T cells persist and mediate more robust recall responses to bacterial infection or peptide vaccination.Importantly,CD8 T cells with enhanced PGC-1αexpression provide stronger antitumor immunity in a mouse melanoma model.Moreover,TILs overexpressing PGC-1αmaintain higher mitochondrial activity and improved expansion when rechallenged in a tumor-free host.Altogether,our findings indicate that enforcing mitochondrial biogenesis promotes CD8 T cell memory formation,metabolic fitness,and antitumor immunity in vivo.Nina Dumauthioz Benjamin Tschumi Mathias Wenes Bastien Marti Haiping Wang Fabien Franco Wenhui Li Isabel CLopez-Mejia Lluis Fajas Ping-Chih Ho Alena Donda Pedro Romero Lianjun Zhang 2021Cellular & Molecular Immunology2021,18,7:3
6Mortality of therapeutic fish Garra rufa caused by Aeromonas sobria显示文摘Objective:To investigate a case of mass mortality of Garra rufa(G.rufa)from a fish hatchery farm in Slovakia.Methods:Causative bacterial agent was swabbing out of affected fish skin area and subsequently identified using commercial test system.Antibiotic susceptibility was determined by the disk diffusion method.Results:Infected G.rufa was characterized by abnormal swimming behaviour,bleeding of skin lesions and local haemorrhages.Despite of using recommended aquatic antibiotic treatment no improvement was achieved and Aeromonas sobria{A.sobria)was identified as a causative agent of fish mortality.Due to massive fish mortality,antibiotic susceptibility of pure isolated culture of A.sobria was evaluated employing eight antibiotics against human infections.A.sobria was resistant only against one antibiotic,namely ampicilin.Conclusions:These results indicate that A.sobria can act as a primary pathogen of G.rufa and may be a potential risk factor for immunodeficient or immunoincompetent patients during the ichthyotherapy.Juraj Majtán Jaroslav erny Alena Ofúkaná Peter Taká Milan Kozánek 2012Asian Pacific Journal of Tropical Biomedicine2012,2,2:3
7Epithelial markers of colorectal carcinogenesis in ulcerative colitis and primary sclerosing cholangitis显示文摘AIM:To evaluate the expression of epithelial markers of colorectal carcinogenesis in patients with long-term ulcerative colitis(UC) and primary sclerosing cholangitis(PSC) before and after transplantation.METHODS:Eight patients with UC and PSC prior to liver transplantation(PSC-UC),22 patients with UC after liver transplantation for PSC(OLT),9 patients with active ulcerative colitis without PSC(UCA),7 patients withUC in remission(UCR) and 10 controls(N) underwent colonoscopy with multiple biopsies.Specimens were analysed histologically and semi-quantitatively immunohistochemically for p53,Bcl-2 and cyclooxygenase-2(COX-2) markers.Statistical analysis was performed by Kruskal-Wallis and Fisher's exact tests.RESULTS:PSC-UC had a statistically significantly higher expression of p53 in the nondysplastic mucosa as compared to OLT,UCA,UCR and N(P < 0.05).We also found a statistically significant positive correlation between the incidence of PSC and the expression of p53(P < 0.001).UCA had a higher p53 expression as compared to UCR.OLT had a significantly lower expression of p53 as compared with PSC-UC(P < 0.001).Bcl-2 had a significant higher bcl-2 expression as compared with controls.No difference in COX-2 expression between PSC-UC,UCR and UCA was found.UCA had higher COX-2 expression as compared to UCR.We also found a statistically significant positive correlation between the expression of COX-2 and p53.Patients after liver transplantation for PSC had a statistically significantly lower expression of the p53 compared with PSCUC(P < 0.001).PSC-UC had the same inflammatory endoscopic activity as OLT and UCR when evaluated with the Mayo score.CONCLUSION:Our study shows that the nondysplatic mucosa of UC patients with PSC is characterised by a higher expression of the tumour suppressor gene p53,suggesting a higher susceptibility of cancer.This p53 overexpression correlates with the presence of PSC whilst it is not present in patients with UC after liver transplantation for PSC.Pavel Wohl Tomas Hucl Pavel Drastich David Kamenar Julius Spicak Eva Honsova Eva Sticova Alena Lodererova Jan Matous Martin Hill Petr Wohl Milos Kucera 2013World Journal of Gastroenterology2013,19,14:2
8TNF-α gene polymorphisms: association with age-related macular degeneration in Russian population显示文摘AIM: To study polymorphisms in promotor regions of tumor necrosis factor(TNF)-α TNF-863 A/C(rs1800630), TNF-308 A/G(rs1800629), and TNF-238 A/G(rs361525) in patients with age-related macular degeneration(AMD) and associations of complex TNF-α genotypes with AMD. METHODS: One hundred and two patients(82 women, 20 men; mean age 64.2±1.2 y) with AMD and 100 healthy age-and sex-matched controls(82 women, 18 men; 60±1.4 y) were included in the study. All subjects were Caucasian, all subjects and their parents were inhabitants of Russia. Genomic DNA was obtained from EDTA-preserved blood using the standard phenol-chloroform method. Polymorphisms were detected by polymerase chain reaction followed by the restriction fragment length polymorphism method. The following TNF-α genotypes were studied: TNF-α-238 AA, GA, GG, TNF-α-308 AA, GA, GG, TNF-α-863 AA, CA, CC. RESULTS: Differences in TNF-α-863 and TNF-α-238 genotypes frequencies in patients with AMD and healthy controls were not found. The distribution of TNF-α-308 AA and TNF-α-308 GA genotypes was significantly different between the studied group and the controls [odds ratios(OR) =0.22, P=0.0287 and OR=2.91, P=0.0063, respectively]. TNF-863 CC/TNF-308 GA and TNF-308 GA/TNF-238 GG genotypes were associated with the increased risk of AMD(OR=2.48, P=0.0332 and OR=2.51, P=0.0187, respectively). Five genotypes combinations appeared to be protective. CONCLUSION: In the present study, single nucleotide polymorphisms and complex polymorphisms of one of the key inflammatory cytokines TNF-α, and a number of significant associations of these polymorphisms with AMD in Russian population have been shown. 9Organelle Extensions in Plant Cells显示文摘Cell walls lock each cell in a specific position within the supraorganization of a plant.Despite its fixed location,each cell must be able to sense alterations in its immediate environment and respond rapidly to ensure the optimal functioning,continued growth and development,and eventual long-term survival of the plant.The ultra-structural detail that underlies our present understanding of the plant cell has largely been acquired from fixed and processed material that does not allow an appreciation of the dynamic nature of sub-cellular events in the cell.In recent years,fluorescent proteinaided imaging of living plant cells has added to our understanding of the dynamic nature of the plant cell.One of the major outcomesof live imaging of plant cells is the growing appreciation that organelle shapes are not fixed,and many organelles extend their surface transiently in rapid response to environmental stimuli.In many cases,the extensions appear as tubules extending from the main organelle.Specific terms such as stromules from plastids,matrixules from mitochondria,and peroxules from peroxisomes have been coined to describe the extensions.Here,we review our present understanding of organelle extensions and discuss how they may play potential roles in maintaining cellular homeostasis in plant cells.Jaideep Mathur Alena Mammone Kiah A.Barton 2012Journal of Integrative Plant Biology2012,54,11:2
10Relation between basin morphometric features and dynamic characteristics of debris flows——a case study in Siberia, Russia显示文摘Extreme and prolonged rainfall in the Tunka Ridge caused several debris flows in the vicinity of the Arshan village(Siberia, Russia) on June 28, 2014. These debris flows, in spite of similar geological conditions, had different velocity, peak discharge and alluvial fan volume values. The flow velocity was from 3.5 m/s to 19.6 m/s, the peak discharge ranged from 63 m3/s to 13566 m3/s, and the alluvial fan volume varied from 4.13×103 to 8.45×105 m3. Such a great range of values is due to the morphometric parameters of the debris flow basins. The article deals with the influence of morphometric parameters of debris flow basins, such as the basin area, the average slope, Melton ratio, relief ratio on the debris flow velocity, peak discharge and volume of alluvial fans. In this debris flow event the average values of slope angle and total basin relief of the debris flow basins did not affect the values of debris flow velocity, peak discharge and alluvial fan volume. The highest correlations were observed with the debris flow basin area that was connected with the water inflow volume into the debris flow basins during the rainfall. 11Cytokine interplay among the diseased retina, inflammatory cells and mesenchymal stem cells-a clue to stem cell-based therapy显示文摘Retinal degenerative disorders,such as diabetic retinopathy,retinitis pigmentosa,age-related macular degeneration or glaucoma,represent the most common causes of loss of vision and blindness.In spite of intensive research,treatment options to prevent,stop or cure these diseases are limited.Newer therapeutic approaches are offered by stem cell-based therapy.To date,various types of stem cells have been evaluated in a range of models.Among them,mesenchymal stem/stromal cells(MSCs)derived from bone marrow or adipose tissue and used as autologous cells have been proposed to have the potential to attenuate the negative manifestations of retinal diseases.MSCs delivered to the vicinity of the diseased retina can exert local anti-inflammatory and repairpromoting/regenerative effects on retinal cells.However,MSCs also produce numerous factors that could have negative impacts on retinal regeneration.The secretory activity of MSCs is strongly influenced by the cytokine environment.Therefore,the interactions among the molecules produced by the diseased retina,cytokines secreted by inflammatory cells and factors produced by MSCs will decide the development and propagation of retinal diseases.Here we discuss the interactions among cytokines and other factors in the environment of the diseased retina treated by MSCs,and we present results supporting immunoregulatory and trophic roles of molecules secreted in the vicinity of the retina during MSC-based therapy.Vladimir Holan Barbora Hermankova Magdalena Krulova Alena Zajicova 2019World Journal of Stem Cells2019,11,11:2
