1体外模拟消化对藜麦抗氧化活性、α-葡萄糖苷酶和α-淀粉酶抑制活性影响研究显示文摘以藜麦为研究对象,研究体外模拟消化对藜麦总酚和总黄酮含量、生物利用率、抗氧化活性以及对α-葡萄糖苷酶和α-淀粉酶抑制作用的影响。结果表明,模拟消化后,总酚和总黄酮含量分别提高了278%~370%和320%~455%,生物利用度分别提高到17.50%~20.19%和32%~32.14%,而1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除能力、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(2,2'-Azinobis-(3-ethylbenzthiazoline-6-sulphonate),ABTS)自由基清除能力和铁离子还原力(ferric reducing ability of plasma,FRAP)三种方法测得的抗氧化活性分别提高了185.2%~224.3%、137.4%~186.4%和154%~171.2%。同时,与未消化组相比,消化组表现出较高的α-葡萄糖苷酶和α-淀粉酶的抑制活性,半抑制浓度(50%inhibiting concentration,IC50)分别为16.24~19.83 mg/mL和8.17~10.67 mg/mL。王静 刘丁丽 罗丹 成婧 陆俊 Otobong Donald Akan 罗非君 2021中国粮油学报2021,36,4:15
2Biomarkers in autoimmune pancreatitis and immunoglobulin G4-related disease显示文摘Solitary organ autoimmune disorders,formerly known as autoimmune pancreatitis(AIP),autoimmune sialadenitis,and autoimmune sclerosing cholangitis,are now considered organ-specific manifestations of systemic immunoglobulin G4-related disease(IgG4-RD).AIP and IgG4-RD are characterized by elevated serum concentration of IgG4 antibody(Ab),accumulation of IgG4-expressing plasmacytes in the affected organs,and involvement of multiple organs.It is well established that enhanced IgG4 Ab responses are a hallmark of AIP and IgG4-RD for diagnosis and monitoring disease activity.However,a significant fraction of patients with AIP and IgG4-RD who develop chronic fibroinflammatory responses have normal serum concentrations of this IgG subtype.In addition,disease flare-up is sometimes seen even in the presence of normalized serum concentrations of IgG4 Ab after successful induction of remission by prednisolone.Therefore,it is necessary to identify new biomarkers based on the understanding of the pathophysiology of AIP and IgG4-RD.Recently,we found that activation of plasmacytoid dendritic cells producing both interferon-α(IFN-α)and interleukin-33(IL-33)mediate murine AIP and human IgG4-RD.More importantly,we provided evidence that serum concentrations of IFN-αand IL-33 could be useful biomarkers for the diagnosis and monitoring of AIP and IgG4-RD activity after induction of remission in these autoimmune disorders.In this Frontier article,we have summarized and discussed biomarkers of AIP and IgG4-RD,including Igs,autoAbs,and cytokines to provide useful information not only for clinicians but also for researchers.Akane Hara Tomohiro Watanabe Kosuke Minaga Tomoe Yoshikawa Ken Kamata Masatoshi Kudo 2021World Journal of Gastroenterology2021,27,19:8
3牡丹籽油抑制结肠癌细胞HCT116的体外增殖和调控细胞周期作用显示文摘以牡丹籽油为研究对象,结肠癌细胞HCT116为模型,评估了牡丹籽油对HCT116结肠癌细胞体外增殖和周期的影响,并对其分子机理进行了初步的研究。利用MTS实验、蛋白蛋印迹等检测了结肠癌细胞在增殖、迁移和分子水平上的变化。结果表明:牡丹籽油能显著抑制HCT116细胞的增殖和迁移能力;当牡丹籽油的浓度达到100μg/m L时,细胞周期相关基因p15、p21、p53的m RNA相对表达量相比对照组(Con)分别上升了5.49、4.56和3.73倍,具有极显著性差异(p<0.01)。提示牡丹籽油能调控细胞周期G1/S期关键靶基因的表达,使细胞周期阻滞在G1/S期。牡丹籽油也能抑制MAPK信号通路的激活,且能显著抑制其下游转录因子AP-1和NF-κB的活性。结果提示牡丹籽油可能通过抑制MAPK信号通路/AP-1和NF-κB活性下调,调控细胞周期相关基因的表达,从而抑制结直肠癌细胞的增殖,课题研究将为功能性抗癌类食用植物油的开发提供科学的理论基础。邹佳文 罗依 Akan OD 周潇恬 徐魏 白婕 刘津良 胡作民 周亚萍 罗非君 2020现代食品科技2020,36,5:5
4Lead Removal from Aqueous Solutions Using Novel Gel Adsorbent Synthesized from Natural Condensed Tannin显示文摘Lead has caused serious environmental pollution due to its toxicity, accumulation in food chains and persistence in nature. In this paper, removal of lead from aqueous solutions is investigated using a novel gel adsorbent synthesized from natural condensed tannin. The novel adsorbent performs in aqueous solutions as a weak base with valid basic groups of 1.2mmol·g^-1 tannin gel particles and therefore results in the elevation of pH value of aqueous solutions. Even when initial pH is 3.6, final pH at equilibrium can climb up to 6.5 that is above the pH value for Pb(OH)2 precipitation formation and then lead can be removed from wastewater by this so-called surface precipitation. The adsorption isotherm can be expressed by the Langmuir equation and the maximum capacity for adsorption of Pb is up to 92mg·g^-l(based on dry adsorbent) when initial pH value is 3.6. Hence, the adsorbent does offer favorable properties in lead removal with respect to its high adsorption capacity at low initial pH value,which is advantageous to lead removal from acidic wastewater. 5Preparation,characterization and catalytic behavior of Pt-Cu nanoparticles in methane combustion显示文摘Fine and well dispersed Pt-Cu bimetallic nanoparticles stabilized by polyvinyl pyrrolidone (PVP) were synthesized by alkaline polyol method. The molar ratio of Pt to Cu was 1 : 1. Further, the Pt-Cu bimetallic nanoparticles were supported on alumina and their catalytic behavior in methane combustion was investigated. The as-prepared as well as the supported Pt-Cu nanoparticles were characterized by transmission elec-tron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), fractal analysis and X-ray diffraction (XRD). 6Identification of arylamine N-acetyltransferase inhibitors as an approach towards novel anti-tuberculars显示文摘New anti-tubercular drugs and drug targets are urgently needed to reduce the time for treatment and also to identify agents that will be effective against Mycobacterium tuberculosis persisting intracellularly.Mycobacteria have a unique cell wall.Deletion of the gene for arylamine N-acetyltransferase(NAT)decreases mycobacterial cell wall lipids,particularly the distinctive mycolates,and also increases antibiotic susceptibility and killing within macrophage of Mycobacterium bovis BCG.The nat gene and its associated gene cluster are almost identical in sequence in M.bovis BCG and M.tuberculosis.The gene cluster is essential for intracellular survival of mycobacteria.We have therefore used pure NAT protein for high-throughput screening to identify several classes of small molecules that inhibit NAT activity.Here,we characterize one class of such molecules—triazoles—in relation to its effects on the target enzyme and on both M.bovis BCG and M.tuberculosis.The most potent triazole mimics the effects of deletion of the nat gene on growth,lipid disruption and intracellular survival.We also present the structure-activity relationship between NAT inhibition and effects on mycobacterial growth,and use ligand-protein analysis to give further insight into the structure-activity relationships.We conclude that screening a chemical library with NAT protein yields compounds that have high potential as anti-tubercular agents and that the inhibitors will allow further exploration of the biochemical pathway in which NAT is involved.Isaac M.Westwood Sanjib Bhakta Angela J.Russell Elizabeth Fullam Matthew C.Anderton Akane Kawamura Andrew W.Mulvaney Richard J.Vickers Veemal Bhowruth Gurdyal S.Besra Ajit Lalvani Stephen G.Davies Edith Sim 2010Protein & Cell2010,1,1:3
7Osteopontin is a biomarker for early autoimmune uveoretinitis显示文摘Osteopontin(OPN)is an extracellular matrix protein with a diverse range of functions,including roles in cell adhesion,migration,and immunomodulation,which are associated with the modulation of neuroinflammation in the central nervous system.The present study was performed to evaluate the involvement of OPN in the eyes of an experimental autoimmune uveoretinitis(EAU)model.The EAU model was developed by immunization of Lewis rats with interphotoreceptor retinoid-binding protein.The results showed the OPN level was remarkably upregulated in the eye of EAU rats on day 9 post-immunization.The level of CD44,a ligand of OPN,was increased in the ciliary body of EAU rats.Furthermore,OPN was also detected in the ciliary body and activated microglia/macrophages in the EAU retina.The results suggest that OPN was significantly upregulated in the eyes of EAU rats,and that it may be useful as an early biomarker of ocular autoimmune diseases.All animal experiments were approved by the Institutional Animal Care and Use Committee of Jeju National University(approval No.2020-0012)on March 11,2020.Jeongtae Kim Meejung Ahn Yuna Choi Jiyoon Chun Kyungsook Jung Akane Tanaka Hiroshi Matsuda Taekyun Shin 2022Neural Regeneration Research2022,17,7:3
