| 1 | Phytochemical analysis and antioxidants activities of aqueous stem bark extract of Schotia latifolia Jacq显示文摘Objective:To evaluate the phytochemical constituents and antioxidant activities of aqueous extract of Scholia latifolia(S.latifolia)bark locally used for the treatment of oxidative stress-induced ailments in South Africa.Methods:The antioxidant and free radical scavenging activity of aqueous extract of the plant was assessed against 1,1-diphenyl-2-pierylhydrazyl(DPPH),nitric oxide(NO),2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)and the ferric reducing agent.Total phenolics,flavonoids,flavonols and proanthocyanidins were also determined to assess their corresponding effect on the antioxidant activity of this plant.Results:The activities of plant extract against DPPH,ABTS and NO radicals were concentration dependent with IC_(50)value of 0.06,0.05 and 0.05 mg/mL,respectively.The reducing power of the extract was greater than that of butylated hydroxyl toluene(BHT)and ascorbic acid which were used as standard drugs in a concentration dependent manner.The total phenolics content of the aqueous bark extract was(193.33±0.03 TE/g),followed by flavonoids(72.70±0.01 QE/g),proanthocyanidins|48.76±0.00 CE/g)and flavonols(47.76±0.21 QE/g).Phytochemical analysis revealed the presence of percentage tannin(11.40±0.02),alkaloid(9.80±0.01),steroids(18.20±0.01),glycosides(29.80±0.01)and saponins(6.80±0.00).The results exhibited a positive linear correlation between these polyphenols and the free radical scavenging activities.Conclusions:Our findings provide evidence that the crude aqueous extract of S.latifolia is a potential source of natural antioxidants and this justifies its uses in folkloric medicines. | Mbaebie BO Edeoga HO Afolayan AJ | 2012 | Asian Pacific Journal of Tropical Biomedicine2012,2,2: | 15 |
| 2 | Antibacterial and antioxidant activities of hydroalcoholic stem bark extract of Schotia latifolia Jacq显示文摘Objective:To investigate the antibacterial and antioxidant activities of hydroalcoholic extract of Scholia latifolia(S.latifolia) bark commonly used in South Africa traditional medicine for the treatment of various ailments.Methods:The antibacterial test and MIC was determined by using agar well diffusion and dilution methods respectively against eight strains of bacteria. The total phenol,proanthocyanidin and flavonoid contents of 5.latifolia were assessed using Standard methods.The antioxidant activity of the extract was evaluated using ferric reducing power and the free radical scavenging activity against 1.1-diphenyl-2-picrylhydrazyl(DPPH), 2.2’-azino-bis-3-ethylbenzothiazoline-6-sulfonic-acid(ABTS),nitric oxide(NO),hydrogen peroxide(H2O2) and lipid oxidation(LO).Results:The antibacterial activity demonstrated an appreciable effect against all the gram positive bacteria at MIC between 0.016 and 10 mg/ml. while that of gram negative bacteria was above 10 mg/mL.The plant extract exhibited high concentration of proanthocyanidin[(300.00±0.10) mg CE/g].followed by flavonoid[(12.46±0.04 mg) TE/g]and phenol[(1 l.06±0.03) mg QE/g]contents.Similarly,the extract at 0.5 mg/mL scavenges DPPH.ABTS.H2O2.LO and NO by 87.55%.89.47%.77.15%.86.48%and 77.75%of the radicals respectively.The reducing power was also found to be concentration dependent Conclusions: Our data suggest that S.latifolia extract has antibacterial and antioxidants activity and thus could be used as alternative therapv against antibiotic resistance bacteria and to prevent man) radical related diseases. | Sunday Oyewole Oyedemi Anthony Jide Afolayan | 2011 | Asian Pacific Journal of Tropical Medicine2011,4,12: | 9 |

| 6 | Abortifacient Potential of Aqueous Extract of Senna alata Leaves in Rats显示文摘Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar rats.Methods Pregnant rats weighing 143.65 ± 13.11 g were completely randomized into 5 groups(A-E).Rats in group A(negative control) were orally administered,once daily with 0.5 ml of distilled water on days 10-18 post-coitum while those in groups B,C,D,and E were treated exactly like the negative control except they received the same volume containing 2.85 mg/kg body weight of mifepristone(group B,reference drug),250,500 and 1 000 mg/kg body weight of the extract respectively(groups C-E).Results Phytochemical screening of the extract showed positive results for saponins(1.22%),flavonoids(1.06%),cardiac glycosides(0.20%),cardenolides and dienolides(0.18%),phenolics(0.44%) and alkaloids(0.52%).Respiratory distress,salivation,diarrhoea,changes in the appearance of hair as well as maternal mortality were not observed at any time during the exposure period except in the mifepristone-treated animals where there was episode of diarrhoea and tiredness.The extract significantly reduced(P<0.05) the number of life foetus,weight and survival ratio of the foetus,numbers of implantations and Corpora lutea,implantation index,progesterone,prolactin,estradiol,follicle stimulating and luteinizing hormones whereas the number of dead foetus,number and percentage of rats that aborted,percentage vaginal opening,resorption index,pre-and post-implantation losses increased significantly.In addition,mifepristone-treated animals produced resorption index that compared well with the distilled water control.There was also no dead or life foetus and serum progesterone concentration was increased in the animals treated with mifepristone.All cases of abortion were accompanied with vaginal bleeding.Although,the final weight of the rats increased significantly,the feed and water intake were not significantly altered in all the treatment groups.The weight of the uterus,uterine-body weight ratio,length of the right uterus horn and uterine cholesterol decreased significantly in all the treatment groups.The uterine alkaline phosphatase activity and glucose concentration increased in only the extract-treated animals whereas mifepristone decreased the uterine alkaline phosphatase activity and glucose content of the animals.Conclusion This study has provided evidence to the age-long claim of S.alata leaves in 'washing the uterus'.The abortifacient properties were most pronounced at 500 and 1 000 mg/kg body weight of the extract and were similar to the animals treated with 2.85 mg/kg body weight of mifepristone.Hormonal influence,changes in implantation site,estrogenicity and uterogenicity are suggested as possible mechanism of abortifacient activity of aqueous extract of S.alata leaves.Overall,the extract may be used as an abortifacient especially at 500 and 1 000 mg/kg body weight and therefore not safe for consumption as oral remedy during pregnancy. | Yakubu MT Adeshina AO Oladiji AT Akanji MA Oloyede OB Jimoh GA Olatinwo AWO Afolayan AJ | 2010 | Journal of Reproduction and Contraception2010,21,3: | 2 |

| 16 | Phytochemical and antioxidant activities of Rumex crispus L. in treatment of gastrointestinal helminths in Eastern Cape Province, South Africa显示文摘Objective: To evaluate the antioxidant activities and phytochemical content of the leaf and root extracts of Rumex crispus using the solvents extraction; methanol extract,ethanol extract, acetone extract(ACE), and water extract.Methods: Total flavonoids content, total phenolic content, and total proanthocyanidin were evaluated using spectrophotometric equivalents of the standards, quercetin, gallic acid and catechin respectively. The antioxidant activities of the plant extracts were determined using ABTS, DPPH, ferric reducing antioxidant power, total antioxidant capacity and nitric oxide scavenging assays.Results: The flavonoids and phenols contents of the extracts were in the range of(19.39 ± 4.08) to(526.23 ± 17.52) mg QE/g and(16.95 ± 12.03) to(240.68 ± 3.50) mg GAE/g, respectively. ACE of the leaf has the highest value of total flavonoids content(526.23 ± 17.52) mg QE/g while ACE of the root has the highest value of total phenolic content(240.68 ± 3.50) mg GAE/g. The highest content of total proanthocyanidin(645.38 ± 1.33) mg CE/g was in ACE of the root. Significant amounts of saponin and alkaloid were also present in the root and leaf extracts. All solvent fractions showed significant antioxidant activities(P < 0.05) with ACE of the root having the highest scavenging value as shown in DPPH, ABTS, total antioxidant capacity, nitric oxide and ferric reducing antioxidant power(IC50= 0.014 mg/m L, <0.005 mg/m L, 0.048 mg/m L,0.067 mg/m L, and 0.075 mg/m L, respectively).Conclusions: In this study, the mean phytochemical content of the root of Rumex crispus is higher than that of the leaf and this may have contributed to its high antioxidant activities. | 19 | Antioxidant, antibacterial and phytochemical properties of two medicinal plants against the wound infecting bacteria显示文摘Objective: To investigate the antioxidant, antibacterial and phytochemical properties of ethanol extracts of Brachylaena elliptica and Brachylaena ilicifolia against wound infecting bacteria normally found in diabetic patients.Methods: The in vitro antioxidant activity of the two plants extracts were investigated spectrophotometrically using 1,1-diphenyl-2-picrylhydrazyl, nitric oxide, azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt, hydrogen peroxide(H_2O_2) and ferric reducing power. The antibacterial assay and minimum inhibitory concentration(MIC) was determined using the agar dilution method against five bacteria strains using amoxycillin and ciprofloxacin as positive control. The phytochemical analyses(tannins,total phenol, flavonoids, flavonols, proanthocyanidin, alkaloids and saponins) were assessed using standard methods.Results: The ethanol extract of both plants exhibited strong antioxidant activities in some cases when compared to the standards(vitamin C and BHT). The antibacterial activity of both plants showed an appreciable broad spectrum activity against these wound pathogens with MIC value ranges between 0.3 mg/m L and 5 mg/m L. Tannins,phenols, flavanols, proanthocyanidins and alkaloids content of B. ilicifolia were significantly higher than those in B. elliptica.
