2All-optical forward-viewing photoacoustic probe for high-resolution 3D endoscopy显示文摘A miniature forward-viewing endoscopic probe that provides high-resolution 3D photoacoustic images is demonstrated.The probe is of outer diameter 3.2mm and comprised of a transparent Fabry-Pérot(FP)polymer-film ultrasound sensor that is located at the distal end of a rigid optical fiber bundle.Excitation laser pulses are coupled simultaneously into all cores of the bundle and are transmitted through the FP sensor to provide wide-field tissue illumination at the distal end.The resulting photoacoustic waves are mapped in 2D by sequentially scanning the input end of the bundle with an interrogation laser beam in order to individually address different points on the FP sensor.In this way,the sensor acts as a high-density ultrasound array that is comprised of 50,000 individual elements,each of which is 12μm in diameter,within the 3.2mm diameter footprint of the probe.The fine spatial sampling that this affords,along with the wide bandwidth(f_(-3dB)=34 MHz)of the sensor,enables a high-resolution photoacoustic image to be reconstructed.The measured on-axis lateral resolution of the probe was depth-dependent and ranged from 45-170μm for depths between 1 and 7 mm,and the vertical resolution was 31μm over the same depth range.The system was evaluated by acquiring 3D images of absorbing phantoms and the microvascular anatomies of a duck embryo and mouse skin.Excellent image fidelity was demonstrated.It is anticipated that this type of probe could find application as a tool for guiding laparoscopic procedures,fetal surgery and other minimally invasive interventions that require a millimeter-scale forward-viewing 3D photoacoustic imaging probe.Rehman Ansari Edward Z.Zhang Adrien E.Desjardins Paul C.Beard 2018Light(Science & Applications)2018,7,1:11

4The interferon inducing pathways and the hepatitis C virus显示文摘The innate immune response is triggered by a variety of pathogens, including viruses, and requires rapid induction of typeⅠ?interferons (IFN), such as IFNβ and IFNα. IFN induction occurs when specific pathogen motifs bind to specific cellular receptors. In non-professional immune, virally-infected cells, IFN induction is essentially initiated after the binding of dsRNA structures to TLR3 receptors or to intracytosolic RNA helicases, such as RIG-Ⅰ/MDA5. This leads to the recruitment of specific adaptors, such as TRIF for TLR3 and the mitochondrial-associated IPS-1/VISA/MAVS/CARDIF adapter protein for the RNA helicases, and the ultimate recruitment of kinases, such as MAPKs, the canonical IKK complex and the TBK1/IKKε kinases, which activate the transcription factors ATF-2/ c-jun, NF-κB and IRF3, respectively. The coordinated action of these transcription factors leads to induction of IFN and of pro-inflammatory cytokines and to the establishment of the innate immune response. HCV can cleave both the adapters TRIF and IPS-1/VISA/MAVS/ CARDIF through the action of its NS3/4A protease. 5Through-needle all-optical ultrasound imaging in vivo: a preclinical swine study显示文摘High-frequency ultrasound imaging can provide exquisite visualizations of tissue to guide minimally invasive procedures.Here,we demonstrate that an all-optical ultrasound transducer,through which light guided by optical fibers is used to generate and receive ultrasound,is suitable for real-time invasive medical imaging in vivo.Broad-bandwidth ultrasound generation was achieved through the photoacoustic excitation of a multiwalled carbon nanotube-polydimethylsiloxane composite coating on the distal end of a 300-μm multi-mode optical fiber by a pulsed laser.The interrogation of a high-finesse Fabry–Pérot cavity on a single-mode optical fiber by a wavelength-tunable continuous-wave laser was applied for ultrasound reception.This transducer was integrated within a custom inner transseptal needle(diameter 1.08 mm;length 78 cm)that included a metallic septum to acoustically isolate the two optical fibers.The use of this needle within the beating heart of a pig provided unprecedented realtime views(50 Hz scan rate)of cardiac tissue(depth:2.5 cm;axial resolution:64μm)and revealed the critical anatomical structures required to safely perform a transseptal crossing:the right and left atrial walls,the right atrial appendage,and the limbus fossae ovalis.This new paradigm will allow ultrasound imaging to be integrated into a broad range of minimally invasive devices in different clinical contexts.Malcolm C Finlay Charles A Mosse Richard J Colchester Sacha Noimark Edward Z Zhang Sebastien Ourselin Paul C Beard Richard J Schilling Ivan P Parkin Ioannis Papakonstantinou Adrien E Desjardins 2017Light(Science & Applications)2017,6,1:6
6Modeling strain rate sensitivity and high temperature deformation of Mg-3Al-lZn alloy显示文摘Increasing the plastic deformation temperature of Mg alloys results in higher strain rate sensitivity,easier activation of secondary slip modes,and impeded twinning.In this study,the strain rate sensitivity is estimated for each deformation mode,and visco-plastic self-consistent modeling is used to reproduce the plastic deformation behavior of an Mg-3Al-lZn O-temper plate from 150 to 450℃.Twinning and basal slip have relatively low strain rate sensitivity,whereas secondary slip modes are highly strain rate sensitive at high temperature.The texture evolution and plastic anisotropy are modeled at different temperatures and strain rates.Results show that when the strain rate sensitivity is taken into account,compared with rate independent critical resolved shear stresses,the material parameters and predictions are different.In particular,this study shows that,for hot deformation,there is a critical strain rate above which secondary slip modes predominate,and beyond which tension twinning is activated.A similar transition is expected for modes that have different strain rate sensitivity.Adrien Chapuis Qing Liu 2019Journal of Magnesium and Alloys2019,7,3:6

8Simulation of texture evolution and deformation mechanism in Mg-3Al-1Zn alloy during uniaxial compression显示文摘The effects of starting texture on he flow curves, and microstructure and texture evolutions of AZ31(Mg-3Al-1Zn-0.3Mn in wt.%) alloy during uniaxial compression at room temperature have been investigated by experiments and simulations. Two kinds of cylindrical compression samples were cut from the AZ31 extruded rod: one was cut with the compression direction parallel to the extruded direction(ED), and the other was cut with the compression axis perpendicular to the ED. The samples were termed as C//ED sample and C⊥ED sample, respectively. The results indicate that the general characteristics of flow curves and texture evolutions in both C⊥ED and C//ED samples during compression can be well explained by the relative activities of deformation mechanisms, which show a strong orientation dependence on the initial textures and the current texture. A significant yield behavior can be induced by {10-12} twinning at the starting deformation, and the lower yield stress in C⊥ED sample can be enhanced by the high activity of basal slip. The work hardening behavior is related to the activities of basal slip, pyramidal slip, {10-12} twinning, as well as {10-11} twinning. 10Current view and perspectives in amyotrophic lateral sclerosis显示文摘Amyotrophic lateral sclerosis(ALS), identified as a distinct clinical entity by Charcot since the end of the nineteenth century, is a devastating and fatal neurodegenerative disorder that affects motor neurons in the brain, brainstem and spinal cord. Survival of patients with ALS is associated with several factors such as clinical phenotype, age at onset, gender, early presence of respiratory failure, weight loss and treatment with Riluzole(the only disease-modifying drug approved for this disease). Nowadays, there is still no curative treatment for ALS: palliative care and symptomatic treatment are therefore essential components in the management of these patients. Nevertheless, the scientific knowledge in the field of ALS motor neuron degeneration is growing, with the prospect of new treatments. Based on this physiopathological knowledge, several new therapeutic targets are being studied, involving various mechanisms such as excitotoxicity, neuroinflammation, mitochondrial dysfunction, oxidative stress, RNA metabolism and other attractive concepts. Moreover, it is also important to identify reliable biomarkers that will be essential components for future therapeutic development and study design in ALS. 11不同病因致黄斑囊样水肿患者视力下降与水肿形态的相关性分析(英文)显示文摘目的:探讨囊样黄斑水肿(CME)患者光学相干断层扫描(OCT)形态,病因学和视力之间的关系。方法:本研究共搜集了本院门诊CME患者52例57眼,患者均应用OCT确诊为黄斑囊样水肿,且在同1d进行眼底及最佳矫正视力检查。最佳矫正视力(BCVA)检查使用E视力表和对数视力表(logMAR),视力范围2.3 logM AR^0logM AR(均数1.11±0.57)。通过患者病史和眼底照相进行病因诊断,根据黄斑囊样水肿最大垂直直径和黄斑厚度比率将OCT分为四级(一级<30%,30%≤二级<60%,60%≤三级<90%,四级≥90%),相关性是通过计算皮尔逊相关系数'r'值(P<0.05时有统计学意义)。结果:OCT IV级患者视力最差(平均视力1.96±0.23logMAR),此时OCT分级与视力之间有显著正相关,统计学有显著差异(r=0.729,P<0.001)。视网膜中央静脉阻塞(CRVO)是引起视力严重下降最常见的疾病,(r=0.375,P=0.004)。另外根据OCT的形态,IS/OS层(inner segment/outer segment)和ELM(external limiting membrane)均破环的患者视力显著下降(平均视力1.11±0.57 logM AR),二者间具有统计学差异(r=-0.346,P=0.008)。玻璃体视网膜牵拉和外视网膜层破坏均与OCTⅣ级和(r=0.390,P=0.003)CRVO呈正相关(r=0.362,P=0.006),差异有统计学意义。结论:当CME的OCT形态表现Ⅳ级时CRVO患者视力最差。Adrien Bironkwaninguvu 姜伟 王玉清 2016国际眼科杂志2016,16,8:3

14Bimanual motor skill learning and robotic assistance for chronic hemiparetic stroke: a randomized controlled trial显示文摘Using robotic devices might improve recovery post-stroke, but the optimal way to apply robotic assistance has yet to be determined. The current study aimed to investigate whether training under the robotic active-assisted mode improves bimanual motor skill learning(biMSkL) more than training under the active mode in stroke patients. Twenty-six healthy individuals(HI) and 23 chronic hemiparetic stroke patients with a detectable lesion on MRI or CT scan, who demonstrated motor deficits in the upper limb, were randomly allocated to two parallel groups. The protocol included a two-day training on a new bimanual cooperative task, LIFT-THE-TRAY, under either the active or activeassisted modes(where assistance decreased in a pre-determined stepwise fashion) with the bimanual version of the REAplan? robotic device. The hypothesis was that the active-assisted mode would result in greater biMSkL than the active mode. The biMSkL was quantified by a speed-accuracy trade-off(SAT) before(T1) and immediately after(T2) training on days 1 and 2(T3 and T4). The change in SAT after 2 days of training(T4/T1) indicated that both HI and stroke patients learned and retained the bimanual cooperative task. After 2 days of training, the active-assisted mode did not improve biMSkL more than the active mode(T4/T1) in HI nor stroke patients. Whereas HI generalized the learned bimanual skill to different execution speeds in both the active and active-assisted subgroups, the stroke patients generalized the learned skill only in the active subgroup. Taken together, the active-assisted mode, applied in a pre-determined stepwise decreasing fashion, did not improve biMSkL more than the active mode in HI and stroke subjects. Stroke subjects might benefit more from robotic assistance when applied 'as-needed.' 15Precision micro-mechanical components in single crystal diamond by deep reactive ion etching显示文摘The outstanding material properties of single crystal diamond have been at the origin of the long-standing interest in its exploitation for engineering of high-performance micro-and nanosystems.In particular,the extreme mechanical hardness,the highest elastic modulus of any bulk material,low density,and the promise for low friction have spurred interest most notably for micro-mechanical and MEMS applications.While reactive ion etching of diamond has been reported previously,precision structuring of freestanding micro-mechanical components in single crystal diamond by deep reactive ion etching has hitherto remained elusive,related to limitations in the etch processes,such as the need of thick hard masks,micromasking effects,and limited etch rates.In this work,we report on an optimized reactive ion etching process of single crystal diamond overcoming several of these shortcomings at the same time,and present a robust and reliable method to produce fully released micro-mechanical components in single crystal diamond.Using an optimized Al/SiO_(2) hard mask and a high-intensity oxygen plasma etch process,we obtain etch rates exceeding 30μm/h and hard mask selectivity better than 1:50.We demonstrate fully freestanding micro-mechanical components for mechanical watches made of pure single crystal diamond.The components with a thickness of 150μm are defined by lithography and deep reactive ion etching,and exhibit sidewall angles of 82°–93°with surface roughness better than 200 nm rms,demonstrating the potential of this powerful technique for precision microstructuring of single crystal diamond.Adrien Toros Marcell Kiss Teodoro Graziosi Hamed Sattari Pascal Gallo Niels Quack 2018Microsystems & Nanoengineering2018,4,1:2

18Kupffer cell restoration after partial hepatectomy is mainly driven by local cell proliferation in IL-6-dependent autocrine and paracrine manners显示文摘Kupffer cells(KCs),which are liver-resident macrophages,originate from the fetal yolk sac and represent one of the largest macrophage populations in the body.However,the current data on the origin of the cells that restore macrophages during liver injury and regeneration remain controversial.Here,we address the question of whether liver macrophage restoration results from circulating monocyte infiltration or local KC proliferation in regenerating livers after partial hepatectomy(PHx)and uncover the underlying mechanisms.By using several strains of genetically modified mice and performing immunohistochemical analyses,we demonstrated that local KC proliferation mainly contributed to the restoration of liver macrophages after PHx.Peak KC proliferation was impaired in Il6-knockout(KO)mice and restored after the administration of IL-6 protein,whereas KC proliferation was not affected in Il4-KO or Csf2-KO mice.The source of IL-6 was identified using hepatocyte-and myeloid-specific Il6-KO mice and the results revealed that both hepatocytes and myeloid cells contribute to IL-6 production after PHx.Moreover,peak KC proliferation was also impaired in myeloid-specific Il6 receptor-KO mice after PHx,suggesting that IL-6 signaling directly promotes KC proliferation.Studies using several inhibitors to block the IL-6 signaling pathway revealed that sirtuin 1(SIRT1)contributed to IL-6-mediated KC proliferation in vitro.Genetic deletion of the Sirt1 gene in myeloid cells,including KCs,impaired KC proliferation after PHx.In conclusion,our data suggest that KC repopulation after PHx is mainly driven by local KC proliferation,which is dependent on IL-6 and SIRT1 activation in KCs.Yeni Ait Ahmed Yaojie Fu Robim M.Rodrigues Yong He Yukun Guan Adrien Guillot Ruixue Ren Dechun Feng Juan Hidalgo Cynthia Ju Fouad Lafdil Bin Gao 2021Cellular & Molecular Immunology2021,18,9:2
19Inverse analysis of coupled carbon-nitrogen cycles against multiple datasets at ambient and elevated CO_(2)显示文摘Aims Carbon(C)sequestration in terrestrial ecosystems is strongly regulated by nitrogen(N)processes.However,key parameters that determine the degree of N regulation on terrestrial C sequestration have not been well quantified.Methods Here,we used a Bayesian probabilistic inversion approach to estimate 14 target parameters related to ecosystem C and N interactions from 19 datasets obtained from Duke Forests under ambient and elevated carbon dioxide(CO_(2)).Important FindingsOur results indicated that 8 of the 14 target parameters,such as C:N ratios in most ecosystem compartments,plant N uptake and external N input,were well constrained by available datasets whereas the others,such as N allocation coefficients,N loss and the initial value of mineral N pool were poorly constrained.Our analysis showed that elevated CO_(2)led to the increases in C:N ratios in foliage,fine roots and litter.Moreover,elevated CO_(2)stimulated plant N uptake and increased ecosystem N capital in Duke Forests by 25.2 and 8.5%,respectively.In addition,elevated CO_(2)resulted in the decrease of C exit rates(i.e.increases in C residence times)in foliage,woody biomass,structural litter and passive soil organic matter,but the increase of C exit rate in fine roots.Our results demonstrated that CO_(2)enrichment substantially altered key parameters in determining terrestrial C and N interactions,which have profound implications for model improvement and predictions of future C sequestration in terrestrial ecosystems in response to global change.Zheng Shi Yuanhe Yang Xuhui Zhou Ensheng Weng Adrien C.Finzi Yiqi Luo 2016Journal of Plant Ecology2016,9,3:2
