| 1 | Developmental dysplasia of the hip in the newborn: A systematic review显示文摘Developmental dysplasia of the hip(DDH) denotes a wide spectrum of conditions ranging from subtle acetabular dysplasia to irreducible hip dislocations. Clinical diagnostic tests complement ultrasound imaging in allowing diagnosis, classification and monitoring of this condition. Classification systems relate to the alpha and beta angles in addition to the dynamic coverage index(DCI). Screening programmes for DDH show considerable geographic variation; certain risk factors have been identified which necessitate ultrasound assessment of the newborn. The treatment of DDH has undergone significant evolution, but the current gold standard is still the Pavlik harness. Duration of Pavlik harness treatment has been reported to range from 3 to 9.3 mo. The beta angle, DCI and the superior/lateral femoral head displacement can be assessed via ultrasound to estimate the likelihood of success. Success rates of between 7% and 99% have been reported when using the harness to treat DDH. Avascular necrosis remains the most devastating complication of harness usage with a reported rate of between 0% and 28%. Alternative non-surgical treatment methods used for DDH include devices proposed by LeD amany, Frejka, Lorenz and Ortolani. The Rosen splint and Wagner stocking have also been used for DDH treatment. Surgical treatment for DDH comprises open reduction alongside a combination of femoral or pelvic osteotomies. Femoral osteotomies are carried out in cases of excessive anteversion or valgus deformity of the femoral neck. The two principal pelvic osteotomies most commonly performed are the Salter osteotomy and Pemberton acetabuloplasty. | 3 | N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. | 4 | Impact of a single bout of high-intensity interval exercise and short-term interval training on interleukin-6, FNDC5, and METRNL mRNA expression in human skeletal muscle显示文摘Background: Exercise promotes numerous phenotypic adaptations in skeletal muscle that contribute to improved function and metabolic capacity. An emerging body of evidence suggests that skeletal muscle also releases a myriad of factors during exercise, termed 'myokines'. The purpose of this study was to examine the effects of high-intensity interval training(HIIT) on the acute regulation of the mRNA expression of several myokines, including the prototypical myokine interleukin-6(IL-6), and recently identified myokines fibronectin type III domain-containing protein 5(FNDC5)(irisin) and meteorin-like protein(METRNL).Methods: Both before and after a 20-day period of twice-daily high-volume HIIT, 9 healthy males(20.5 ± 1.5 years performed a standardized bout of high-intensity interval exercise(HIIE; 5 × 4 min at ~80% pretraining peak power output) with skeletal muscle biopsy samples(vastus lateralis) obtained at rest, immediately following exercise, and at 3 h recovery.Results: Before training, a single bout of HIIE increased IL-6(p < 0.05) and METRNL(p < 0.05) mRNA expression measured at 3 h recovery when compared to rest. Following 20 days of HIIT, IL-6 and FNDC5 mRNA were increased at 3 h recovery from the standardized HIIE bout when compared to rest(both p < 0.05). Resting METRNL and FNDC5 mRNA expression were higher following training(p < 0.05), and there was an overall increase in FNDC5 mRNA post-training(main effect of training, p < 0.05).Conclusion: In human skeletal muscle(1) an acute bout of HIIE can induce upregulation of skeletal muscle IL-6 mRNA both before and after a period of intensified HIIT;(2) Resting and overall FNDC5 mRNA expression is increased by 20 days of HIIT; and(3) METRNL mRNA expression is responsive to both acute HIIE and short-term intense HIIT. | 9 | Acute inflammatory demyelinating polyneuropathy associated with pegylated interferon α 2a therapy for chronic hepatitis C virus infection显示文摘The combination of pegylated interferon (Peg-IFN) and ribavirin is the standard of care for chronic hepatitis C virus (HCV) infection treatment. In general, common side effects related to this combination therapy are mild and are very well tolerated. However, peripheral neuropathy including demyelinating polyneuropathy related to Peg-IFN is extremely rare. We present the first case of an acute inflammatory demyelinating polyneuropathy (AIDP) associated with Peg-IFN-α 2a (Pegasys) after 16 wk of a combination therapy with Pegasys and ribavirin in a 65-year-old woman with chronic HCV infection. She developed tingling, numbness, and weakness of her upper and lower extremities and was hospitalized for acute neurological deficits. Her clinical course, neurological findings, an electromyogram (EMG), nerve conductions studies (NCS), muscle biopsy, and a sural nerve biopsy were all consistent with AIDP likely related to Pegasys use. The patient recovered completely with the use of intravenous immunoglobulin (IVIG) including physical therapy and neurological rehabilitation. | 11 | Abnormal ground reaction forces lead to a general decline in gait speed in knee osteoarthritis patients显示文摘AIM To analyse ground reaction forces at higher speeds using another method to be more sensitive in assessing significant gait abnormalities. METHODS A total of 44 subjects, consisting of 24 knee osteoarthritis(OA) patients and 20 healthy controls were analysed. The knee OA patients were recruited from an orthopaedic clinic that were awaiting knee replacement. All subjects had their gait patterns during stance phase at top walking speed assessed on a validated treadmill instrumented with tandem force plates. Temporal measurements and ground reaction forces(GRFs) along with a novel impulse technique were collected for both limbs and a symmetry ratio was applied to all variables to assess inter-limb asymmetry. All continuous variables for each group were compared using a student t-test and χ2 analysis for categorical variables with significance set at α = 0.05. Receiver operator characteristics curves were utilised to determine best discriminating ability.RESULTS The knee OA patients were older(66 ± 7 years vs 53 ± 9 years, P = 0.01) and heavier(body mass index: 31 ± 6 vs 23 ± 7, P < 0.001) but had a similar gender ratio when compared to the control group. Knee OA patients were predictably slower at top walking speed(1.37 ± 0.23 m/s vs 2.00 ± 0.20 m/s, P < 0.0001) with shorter mean step length(79 ± 12 cm vs 99 ± 8 cm, P < 0.0001) and broader gait width(14 ± 5 cm vs 11 ± 3 cm, P = 0.015) than controls without any known lower-limb joint disease. At a matched mean speed(1.37 ± 0.23 vs 1.34 ± 0.07), ground reaction results revealed that pushoff forces and impulse were significantly(P < 0.0001) worse(18% and 12% respectively) for the knee OA patients when compared to the controls. Receiver operating characteristic curves analysis demonstrated total impulse to be the best discriminator of asymmetry, with an area under the curve of 0.902, with a cut-off of-3% and a specificity of 95% and sensitivity of 88%.CONCLUSION Abnormal GRFs in knee osteoarthritis are clearly evident at higher speeds. | 12 | Development of image-based wheat spike counter through a Faster R-CNN algorithm and application for genetic studies显示文摘Spike number(SN) per unit area is one of the major determinants of grain yield in wheat. Development of high-throughput techniques to count SN from large populations enables rapid and cost-effective selection and facilitates genetic studies. In the present study, we used a deep-learning algorithm, i.e., Faster Region-based Convolutional Neural Networks(Faster R-CNN) on Red-Green-Blue(RGB) images to explore the possibility of image-based detection of SN and its application to identify the loci underlying SN. A doubled haploid population of 101 lines derived from the Yangmai 16/Zhongmai 895 cross was grown at two sites for SN phenotyping and genotyped using the high-density wheat 660 K SNP array.Analysis of manual spike number(MSN) in the field, image-based spike number(ISN), and verification of spike number(VSN) by Faster R-CNN revealed significant variation(P < 0.001) among genotypes, with high heritability ranged from 0.71 to 0.96. The coefficients of determination(R^(2)) between ISN and VSN was 0.83, which was higher than that between ISN and MSN(R^(2)= 0.51), and between VSN and MSN(R^(2)= 0.50). Results showed that VSN data can effectively predict wheat spikes with an average accuracy of 86.7% when validated using MSN data. Three QTL Qsnyz.caas-4 DS, Qsnyz.caas-7 DS, and QSnyz.caas-7 DL were identified based on MSN, ISN and VSN data, while QSnyz.caas-7 DS was detected in all the three data sets. | 13 | Lipid dysregulation in hepatitis C virus, and impact of statin therapy upon clinical outcomes显示文摘The hepatitis C virus(HCV) is one of the most common causes of chronic liver disease and the leading indication for liver transplantation worldwide. Every aspect of the HCV life cycle is closely tied to human lipid metabolism. The virus circulates as a lipid-rich particle, utilizing lipoprotein cell receptors to gain entry into the hepatocyte. It has also been shown to upregulate lipid biosynthesis and impair lipid degradation, resulting in significant intracellular lipid accumulation and circulating hypocholesterolemia. Patients with chronic hepatitis C(CHC) are at increased risk of hepatic steatosis, fibrosis, and cardiovascular disease including accelerated atherosclerosis. HMG Co A Reductase inhibitors, or statins, have been shown to play an important role in the modulation of hepatic steatosis and fibrosis, and recent attention has focused upon their potential therapeutic role in CHC. This article reviews the hepatitis C viral life cycle as it impacts host lipoproteins and lipid metabolism. | 14 | Formation of Poly(ε-caprolactone) Networks via Supramolecular Hydrogen Bonding Interactions显示文摘In this contribution, we reported a novel approach to crosslink poly(ε-caprolactone)(PCL) via supramolecular hydrogen bonding interactions. First, a series of octa-armed poly(ε-caprolactone) stars with polyhedral oligomeric silsesquioxane(POSS) cores were synthesized via the ring-opening polymerizations. Thereafter, the arm ends of organic-inorganic star-shaped PCLs were reacted with 2-(6-isocyanatohexylaminocarbonylamino)-6-methyl-4[1 H]-pyrimidinone to obtain 2-ureido-4[1 H]-pyrimidone(UPy)-terminated PCL stars.Notably, the UPy-terminated PCL stars were physically crosslinked, which was evidenced by means of dynamic mechanical thermal analysis(DMTA) and rheological analysis. | 19 | Review:Botnet detection techniques: review, future trends, and issue显示文摘In recent years, the Internet has enabled access to widespread remote services in the distributed computing environment; however, integrity of data transmission in the distributed computing platform is hindered by a number of security issues. For instance, the botnet phenomenon is a prominent threat to Internet security, including the threat of malicious codes. The botnet phenomenon supports a wide range of criminal activities, including distributed denial of service(DDoS) attacks, click fraud, phishing, malware distribution, spam emails, and building machines for illegitimate exchange of information/materials. Therefore, it is imperative to design and develop a robust mechanism for improving the botnet detection, analysis, and removal process. Currently, botnet detection techniques have been reviewed in different ways; however, such studies are limited in scope and lack discussions on the latest botnet detection techniques. This paper presents a comprehensive review of the latest state-of-the-art techniques for botnet detection and figures out the trends of previous and current research. It provides a thematic taxonomy for the classification of botnet detection techniques and highlights the implications and critical aspects by qualitatively analyzing such techniques. | 20 | Mechanisms and molecular approaches for heat tolerance in rice (Oryza sativa L.) under climate change scenario显示文摘Rice, a staple cereal crop in many parts of the world, has been confronted with multiple environmental stresses including high temperature, negatively impacts the booting as well as anthesis growth stages. The situation is further complicated by the changing climatic conditions, resulting in gradual escalation of temperature as well as changing the rainfall pattern and frequency, thus raising a concern of food security worldwide. The situation can be combat by developing rice varieties with excellent genetics with improved morpho-physiological, biochemical, and molecular mechanisms, together can minimize the adverse effects of heat stress. Here, several strategies(encompassing genetic and genomic, and mechanisms involved) for mitigating the impact of high temperature on rice have been discussed.
