| 1 | Reversibility and heritability of liver fibrosis:Implications for research and therapy显示文摘Liver fibrosis continues to be a major health problem worldwide due to lack of effective therapy.If the etiology cannot be eliminated,liver fibrosis progresses to cirrhosis and eventually to liver failure or malignancy;both are associated with a fatal outcome.Liver transplantation,the only curative therapy,is still mostly unavailable.Liver fibrosis was shown to be a reversible process;however,complete reversibility remains debatable.Recently,the molecular markers of liver fibrosis were shown to be transmitted across generations.Epigenetic mechanisms including DNA methylation,histone posttranslational modifications and noncoding RNA have emerged as major determinants of gene expression during liver fibrogenesis and carcinogenesis.Furthermore,epigenetic mechanisms have been shown to be transmitted through mitosis and meiosis to daughter cells and subsequent generations.However,the exact epigenetic regulation of complete liver fibrosis resolution and inheritance has not been fully elucidated.This communication will highlight the recent advances in the search for delineating the mechanisms governing resolution of liver fibrosis and the potential for multigenerational and transgenerational transmission of fibrosis markers.The fact that epigenetic changes,unlike genetic mutations,are reversible and can be modulated pharmacologically underscores the unique opportunity to develop effective therapy to completely reverse liver fibrosis,to prevent the development of malignancy and to regulate heritability of fibrosis phenotype. | Hussein M Atta | 2015 | World Journal of Gastroenterology2015,21,17: | 23 |
| 2 | Prevention of peritoneal adhesions:A promising role for gene therapy显示文摘Adhesions are the most frequent complication of abdominopelvic surgery,yet the extent of the problem,and its serious consequences,has not been adequately recognized.Adhesions evolved as a life-saving mecha-nism to limit the spread of intraperitoneal inflammatory conditions.Three different pathophysiological mechanisms can independently trigger adhesion formation.Mesothelial cell injury and loss during operations,tissue hypoxia and inflammation each promotes adhesion formation separately,and potentiate the effect of each other.Studies have repeatedly demonstrated that interruption of a single pathway does not completely prevent adhesion formation.This review summarizes the pathogenesis of adhesion formation and the results of single gene therapy interventions.It explores the prom-ising role of combinatorial gene therapy and vector modif ications for the prevention of adhesion formation in order to stimulate new ideas and encourage rapid advancements in this field. | Hussein M Atta | 2011 | World Journal of Gastroenterology2011,17,46: | 10 |
| 3 | Updates in the pathophysiological mechanisms of Parkinson's disease: Emerging role of bone marrow mesenchymal stem cells显示文摘AIM: To explore the approaches exerted by mesenchymal stem cells(MSCs) to improve Parkinson's disease(PD) pathophysiology.METHODS: MSCs were harvested from bone marrowof femoral bones of male rats, grown and propagated in culture. Twenty four ovariectomized animals were classified into 3 groups: Group(1) was control, Groups(2) and(3) were subcutaneously administered with rotenone for 14 d after one month of ovariectomy for induction of PD. Then, Group(2) was left untreated, while Group(3) was treated with single intravenous dose of bone marrow derived MSCs(BM-MSCs). SRY gene was assessed by PCR in brain tissue of the female rats. Serum transforming growth factor beta-1(TGF-β1), monocyte chemoattractant protein-1(MCP-1) and brain derived neurotrophic factor(BDNF) levels were assayed by ELISA. Brain dopamine DA level was assayed fluorometrically, while brain tyrosine hydroxylase(TH) and nestin gene expression were detected by semi-quantitative real time PCR. Brain survivin expression was determined by immunohistochemical procedure. Histopathological investigation of brain tissues was also done.RESULTS: BM-MSCs were able to home at the injured brains and elicited significant decrease in serum TGF-β1(489.7 ± 13.0 vs 691.2 ± 8.0, P < 0.05) and MCP-1(89.6 ± 2.0 vs 112.1 ± 1.9, P < 0.05) levels associated with significant increase in serum BDNF(3663 ± 17.8 vs 2905 ± 72.9, P < 0.05) and brain DA(874 ± 15.0 vs 599 ± 9.8, P < 0.05) levels as well as brain TH(1.18 ± 0.004 vs 0.54 ± 0.009, P < 0.05) and nestin(1.29 ± 0.005 vs 0.67 ± 0.006, P < 0.05) genes expression levels. In addition to, producing insignificant increase in the number of positive cells for survivin(293.2 ± 15.9 vs 271.5 ± 15.9, P > 0.05) expression. Finally, the brain sections showed intact histological structure of the striatum as a result of treatment with BM-MSCs. | 4 | 不同灌溉和施肥水平对东尼罗河三角洲地区甜菜产量、品质及部分水分指标的影响(英文)显示文摘【目的】2008~2009、2009~2010年分别在埃及El-Sharkia省Zankalon研究站进行不同灌溉和氮肥水平对甜菜产量、品质及水分指标的影响试验。【方法】采用裂区设计,分别设3个灌溉处理即I1(60%田间持水量)、I2(80%田间持水量)和I3(100%田间持水量)和3个施肥处理即N1(50kgN/fed)、N2(70kgN/fed)和N3(90kgN/fed)。【结果】灌水处理I1的甜菜块根和糖产量最高、根径最大,其次为处理I2,处理I3的最低。随着灌溉水平的提高,甜菜块根长和蔗糖含量明显下降。当施用90kgN/fed氮肥,甜菜根长、根径、块根和糖产量明显提高,但糖分则有所降低。灌溉和施氮肥处理均对蔗糖纯度没有明显影响。3个灌溉处理I1、I2和I3的水分利用量分别为3579.7、3042.0和2504.0m2/fed,季节耗水量分别为58.12、52.06和47.29cm,而甜菜块根产量、糖产量的水分利用率分别为15.86、13.78、13.35kg块根/m3水和2.73、2.61and2.56kg糖/m3水。随着施氮水平的提高,获得一定甜菜根产量和糖产量,所需的实际耗水量和水分利用率也不断提高。在东尼罗河三角洲地区的甜菜平均季节作物系数为0.87。【结论】可在东尼罗河三角洲地区推荐使用Kc经验值来计算耗水量。 | Abd El-Razek A M Atta Y I Hassan A F | 2011 | 南方农业学报2011,42,8: | 7 |
| 5 | Gene therapy for liver regeneration:Experimental studies and prospects for clinical trials显示文摘The liver is an exceptional organ,not only because of its unique anatomical and physiological characteristics,but also because of its unlimited regenerative capacity.Unfolding of the molecular mechanisms that govern liver regeneration has allowed researchers to exploit them to augment liver regeneration.Dramatic progress in the field,however,was made by the introduction of the powerful tool of gene therapy.Transfer of genetic materials,such as hepatocyte growth factor,using both viral and non-viral vectors has proved to be successful in augmenting liver regeneration in various animal models.For future clinical studies,ongoing research aims at eliminating toxicity of viral vectors and increasing transduction efficiency of non-viral vectors,which are the main drawbacks of these systems.Another goal of current research is to develop gene therapy that targets specific liver cells using receptors that are unique to and highly expressed by different liver cell types.The outcome of such investigations will,undoubtedly,pave the way for future successful clinical trials. | Hussein M Atta | 2010 | World Journal of Gastroenterology2010,16,32: | 3 |
