| 1 | Antioxidant Effect of Sepia Ink Extract on Extrahepatic Cholestasis Induced by Bile Duct Ligation in Rats显示文摘Objective The aim of our study was to assess the complications of hepatic fibrosis associated with bile duct ligation and the potential curative role of sepia ink extract in hepatic damage induced by bile duct ligation. Methods Rattus norvegicus rats were divided into 3 groups: Sham-operated group, model rats that underwent common bile duct ligation(BDL), and BDL rats treated orally with sepia ink extract(200 mg/kg body weight) for 7, 14, and 28 d after BDL. Results There was a significant reduction in hepatic enzymes, ALP, GGT, bilirubin levels, and oxidative stress in the BDL group after treatment with sepia ink extract. Collagen deposition reduced after sepia ink extract treatment as compared to BDL groups, suggesting that the liver was repaired. Histopathological examination of liver treated with sepia ink extract showed moderate degeneration in the hepatic architecture and mild degeneration in hepatocytes as compared to BDL groups. | 2 | Potential predictive factors for pathologic complete response after the neoadjuvant treatment of rectal adenocarcinoma:a single center experience显示文摘Objective:To assess the response rate of patients with rectal adenocarcinoma to neoadjuvant therapy and to identify the predictors of histological regression after neoadjuvant radiotherapy(RT)or concurrent chemoradiotherapy(CCRT).Methods:This study recruited 64 patients.The patients had resectable cancer of the lower and the middle rectum(T3/T4 and/or N+)without distant metastasis and received neoadjuvant RT or CCRT followed by radical surgery with total mesorectal excision(TME)between January 2006 and December 2011.The patients were classified into non-response(NR),partial response(PR),and pathologic complete response(p CR)based on the Dworak tumor regression grading system.Results:The median age of patients was 57 years(ranging from 22 to 85).A total of 24 patients were treated with neoadjuvant CCRT,whereas 40 patients were treated with RT alone.Abdominoperineal resection(APR)was performed on 29 patients(45%).Anterior resection with TME was performed on 34 patients(53%).One patient had local resection.Histologically,12(19%),24(73%),and 28(44%)patients exhibited p CR,PR,and NR,respectively.Univariate analysis revealed that the predictors of tumor regression were as follows:the absence of lymph node involvement from initial imaging(c N0)(P=0.021);normal initial carcinoembryonic antigen(CEA)level(P=0.01);hemoglobin level≥12 g/dl(P=0.009);CCRT(P=0.021);and tumor downstaging in imaging(P=0.001).Multivariate analysis showed that the main predictors of p CR were CT combined with neoadjuvant RT,c N0stage,and tumor regression on imaging.Conclusions:Identifying the predictors of p CR following neoadjuvant therapy aids the selection of responsive patients for nonaggressive surgical treatment and possible surveillance. | Feryel Letaief Meher Nasri Mouna Ayadi Khedija Meddeb Amina Mokrani Yosra Yahyaoui Nesrine Chraiet Henda Raies Amel Mezlini | 2017 | Cancer Biology & Medicine2017,14,3: | 3 |

| 5 | Self-compatibility in peach[Prunus persica(L.)Batsch]:patterns of diversity surrounding the S-locus and analysis of SFB alleles显示文摘Self-incompatibility(SI)to self-compatibility(SC)transition is one of the most frequent and prevalent evolutionary shifts in flowering plants.Prunus L.(Rosaceae)is a genus of over 200 species most of which exhibit a Gametophytic SI system.Peach[Prunus persica(L.)Batsch;2n=16]is one of the few exceptions in the genus known to be a fully selfcompatible species.However,the evolutionary process of the complete and irreversible loss of SI in peach is not well understood and,in order to fill that gap,in this study 24 peach accessions were analyzed.Pollen tube growth was controlled in self-pollinated flowers to verify their self-compatible phenotypes.The linkage disequilibrium association between alleles at the S-locus and linked markers at the end of the sixth linkage group was not significant(P>0.05),except with the closest markers suggesting the absence of a signature of negative frequency dependent selection at the S-locus.Analysis of SFB1 and SFB2 protein sequences allowed identifying the absence of some variable and hypervariable domains and the presence of additionalα-helices at the C-termini.Molecular and evolutionary analysis of SFB nucleotide sequences showed a signature of purifying selection in SFB2,while the SFB1 seemed to evolve neutrally.Thus,our results show that the SFB2 allele diversified after P.persica and P.dulcis(almond)divergence,a period which is characterized by an important bottleneck,while SFB1 diversified at a transition time between the bottleneck and population expansion. | Donia Abdallah Ghada Baraket Veronica Perez Amel Salhi Hannachi Jose I.Hormaza | 2020 | Horticulture Research2020,7,1: | 3 |
| 6 | Cytogenetic and Gene Expression Study in Radiation Workers Occupationally Exposed to Low Levels of Ionizing Radiation显示文摘Background:Exposure to IR(ionizing radiation)causes damage to living cells,especially to DNA,the degree of cellular damage depends on the radiation amount.Humans are naturally exposed to IR from cosmic rays,and artificially through diagnostic procedures,medical treatments or occupationally during work shifts.Cytogenetic assay of peripheral blood lymphocytes can provide a biological estimation of the dose received in IR exposures.The CBMN(cytokinesis-blocked micronucleus)assay is a widely used,since it represents a reliable test to detect radiation-induced chromosome aberration and it is a valuable biomarker in many biomonitoring studies on human populations occupationally exposed to IR.The NDI(nuclear division index)is a cell proliferation marker in cultures which is considered a measure of general cytotoxicity,the relative frequencies of the cells may be used to define cell cycles progression of the lymphocyte after mitogenic stimulation.Nevertheless the assay is frequently employed as a useful research tool for understanding the cell cycling kinetics of the cultures.Many gene expression studies demonstrated an up-regulation of genes involved in the processes of signal transduction,control of cell cycle,DNA repair and apoptosis after exposure of IR in different mammalian cell types.Altered expression of a few genes plays specific roles in DNA repair/cell cycle control such as CDKN1A,XPC,GADD45A,DDB2 and PCNA and cell cycle regulation/proliferation such as IL16,CABLES2,TGFB2 and RHOA.Objective:The present study aims to use the MN,NDI and gene expression analysis as biomarkers for investigation of the effects of IR exposure in some radiation workers occupationally exposed to low IR in Al-Tuwaitha site,Baghdad,Iraq.Also,authors assessed the effect of IR on the expression of some gene marker such as:RHOA,CDKN1A,GADD45A and RAD52.Methods:This study was carried out on thirty Iraqi male radiation workers occupationally exposed to low levels of IR at Al-Tuwaitha site,Baghdad,non-smokers and non-alcoholic,aged 30-59 years,as well as thirty apparently healthy individuals males collected randomly from population living in Baghdad,aged 30-59 years who are non-smokers,non-alcoholic as control group.Three molecular genetic parameters were employed such as CBMN,NDI and gene expression assay.The MN and NDI assay were performed according to the description by IAEA,2001.Total RNA was isolated from blood for radiation worker and control group.The RNA concentration was determined by measuring their absorbance that depends on the ratio A260/A280 of the wavelength,which leads to the determination of RNA purity,which ranged from 1.79-2.1 in two groups.Using RT-PCR for study gene expression,four types of specialized primer genes were selected for the mRNA genes:RHOA,CDKN1A,GADD45A and RAD52 which have a relation with IR in addition to the primers for internal control.Housekeeping gene((β-actin)was used as a reference gene to normalize the quantity of the target genes.Results:The results of cytogenetic analysis showed that micronuclei frequencies were significantly higher(p<0.01)in the radiation workers group in Al-Tuwaithasite,Baghdadas compared with the control group,and showed significant decrease(p>0.01)in the NDI in radiation workers as compared with the control group.Total RNA was isolated from blood for the radiation workers and control groups mentioned.Using RT-PCR for study gene expression,four types of specialized primer genes were selected for the mRNA:genes RHOA,CDKN1A,GADD45A and RAD52 which have a relation with IR in addition to the primers for internal control(β-actin)gene.The products of replicated specialized primers for the genes concerned and the cDNA for the studied samples were electrophoretically separated in agarose gels.The banding profiles were visualized by ethidium bromide staining,as the molecular weight were 135 bp,165 bp,185 bp and 470 bp(nitrogen-base pair)for RHOA,CDKN1A,GADD45A and RAD52 genes,respectively.Gene expression analysis revealed statistically significant(ΔΔCt comparative Ct method)transcriptional changes in three genes RHOA,GADD45A,CDKN1A up-regulated while the RAD52 gene down-regulated.Conclusions:The results indicated that there is a possibility of using the changes in the MN,NDI and genes expression such as RHOA,CDKN1A,GADD45A and RAD52 as useful biomarkers for detection of IR in radiation workers occupationally exposed to low levels of IR. | Abdulsahib.K.Ali.Al-Ziyad Amel J.Mutar Zainab Abbas Ouda Shatha K.Khyon | 2021 | Journal of US-China Medical Science2021,18,1: | 2 |

| 8 | Geochemical characterization of the salinity of irrigated soils in arid regions(Biskra, SE Algeria)显示文摘The agriculture in Biskra,southeastern Algeria,is based on traditional practices and characterized by small irrigated fields.In the last decades,the increasing demand in water as well as the scarcity of rainfall has forced many farmers to use groundwater of low quality to maintain the profitability of their crops.Unfortunately,this practice seems to be the main harmful factor for soil quality in the region since it is responsible for the salinization of the irrigated areas.Aiming to assess the impact of this phenomenon,the soils of the irrigated perimeter of El Ghrous-a representative rural community located in the west of Biskra-have been analyzed.A set of 82 soil samples was collected from top and subsoil(0-15 and15-35 cm respectively),on which the following physicochemical analyzes were performed:Ca^(2+),Mg^(2+),K^(+),Na^(+),Cl^(-),SO_(4)^(2-),HCO_(3)^(-),NO_(3)^(-),pH,electrical conductivity(EC)and sodium adsorption ratio.A Principal Component Analysis was performed to individuate the geochemical processes that influenced significantly the evolution of soil salinity and its pathways.The results showed a calcium sulfate(CaSO_(4))facies with a high risk of salinity and a low to medium risk of alkalinity.The calcite residual alkalinity and generalized residual alkalinity decreased as the solutions became more concentrated.Most of the samples were oversaturated in carbonate minerals(aragonite,calcite,and dolomite)and under saturated in evaporitic minerals(anhydrite,gypsum,and halite).Finally,two multiple linear regressions(using cations and anions as independent variables)have been proposed to quantify soil salinity.These equations,with an accuracy of 85%,can represent a time and money-saving tool for managers and farmers to estimate the EC,in comparison to the traditional estimation methods. | Mohamed Amine Abdennour Abdelkader Douaoui Jesus Barrena Manuel Pulido Abdelhamid Bradai Amel Bennacer Chiara Piccini Alberto Alfonso-Torreno | 2021 | Acta Geochimica2021,40,2: | 2 |
