1套式PCR检测恶性疟原虫Pfcrt基因76号编码多态性及RFLP分析显示文摘目的检测恶性疟原虫氯喹抗性转运基因 ( Pfcrt) 76编码多态性及其用该多态性显示恶性疟原虫对氯喹反应性进行评价。方法用套式 PCR/RFLP技术检测现场采集的滤纸血滴上恶性疟原虫 crt基因 76编码的赖氨酸野生型多态性及苏氨酸突变型多态性 ,并对恶性疟原虫的氯喹反应性进行体内法测定。结果 34份滤纸血样中 ,32份的恶性疟原虫 crt基因 76号编码为苏氨酸 (突变型 ) ,突变型比例为 94 .1 2 % ;34份血样中 2 1份做氯喹抗性体内观察 ,该 2 1个病例中 1 4份血样恶性疟原虫 crt基因 76号编码为突变型 ,体内法观察同样显示恶性疟原虫对氯喹有抗性 ,两种方法显示恶性疟原虫对氯喹有抗性的符合率为 66.67%。结论套式 PCR/RFLP检测 Pfcrt基因 76号编码多态性 ,其方法简便、快速 ,但其多态性对氯喹反应性的提示尚需进一步评价。董莹 张再兴 Adagu IS 2001实用寄生虫病杂志2001,9,3:3
2云南边境地区氯喹及与青蒿琥酯伍用治疗前后恶性疟原虫pfcrt和pfmdr1抗药性有关基因点突变的变化特征(英文)显示文摘目的 了解氯喹单用及与青蒿琥脂伍用治疗恶性疟前后 ,pfcrt和 pfmdr1抗药性有关基因的点突变变化特征。 方法 使用PCR RFLP技术检测基因点突变。 结果 氯喹及与青蒿琥脂伍用治疗前后的所有样本都发现有恶性疟原虫pfcrt基因氨基酸编码 76突变为苏氨酸的特征。但是 ,氯喹治疗前 ,5 0 % pfmdr1基因氨基酸编码 86为天冬酰氨酸 (野生型 ) ,而剩余的 5 0 %为野生型和突变型 (苏氨酸 )的特征。氯喹治疗后 ,在 18个复燃的病例中 ,83 .3 %的 pfmdr1基因 86位点为野生型 ,剩余的 16.7%是混合型。氯喹与青蒿琥脂伍用治疗前 ,3个样本携带混合型基因型 ,剩余的 (86% )为野生型 ,但治疗后 ,所有样本只携带野生型。 结论 这些结果可能支持这样的假说 :pfcrt基因突变起主导作用 ,但 pfmdr1基因突变增强了氯喹抗药性的效果。杨亚明 Adagu IS 张再兴 Warhurst DC 2003中国寄生虫病防治杂志2003,16,6:2

18云南省恶性疟原虫多药抗性基因^(Asn)86^(Tyr)多态性调查显示文摘目的检测云南恶性疟原虫多药抗性基因 (Pfmdr1 ) Asn 86Tyr的多态性 ,探索其与氯喹敏感性表现型的关系。方法用等位基因特异 PCR和限制性酶切片段长度分析技术 (AS-PCR/RFLP)。结果在 86% (1 9/2 2 )的氯喹抗性分离株中检测到 Pfmdr1基因的 Asn86Tyr两种多态性 ,携带编码 86Tyr多态变异的占 50 % ,而含编码 Asn86多态的也是 50 %。结论在云南氯喹抗性高度流行区 ,Pfmdr1基因杨亚明 Adagu IS 高白荷 2001实用寄生虫病杂志2001,9,2:1
