| 7 | Comparative and population genomics of buckwheat species reveal key determinants of flavor and fertility显示文摘Common buckwheat(Fagopyrum esculentum)is an ancient crop with a world-wide distribution.Due to its excellent nutritional quality and high economic and ecological value,common buckwheat is becoming increasingly important throughout the world.The availability of a high-quality reference genome sequence and population genomic data will accelerate the breeding of common buckwheat,but the high heterozygosity due to the outcrossing nature has greatly hindered the genome assembly.Here we report the assembly of a chromosome-scale high-quality reference genome of F.esculentum var.homotropicum,a homozygous self-pollinating variant of common buckwheat.Comparative genomics revealed that two cultivated buckwheat species,common buckwheat(F.esculentum)and Tartary buckwheat(F.tataricum),underwent metabolomic divergence and ecotype differentiation.The expansion of several gene families in common buckwheat,including FhFAR genes,is associated with its wider distribution than Tartary buckwheat.Copy number variation of genes involved in the metabolism of flavonoids is associated with the difference of rutin content between common and Tartary buckwheat.Furthermore,we present a comprehensive atlas of genomic variation based on whole-genome resequencing of 572 accessions of common buckwheat.Population and evolutionary genomics reveal genetic variation associated with environmental adaptability and floral development between Chinese and non-Chinese cultivated groups.Genome-wide association analyses of multi-year agronomic traits with the content of flavonoids revealed that Fh05G014970 is a potential major regulator of flowering period,a key agronomic trait controlling the yield of outcrossing crops,and that Fh06G015130 is a crucial gene underlying flavor-associated flavonoids.Intriguingly,we found that the gene translocation and sequence variation of FhS-ELF3 contribute to the homomorphic self-compatibility of common buckwheat.Collectively,our results elucidate the genetic basis of speciation,ecological adaptation,fertility,and unique flavor of common buckwheat,and provide new resources for future genomics-assisted breeding of this economically important crop. | Kaixuan Zhang Yuqi He Xiang Lu Yaliang Shi Hui Zhao Xiaobo Li Jinlong Li Yang Liu Yinan Ouyang Yu Tang Xue Ren Xuemei Zhang Weifei Yang Zhaoxia Sun Chunhua Zhang Muriel Quinet Zlata Luthar Mateja Germ Ivan Kreft Dagmar Janovska Vladimir Meglic Barbara Pipan Milen I.Georgiev Bruno Studer Mark A.Chapman Meiliang Zhou | 2023 | Molecular Plant2023,16,9: | 0 |

| 11 | A 40-year evaluation of drivers of African rainforest change显示文摘Background:Tropical forests are repositories of much of the world’s biodiversity and are critical for mitigation of climate change.Yet,the drivers of forest dynamics are poorly understood.This is in large part due to the lack of long-term data on forest change and changes in drivers.Methodology:We quantify changes in tree abundance,diversity,and stand structure along transects first enumerated in 1978 and resampled 2019 in Kibale National Park,Uganda.We tested five predictions.First,based on the purported role of seed dispersal and herbivory and our quantification of changes in the abundance of frugivores and herbivores,we tested two predictions of how faunal change could have influenced forest composition.Second,based on an evaluation of life history strategies,we tested two predictions concerning how the forest could have changed following disturbance that happened prior to written history.Finally,based on a 50-year climate record,we evaluate the possible influence of climate change on forest dynamics.Results:More trees were present on the assessed transects in 2019(508)than in 1978(436),species richness remained similar,but diversity declined as the number of dominant species increased.Rainfall increased by only 3 mm over the 50 years but this had not significant effect on forest changes measured here.Annual average monthly maximum temperature increased significantly by 1.05℃ over 50 years.The abundance of frugivorous and folivorous primates and elephants increased over the 50 years of monitoring.Neither the prediction that an increase in abundance of seed dispersing frugivores increases the abundance of their preferred fruiting tree species,nor that as an increase in folivore abundance causes a decline in their preferred species were supported.As predicted,light-demanding species decreased in abundance while shade-tolerant species increased as expected from Kibale being disturbed prior to historical records.Finally,while temperature increased over the 50 years,we found no means to predict a priori how individual species would respond.Conclusions:Our study revealed subtle changes in the tree community over 40 years,sizable increases in primate numbers,a substantial increase in the elephant population and an increase in local temperature.Yet,a clear picture of what set of interactions impact the change in the tree community remains elusive.Our data on tree life-history strategies and frugivore/herbivore foraging preferences suggest that trees species are under opposing pressures. | Colin A.Chapman Carmen Galan-Acedo Jan F.Gogarten Rong Hou Michael J.Lawes Patrick A.Omeja Dipto Sarkar Anna Sugiyama Urs Kalbitzer | 2021 | Forest Ecosystems2021,8,4: | 0 |
