| 1 | Analysis of sea surface temperature fronts in the Taiwan Strait and its adjacent area using an advanced edge detection method显示文摘A morphology-based edge detection method has been used to study sea surface temperature (SST) fronts in the Taiwan Strait and its adjacent area. The method is based on mathematical morphology with multi-dimensional and multi-structural elements. Using six years’ SST data from September 2002 to August 2008, we distinguished the large SST front like Kuroshio Front as well as the smaller ones: namely Taiwan Bank Front, Zhe-Min Coastal Front and Zhang-Yun Ridge Front. The seasonal and monthly variations of these fronts were also studied. Generally, the SST fronts are stronger in winter but weaker in summer. | 2 | Activation of JNK by TPA promotes apoptosis via PKC pathway in gastric cancer cells显示文摘AIM: JNK cascade plays an important role in cell proliferation, differentiation and apoptosis. However, the exact function of JNK cascade for apoptosis induction remains largely unknown. In this study, the role of JNK activation stimulated by TPA in the process of apoptosis induction and its signaling transduction pathway in gastric cancer cells were investigated and determined.METHODS: Expressions of mRNA and protein were detected by Northern blot and Western blot. Transcription activity was measured by transient transfection and CAT assay. Apoptotic cells were displayed through staining the nucleus with DAPI and were observed under fluorescence microscope. The apoptotic index was determined by counting 1000 cells randomly.RESULTS: JNK protein was stimulated rapidly by TPA, and reached its highest peak within 3 hr, then decreased in a time-dependent manner, but the expression level of JNK protein induced by TPA was always keeping higher than that in untreated cells. Similar pattern was seen in c-jun mRNA level induced by TPA. TPA significantly activated the transcriptional activity of activator protein-1 with a TPA-closedependent manner. Furthermore, activation of JNK was mediated through PKC pathway. Treatment of cells with PKC specific inhibitor, Wortmannin, led to repression of JNK even in the presence of TPA. More importantly, all these effects were associated with induction of apoptosis in gastric cancer cells. TPA inducted apoptosis obviously in gastric cancer cells. The apoptotic cells became smaller and rounded, and their nuclei became condensation and fragmentation with brightly stained chromatin. However, suppression of JNK by PKC specific inhibitor, Wortmannin, resulted in the decrease of apoptosis induced by TPA in a time-dependent manner, apoptotic index dramatically decreased from 32.56 % to 8.71%.CONCLUSION: TPA stimulates JNK cascade, including upregulation of JNK protein expression level and c-jun mRNA expression level, and activation of activator protein-1 transcriptional activity. Activation of JNK is mediated through PKC pathway, which has an association with induction of apoptosis by TPA. | 3 | Impacts of increased atmospheric CO_2 concentration on photosynthesis and growth of micro-and macro-algae显示文摘Marine photosynthesis drives the oceanic biological CO2 pump to absorb CO2 from the atmosphere, which sinks more than one third of the industry-originated CO2 into the ocean. The increasing atmos-pheric CO2 and subsequent rise of pCO2 in seawater, which alters the carbonate system and related chemical reactions and results in lower pH and higher HCO3- concentration, affect photosynthetic CO2 fixation processes of phytoplanktonic and macroalgal species in direct and/or indirect ways. Although many unicellular and multicellular species can operate CO2-concentrating mechanisms (CCMs) to util-ize the large HCO3- pool in seawater, enriched CO2 up to several times the present atmospheric level has been shown to enhance photosynthesis and growth of both phytoplanktonic and macro-species that have less capacity of CCMs. Even for species that operate active CCMs and those whose photo-synthesis is not limited by CO2 in seawater, increased CO2 levels can down-regulate their CCMs and therefore enhance their growth under light-limiting conditions (at higher CO2 levels, less light energy is required to drive CCM). Altered physiological performances under high-CO2 conditions may cause genetic alteration in view of adaptation over long time scale. Marine algae may adapt to a high CO2 oceanic environment so that the evolved communities in future are likely to be genetically different from the contemporary communities. | 4 | Properties and characterization of Au^(3+)-adsorption by mycelial waste of Streptomyces aureofaciences显示文摘Mycelial waste of Streptomyces aureofaciences procured from the aureomycin fermentation industry is used as biosorbent for Au3+. The properties of Au3+ adsorption by the mycelial waste are studied. The results indicate that the optimum pH value of Au3+ adsorption is 3.5. The biosorption is a rapid and non-temperature-dependent process. The bio-sorptive capacity with 45.6 mg/g and efficiency with 91.2% are achieved under the conditions of pH 3.5 and 30℃ for 45 min, in which the ratio is 50 mg/g dry weight for the concentrations of initial Au3+ and the mycelial waste. The Au3+ ions adsorbed on the mycelial waste can be eluted. The observation in a transmission electron microscope shows that the Au3+ ions can be reduced to Au particles by the mycelial waste and the Au can become gold crystals with differentforms and sizes. | 5 | Studies on the role of marine bacteria in the upwelling ecosystem——Ⅰ.Bacterial biomass in the upwelling regions显示文摘-Six cruises were carried out off the south bank of Fujian - Taiwan during the period of December 21, 1987 to November 15, 1988 to estimate the contribution of bacterial biomass carbon (BBC) to the totai particulate organic pools using epifluorescent microscopic technique. The results show that the standing crop of bacteria fluctuated from 0. 95 to 66. 60 mg /m3 (dry weight). Upwelling phenomena appeared in the region around Nanpeng Island in summer while in the region of Waixie in all seasons. The average value of BBC was 27. 60(±6. 08)mg/m3and 21. 32 (±2. 34) mg/m3 respectively. The seasonal and spatial distribution is discussed in relation to environmental factors as well as upwelling phenomena. | 6 | Molecular phylogenetic relationships of China Seas groupers based on cytochrome b gene fragment sequences显示文摘The classification and evolutionary relationships are important issues in the study of the groupers. Cytochrome b gene fragment of twenty-eight grouper species within six genera of subfamily Epinephelinae was amplified using PCR techniques and the sequences were analyzed to derive the phylogenetic relationships of the groupers from the China Seas. Genetic information indexes, including Kimura-2 parameter genetic distance and Ts/Tv ratios, were generated by using a variety of biology softwares. With Niphon spinosus, Pagrus major and Pagrus auriga as the designated outgroups, phylogenetic trees, which invoke additional homologous sequences of other Epinephelus fishes from GenBank, were constructed based on the neighbor-joining (NJ), maximum-parsimony (MP), maximum-likelihood (ML) and minimum-evolution (ME) methods. | 8 | Primary structure and configuration of tea polysaccharide显示文摘The monosaccharide composition of a tea polysaccharide (TGC) was determined by GC-MS method. Furthermore, the primary structure of tea polysaccharide and its configura-tion in the aqueous solution were investigated utilizing a combination of classical chemical methods and modern instrumental techniques including GC-MS, Proton NMR, UV and CD. The results indicate that TGC consists of 6 monosaccharides: Rha, Ara, Xyl, Glu, Man and Gal. The configuration of TGC in water solution is proposed to be an ordered helix. The possible primary structure of TGC was outlined as below: the basic structure of the main chain consists of Rha, Glu and Gal units. All three monosaccharides can potentially be connected to branch chains consisting of mainly Ara, and the linkages could be in b1→2, b 1→3, b 2→3 forms. | 9 | Species composition and distribution of marine nematode community in the North Taiwan Strait显示文摘Marine free-living nematodes were investigated at 13 sampling stations divided into three transects in the northern Taiwan Strait in February 1998. One hundred species of marine nematodes belonging to 91 Genera 28 Families 3 Orders were identified and were first recorded in the northern Taiwan Strait. The dominant species were Vasostoma sp., Sabatieria sp. 1, Linhystera sp. 1, Spilophorella sp., Daptonema sp., Halalaimus sp. and Dorylaimopsis variabilis. Their main densities were all over 4 950 ind./m2. According to mean density at transects, marine nematode density decreased from coastal Weitou to off Minjiang Estuary, which was similar to polychaete distribution in northern Taiwan Strait. The selective deposit feeder (1A) was the dominant food type of marine nematodes in the northern Taiwan Strait, but non-selective deposit feeders (1B) and epigrowth feeders (2A) occupied high proportion, indicating diverse feeding types of marine nematodes in the northern Taiwan Strait. | 10 | Characteristics of pCO_(2) in surface water of the Bering Abyssal Plain and their effects on carbon cycle in the western Arctic Ocean显示文摘Characteristics of the pCO2 distribution in surface water of the Bering Abyssal Plain and their relationships with the ambient hydrological conditions were discussed using variations of the partial pressure of CO2 in surface water of the Bering Abyssal Plain and the Chukchi Sea. Data in this study are from a field investigation during the First Chinese National Arctic Research Expedition in 1999. Compared to the high productivity in the Bering Continental Shelf, much lower levels of chlorophyll a were observed in the Bering Abyssal Plain. The effect of hydrological factors on the pCO2 distribution in surface seawater of the Plain in summer has become a major driving force and dominated over biological factors. The Plain also presents a High Nutrient Low Chlorophyll (HNLC). In addition, the pCO2 distribution in the Bering Abyssal Plain has also been found to be influenced from the Bering Slope Current which would transform to the Anadyr Cur- rent when it inflows northwestward over the Plain. The Anadyr Current would bring a high nutrient water to the western Arctic Ocean where local nutrients are almost depleted in the surface water during the summer time. Resupplying nutrients would stimulate the growth of phytoplankton and enhance capacity of absorbing atmospheric CO2 in the surface water. Otherwise, in the Bering Sea the dissolved inorganic carbon brought from freshwater are not deposited down to the deep sea water but most of them would be transported into the western Arctic Ocean by the Alaska Coastal Current to form a carbon sink there. | 14 | Roles of PLC-γ2 and PKCα in TPA-induced apoptosis of gastric cancer cells显示文摘AIM: To investigate the roles of PLCγ2 and PKCα in TPA-induced apoptosis of gastric cancer cells.METHODS: Human gastric cancer cell line MGC80-3 was used. Protein expression levels of PLCγ2 and PKCα were detected by Western blot. Protein localization of PLCγ2 and PKCα was shown by immunofluoscence analysis under laserscanning confocal microscope. Apoptotic morphology was observed by DAPI fluorescence staining, and apoptotic index was counted among 1 000 cells randomly.RESULTS: Treatment of gastric cancer cells MGC80-3 with TPA not only up-regulated expression of PLC-γ2 protein,but also induced PLC-γ2 translocation from the cytoplasm to the nucleus. However, this process was not directly associated with apoptosis induction. Further investigation showed that PKCa translocation from the cytoplasm to the nucleus was correlated with initiation of apoptosis. To explore the inevitable linkage between PLC-γ2 and PKCα during apoptosis induction,PLC inhibitor U73122 was used to block PLC-γ2 translocation,in which neither stimulating PKCα translocation nor inducing apoptosis occurred in MGC80-3 cells. However, when U73122treated cells were exposed to TPA, not only PLC-γ2, but also PKCα was redistributed. On the other hand, when cells were treated with PKC inhibitor alone, PLC-γ2 protein was still located in the cytoplasm. However, redistribution of PLC-γ2 protein occurred in the presence of TPA, no matter whether PKC inhibitor existed or not.CONCLUSION: PLC-γ2 translocation is critical in transmitting TPA signal to its downstream molecule PKCα. | 15 | Novel Ru - K/Carbon Nanotubes Catalyst for Ammonia Synthesis显示文摘A novel ammonia synthesis catalyst, potassium-promoted ruthenium supported on carbon nanotubes, was developed. It was found that the Ru-K/carbon nanotubes catalyst had higher activity for ammonia synthesis (20.85 ml NH3/h/g-cat ) than the Ru-K/fullerenes (13.3 ml NH3/h/g-cat ) at atmospheric pressure and 623 K. The catalyst had activity even at 473 K. and had the highest activity (23.46 ml NH3/h/g-cat ) at 643 K. | 16 | Controllable incorporation of CdS nanoparticles into TiO_2 nanotubes for highly enhancing the photocatalytic response to visible light显示文摘A constant current electrochemical deposition was employed to incorporate CdS nanoparticles into the TiO2 nanotube arrays (TiO2NTs). The size and amount of CdS nanoparticles in TiO2NTs (CdS@TiO2NTs) were controllable via modulating current,deposition time and electrolyte concentration. It was revealed,from the scanning electron microscopy (SEM) images and X-ray photoelectron spectroscopy (XPS) in depth profile,that CdS nanoparticles were filled into TiO2 nanotubes. A shift of the absorption edge toward the visible region under the optimal electrodeposition condition was observed with the diffuse reflectance spectroscopy (DRS). A 5-fold enhancement in the photocurrent spectrum for TiO2NTs was observed and the photocurrent response range was significantly extended into the visible region because of the CdS incorporation. | 17 | High-yield expression of recombinant SARS coronavirus nudeocapsid protein in methylotrophic yeast Pichia pastoris显示文摘AIM: Nucleocapsid (N) protein plays an important role in reproduction and pathological reaction of severe acute respiratory syndrome (SARS) coronavirus (SCoV), the antigenicity of the protein is better than spike (S) protein.This study was to find a highly specific and antigenic recombinant SCoV nucleocapsid (rSCoVN) protein, and to provide a basis for further researches on early diagnosis of SARS.METHODS: Full length cDNA of SCoV nucleocapsid (SCoVN) protein was amplified through polymerase chain reaction (PCR) and cloned into yeast expression vector pPIC3.5K to construct plasmid of pPIC3.5K-SCoVN. The plasmid was linearized and then transformed into Pichia pastoris (P.pastoris) GS115 (His^+ Mut^+) by electroporation. His^+Mut^+ recombinant strains were identified by PCR and cultivated on MM/MD plates. The influence of different factors on biomass and rSCoVN protein production during induction phase, such as various induction media, dissolved oxygen (DO) and different final concentrations of methanol, was subsequently studied. The expression level and activation were detected by SDS-PAGE and Western-blot respectively.RESULTS: All of the recombinants were His^+Mut^+ after transformation of P.pastoris with linearized plasmids. The BMMY medium was optimal for recombinant ScoVN (rSCoVN) protein expression and growth of the recombinant strains.The final optimal concentration of methanol was 20 ml_/L,the DO had a significant effect on rSCoVN protein expression and growth of recombinant strains. The rSCoVN protein expressed in recombinant strains was about 8% of the total cell protein, 520 mg/L of rSCoVN protein was achieved,and a maximum cell A at 600 nm of 62 was achieved in shake flask culture. The rSCoVN protein had a high specificity against mouse-anti-SARS-CoVN-mAb and SARS positive sera, but had no cross-reaction with normal human serum.The biological activity of rSCoVN expressed in P.pastoris was about 4-fold higher than that expressed in Ecoliwhen the same rSCoVN protein quantity was used.CONCLUSION: Active recombinant severe acute respiratory syndrome (SARS) coronavirus nucleocapsid (rSCoVN) protein can be successfully expressed in recombinant methylotrophic yeast P.pastoris GSl15. | 18 | The effect pathway of retinoic acid through regulation of retinoic acid receptor α in gastric cancer cells显示文摘AIM To evaluate the role of RARa gene in mediating the growth inhibitory effect of ail-trans retinoic acid (ATRA)on gastric cancer cells.``METHODS The expression levels of retinoic acid receptors (RARs) in gastric cancer cells were detected by Northern blot. Transient transfection and chlorophenicol acetyl transferase (CAT) assay were used to show the transcriptional activity of β retinoic acid response element (βRARE) and AP-l activity. Cell growth inhibition was determined by MTT assay and anchorage-independent growth assay, respectively. Stable transfection was performed by the method of Lipofectamine, and the cells were screened by G418.``RESULTS ATRA could induce expression level of RARα in MGC80-3, BGCC8823 and SGC-7901 cells obviously,resulting in growth inhibition of these cell lines. After sense RARa gene was transfected into MKN-45 cells that expressed rather Iow level of RARα and could not be induced by ATRA, the cell growth was inhibited by ATRA markedly. In contrast, when antisense RARα gene was transfected into BGC-825 cells, a little inhibitory effect by ATRA was seen, compared with the parallel BGC-823cells. In transient transfection assay, ATRA effectively induced transcriptional activity of βRARE in MGC80-3,BGC.823, SGC-7902 and MKN/RARa cell lines, but not in MKN-45 and BGC/aRARa cell lines. | 20 | The input/removal rates of size-fractionated ^210Po in the Jiulongjiang estuary and its implications显示文摘The geochemical behaviors of dissolved (<0.4 mm), low molecular weight (<10 kDa) and colloidal (10 kDa- 0.4 mm) 210Po in surface waters from the Jiulongjiang estuary were examined and the input/removal rates of different 210Po species were estimated. Both dissolved and low molecular weight 210Po were considerably removed during estuarine mixing. However, colloidal 210Po was removed only at low salinity regions (S <15) but had input behavior at high salin-ity surface waters (S >15). The input/removal rates of three species indicate that 210Po was removed rapidly in the Jiu-longjiang estuary. The discrepancy of input/removal rates between 210Po species suggests different geochemical be-havior and a dynamic transformation between 210Po species.
