The biological stress of early weaned piglets显示文摘Pigs experience biological stress such as physiological, environmental, and social challenges when weaned from the sow. The process of weaning is one of the most stressful events in the pig's life that can contribute to intestinal and immune system dysfunctions that result in reduced pig health, growth, and feed intake, particularly during the first week after weaning. Technological improvements in housing, nutrition, health, and management have been used to minimize some of the adverse effects of weaning stress, but a greater understanding of the biological impact of stress is needed to improve strategies to overcome weaning stress. The focus of this review paper is to briefly describe how the biological stress associated with weaning impacts intestinal morphology, structure, physiology, and intestinal immune responses that can impact subsequent production efficiencies such as growth, intake, morbidity, and mortality. | Joy M Campbell Joe D Crenshaw Javier Polo | 2013 | Journal of Animal Science and Biotechnology2013,4,2: | 83
Fermented liquid feed for pigs: an ancient technique for the future显示文摘Fermented liquid feed is feed that has been mixed with water at a ratio ranging from 1:1.5 to 1:4.By mixing with water, lactic acid bacteria and yeasts naturally occurring in the feed proliferate and produce lactic acid, acetic acid and ethanol which reduces the pH of the mixture. This reduction in pH inhibits pathogenic organisms from developing in the feed.In addition, when this low pH mixture is fed,it reduces the pH in the stomach of pigs and prevents the proliferation of pathogens such as coliforms and Salmonella in the gastrointestinal tract. For piglets, the use of fermented liquid feed offers the possibility of simultaneously providing feed and water, which may facilitate an easier transition from sow's milk to solid feed.Secondly, offering properly produced fermented liquid feed may strengthen the role of the stomach as the first line of defense against possible pathogenic infections by lowering the pH in the gastrointestinal tract thereby helping to exclude enteropathogens. Finally, feeding fermented liquid feed to pigs has been shown to improve the performance of suckling pigs, weaner pigs and growing-finishing pigs.In this review, current knowledge about the use of fermented liquid feed in pig diets will be discussed. This will include a discussion of the desirable properties of fermented liquid feed and factors affecting fermentation. In addition, advantages and disadvantages of fermented liquid feed will be discussed including its effects on gastrointestinal health, intestinal pH and the types of bacteria found in the gastrointestinal tract as well as the effects of fermented liquid feeds on pig performance. | Joris AM Missotten Joris Michiels Jeroen Degroote Stefaan De Smet | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 67
Synthesis and paste properties of octenyl succinic anhydride modified early Indica rice starch显示文摘Octenyl succinic anhydride (OSA) modified early Indica rice starch was prepared in aqueous slurry systems using response surface methodology. The paste properties of the OSA starch were also investigated. Results indicated that the suitable parameters for the preparation of OSA starch from early Indica rice starch were as follows: reaction period 4 h, reaction tem- perature 33.4 °C, pH of reaction system 8.4, concentration of starch slurry 36.8% (in proportion to water, w/w), amount of OSA 3% (in proportion to starch, w/w). The degree of substitution was 0.0188 and the reaction efficiency was 81.0%. The results of paste properties showed that with increased OSA modification, the starch derivatives had higher paste clarity, decreased retro- gradation and better freeze-thaw stability. | SONG Xiao-yan CHEN Qi-he RUAN Hui HE Guo-qing XU Qiong | 2006 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2006,7,10: | 51

Relationship between hyperuricemia and metabolic syndrome显示文摘Objective: To investigate the relationship between metabolic syndrome and hyperuricemia. Methods: A total of 2 374 subjects who received health examination in our hospital from Jan. 2004 to Dec. 2006 were enrolled in our study. Hyperuricemia is defined as ≥7 mg/dl (in men) or ≥6.0 mg/dl (in women). Metabolic syndrome was defined using AHA/NHLBI (American Heart Association/National Heart, Lung, and Blood Institute) criteria. Results: (1) The overall prevalence of hyperuricemia was 13.10%. The condition was more common in men than in women (19.07% vs 3.42%). (2) Among men, uric acid concentration is statistically significantly positively correlated with waist circumference, blood pressure, and triglyceride. Uric acid is negatively correlated with serum high-density lipoprotein-cholesterol (HDL-C). Uric acid concentration is most strongly correlated with serum triglyceride (r=0.379) and waist circumference (r=0.297). Among women, statistically significant positive correlations were noted for the serum uric acid concentrations with waist circumference, triglyceride and fasting plasma glucose. Serum triglyceride (r=0.329) and waist circumference (r=0.234) are most strongly correlated with uric acid concentrations. (3) Men with hyperuricemia had a 1.634-fold increased risk of metabolic syndrome as compared with those without hyperuricemia odds ratio (OR)=1.634, P=0.000. Women with hyperuricemia had a 1.626-fold increased risk of metabolic syndrome (OR=1.626, P=0.000) as compared with those without hyperuricemia. Conclusion: Hyperuricemia is prevalent among Chinese population. Additionally, serum uric acid is positively associated with metabolic syndrome. | CHEN Li-ying ZHU Wen-hua CHEN Zhou-wen DAI Hong-lei REN Jing-jing CHEN Jian-hua CHEN Lei-qian FANG Li-zheng | 2007 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2007,8,8: | 54
WOMBAT—A tool for mixed model analyses in quantitative genetics by restricted maximum likelihood (REML)显示文摘WOMBAT is a software package for quantitative genetic analyses of continuous traits, fitting a linear, mixed model; estimates of covariance components and the resulting genetic parameters are obtained by restricted maximum likelihood. A wide range of models, comprising numerous traits, multiple fixed and random effects, selected genetic covariance structures, random regression models and reduced rank estimation are accommodated. WOMBAT employs up-to-date numerical and computational methods. Together with the use of efficient compilers, this generates fast executable programs, suitable for large scale analyses. Use of WOMBAT is illustrated for a bivariate analysis. | 7 | Review:Mechanism of acute pancreatitis complicated with injury of intestinal mucosa barrier显示文摘Acute pancreatitis (AP) is a common acute abdomen in clinic with a rapid onset and dangerous pathogenetic condition. AP can cause an injury of intestinal mucosa barrier, leading to translocation of bacteria or endotoxin through multiple routes, bacterial translocation (BT), gutorigin endotoxaemia, and secondary infection of pancreatic tissue, and then cause systemic in- flammatory response syndrome (SIRS) or multiple organ dysfunction syndrome (MODS), which are important factors influencing AP’s severity and mortality. Meanwhile, the injury of intestinal mucosa barrier plays a key role in AP’s process. Therefore, it is clinically important to study the relationship between the injury of intestinal mucosa barrier and AP. In addition, many factors such as microcirculation disturbance, ischemical reperfusion injury, excessive release of inflammatory mediators and apoptosis may also play important roles in the damage of intestinal mucosa barrier. In this review, we summarize studies on mechanisms of AP. | ZHANG Xi-ping ZHANG Jie SONG Qiao-ling CHEN Han-qin | 2007 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2007,8,12: | 44
Postconditioning of sevoflurane and propofol is associated with mitochondrial permeability transition pore显示文摘Background: Sevoflurane and propofol are effective cardioprotective anaesthetic agents, though the cardioprotection of propofol has not been shown in humans. Their roles and underlying mechanisms in anesthetic postconditioning are unclear. Mitochondrial permeability transition pore (MPTP) opening is a major cause of ischemia-reperfusion injury. Here we investigated sevoflurane- and propofol-induced postconditioning and their relationship with MPTP. Methods: Isolated perfused rat hearts were exposed to 40 min of ischemia followed by 1 h of reperfusion. During the first 15 min of reperfusion, hearts were treated with either control buffer (CTRL group) or buffer containing 20 μmol/L atractyloside (ATR group), 3% (v/v) sevoflurane (SPC group), 50 μmol/L propofol (PPC group), or the combination of atractyloside with respective anesthetics (SPC+ATR and PPC+ATR groups). Infarct size was determined by dividing the total necrotic area of the left ventricle by the total left ventricular slice area (percent necrotic area). Results: Hearts treated with sevoflurane or propofol showed significantly better recovery of coronary flow, end-diastolic pressures, left ventricular developed pressure and derivatives compared with controls. Sevoflurane resulted in more protective alteration of hemodynamics at most time point of reperfusion than propofol. These improvements were paralleled with the reduction of lactate dehydrogenase release and the decrease of infarct size (SPC vs CTRL: (17.48±2.70)% vs (48.47±6.03)%, P<0.05; PPC vs CTRL: (35.60±2.10)% vs (48.47±6.03)%, P<0.05). SPC group had less infarct size than PPC group (SPC vs PPC: (17.48±2.70)% vs (35.60±2.10)%, P<0.05). Atractyloside coadministration attenuated or completely blocked the cardioprotective effect of postconditioning of sevoflurane and propofol. Conclusion: Postconditioning of sevoflurane and propofol has cardio-protective effect against ischemia-reperfusion injury of heart, which is associated with inhibition of MPTP opening. Compared to propofol, sevoflurane provides superior protection of functional recovery and infarct size. | Wei HE Feng-jiang ZHANG Shao-ping WANG Gang CHEN Cong-cong CHEN Min YAN | 2008 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,2: | 48
Relationship between alanine aminotransferase levels and metabolic syndrome in nonalcoholic fatty liver disease显示文摘Objective:To investigate the relationship between alanine aminotransferase (ALT) levels and metabolic syndrome (MS) in nonalcoholic fatty liver disease (NAFLD). Methods: A total of 26527 subjects who received medical health checkup in our hospital from January 2005 to July 2007 were enrolled in the study. The diagnosis of fatty liver was based on ultrasound imaging. MS was defined according to the criteria of the Adult Treatment Panel III. ALT, triglyceride (TG), high density lipo-protein cholesterol (HDL-c), fasting plasma glucose (FPG), height, weight, waist circumference (WC), systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured in each subject to analyze the relationship between MS and ALT activity. Results: (1) The prevalence of NAFLD in men (30.94%) was significantly higher than that in women (15.65%); (2) The incidence of MS in NAFLD (33.83%) was significantly greater than that in non-NAFLD (10.62%); (3) Of the 6470 subjects with NAFLD, in the age-adjusted partial correlation analysis, there were statistically significant correlations between the ALT levels and most metabolic risk factors in each sex (P<0.01), except that ALT levels had no correlation with HDL-c in women. Moreover, in the multiple stepwise regression analysis, SBP lost its significance, and WC, body mass index (BMI), age, DBP, TG and FPG were independently associated with ALT levels in both sexes (P<0.05). HDL-c remained significant and was independently related to ALT levels in men; (4) ALT levels were significantly higher in subjects with MS compared to those without MS (P<0.001). Mean ALT levels increased with the number of MS components in each sex (P<0.05 for trend). Conclusion: We found a strong rela-tionship between ALT levels and MS in NAFLD and revealed that the cluster of MS components might be the predictor for ALT elevations. | Zhou-wen CHEN Li-ying CHEN Hong-lei DAI Jian-hua CHEN Li-zheng FANG | 2008 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,8: | 45

Effects of dietary supplementation of probiotic,Clostridium butyricum,on growth performance, immune response, intestinal barrier function, and digestive enzyme activity in broiler chickens challenged with Escherichia coli K88显示文摘Background:Colibacillosis caused by enterotoxigenic Escherichia coli(E.coli) results in economic losses in the poultry industry.Antibiotics are usually used to control colibacillosis, however,E.coli has varying degrees of resistance to different antibiotics. Therefore the use of probiotics is becoming accepted as an alternative to antibiotics. In this study,we evaluated the effects of Clostridium butyricum(C.butyricum) on growth performance, immune response, intestinal barrier function, and digestive enzyme activity in broiler chickens challenged with Escherichia coli(E.coif) K88.Methods:The chickens were randomly divided into four treatment groups for 28 days.Negative control treatment(NC) consisted of birds fed a basal diet without E.coli K88 challenge and positive control treatment(PC) consisted of birds fed a basal diet and challenged with E.coli K88.C.butyricum probiotic treatment(CB) consisted of birds fed a diet containing 2×10~7 cfu C butyricum/kg of diet and challenged with E.coli K88.Colistin sulfate antibiotic treatment(CS)consisted of birds fed a diet containing 20 mg colistin sulfate/kg of diet and challenged with E.coli K88.Results:The body weight(BW) and average day gain(ADG) in the broilers of CB group were higher(P< 0.05) than the broilers in the PC group overall except the ADG in the 14-21 d post-challenge.The birds in CB treatment had higher(P < 0.05) concentration of tumor necrosis factor-α(TNF-α) at 3 and 7 d post-challenge,and higher(P < 0.05)concentration of interleukin-4(IL-4) at 14 d post-challenge than those in the PC treatment group. The concentration of serum endotoxin in CB birds was lower(P<0.05) at 21 d post-challenge,and the concentrations of serum diamine oxidase in CB birds were lower(P < 0.05) at 14 and 21 d post-challenge than in PC birds.Birds in CB treatment group had higher(P< 0.05) jejunum villi height than those in PC,NC,or CS treatment at 7,14,and 21 d post-challenge.In comparison to PC birds, the CB birds had lower(P < 0.05) jejunum crypt depth during the whole experiment. The birds in CB or CS treatment group had higher(P < 0.05) activities of amylase and protease at 3,7,and 14 d post-challenge,and higher(P< 0.05) activity of lipase at 3,7 d post-challenge than PC birds. Conclusions:In all, these results indicate that dietary supplementation with C.butyricum promotes immune response, improves intestinal barrier function, and digestive enzyme activities in broiler chickens challenged with E.coli K88.There is no significant difference between the C.butyricum probiotic treatment and the colistin sulfate antibiotic treatment. Therefore, the C.butyricum probiotic may be an alternative to antibiotic for broiler chickens. | Ling Zhang Lingling Zhang Xiu'an Zhan Xinfu Zeng Lin Zhou Guangtian Cao An'guo Chen Caimei Yang | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 43
Effects of thymol and carvacrol supplementation on intestinal integrity and immune responses of broiler chickens challenged with Clostridium perfringens显示文摘Background: Necrotic enteritis caused by Clostridium perfringens infection leads to serious economic losses in the global poultry production. In the present study, we investigated the protective effects of essential oils(EO, which contained 25 % thymol and 25 % carvacrol as active components) supplementation on growth performance, gut lesions, intestinal morphology, and immune responses of the broiler chickens infected with C. perfringens. A total of448 1-day-old male broiler chicks were allocated into eight treatment groups following a 4 × 2 factorial arrangement with four dietary EO dosages(0, 60, 120, or 240 mg/kg) and two infection status(with or without C. perfringens challenge from d 14 to 20).Results: The challenge did not impair the growth performance of birds, but induced gut lesions and increased crypt depth in the ileum(P ≤ 0.05). It also down-regulated the claudin-1 and occludin m RNA expression(P ≤ 0.05),up-regulated the m RNA expression of interleukin-1β(P ≤ 0.05), tended to increase the toll-like receptor(TLR)2 m RNA expression(P < 0.10) in the ileum, and enhanced the mucosal secretory Ig A production(P ≤ 0.05). In the challenged birds, dietary EO supplementation linearly alleviated the gut lesions and improved the ratio of vil us height to crypt depth(P ≤ 0.05), and the supplementation of 120 and 240 mg/kg EO increased the serum antibody titers against Newcastle disease virus(P ≤ 0.05). Regardless of challenge, the EO supplementation showed a tendency to linearly elevate the feed conversion efficiency between 14 and 28 d of age as well as the occludin m RNA expression(P < 0.10), and linearly inhibited the m RNA expression of TLR2 and tumor necrotic factor-α in the ileum(P ≤ 0.05).Conclusions: The dietary supplementation of EO could alleviate the intestinal injury by improving intestinal integrity and modulating immune responses in the C. perfringens-challenged broiler chickens. | Encun Du Weiwei Wang Liping Gan Zhui Li Shuangshuang Guo Yuming Guo | 2016 | Journal of Animal Science and Biotechnology2016,7,4: | 43
Solid-state fermentation of corn-soybean meal mixed feed with Bacillus subtilis and Enterococcus faecium for degrading antinutritional factors and enhancing nutritional value显示文摘Background: Corn and soybean meal(SBM) are two of the most common feed ingredients used in pig feeds. However, a variety of antinutritional factors(ANFs) present in corn and SBM can interfere with the bioavailability of nutrients and have negative health effects on the pigs. In the present study, two-stage fermentation using Bacillus subtilis followed by Enterococcus faecium was carried out to degrade ANFs and improve the nutritional quality of corn and SBM mixed feed. Furthermore, the microbial composition and in vitro nutrient digestibility of inoculated mixed feed were determined and compared those of the uninoculated controls. Results: During the fermentation process, B. subtilis and lactic acid bacteria(LAB) were the main dominant bacteria in the solid-state fermented inoculated feed, and fermentation produced a large amount of lactic acid(170 mmo L/kg),which resulted in a lower pH(5.0 vs. 6.4) than the fermented uninoculated feed. The amounts of soybean antigenic proteins(β-conglycinin and glycinin) in mixed feed were significantly decreased after first-stage fermentation with B. subtilis. Inoculated mixed feed following two-stage fermentation contained greater concentratioin of crude protein(CP), ash and total phosphorus(P) compared to uninoculated feed, whereas the concentrations of neutral detergent fiber(NDF), hemicellulose and phytate P in fermendted inoculated feed declined(P < 0.05) by 38%, 53%, and 46%,respectively. Notably, the content of trichloroacetic acid soluble protein(TCA-SP), particularly that of small peptides and free amino acids(AA), increased 6.5 fold following two-stage fermentation. There was no difference in the total AA content between fermented inoculated and uninoculated feed. However, aromatic AAs(Phe and Tyr) and Lys in inoculated feed increased, and some polar AAs, including Arg, Asp, and Glu, decreased compared with the uninoculated feed. In vitro dry matter and CP digestibility of inoculated feed improved(P < 0.05) compared with the uninoculated feed. Conclusions: Our results suggest that two-stage fermentation using B. subtilis followed by E. faecium is an effective approach to improve the quality of corn-soybean meal mixed feed. | Changyou Shi Yu Zhang Zeqing Lu Yizhen Wang | 2017 | Journal of Animal Science and Biotechnology2017,8,4: | 44
Effect of chlorogenic acid on antioxidant activity of Flos Lonicerae extracts显示文摘Flos Lonicerae is a medically useful traditional Chinese medicine herb. However, little is known about the antioxidant properties of Flos Lonicerae extracts. Here the antioxidant capacity of water, methanolic and ethanolic extracts prepared from Flos Lonicerae to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and reduce Fe3+ to Fe2+ is examined. Chlorogenic acid, a major component of Flos Lonicerae, is identified and further purified from 70% ethanolic extract with high performance liquid chromatography (HPLC) and its antioxidant capacity is characterized. The total phenolic compounds and chlorogenic acid con-tents in Flos Lonicerae are determined. The present results demonstrate that the Flos Lonicerae extracts exhibit antioxidant ac-tivity and chlorogenic acid is a major contributor to this activity. | WU Lan | 2007 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2007,8,9: | 36
Differential response of root morphology to potassium deficient stress among rice genotypes varying in potassium efficiency显示文摘Disparity in the root morphology of six rice(Oryza sativa L.) genotypes varying in potassium(K) efficiency was studied with three K levels:5 mg/L(low),10 mg/L(moderate) and 40 mg/L(adequate) in hydroponic culture. Morphological parameters included root length, surface area, volume and count of lateral roots,as well as fine(diameter<0.2 mm) and thick(diameter>0.2 mm) roots. The results indicate that the root growth of all genotypes was reduced under low K,but moderate K deficiency increased the root length of the efficient genotypes. At deficient and moderate K levels, all the efficient rice genotypes developed more fine roots(diameter<0.2 mm) than the inefficient ones. Both fine root count and root surface area were found to be the best parameters to portray K stress in rice. In accordance with the root morphology, higher K concentrations were noted in shoots of the efficient genotypes when grown at moderate and deficient K levels, indicating that root morphology parameters are involved in root uptake for K and in the translocation of K up to shoots. K deficiency affected not only the root morphology, but also the root ultra-structure. The roots of high-efficient genotypes had stronger tolerance to K deficient stress for root membrane damage, and could maintain the developed root architecture to adapt to the low K growth medium. | Yan-bo JIA Xiao-e YANG Ying FENG Ghulam JILANI | 2008 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,5: | 36
Sevoflurane postconditioning reduces myocardial reperfusion injury in rat isolated hearts via activation of PI3K/Akt signaling and modulation of Bcl-2 family proteins显示文摘Sevoflurane postconditioning reduces myocardial infarct size. The objective of this study was to examine the role of the phosphatidylinositol-3-kinase(PI3K)/Akt pathway in anesthetic postconditioning and to determine whether PI3K/Akt signaling modulates the expression of pro-and antiapoptotic proteins in sevoflurane postconditioning.Isolated and perfused rat hearts were prepared first, and then randomly assigned to the following groups:Sham-operation(Sham),ischemia/reperfusion(Con),sevoflurane postconditioning(SPC),Sham plus 100 nmol/L wortmannin(Sham+Wort),Con+Wort,SPC+Wort, and Con+dimethylsulphoxide(DMSO).Sevoflurane postconditioning was induced by administration of sevoflurane(2.5%,v/v) for 10 min from the onset of reperfusion.Left ventricular developed pressure(LVDP),left ventricular end-diastolic pressure(LVEDP),maximum increase in rate of LVDP(+dP/dt),maximum decrease in rate of LVDP(?dP/dt),heart rate(HR),and coronary flow(CF) were measured at baseline,R30 min(30 min of reperfusion),R60 min,R90 min,and R120 min.Creatine kinase(CK) and lactate dehydrogenase(LDH) were measured after 5 min and 10 min reperfusion.Infarct size was determined by triphenyltetrazolium chloride staining at the end of reperfusion.Total Akt and phosphorylated Akt(phospho-Akt),Bax,Bcl-2,Bad,and phospho-Bad were determined by Western blot analysis.Analysis of variance(ANOVA) and Student-Newman-Keuls' test were used to investigate the significance of differences between groups. The LVDP,±dP/dt,and CF were higher and LVEDP was lower in the SPC group than in the Con group at all points of reperfusion(P<0.05).The SPC group had significantly reduced CK and LDH release and decreased infarct size compared with the Con group [(22.9±8)% vs.(42.4±9.4)%,respectively;P<0.05].The SPC group also had increased the expression of phospho-Akt,Bcl-2,and phospho-Bad,and decreased the expression of Bax.Wortmannin abolished the cardioprotection of sevoflurane postconditioning.Sevoflurane postconditioning may protect the isolated rat heart.Activation of PI3K and modulation of the expression of pro-and antiapoptotic proteins may play an important role in sevoflurane-induced myocardial protection. | Li-na YU Jing YU Feng-jiang ZHANG Mei-juan YANG Ting-ting DING Jun-kuan WANG Wei HE Tao FANG Gang CHEN Min YAN | 2010 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,9: | 36
