2Hydrogen Sulfide Promotes Wheat Seed Germination and Alleviates Oxidative Damage against Copper Stress显示文摘With the enhancement of copper(Cu) stress,the germination percentage of wheat seeds decreased gradually. Pretreatment with sodium hydrosulfide(NaHS),hydrogen sulfide(H2S) donor alleviated the inhibitory effect of Cu stress in a dose-dependent manner; whereas little visible symptom was observed in germinating seeds and radicle tips cultured in NaHS solutions. It was verified that H2S or HS-rather than other sulfur-containing components derived from NaHS attribute to the potential role in promoting seed germination against Cu stress. Further studies showed that NaHS could promote amylase and esterase activities,reduce Cu-induced disturbance of plasma membrane integrity in the radicle tips,and sustain lower levels of malondialdehyde and H2O2 in germinating seeds. Furthermore,NaHS pretreatment increased activities of superoxide dismutase and catalase and decreased that of lipoxygenase,but showed no significant effect on ascorbate peroxidase. Alternatively,NaHS prevented uptake of Cu and promoted the accumulation of free amino acids in seeds exposed to Cu. In addition,a rapid accumulation of endogenous H2S in seeds was observed at the early stage of germination,and higher level of H2S in NaHS-pretreated seeds. 4Epidemiology of ovarian cancer:a review显示文摘Ovarian cancer(OC) is the seventh most commonly diagnosed cancer among women in the world and the tenth most common in China. Epithelial OC is the most predominant pathologic subtype, with five major histotypes that differ in origination,pathogenesis, molecular alterations, risk factors, and prognosis. Genetic susceptibility is manifested by rare inherited mutations with high to moderate penetrance. Genome-wide association studies have additionally identified 29 common susceptibility alleles for OC, including 14 subtype-specific alleles. Several reproductive and hormonal factors may lower risk, including parity, oral contraceptive use, and lactation, while others such as older age at menopause and hormone replacement therapy confer increased risks. These associations differ by histotype, especially for mucinous OC, likely reflecting differences in etiology. Endometrioid and clear cell OC share a similar, unique pattern of associations with increased risks among women with endometriosis and decreased risks associated with tubal ligation. OC risks associated with other gynecological conditions and procedures, such as hysterectomy,pelvic inflammatory disease, and polycystic ovarian syndrome, are less clear. Other possible risk factors include environmental and lifestyle factors such as asbestos and talc powder exposures, and cigarette smoking. 5WRKY transcription factors in plant responses to stresses显示文摘The WRKY gene family is among the largest families of transcription factors(TFs) in higher plants.By regulating the plant hormone signal transduction pathway,these TFs play critical roles in some plant processes in response to biotic and abiotic stress.Various bodies of research have demonstrated the important biological functions of WRKY TFs in plant response to different kinds of biotic and abiotic stresses and working mechanisms.However,very little summarization has been done to review their research progress.Not just important TFs function in plant response to biotic and abiotic stresses,WRKY also participates in carbohydrate synthesis,senescence,development,and secondary metabolites synthesis.WRKY proteins can bind to W-box(TGACC(A/T)) in the promoter of its target genes and activate or repress the expression of downstream genes to regulate their stress response.Moreover,WRKY proteins can interact with other TFs to regulate plant defensive responses.In the present review,we focus on the structural characteristics of WRKY TFs and the research progress on their functions in plant responses to a variety of stresses.Jingjing Jiang Shenghui Ma Nenghui Ye Ming Jiang Jiashu Cao Jianhua Zhang 2017Journal of Integrative Plant Biology2017,59,2:75
6Epidemiology,etiology,and prevention of esophageal squamous cell carcinoma in China显示文摘Esophageal cancer is one of the most fatal diseases worldwide mainly because of its rapid progression and poor prognosis.Although the incidence of esophageal adenocarcinoma has markedly risen in North America and Europe in the past several decades, esophageal squamous cell carcinoma is still the predominant subtype of esophageal cancer, especially in China. It accounts for more than 90% of all esophageal squamous cell carcinoma cases in China. Geographical differentiation is one of the most distinctive characteristics of esophageal cancer. The progression, risk factors, and prognosis of these two subtypes of esophageal cancer differ. 7Unraveling salt stress signaling in plants显示文摘Salt stress is a major environmental factor limiting plant growth and productivity. A better understanding of the mechanisms mediating salt resistance will help researchers design ways to improve crop performance under adverse environmental conditions. Salt stress can lead to ionic stress, osmotic stress and secondary stresses, particularly oxidative stress, in plants. Therefore,to adapt to salt stress, plants rely on signals and pathways that re-establish cellular ionic, osmotic, and reactive oxygen species(ROS) homeostasis. Over the past two decades, genetic and biochemical analyses have revealed several core stress signaling pathways that participate in salt resistance. The Salt Overly Sensitive signaling pathway plays a key role in maintaining ionic homeostasis,via extruding sodium ions into the apoplast. Mitogenactivated protein kinase cascades mediate ionic, osmotic,and ROS homeostasis. SnR K2(sucrose nonfermenting1-related protein kinase 2) proteins are involved in maintaining osmotic homeostasis. In this review, we discuss recent progress in identifying the components and pathways involved in the plant's response to salt stress and their regulatory mechanisms. 8Cancer burden in China:trends,risk factors and prevention显示文摘As the most populous country in the world,China has made strides in health promotion in the past few decades.With the aging population,the burden of cancer in China continues to grow.Changes in risk factors for cancer,especially diet,obesity,diabetes,and air pollution,continue to fuel the shift of cancer transition in China.The burden of upper gastrointestinal cancer in China is decreasing,but still heavy.The rising burden of colorectal,prostate,and breast cancers is also significant.Lung cancer became the top cause of cancer-related deaths,together with smoking as the most important contributor to cancer deaths.The Chinese government has taken several approaches to control cancer and cancer-related risk factors.Many achievements have been made,but some challenges remain.Health China 2030 is ambitious and depicts a bright vision of the future for cancer control in China.The decrease in the cancer burden in China will require cross-sector collaboration and coordinated efforts on primary and secondary preventions by governments,public health organizations,and individuals.In this review,we describe the trends of cancer burden and discuss cancer-related risk factors in China,identifying strategies to reduce the burden of cancer in China.Dianqin Sun He Li Maomao Cao Siyi He Lin Lei Ji Peng Wanqing Chen 2020Cancer Biology & Medicine2020,17,4:62
9Volatile Organic Compounds and Their Roles in Bacteriostasis in Five Conifer Species显示文摘In order to make clear the functions of plant volatile organic compounds (VOCs) on bacteriostasis and air decontamination, we analyzed the composition and content of VOCs in Pinus tabulaeformis Carr., P. bungeana Zucc., Sabina chinensis Antoine, Picea koraiensis Nakai, and Cedrus deodara G. Don under near-natural conditions using the thermal-desorption cold trap gas chromatography/mass spectrometer technique. The effects of the VOCs on airborne microorganisms were investigated using the method of natural sedimentation. Results showed that the major VOCs were as follows: limonene, β-pinene, α-pinene,and α-caryophyllene in Pinus tabulaeformis and P. bungeana; limonene, borneol acetate, β-pinene, myrcene,and tricylene in S. chinensis; limonene, α-pinene, myrcene, camphene, and β-pinene in Picea koraiensis;and limonene, 2, (10)-pinene, α-pinene, and myrcene in C. deodara. These VOCs and the corresponding foliar extracts inhibited the growth of bacteria and stimulated the growth of fungi. Experimental data using monomers of the VOCs demonstrated that limonene, β-pinene, and three aldehydes could significantly inhibit bacterial growth, suggesting an inhibitory effect of VOCs on the growth of airborne microorganisms in the five conifer species. 10Promotion by 5-Aminolevulinic Acid of Germination of Pakchoi (Brassica campestris ssp. chinensis var. communis Tsen et Lee) Seeds Under Salt Stress显示文摘The seed germination and seedling growth of pakchoi (Brassica campestris ssp. chinensis var.communis Tsen et Lee cv. Hanxiao) were not significantly inhibited until the concentration of NaCl was increased to150 mmol/L. Treatment of pakchoi seeds with exogenous 5-aminolevulinic acid (ALA), at concentrations ranging from 0.01 to 10.00 mg/L, promoted seed germination when seeds were stressed by salinity, whereas levulinic acid (LA), an inhibitor of ALA dehydrase, significantly inhibited seed germination and seedling growth, suggesting that metabolism of ALA into porphyrin compounds was necessary for seed germination and seedling growth. Determination of respiratory rate during seed germination showed that ALA increased seed respiration under both normal conditions and salt stress. Furthermore, salt stress decreased levels of endogenous ALA, as well as heme, in etiolated seedlings. 12Hydrogen Peroxide in Plants:a Versatile Molecule of the Reactive Oxygen Species Network显示文摘Plants often face the challenge of severe environmental conditions,which include various biotic and abiotic stresses that exert adverse effects on plant growth and development. During evolution,plants have evolved complex regulatory mechanisms to adapt to various environmental stressors. One of the consequences of stress is an increase in the cellular concentration of reactive oxygen species(ROS),which are subsequently converted to hydrogen peroxide(H2O2). Even under normal conditions,higher plants produce ROS during metabolic processes. Excess concentrations of ROS result in oxidative damage to or the apoptotic death of cells. Development of an antioxidant defense system in plants protects them against oxidative stress damage. These ROS and,more particularly,H2O2,play versatile roles in normal plant physiological processes and in resistance to stresses. Recently,H2O2 has been regarded as a signaling molecule and regulator of the expression of some genes in cells. 13How Plants Cope with Cadmium: Staking All on Metabolism and Gene Expression显示文摘Environmental pollution is one of the major problems for human health. Toxic heavy metals are normally present as soil constituents or can also be spread out in the environment by human activity and agricultural techniques. Soil contamination by heavy metals as cadmium, highlights two main aspects: on one side they interfere with the life cycle of plants and therefore reduce crop yields, and on the other hand, once adsorbed and accumulated into the plant tissues, they enter the food chain poisoning animals and humans. Considering this point of view, understanding the mechanism by which plants handle heavy metal exposure, in particular cadmium stress, is a primary goal of plant-biotechnology research or plant breeders whose aim is to create plants that are able to recover high amounts of heavy metals, which can be used for phytoremediation, or identify crop varieties that do not accumulate toxic metal in grains or fruits. In this review we focus on the main symptoms of cadmium toxicity both on root apparatus and shoots. We elucidate the mechanisms that plants activate to prevent absorption or to detoxify toxic metal ions, such as synthesis of phytochelatins, metallothioneins and enzymes involved in stress response. 14Hydrogen Sulfide Promotes Root Organogenesis in Ipomoea batatas, Salix matsudana and Glycine max显示文摘In this report, we demonstrate that sodium hydrosulfide (NaHS), a hydrogen sulfide (H2S) donor, promoted adventitious root formation mediated by auxin and nitric oxide (NO). Application of the H2S donor to seedling cuttings of sweet potato (Ipomoea batatas L.) promoted the number and length of adventitious roots in a dose-dependent manner. It was also verified that H2S or HS- rather than other sulfur-containing components derived from NaHS could be attributed to the stimulation of adventitious root formation. A rapid increase in endogenous H2S, indole acetic acid (IAA) and NO were sequentially observed in shoot tips of sweet potato seedlings treated with HaHS. Further investigation showed that H2S-mediated root formation was alleviated by N-1-naphthylphthalamic acid (NPA), an IAA transport inhibitor, and 2-(4-carboxyphenyl)- 4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO), an NO scavenger. Similar phenomena in H2S donor-dependent root organogenesis were observed in both excised willow (Salix matsudana var. tortuosa Vilm) shoots and soybean (Glycine max L.) seedlings. 16Comparative study of CEA and CA19-9 in esophageal,gastric and colon cancers individually and in combination(ROC curve analysis)显示文摘Objective:To determine the clinical serum levels of carcinoembryonic antigen(CEA) and carbohydrate antigen 19-9(CA19-9),individually and in combination,for the diagnosis of 50 healthy subjects and 150 cases of esophageal,gastric,and colon cancers.Methods:The sensitivities of the two markers were compared individually and in combination,with specificity set at 100%.Receiver operating characteristic(ROC) curves were plotted.Results:Serum CEA levels were significantly higher in cancer patients than in the control group.The sensitivity of CEA was determined:in esophageal cancer,sensitivity=28%,negative predictive value(NPV)=61.72%,and AUC=0.742(SE=0.05),with a significance level of P<0.0001;in gastric cancer,sensitivity=30%,NPV=58.82%,and AUC=0.734(SE=0.05),with a significance level of P<0.0001;in colon cancer,sensitivity=74%,NPV=79.36%,and AUC=0.856(SE=0.04),with a significance level of P<0.0001.The sensitivity of CA19-9 was also evaluated:in esophageal cancer,sensitivity=18%,NPV=54.94%,and AUC=0.573(SE=0.05),with a significance level of P=0.2054.In gastric cancer,sensitivity=42%,NPV=63.29%,and AUC=0.679(SE=0.05),with a significance level of P<0.0011.In colon cancer,sensitivity=26%,NPV=57.47%,and AUC=0.580(SE=0.05),with a significance level of P=0.1670.The following were the sensitivities of CEA/CA19-9 combined:in esophageal cancer,sensitivity=42%,NPV=63.29%,SE=0.078(95% CI:0.0159-0.322);gastric cancer,sensitivity=58%,NPV=70.42%,SE=0.072(95% CI:-0.0866-0.198);and colon cancer,sensitivity=72%,NPV=78.12%,SE=0.070(95% CI:0.137-0.415).Conclusion:CEA exhibited the highest sensitivity for colon cancer,and CA19-9 exhibited the highest sensitivity for gastric cancer.Combined analysis indicated an increase in diagnostic sensitivity in esophageal and gastric cancer compared with that in colon cancer.Bhawna Bagaria Sadhna Sood Rameshwaram Sharma Soniya Lalwani 2013Cancer Biology & Medicine2013,10,3:53
17HOTAIR:an oncogenic long non-coding RNA in different cancers显示文摘Long non-coding RNAs(lncRNAs) refer to a group of RNAs that are usually more than 200 nucleotides and are not involved in protein generation. Instead, lncRNAs are involved in different regulatory processes, such as regulation of gene expression. Different lncRNAs exist throughout the genome. Lnc RNAs are also known for their roles in different human diseases such as cancer. HOTAIR is an lnc RNA that plays a role as an oncogenic molecule in different cancer cells, such as breast, gastric, colorectal, and cervical cancer cells. Therefore, HOTAIR expression level is a potential biomarker for diagnostic and therapeutic purposes in several cancers. This RNA takes part in epigenetic regulation of genes and plays an important role in different cellular pathways by interacting with Polycomb Repressive Complex 2(PRC2). 18Risk factors for cervical lymph node metastasis in papillary thyroid microcarcinoma: a study of 1,587 patients显示文摘Objective: The purposes of this study were to identify risk factors for cervical lymph node metastasis and to examine the association between BRAF^(V600E) status and clinical features in papillary thyroid microcarcinoma(PTMC).Methods: A total of 1,587 patients with PTMC, treated in Tianjin Medical University Cancer Institute and Hospital from January2011 to March 2013, underwent retrospective analysis. We reviewed and analyzed factors including clinical results, pathology records, ultrasound results, and BRAF^(V600E) status.Results: Multivariate logistic regression analyses demonstrated that gender(male) [odds ratio(OR) = 1.845, P = 0.000], age(< 45 years)(OR = 1.606, P = 0.000), tumor size(> 6 mm)(OR = 2.137, P = 0.000), bilateralism(OR = 2.011, P = 0.000) and extrathyroidal extension(OR = 1.555, P = 0.001) served as independent predictors of central lymph node metastasis(CLNM).Moreover, CLNM(OR = 29.354, P = 0.000) served as an independent predictor of lateral lymph node metastasis(LLNM). Among patients with a solitary primary tumor, those with tumor location in the lower third of the thyroid lobe or the isthmus were more likely to experience CLNM(P < 0.05). Univariate analyses indicated that CLNM, LLNM, extrathyroidal extension, and multifocality were not significantly associated with BRAF^(V600E) mutation.Conclusions: The present study suggested that prophylactic neck dissection of the central compartment should be considered in patients with PTMC, particularly in men with tumor size greater than 6 mm, age less than 45 years, extrathyroidal extension, and tumor bilaterality. Among patients with PTMC, BRAF^(V600E) mutation is not significantly associated with prognostic factors. 19Reactive capillary hemangiomas: a novel dermatologic toxicity following anti-PD-1 treatment with SHR-1210显示文摘Objective: SHR-1210 is a new and promising anti-PD-1 agent for solid tumors. During the phase I study of SHR-1210, we encountered a novel but prevalent immune-related dermatologic toxicity: reactive capillary hemangiomas(RCHs). Thus we tried to summarize the features of RCHs and estimate their relationship with tumor response.Methods: This prospective observational study systematically enrolled 98 patients with advanced solid tumors from April 27th,2016 to June 8th, 2017 in the context of the phase I clinical study of SHR-1210. This report focused on the skin toxicities. Patients underwent entire skin inspection every two weeks while taking medication. The clinical course of RCHs was recorded and their association with tumor response was estimated. The data cut-off date was November 15th, 2017.Results: After a median follow-up of 242(range, 29–567) days, RCHs were observed in 85.7%(84/98) of patients on cutaneous/mucosal surfaces; 84.5%(71/84) of the RCHs were evaluated as grade 1 adverse events. No grade 3 or 4 RCHs were observed. The time of onset of RCHs was dose dependent and shortest in the 400 mg-dose cohort(P < 0.001). Spontaneous and complete regression of RCHs was observed both during and after treatment. The objective response rate of tumors for patients with RCHs was 28.9%(24/83). However, no responders were observed among the patients without RCHs.Conclusions: RCHs were prevalent but manageable during treatment with SHR-1210. 20Biological and clinical aspects of HPV-related cancers显示文摘Cancer-related diseases represent the second overall cause of death worldwide.Human papilloma virus(HPV)is an infectious agent which is mainly sexually transmitted and may lead to HPV-associated cancers in both men and women.Almost all cervical cancers are HPV-associated,however,an increasing number of head and neck cancers(HNCs),especially oropharyngeal cancer,can be linked to HPV infection.Moreover,anogenital cancers,including vaginal,vulvar,penial,and anal cancers,represent a subset of HPVrelated cancers.Whereas testing and prevention of cervical cancer have significantly improved over past decades,anogenital cancers remain more difficult to confirm.Current clinical trials including patients with HPV-related cancers focus on finding proper testing for all HPV-associated cancers as well as improve the currently applied treatments.The HPV viral oncoproteins,E6 and E7,lead to degradation of,respectively,p53 and pRb resulting in entering the S phase without G1 arrest.These high-risk HPV viral oncogenes alter numerous cellular processes,including DNA repair,angiogenesis,and/or apoptosis,which eventually result in carcinogenesis.Additionally,a comprehensive analysis of gene expression and alteration among a panel of DNA double strand breaks(DSB)repair genes in HPV-negative and HPV-positive HNC cancers reveals differences pointing to HPV-dependent modifications of DNA repair processes in these cancers.In this review,we discuss the current knowledge regarding HPV-related cancers,current screening,and treatment options as well as DNA damage response-related biological aspects of the HPV infection and clinical trials.Klaudia Anna Szymonowicz Junjie Chen 2020Cancer Biology & Medicine2020,17,4:54
