| 7 | Antimicrobial activity of Cannabis sativa,Thuja orientalis and Psidium guajava leaf extracts against methicillin-resistant Staphylococcus aureus显示文摘Objective: This study examined the antimicrobial activity of Cannabis sativa, Thuja orientalis and Psidium guajava against methicillin-resistant Staphylococcus aureus(MRSA) and used a standardized purification protocol to determine the presence and abundance of bioactive compounds in the leaf extracts.Methods: In vitro antimicrobial activities of the ethanolic extracts of C. sativa, T. orientalis and P. guajava were tested against MRSA. The presence of bioactive molecules in these three leaves was evaluated using biochemical assays and high-performance thin-layer chromatography(HPTLC).Results: Resistance to methicillin, penicillin, oxacillin and cefoxitin was observed in each of the clinical and nonclinical MRSA isolates. However, they were still vulnerable to vancomycin. Used individually,the 50% extract of each plant leaf inhibited MRSA growth. A profound synergism was observed when C. sativa was used in combination with T. orientalis(1:1) and when P. guajava was used in combination with T. orientalis(1:1). This was shown by larger zones of inhibition. This synergism was probably due to the combined inhibitory effect of phenolics present in the leaf extracts(i.e., quercetin and gallic acid)and catechin, as detected by HPTLC.Conclusion: The leaf extracts of C. sativa, T. orientalis and P. guajava had potential for the control of both hospital-and community-acquired MRSA. | 8 | Chromatographic finger print analysis of steroids in Aerva lanata L by HPTLC technique显示文摘Objective:To determine the chemical profile and steroids composition of the medicinally important plant Aerva lanata(A.lanata) L.Methods:Preliminary phytochemicul screening was done by the method as Harborne described.HPTLC studies were canied out as Harborne and Wagner et al described.The Ethyl acetate-ethanol-water(8:2:1.2) was employed as mobile phase for glycosides.Results:The desired aim was achieved using Chloroform-acetone(8:2) as the mobile phase.The methanolic extract of stem,leaves,root,flower and seeds of A.lanata showed the presence of 30 different types of steroids with 30 different Rf values from 0.04 to 0.97. | 13 | Preparation of κ-carra-Oligosaccharides with Microwave Assisted Acid Hydrolysis Method显示文摘A rapid method of microwave assisted acid hydrolysis was established to prepare κ-carra-oligosaccharides. The optimal hydrolysis condition was determined by an orthogonal test. The degree of polymerization(DP) of oligosaccharides was detected by high performance thin layer chromatography(HPTLC) and polyacrylamide gel electrophoresis(PAGE). Considering the results of HPTLC and PAGE, the optimum condition of microwave assisted acid hydrolysis was determined. The concentration of κ-carrageenan was 5 mg m L-1; the reaction solution was adjusted to p H 3 with diluted hydrochloric acid; the solution was hydrolyzed under microwave irradiation at 100 for 15℃ min. Oligosaccharides were separated by a Superdex 30 column(2.6 cm × 90 cm) using AKTA Purifier UPC100 and detected with an online refractive index detector. Each fraction was characterized by electrospray ionization mass spectrometry(ESI-MS). The data showed that odd-numbered κ-carra-oligosaccharides with DP ranging from 3 to 21 could be obtained with this method, and the structures of the oligosaccharides were consistent with those obtained by traditional mild acid hydrolysis. The new method was more convenient, efficient and environment-friendly than traditional mild acid hydrolysis. | 20 | Morpho-anatomical and physicochemical studies of Fumaria indica(Hausskn.)Pugsley显示文摘Objective:To study morpho-anatomical characters and physicochemical analysis of Fumaria indica(F.indica)(Hausskn.)Pugsley,(Fumariaceae),an important medicinal plant used extensively for treating a variety of ailments in various system of indigenous medicine.Methods:Evaluation of the different parts of the plant was carried out to determine the morphoanatomical,physicochemical,phytochemical and HPTLC fingerprinting profile of F.indica and other WHO recommended methods were performed for standardization.Results:Morphoanatomical studies showed compound and pinnatifid leaf,4 to 6 cm in length,linear and oblong in shape and anomocytic arrangement of stomata,thin walled parenchymatous cells,scattered,sclerenchymatous,capped vascular bundles and radiating medullary rays.Physicochemical studies showed foreign matter 0.2%,loss on drying 6.8%,total ash 16.77%,alcohol and water soluble extractives 8.92%and 20.26%,respectively,sugar 17.75%,starch 22.97%and tannins 2.37%.Phytochemical evaluation revealed the presence of carbohydrate,alkaloids,flavonoids,saponins,tannins and sterol.Thin layer chromatography was carried out with different solvents and the best solvent system was chloroform and methanol in 80:20 ratio and revealed 12 spots with different R_f value under UV light 366λ.Conclusions:The results of the study can serve as a valuable source of information and provide suitable standards for identification of this plant material for future investigations and applications. | Prakash Chandra Gupta Ch V Rao | 2012 | Asian Pacific Journal of Tropical Biomedicine2012,2,10: | 2 |
