| 1 | 中药多成分药代动力学:发现与中药安全性和有效性关联的物质并揭示其药代特征显示文摘中医药对中华民族健康和国家稳定发挥了重要作用,揭示决定中药有效性和安全性的物质是推进中药现代化的一项重要工作。对于化学组成复杂的中药,可通过开展多成分药代研究,根据给药后中药成分能否以某种形式被机体利用产生显著的体内暴露(以成分原形和/或代谢物形式),选拔出用于考察药效活性的中药物质,研究物质产生疗效的体内过程和药代特征,由此为揭示决定中药药效作用的物质创造条件。此外,这类多成分药代研究还可用于揭示与中药不良反应或联合用药风险关联的中药物质。经过十多年的努力,中药多成分药代研究在理论、方法、技术、应用上已取得突破,成为药代动力学的一个新分支。本文系统阐述了中药多成分药代动力学的研究方法、技术要求和分析技术,并用一类活性中药成分(三七的皂苷类成分)的研究和围绕一种已上市中药制剂(连花清瘟胶囊)的研究介绍了两类多成分药代研究实例,最后讨论了中药多成分药代研究的进一步发展。 | 李川 程晨 贾伟伟 杨军令 余玄 Olajide E.OLALEYE | 2021 | 药学学报2021,56,9: | 7 |
| 2 | 基于UHPLC-Q-Exactive Orbitrap MS技术分析不同干燥方法对人参皂苷类成分的影响显示文摘对鲜人参中的三萜皂苷类成分进行快速分析鉴定,并阐明鲜人参中不同干燥方法(50℃烘干、80℃烘干以及-70℃冷冻干燥)对其所含的三萜皂苷类成分的影响差异性。制备3种不同的人参干燥品,应用UHPLC-Q-Exactive Orbitrap高分辨液质联用技术,以水-乙腈作为流动相进行梯度洗脱,并利用X Calibur 2.2软件对在负离子模式下采集的数据进行分析。结果显示:新鲜人参中含有57种皂苷类成分;与新鲜人参相比,50℃烘干品中的人参皂苷类成分损失较少,-70℃冷冻干燥品次之,80℃烘干品损失最多。该研究阐明了不同干燥方法对人参皂苷类成分的种类和相对含量的影响,可为人参药材的产地加工提供借鉴。 | 王红 崔议方 姜慧祯 王少平 宋晓光 韩丽娟 代龙 李宁 张加余 | 2022 | 中国中药杂志2022,47,19: | 6 |
| 3 | 益气养阴法对重症监护室耐药菌肺炎患者临床疗效、病原菌清除率及炎症反应的影响显示文摘目的:探讨益气养阴法治疗重症监护室(ICU)耐药菌肺炎患者的临床疗效及对病原菌清除率、白细胞计数(WBC)、超敏C反应蛋白(hs-CRP)、降钙素原(PCT)的影响。方法:选择30例于2016年8月至2018年5月期间在安徽中医药大学第一附属医院住院的ICU耐药菌肺炎患者,按照随机数字表法分为观察组(n=16)和对照组(n=14),两组均常规给予常规抗感染治疗以及对症支持治疗,观察组在此基础上采用参麦注射液的益气养阴法。分别观察两组患者的临床疗效,对比两组病原菌清除率及治疗前后WBC、PCT、hs-CRP水平。结果:观察组临床疗效为93.75%(15/16),高于对照组的64.29%(9/14)(P<0.05)。观察组的病原菌清除率为68.75%(11/16),高于对照组的42.85%(6/14),但差异无统计学意义(P>0.05)。治疗前,两组WBC、PCT、hs-CRP水平比较无统计学差异(P>0.05),治疗后,观察组和对照组WBC、PCT、hs-CRP水平均较治疗前降低(P<0.05),与对照组相比,观察组治疗后WBC、PCT、hs-CRP水平更低(P<0.05)。结论:采用参麦注射液的益气养阴法可控制ICU耐药菌肺炎,临床疗效确切,提高抗菌药物的杀菌效果,减轻肺部炎症反应。 | 杨明 舒婵娟 俞兴群 高志凌 聂卫群 | 2020 | 现代生物医学进展2020,20,4: | 5 |
| 4 | Shenmai injection enhances cisplatin-induced apoptosis through regulation of Mfn2-dependent mitochondrial dynamics in lung adenocarcinoma A549/DDP cells显示文摘Aim:Chemoresistance is the biggest obstacle in cancer treatment.Our previous study demonstrated that Shenmai injection(SMI),a Chinese herbal medicine,enhanced the antitumor effect of cisplatin via glucose metabolism reprogramming.This study aimed to further determine whether the SMI sensitizes the non-small cell lung cancer(NSCLC)cells to cisplatin through regulation mitochondrial dynamics.Methods:The Kaplan-Meier Plotter database was used to investigate the relationship between mRNA expression of mitofusin-2(Mfn2)and the survival analysis of NSCLC patients.The protein expression of Mfn2 in a lung adenocarcinoma tissue chip was detected by immunohistochemistry staining.The expression of Mfn2 and ATAD3A were compared between cisplatin-sensitive A549 and cisplatin-resistant A549/DDP cells.Additionally,A549/DDP cells were co-treated with cisplatin and SMI to detect mitochondrial morphology by fluorescent staining,apoptosis-related protein expression with Western blotting,and mitochondrial membrane potential(ΔΨm)with flow cytometry analysis.Results:The mean survival time of the Mfn2^(low) group was significantly lower than that of the Mfn2^(high) group by Kaplan-Meier Plotter database analysis,and the Mfn2 protein expression level was lower in cancer tissues than in adjacent tissues.The combination of SMI and cisplatin induced dynamic changes in A549/DDP cells,with increased mitochondrial fusion followed by upregulation of Mfn2 and downregulation of ATAD3A and reduced mitochondrial mass and ΔΨm.Moreover,SMI significantly enhanced cisplatin-induced A549/DDP apoptosis,upregulated Bax and the active subunit of caspase-3,and downregulated Bcl-2 expression,as shown via Hoechst staining and flow cytometry analysis.Conclusion:Our findings suggest SMI enhances cisplatin-induced apoptosis through regulation of Mfn2-dependent mitochondrial dynamics in cisplatin-resistant lung adenocarcinoma cells. | Yushi Chen Ye Sun Qiuyu Zhao Chunying Liu Chun Wang | 2021 | Cancer Drug Resistance2021,4,4: | 2 |
