| 1 | Simultaneous determination of 15 flavonoids from different parts of Scutellaria baicalensis and its chemometrics analysis显示文摘Objective: The aerial parts of Scutellaria baicalensis were used as Huangqin Tea for thousands of years and mainly contain flavonoids which contribute to its bioactivities. However, there is no appropriate quality evaluation method of Huangqin Tea, and three flavanones of isocarthamidin-7-O-β-D-glucuronide,carthamidin-7-O-β-D-glucuronide, and isoscutellarein-8-O-β-D-glucuronide with high contents in the aerial parts have never been defined quantitatively. Here, an HPLC-DAD method for simultaneous determination of 15 flavonoids and systematically compared their contents and distribution in the roots,stems, leaves, and flowers of S. baicalensis was established.Methods: Under the HPLC-DAD chromatographic conditions, 77 batches of samples of S. baicalensis were analyzed. Meanwhile, the chromatographic fingerprint of different parts of S. baicalensis was established.Subsequently, principal component analysis(PCA), orthogonal projections to latent structures discriminant analysis(OPLS-DA), and clustering heat map were performed based on the contents of 15 flavonoids in different parts of S. baicalensis.Results: The results showed significant differences in the contents and distributions of 15 flavonoids among the different parts of S. baicalensis. The chemical composition of stems showed some similarities to leaves, and their contents were all lower than leaves. The contents of isocarthamidin-7-O-β–glucuronide [(106.66 66 ± 22.68) mg/g], carthamidin-7-O-β-D-glucuronide [(19.82 ± 11.17) mg/g],and isoscutellarein-8-O-β-D-glucuronide [(3.10 ± 1.73) mg/g] were the highest in leaves. The content of apigenin-7-O-β-D-glucopyranoside and chrysin-7-O-β-D-glucuronide were the highest in flowers. The contents of baicalin, baicalein, wogonoside, wogonin, alpinetin, and oroxylin A were higher in roots than in other parts.Conclusion: The method was fully validated and could be effectively used to characterize the contents and distributions of main flavonoids in the different parts of S. baicalensis. | 2 | 雌激素受体介导的柚皮苷抗Aβ_(25-35)损伤PC12细胞凋亡作用研究显示文摘目的探讨雌激素受体介导的柚皮苷(NG)抗β淀粉样蛋白25-35(Aβ_(25-35))诱导大鼠肾上腺嗜铬细胞瘤(PC12)细胞的凋亡作用及其与雌激素受体(ER)信号通路的关系。方法实验分为空白组、Aβ_(25-35)组、E2+Aβ_(25-35)组、NG+Aβ_(25-35)组、ICI182780+E2+Aβ_(25-35)组、ICI182780+NG+Aβ_(25-35)组。实验采用Annexin V-FITC/PI双染法检测细胞凋亡率;蛋白免疫印迹法(WB)检测细胞Bax、Bcl-2和Caspase-3的表达;RT-qPCR法检测细胞凋亡因子mRNA的表达。结果Annexin V-FITC/PI双染色流式细胞术结果显示,与空白组相比,Aβ_(25-35)组细胞凋亡率升高(P<0.01);与Aβ_(25-35)组相比,NG+Aβ_(25-35)组细胞凋亡率降低(P<0.01);与NG+Aβ_(25-35)组相比,ICI182780+NG+Aβ_(25-35)组细胞凋亡率升高(P<0.01)。WB结果显示,与空白组相比,Aβ_(25-35)组细胞Bax和Caspase-3蛋白表达量升高(P<0.01),Bcl-2则相反(P<0.01);与Aβ_(25-35)组相比,E2+Aβ_(25-35)组Bax和Caspase-3蛋白表达量降低(P<0.01),Bcl-2则相反(P<0.01);雌激素受体介导的NG的作用与E2相似,而ICI182780逆转了NG的作用。RT-qPCR结果显示,NG对细胞Caspase-3、Bax和Bcl-2 mRNA表达的影响与蛋白结果一致。说明NG是通过激活ER信号通路发挥抗凋亡作用,作用与E2一致。结论雌激素受体介导的NG有明显的抗凋亡作用,通过提高Bcl-2的正常表达,降低Bax和Caspase-3的正常表达发挥抗凋亡作用,其抗凋亡作用可能经ER介导。 | 王媛 武凤 熊辉 徐艳明 雷霞 徐红丹 孙慧峰 张宁 杨波 | 2023 | 辽宁中医药大学学报2023,25,2: | 3 |
| 3 | Aβ_(25~35)对大鼠海马神经元及小胶质细胞的影响显示文摘目的探讨双侧海马注射Aβ25~35不同时间点对大鼠海马神经元及MG的影响。方法雄性Wistar大鼠随机分为对照组、模型组(注射Aβ后2、7、15、21 d 4个时间点),双侧海马注射Aβ建立大鼠AD模型。水迷宫法观察大鼠学习记忆能力;硫堇染色观察海马CA1区神经元和胶质细胞;Aβ、CD11b免疫组化染色分别观察Aβ沉积和活化MG。结果与对照组相比,模型组大鼠在第3~5天逃避潜伏期均显著延长,学习记忆能力下降(P<0.05);4个时间点模型组大鼠海马都有神经元缺失,胶质细胞增生,2 d、7 d、15 d组大鼠神经元损伤呈加重趋势,但21 d组神经元损伤较15 d组有减轻趋势(P<0.05);4个时间点模型组大鼠都有Aβ沉积和活化的MG,Aβ沉积量及MG活化自第7天后呈下降趋势(P<0.05)。结论 Aβ可以引起神经元损伤,降低大鼠的学习记忆能力;Aβ可以激活MG,引起MG形态和数量改变,MG可吞噬、清除Aβ;Aβ沉积量和MG活化数量变化呈正相关。 | 王爱霞 董雅洁 狄婷婷 申兴斌 王瑞婷 | 2014 | 中国老年学杂志2014,34,24: | 2 |
| 4 | 不同桃品种果肉与叶片总黄酮质量分数变化及相关性研究显示文摘明确不同桃树品种的果肉与叶片中总黄酮质量分数的差异及果实不同发育阶段果肉与叶片中总黄酮质量分数的动态变化及相关性,为科学评价不同品种桃树种质资源的保健育种前景及科学开发利用桃叶资源提供依据。本试验以3大类6个品种桃树果实发育时期的5个阶段的果肉与叶片为试材,采用乙醇超声辅助提取法提取、紫外分光光度计法测定总黄酮质量分数。结果表明:不同品种桃果肉与叶片中总黄酮质量分数差异显著,其中果肉中总黄酮质量分数5批次样品的平均值依次为美蟠777(11.26 mg·g-1)>连黄(10.60mg·g-1)>朝晖(7.58mg·g-1)>瑞蟠4号(6.25mg·g-1)>瑞光19(5.70mg·g-1)和秦光2号(5.08mg·g-1),桃叶中总黄酮质量分数依次为瑞蟠4号(32.18mg·g-1)>秦光2号(31.37mg·g-1)>朝晖(30.55mg·g-1)>瑞光19(28.44mg·g-1)>美蟠777(24.51mg·g-1)>连黄(21.28mg·g-1);6个试验品种中,连黄与美蟠777果肉的总黄酮质量分数较高,具有更好的保健成分育种价值;桃叶总黄酮质量分数在果实发育期均高于相对应的果肉,桃叶与果肉中总黄酮质量分数呈负相关。 | 杨慧珍 慕小倩 王安柱 | 2015 | 西北农业学报2015,24,4: | 1 |
| 5 | β淀粉样蛋白_(25~35)对PC12细胞凋亡相关蛋白表达的影响显示文摘目的探讨β淀粉样蛋白(Aβ)_(25~35)对PC12细胞凋亡相关蛋白表达的影响。方法采用不同浓度的Aβ_(25~35)与PC12细胞共孵育48 h后,用MTT法检测细胞生长活性,Hoechst33258核染色检测细胞凋亡,通过免疫细胞化学检测细胞凋亡相关蛋白Bcl-2、Bax、Caspase-3表达。结果MTT检测显示Aβ_(25~35)抑制PC12细胞生长活性呈浓度依赖性,抑制作用随Aβ_(25~35)浓度的增大而加强;Hoechst33258染色显示Aβ_(25~35)与PC12细胞凋亡呈浓度依赖性关系;免疫细胞化学检测结果显示Bcl-2蛋白表达量随Aβ_(25~35)浓度的增大呈下降趋势,Bax、Caspase-3蛋白表达量随Aβ_(25~35)浓度的增大呈上升趋势。结论 Aβ_(25~35)可剂量依赖性诱导PC12细胞凋亡,使抗凋亡蛋白Bcl-2表达下调,促凋亡蛋白Bax,Caspase-3表达上调。 | 王瑞婷 王爱霞 狄婷婷 张美 申兴斌 | 2017 | 中国老年学杂志2017,37,17: | 1 |
| 6 | 基于网络药理学和分子对接技术研究黄芩治疗糖尿病脑病的作用显示文摘目的采用网络药理学和分子对接技术研究黄芩治疗糖尿病脑病的作用机制。方法通过TCMSP数据库检索黄芩主要活性成分和作用靶点,通过OMIM、GeneCards、TTD及DrugBank等数据库筛选糖尿病和阿尔茨海默病的主要靶点,构建活性成分-靶点网络;利用STRING 11.0进行蛋白质互作分析,构建PPI网络以筛选重要靶点;运用Metascape库对关键交集靶点进行GO和KEGG富集分析;利用Cytoscape 3.7.2将分析得到的重要靶点和通路构建药物成分-疾病靶点-通路网络;进一步对关键活性成分和核心靶点进行分子对接验证。结果筛选出黄芩重要活性成分有汉黄芩素、黄芩素、金合欢素、降穿心莲黄酮等,PPI网络中关键交集靶点61个,核心靶点有AKT1、IL-6、MAPK14、TNF、PRKCA、VEGFA等。GO富集分析得到细胞组成101条,分子功能76条,生物过程1413条;KEGG通路富集分析得到258条,主要涉及AGE-RAGE信号介导的糖尿病并发症通路、IL-17、MAPK、TNF、PI3K-Akt信号通路、胰岛素抵抗通路等。分子对接结果可知,汉黄芩素、金合欢素、黄芩素、红花素、二氢奥洛昔林、黄芩黄酮Ⅱ、5,7,2,5-四羟基-8,6-二甲氧基黄酮、降穿心莲黄酮等活性成分与核心靶点VEGFA、AKT1、TNF、MAPK14、IL-6结合较好。结论黄芩可以通过多成分、多靶点、多通路改善糖尿病脑病。 | 高书春 杜卓琦 马世平 邓雪阳 | 2023 | 中成药2023,45,9: | 0 |
| 7 | 21世纪全球视域下的黄芩科研知识图谱构建及可视化分析显示文摘目的对21世纪以来国内外黄芩Scutellaria baicalensis相关研究进展进行可视化分析,梳理其研究前沿,分析发展方向,为今后黄芩相关研究提供科学依据。方法对Web of Science(WOS)数据库中的英文文献,中国知网、万方、维普中的中文文献进行检索并进行数据清洗。利用VOSviewer、Coocation等软件对发文量、地区/作者合作网络、关键词共现、主题演化路径进行可视化分析,并对黄芩相关的研究前沿热点进行分析。结果纳入英文文献1560篇、中文文献22214篇。黄芩相关领域发文量在2014年以前每年呈现上升趋势,之后每年发文量趋于稳定状态,我国在黄芩相关研究领域占主导地位。合作网络显示国际上中国与美国合作较为紧密,国内各地区合作较为散乱,协作程度较为薄弱。关键词分析表明未来黄芩的研究主要集中在代谢组学、化学成分、作用机制、抗肿瘤、阿尔茨海默症等方向。结论黄芩相关研究进展迅速,近年来对黄芩的研究逐步稳定,2019年后有回温现象。在后续的研究过程中应注重各地区、各机构之间的合作创新。代谢组学、化学成分、作用机制、抗肿瘤、阿尔茨海默症等方向成为近年来黄芩相关领域的研究热点。黄芩不良反应机制研究还不够完善,同时在其质量评价方面需要更深一步的研究。 | 郭旭东 晏伟 席少阳 杨莉萍 姬捷 马晓辉 黄璐琦 晋玲 | 2023 | 中草药2023,54,16: | 0 |
| 8 | Transient axonal glycoprotein-1 induces apoptosisrelated gene expression without triggering apoptosis in U251 glioma cells显示文摘Previous studies show that transient axonal glycoprotein-1, a ligand of amyloid precursor protein, increases the secretion of amyloid precursor protein intracellular domain and is involved in apoptosis in Alzheimer's disease. In this study, we examined the effects of transient axonal glycoprotein-1 on U251 glioma cells. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed that transient axonal glycoprotein-1 did not inhibit the proliferation of U251 cells, but promoted cell viability. The terminal deoxynucleotidyl transferase dUTP nick end labeling assay showed that transient axonal glycoprotein-1 did not induce U251 cell apoptosis. Real-time PCR revealed that transient axonal glycoprotein-1 substantially upregulated levels of amyloid precursor protein intracellular C-terminal domain, and p53 and epidermal growth factor receptor mRNA expression. Thus, transient axonal glycoprotein-1 increased apoptosis-related gene expression in U251 cells without inducing apoptosis.
