| 1 | 脐血干细胞移植2型糖尿病下肢血管病变患者内皮依赖性血管舒张的变化显示文摘背景:研究表明,干细胞在一定诱导条件下可分化成为血管性内皮细胞,促进局部血管再生,生成新的毛细血管网,建立丰富的侧支循环,以达到改善和治疗下肢缺血的目的。目的:观察脐血干细胞对2型糖尿病合并血管病变患者内皮依赖性血管舒张功能的影响。方法:100例2型糖尿病下肢血管病变患者分为两组,试验组60例,对照组40例,试验组静脉输注脐血干细胞,对照组输注生理盐水,4周后比较两组基础指标及内皮依赖性血管舒张功能。结果与结论:试验组治疗后空腹血糖、餐后2 h血糖、糖化血红蛋白、胆固醇、三酰甘油水平较治疗前均明显下降(P<0.05),空腹胰岛素、餐后2 h胰岛素、血管内皮舒张功能较治疗前均明显升高(P<0.05)。对照组空腹血糖、餐后2 h血糖、糖化血红蛋白、胆固醇、三酰甘油、血管内皮舒张功能治疗前后差异无显著性意义(P>0.05)。结果表明脐血干细胞移植可以改善2型糖尿病下肢血管病变患者的内皮功能,促进侧支循环,避免糖尿病足的发生。 | 李翠芳 姚远 | 2014 | 中国组织工程研究2014,18,50: | 3 |
| 2 | 人脐带间充质干细胞修复大鼠坐骨神经损伤的实验模型研究显示文摘目的:探讨人脐带间充质干细胞(HUMSC)修复大鼠坐骨神经损伤的效果。方法:对新鲜的羊膜进行处理,分离纯化 HUMSC后传代培养。取60只大鼠,手术处理成坐骨神经缺损套管模型鼠,随机分为三组:空白对照组(A组)、实验组(B组)、标准对照组(C组)。A组大鼠不采取其他措施,B组大鼠羊膜腔内注入HUMSC悬液,C组大鼠实施自体坐骨神经桥接术。观察记录大鼠一般的身体状态,12周后进行神经电生理检测和腓肠肌湿重称量。结果:术后大鼠术侧拖行,足趾合拢,术侧1~2d后发生红肿和溃疡;术后3~4周,B组和C组的大鼠红肿和溃疡症状渐渐消失,A组大鼠还存在红肿和溃疡;各组大鼠术后术侧腓肠肌都出现不同程度萎缩。B组和C组大鼠的术侧湿重/健侧湿重比值和A组比较,差异具有统计学意义。B组各神经电生理指标和C组比较,潜伏期长,传导速度慢,波幅小,但无统计学差异。结论:HUMSC可以分化为神经细胞,对大鼠坐骨神经损伤有修复作用。 | 韩继明 杨玲 王璐 朱文侠 阮彩莲 | 2014 | 陕西医学杂志2014,43,8: | 1 |
| 3 | Human periodontal ligament stem cells repair mental nerve injury显示文摘Human periodontal ligament stem cells are easily accessible and can differentiate into Schwann cells. We hypothesized that human periodontal ligament stem cells can be used as an alternative source for the autologous Schwann cells in promoting the regeneration of injured peripheral nerve.To validate this hypothesis, human periodontal ligament stem cells(1 × 106) were injected into the crush-injured left mental nerve in rats. Simultaneously, autologous Schwann cells(1 × 106) and PBS were also injected as controls. Real-time reverse transcriptase polymerase chain reaction showed that at 5 days after injection, mRNA expression of low affinity nerve growth factor receptor was significantaly increased in the left trigeminal ganglion of rats with mental nerve injury. Sensory tests,histomorphometric evaluation and retrograde labeling demonstrated that at 2 and 4 weeks after injection, sensory function was significantly improved, the numbers of retrograde labeled sensory neurons and myelinated axons were significantly increased, and human periodontal ligament stem cells and autologous Schwann cells exhibited similar therapeutic effects. | 4 | 右美托咪定对脐血间充质干细胞向神经细胞分化的影响显示文摘背景:大量研究证实右美托咪定在缺血再灌注损伤中具有显著的神经保护作用,但是其对神经系统的作用机制以及影响神经细胞功能的途径和方式鲜见报道。目的:探讨右美托咪定对脐血间充质干细胞增殖及向神经细胞分化的影响。方法:取第3代脐血间充质干细胞分为对照组、低浓度右美托咪定组(1μg/L)和高浓度右美托咪定组(10μg/L),MTT法检测脐血间充质干细胞活性,Western blot法检测增殖期脐血间充质干细胞中ERK1/2磷酸化水平,免疫荧光法检测Neu N/DAPI、GFAP/DAPI和Nestin/DAPI双染细胞比例,观察细胞分化能力。结果与结论:低浓度右美托咪定组干细胞活性及EKR1/2磷酸化水平较对照组明显增加(P<0.05);高浓度右美托咪定组干细胞活性及EKR1/2磷酸化水平较对照组及低浓度右美托咪定组明显降低(P<0.05);高、低浓度右美托咪定组中Neu N、GFAP阳性细胞数较对照组明显增加(P<0.05),而Nestin阳性细胞则显著降低(P<0.05)。结果表明低浓度右美托咪定诱导脐血间充质干细胞增殖而高浓度右美托咪则抑制其增殖,其机制可能通过调节EKR1/2磷酸化而实现的;高、低浓度右美托咪定均诱导脐血间充质干细胞向神经细胞的分化。 | 樊志刚 乔蕾 | 2014 | 中国组织工程研究2014,18,50: | 1 |
