| 2 | 汗孔角化症显示文摘汗孔角化症(porokeratosis)是一种罕见的病因不明表皮角化性疾病,其组织病理学特征是角质层是一列紧密贴合的角化不全的细胞,临床上表现为边缘堤状隆起,中央处皮肤轻度萎缩,可离心扩张。本病病程缓慢,多见于男性,往往无自觉症状,皮损可长期存在,很难痊愈。该疾病属常染色体显性遗传,但其外显不完全,大多数病例是由散发性突变引起的。该病的发病机制不明,但越来越多证据表明甲羟戊酸途径基因突变与汗孔角化症的发病有着密切的关系。该疾病目前暂无特效治疗方法,并且有癌变可能,患者需注意定期随访。 | 孙良丹 | 2020 | 皮肤科学通报2020,37,1: | 3 |
| 3 | IgD对人外周血Th1/Th2、Th17/Treg亚群及核转录因子的影响显示文摘目的:研究免疫球蛋白D(immunoglobulin D,Ig D)对人T细胞亚群Th1/Th2、Th17/Treg细胞平衡的影响。方法:不同浓度的Ig D(终浓度1、3、10μg/m L)刺激人外周血单个核细胞(peripheral blood mononuclear cells,PBMC)24 h后,流式细胞术检测CD69、CD154的表达、Th1、Th2、Th17、Treg细胞亚群的百分比,实时定量PCR(q PCR)技术检测核转录因子T-bet、GATA-3、Foxp3和ROR-γt mRNA的表达。结果:Ig D可增加T细胞表面活化标志物CD69、CD154的表达(P<0.05),显著减少Th1、Treg细胞亚群的百分比且可显著增加Th17的百分比,对Th2细胞亚群无显著影响;Ig D可显著下调核转录因子T-bet、Foxp3mRNA的表达且可显著上调核转录因子ROR-γt mRNA的表达,对核转录因子GATA-3 mRNA的表达无显著影响。结论:Ig D能促进T细胞活化,引起T细胞亚群Th1/Th2、Th17/Treg比例失衡。 | 董小洁 吴育晶 张静 陈文生 黄琼 魏伟 | 2018 | 中国临床药理学与治疗学2018,23,5: | 3 |
| 4 | TAIL-PCR研究进展及其在畜牧兽医中的应用显示文摘热不对称交错PCR(TAIL-PCR)是一种以PCR为基础的染色体步移,主要用于分离已知基因的侧翼序列,具有简便、快速、高效、特异性强等特点。文章从TAIL-PCR的原理、优势及局限性、反应体系优化等方面对TAIL-PCR的研究进展进行综述,其中在畜牧兽医中主要用于已知基因侧翼克隆和转基因动物模型外源基因插入位点确定。但由于基因组DNA的庞大和复杂性,使得从基因组中克隆获得目的基因仍存在诸多限制因素,如特异性引物错配和随机引物结合位点不确定,易造成非特异性产物的产生;模板GC含量异常或存在高度重复序列时,即使使用多种简并引物也难以获得阳性产物。因此,应根据实际情况对TAIL-PCR进行灵活调整。当模板较复杂时,可借鉴连接介导PCR原理,给引物加上接头,提高扩增反应的特异性;当模板较简单或序列较清晰时,可根据模板特点(如高GC含量)或已知模板序列设计引物、调整反应条件,增加操作的简便性;还可参考Suppression-PCR原理,选择引物和反应条件,获得较长的扩增片段。此外,TAIL-PCR可与多种技术联合使用,促使其在畜牧兽医中获得更广泛深入的应用。 | 马玲 王志强 覃绍敏 吴健敏 | 2017 | 南方农业学报2017,48,5: | 1 |
| 5 | IgD-Fc-Ig fusion protein,a new biological agent,inhibits T cell function in CIA rats by inhibiting IgD-IgDR-Lck-NF-κB signaling pathways显示文摘IgD-Fc-Ig fusion protein,a new biological agent,is constructed by linking a segment of human IgD-Fc with a segment of human IgG1-Fc,which specifically blocks the IgD-IgDR pathway and selectively inhibits the abnormal proliferation,activation,and differentiation of T cells.In this study we investigated whether IgD-Fc-Ig exerted therapeutic effects in collagen-induced arthritis(CIA)rats.CIA rats were treated with IgD-Fc-Ig(1,3,and 9 mg/kg)or injected with biological agents etanercept(3 mg/kg)once every 3 days for 40 days.In the PBMCs and spleen lymphocytes of CIA rats,both T and B cells exhibited abnormal proliferation;the percentages of CD3^+total T cells,CD3^+CD4^+Th cells,CD3^+CD4^+CD25^+-activated Th cells,Th1(CD4^+IFN-y^+),and Th17(CD4^+IL-17^+)were significantly increased,whereas the Treg(CD4^+CD25^+Foxp3^+)cell percentage was decreased.IgD-Fc-Ig administration dose-dependently decreased the indicators of arthritis;alleviated the histopathology of spleen and joint;reduced serum inflammatory cytokines levels;decreased the percentages of CD3^+total T cells,CD3^+CD4^+Th cells,CD3^+CD4^+CD25^+-activated Th cells,Th1(CD4^+IFN-y^+),and Th17(CD4^+IL-17^+);increased Treg(CD4^+CD25^+Foxp3^+)cell percentage;and down-regulated the expression of key molecules in IgD-IgDR-Lck-NF-KB signaling(p-Lck,p-ZAP70,p-P38,p-NF-KB65).Treatment of normal T cells with IgD(9μg/mL)in vitro promoted their proliferation.Co-treatment with IgD-Fc-Ig(0.1-10μg/mL)dose-dependently decreased IgD-stimulated T cell subsets percentages and down-regulated the IgD-IgDR-Lck-NF-KB signaling.In summary,this study demonstrates that IgD-Fc-Ig alleviates CIA and regulates the functions of T cells through inhibiting IgD-IgDR-Lck-NF-KB signaling. | Le Han Xian-zheng Zhang Chen Wang Xiao-yu Tang Yue Zhu Xiao-yu Cai Yu-jing Wu Jin-ling Shu Qing-tong Wang Jing-yu Chen Yan Chang Hua-xun WU Ling-ling Zhang Wei Wei | 2020 | Acta Pharmacologica Sinica2020,41,6: | 1 |
| 6 | 汗孔角化症的研究进展显示文摘汗孔角化症是一类较为少见的角化不全性皮肤疾病,其病因不明,病程呈慢性进展性,临床上该病皮损的典型表现为中央轻度萎缩、边缘呈脊状凸起的褐色环形斑疹或斑块,部分类型可表现为角化性丘疹或疣状斑块。目前越来越多证据表明胆固醇代谢过程中相关基因发生突变与汗孔角化症的发生发展有着紧密的关系,并且有相关研究证实针对胆固醇代谢的甲羟戊酸通路进行靶向治疗,对于部分类型汗孔角化症有效,为汗孔角化症的治疗提供了新思路。本文对汗孔角化症的研究进展进行简述。 | 李静 韩建文 | 2023 | 皮肤科学通报2023,40,5: | 0 |
