| 2 | Effect of Lingguizhugan decoction on myocardial Nuclear factor kappa B protein expression in rats with chronic heart failure显示文摘OBJECTIVE: To investigate the effect of Lingguizhugan decoction (LGZGD) on changes of cardiac structure and function, and its putative mechanism of action, by investigating mRNA and protein expression of myocardial nuclear factor kappa B(NF-κB), and the plasma content of NF-κB in rats with chronic heart failure.METHODS: The chronic heart failure (CHF) model in rats was induced by coronary artery ligation.Sham operation was performed in control rats. Six weeks after the procedure, rats were randomly classified into the various treatment groups: model CHF, Captopril (4.4 mg/kg), low LGZGD dose (2.1 g/kg), medium LGZGD dose (4.2 g/kg), and high LG-ZGD dose (8.4 g/kg). Treatments continued for 4 consecutive weeks. Changes of hemodynamic indices were observed by the PowerLab data acquisition and analysis system. Morphological changes of myocardium were observed by hematoxylin and eosin staining, and Masson staining. The mRNA and protein expression of myocardial NF-κB were detected by reverse transcription-polymerase chain reaction and western blotting, respectively. The plasma content of NF-κB was detected by enzyme-linked immuno-sorbent assay.RESULTS: CHF rats showed significant dysfunction in hemodynamic indices and in cardiac structure.Compared with the sham operation group, mRNA expression of myocardial NF-κB and plasma content of NF-κB of the model group was significantly increased. All three doses of LGZGD, and Captopril,improved the hemodynamic dysfunction, and inhibited the change of cardiac structure while significantly improving the survival rate. | 4 | Modeling spatio-temporal distribution of soil moisture by deep learning-based cellular automata model显示文摘Soil moisture content (SMC) is a key hydrological parameter in agriculture,meteorology and climate change,and understanding of spatio-temporal distributions of SMC in farmlands is important to address the precise irrigation scheduling.However,the hybrid interaction of static and dynamic environmental parameters makes it particularly difficult to accurately and reliably model the distribution of SMC.At present,deep learning wins numerous contests in machine learning and hence deep belief network (DBN) ,a breakthrough in deep learning is trained to extract the transition functions for the simulation of the cell state changes.In this study,we used a novel macroscopic cellular automata (MCA) model by combining DBN to predict the SMC over an irrigated corn field (an area of 22 km^2) in the Zhangye oasis,Northwest China.Static and dynamic environmental variables were prepared with regard to the complex hydrological processes.The widely used neural network,multi-layer perceptron (MLP) ,was utilized for comparison to DBN.The hybrid models (MLP-MCA and DBN-MCA) were calibrated and validated on SMC data within four months,i.e.June to September 2012,which were automatically observed by a wireless sensor network (WSN) .Compared with MLP-MCA,the DBN-MCA model led to a decrease in root mean squared error (RMSE) by 18%.Thus,the differences of prediction errors increased due to the propagating errors of variables,difficulties of knowing soil properties and recording irrigation amount in practice.The sequential Gaussian simulation (s Gs) was performed to assess the uncertainty of soil moisture estimations.Calculated with a threshold of SMC for each grid cell,the local uncertainty of simulated results in the post processing suggested that the probability of SMC less than 25% will be difference in different areas at different time periods.The current results showed that the DBN-MCA model performs better than the MLP-MCA model,and the DBN-MCA model provides a powerful tool for predicting SMC in highly non-linear forms.Moreover,because modeling soil moisture by using environmental variables is gaining increasing popularity,DBN techniques could contribute a lot to enhancing the calibration of MCA-based SMC estimations and hence provide an alternative approach for SMC monitoring in irrigation systems on the basis of canals. | SONG Xiaodong ZHANG Ganlin LIU Feng LI Decheng ZHAO Yuguo YANG Jinling | 2016 | Journal of Arid Land2016,8,5: | 20 |

| 6 | Influence of acute hyperglycemia in human sepsis on inflammatory cytokine and counterregulatory hormone concentrations显示文摘AIM: In human sepsis, a prominent component of the hypermetabolite is impaired glucose tolerance (IGT) and hyperglycemia. Elevations in plasma glucose concentration impair immune function by altering cytokine production from macrophages. We assessed the role of glucose in the regulation of circulating levels of insulin, glucagon, cortisol,IL-6 and TNF-α in human sepsis with normal or impaired glucose tolerance.METHODS: According to the results of intravenous glucose tolerance test, forty patients were classified into two groups: control group (n=20) and IGT group (n=20).Plasma glucose levels were acutely raised in two groups and maintained at 15 mmol/L for 3 hours. Plasma insulin,glucagon and cortisol levels were measured by radioimmunoassay, the levels of TNF-α and IL-6 were detected by ELISA.RESULTS: In IGT group, the fasting concentrations of plasma glucose, insulin, glucagon, cortisol, IL-6 and TNFα levels were significantly higher than those in control group (P<0.05). During clamp, the control group had a higher average amount of dextrose infusion than the IGT group (P<0.01). In control group, plasma insulin levels rose from a basal value to a peak at an hour (P<0.05) and maintained at high levels. Plasma glucagon levels descended from a basal value to the lowest level within an hour (P<0.01)and low levels were maintained throughout the clamp. In IGT group, plasma insulin was more significantly elevated (P<0.01), and plasma glucagon levels were not significantly declined. Plasma cortisol levels were not significantly changed in two groups. In control group, plasma IL-6 and TNF-α levels rose (P<0.01) within 2 hours of the clamp and returned to basal values at 3 hours. In IGT group, increased levels of plasma cytokine lasted longer than in control group (3 hours vs. 2 hours, P<0.05), and the cytokine peaks of IGT group were higher (P<0.05) than those of control group.CONCLUSION: Acute hyperglycemia pricks up hyperinsulinemia and increases circulating cytokine concentrations and these effects are more pronounced in sepsis with IGT. | 9 | Quantitative Relationship Between Land Use and Phosphorus Discharge in Subtropical Hilly Regions of China显示文摘The increase of phosphorus concentration is a crucial factor causing the eutrophication of water body,while land use has an important impact on agricultural non-point sources (NPS) phosphorus discharge. Sevensites controlling the water in four sub-watersheds and the main exit of the Meicun Watershed of XuanchengCounty, Anhui Province, were investigated by dynamic monitoring of stream water and nutrient discharge,integrating interpretation of aerial image and GIS analysis to find out how the land use affects phosphorusloss with stream water in typical agriculture-forest watershed in subtropical China. These monitored sitesare different in structure of land use. Phosphorus concentration of the stream water was analyzed everyweek and at the next day of rainfall. The velocity of flow was measured by kinemometer to calculatethe runoff flux and phosphorus discharge. The results showed that the runoff flux and the discharges ofdissolved phosphorus (DP), particle-associated phosphorus (PAP) and total phosphorus (TP) had significantexponential relationships with the area percentages of forest, pond and paddy field. There existed a significantlinear relationship between the TP and PAP concentrations in stream water and the area percentages of forest,pond and paddy field, and the discharge of PAP was also significantly linearly correlated with the dischargeof suspended soil particles. There was a logarithmic linear relationship between DP and PAP discharges. Thestudy indicated that the adjustment of land use patterns and construction of ecologically sound landscapewould be an important measure to reduce the runoff discharge of phosphorus. Evidence shows that phytolith assemblages of the cave deposits are dominated by the cold resistant types with a lower warm index, reflecting an overall cold inclined climate. This possibly connects the cave deposits with glacial climate to a great extent, which is in accordance with thenorthern fauna revealed by fossil mammals and temperateclimate indicated by pollen assemblages. | 15 | Prognosis of microscopic polyangiitis with renal involvement:report of 60 Chinese patients显示文摘With the widespread clinical application of renal biopsy and serum antineutrophil cytoplasmic antibody (ANCA) testing, the percentage of renal injuries caused by systemic small vessel vasculitis (SVV) including microscopic polyangiitis (MPA) and Wegener’ s granulomatosis (WG) is on the rise in China. According to our previous report, SVV constituted 1.09% of diagnoses from all renal biopsies (5. 8% in secondary nephritis) and 14. 4% of that from renal biopsies from patients with chronic renal,failure, among which MPA is the most common type. Many studies reported that SVV with rapid progressive glomerulonephritis had poor prognosis with a 5-year renal survival less than 75%. However, there is a lack of large-sample statistical analysis about the long-term prognosis of SVV in China. Indeed, the clinical manifestations of SVV in China are quite different from those reported in other countries. | 16 | Determination of the controlled landing trajectory of Chang’E-1 satellite and the coordinate analysis of the landing point on the Moon显示文摘Based on the tracking observations of radio ranges and VLBI delays of Chang’E-1 (CE-1) satellite during the controlled landing on the Moon on March 1, 2009, the landing trajectory and the coordinates of the landing point are determined by positioning analysis. It is shown that the landing epoch (the emission epoch of the last signal) of CE-1 satellite on the Moon was at UTC8h13m6.51s. The lunar longitude, latitude and surface height of the landing point in the lunar primary axes frame are respectively 52.2732°, 1.6440° and –3.56 km (the reference lunar radius is 1738 km). The uncertainties are 0.0040°, 0.0168° and 0.18 km. The corresponding uncertainty in the tangential direction of the lunar surface is 0.52 km and the three-dimensional (3D) positioning uncertainty is 0.55 km. It is accordingly deduced that even with the present technical specifications of the radio ranges and VLBI delays, the 1 km 3D positioning precision could be guaranteed for the lander in the second stage of China’s Lunar Explora- tion Project. Concerning the trace determination of the rover on the lunar surface, because only telemetry signal will be emitted, VLBI would be the sole tracking technique from the Earth. The application of the constraint of geocentric distance is shown to be helpful to improving the positioning precision. | 17 | Nutrient Discharges from a Typical Watershed in the Hilly Area of Subtropical China显示文摘Agriculture is still the biggest contributor of non-point source (NPS) pollution to water bodies andrunoff discharges of nutrients and other chemicals are one of the most important pathways. This studywas conducted during 1998~1999 in a typical watershed with complex agriculture and forestry systems aswell as dotted farmer villages, in a transitional region between the mid- and northern subtropical zones ofChina. Continuous sampling of stream water was performed regularly at a weekly frequency, with additionalsampling after all major rainfall events. The discharges of N and P nutrients and suspended materials weremeasured and the total and area-averaged annual discharges of all components were calculated. The resultsshowed an uneven seasonal distribution of nutrient discharges with summer storms contributing most tothe total fluxes. This study demonstrated a high dependence of runoff volume on rainfall but the overallrunoff coefficients were dependent on land use type and watershed size. The area-averaged annual dischargesvaried greatly among the sub-watersheds with different sizes and land use structures. | 18 | Effect of Wuziyanzong Pill on sperm quality and calcium ion content in oligoasthenospermia rats显示文摘OBJECTIVE:To study the effect of Wuziyanzong treatment on the sperm quality and content of calcium ions(Ca 2+) in oligoasthenospermia rats.METHODS:A model of oligoasthenospermia was induced in 50 Sprague Dawley rats by treatment with tripterygium glycosides at 30 mg/kg per day for 8 weeks.They were divided randomly into a model group,a positive group(Huangjingzanyu capsule,3.01 g/kg),and low,medium and high dose Wuziyanzong treatment groups(2.30,4.60,9.20 g/kg crude drug respectively) with 10 in each group.Another 10 rats were used as a control group.The rats in the control and model groups were administered distilled water,while the rats in the remaining groups were administered Wuziyanzong for 30 d.The epididymides were removed,spermatozoa recovered and the sperm density and viability were measured.The spermatozoa were purified and the contents of Ca 2+ in the cytoplasm and mitochondria were detected by flow cytometry and atomic absorption spectrometry,respectively.RESULTS:After 8 weeks of treatment with tripterygium glycosides,the sperm density,sperm activity and the Ca 2 + content of spermatozoa in the model rats were all significantly decreased compared with the control group(all P<0.05).After 30 d treatment,the sperm density and activity improved and the Ca 2 + content of sperm were increased significantly in the medium and high dose Wuziyanzong treatment groups in comparison with the model group(all P<0.05).CONCLUSION:The Wuziyanzong treatment increased sperm density,improved sperm viability and enhanced the content of Ca 2+ in the sperm cytoplasm and mitochondria in this rat model of oligoasthenospermia. | Tongsheng Wang Jinling Huang Deling Wu Qing Li Xiangguo Liu Han Chen Lei Qiao Dongdong Li Jun Li | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 11 |
| 19 | Pathogenesis and treatment of parenteral nutrition-associated liver disease显示文摘BACKGROUND: Parenteral nutrition-associated liver disease (PNALD) has been common in patients who require long-term parenteral nutrition. PNALD develops in 40%-60% of infants on long-term parenteral nutrition compared with 15%-40% of adults on home parenteral nutrition for intestinal failure. The pathogenesis of PNALD is multifactorial and remains unclear There is no specific treatment. Management strategies for its prevention and treatment depend on an understanding of many risk factors. This review aims to provide an update on the pathogenesis and treatment of this disease. DATA SOURCES: A literature search was performed on the MEDLINE and Web of Science databases for articles published up to October 2011, using the keywords: parenteral nutrition associated liver disease, intestinal failure associated liver disease lipid emulsions and fish oil. The available data reported in the relevant literatures were analyzed. RESULTS: The literature search provided a huge amount of evidence about the pathogenesis and management strategies on PNALD. Currently, lack of enteral feeding, extended duration of parenteral nutrition, recurrent sepsis, and nutrient deficiency or excess may play important roles in the pathogenesis of PNALD. Recent studies found that phytosterols present as contaminants in soy-based lipid emulsions, are also an important factor in the pathogenesis. Moreover, the treatment of PNALD is discussed. CONCLUSIONS: The use of lipid emulsions, phytosterols in particular, is associated with PNALD. Management strategies for the prevention and treatment of PNALD include consideration of early enteral feeding, the use of specialized lipid emulsions such as fish oil emulsions, and isolated small bowel or combined liver and small bowel transplantation. A greater understanding of the pathogenesis of PNALD has led to promising interventions to prevent and treat this condition. | 20 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations.
