1Hepatitis B virus,HBx mutants and their role in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is one of the leading causes of death induced by cancer in the modern world and majority of the cases are related to chronic hepatitis B virus(HBV)infection.HBV-encoded X protein(HBx)is known to play a pivotal role in the pathogenesis of viral induced HCC.HBx is a multifunctional protein of17 kDa which modulates several cellular processes by direct or indirect interaction with a repertoire of host factors resulting in HCC.HBX might interfere with several cellular processes such as oxidative stress,DNA repair,signal transduction,transcription,protein degradation,cell cycle progression and apoptosis.A number of reports have indicated that HBx is one of the most common viral ORFs that is often integrated into the host genome and its sequence variants play a crucial role in HCC.By mutational or deletion analysis it was shown that carboxy terminal of HBx has a likely role in protein-protein interactions,transcriptional transactivation,DNA repair,cell,signaling and pathogenesis of HCC.The accumulated evidence thus far suggests that it is difficult to understand the mechanistic nature of HBx associated HCC,and HBx mediated transcriptional transactivation and signaling pathways may be a major determinant.This article addresses the role of HBx in the development of HCC with particular emphasis on HBx mutants and their putative targets.Ashraf Ali Hany Abdel-Hafiz Mohd Suhail Amany Al-Mars Mohammad Khalid Zakaria Kaneez Fatima Sultan Ahmad Esam Azhar Adeel Chaudhary Ishtiaq Qadri 2014World Journal of Gastroenterology2014,20,30:29
2Potential mechanisms of hepatitis B virus induced liver injury显示文摘Chronic active hepatitis(CAH) is acknowledged as an imperative risk factor for the development of liver injury and hepatocellular carcinoma.The histological end points of CAH are chronic inflammation,fibrosis and cirrhosis which are coupled with increased DNA synthesis in cirrhotic vs healthy normal livers.The potential mechanism involved in CAH includes a combination of processes leading to liver cell necrosis,inflammation and cytokine production and liver scaring(fibrosis).The severity of liver damage is regulated by Hepatitis B virus genotypes and viral components.The viral and cellular factors that contribute to liver injury are discussed in this article.Liver injury caused by the viral infection affects many cellular processes such as cell signaling,apoptosis,transcription,DNA repair which in turn induce radical effects on cell survival,growth,transformation and maintenance.The consequence of such perturbations is resulted in the alteration of bile secretion,gluconeogenesis,glycolysis,detoxification and metabolism of carbohydrates,proteins,fat and balance of nutrients.The identification and elucidation of the molecular pathways perturbed by the viral proteins are important in order to design effective strategy to minimize and/or restore the hepatocytes injury.Mohd Suhail Hany Abdel-Hafiz Ashraf Ali Kaneez Fatima Ghazi A Damanhouri Esam Azhar Adeel GA Chaudhary Ishtiaq Qadri 2014World Journal of Gastroenterology2014,20,35:12
3Role of relevant immune-modulators and cytokines in hepatocellular carcinoma and premalignant hepatic lesions显示文摘AIM To assess the levels of different immune modulators in patients with hepatocellular carcinoma(HCC),in relation to other hepatic diseases.METHODS Eighty-eight patients were included in the current study and represented patients with HCC(20),liver cirrhosis(28) and chronic hepatitis(CH;25),and normal controls(NC;15).Peripheral blood was isolated for immunophenotyping of active myeloid dendritic cells(m DCs;CD1 c and CD40),mature inactive myeloid cells(CD1 c and HLA),active plasmacytoid cells(p DCs;CD303 and CD40),mature inactive p DCs(CD30 and HLA),active natural killer(NK) cells(CD56 and CD161),active NK cells(CD56 and CD314) and inactive NK cells(CD56 and CD158) was done by flow cytometry.Serum levels of interleukin(IL)-2,IL-10,IL-12,IL-1β,interferon(IFN)-α,IFN-γ and tumor necrosis factor(TNF)-αR2 were assessed by ELISA.RESULTS Active m DCs(CD1 C+/CD40+) and inactive m DCs(CD1 c+/HLA+) were significantly decreased in HCC patients in relation to NC(P < 0.001).CD40+ expression on active p DCs was decreased in HCC patients(P < 0.001),and its level was not significantly changed among other groups.Inactive p DCs(CD303+/HLA+),inactive NKs(CD56+/CD158+) and active NKs(CD56+/CD161+) were not statistically changed among the four groups studied;however,the latter was increased in CH(P < 0.05).NKG2 D was statistically decreased in HCC,CH and cirrhosis(P < 0.001),and it was not expressed in 63%(12/20) of HCC patients.There was significant decrease of IL-2,IFN-α and IFN-γ(P < 0.001),and a significant increase in IL-10,IL-1β,and TNF-αR2(P <0.01,P < 0.001 and P < 0.001;respectively) in HCC patients.There was inverted correlation between IL-12 and IL-1β in HCC(r =-0.565,P < 0.01),with a strong correlation between p DCs(CD303+/CD40+) and NKs(CD56+/CD161+;r = 0.512,P < 0.05) as well as inactive m DCs(CD1 c+/HLA+) and inactive NK cells(CD56+/CD158+;r = 0.945,P < 0.001).CONCLUSION NKG2 D,CD40,IL-2 and IL-10 are important modulators in the development and progression of HCC.Abdel-Rahman N Zekri Somaya El Deeb Abeer A Bahnassy Abeer M Badr Mona S Abdellateif Gamal Esmat Hosny Salama Marwa Mohanad Ahmed Esam El-dien Shimaa Rabah Assmaa Abd Elkader 2018World Journal of Gastroenterology2018,24,11:6
4Optimization of biliary drainage in inoperable distal malignant strictures显示文摘BACKGROUND Given most patients with distal malignant biliary obstruction present in the nonresectable stage,palliative endoscopic biliary drainage with fully covered metal stent(FCMS)or uncovered metal stent(UCMS)is the only available measure to improve patients’quality of life.Half covered metal stent(HCMS)has been recently introduced commercially.The adverse effects and stent function between FCMS and UCMS have been extensively discussed.AIM To study the duration of stent patency of HCMS and compare it with FCMS and UCMS to optimize biliary drainage in inoperable patients with distal malignant obstruction.Secondary aims in our study included evaluation of patients’survival and the rates of adverse events for each type of stent.METHODS We studied 210 patients and randomized them into three equal groups;HCMS,FCMS and UCMS were inserted endoscopically.RESULTS Stent occlusion occurred in(18.6%,17.1%and 15.7%in HCMS,FCMS and UCMS groups,respectively,P=0.9).Stent migration occurred only in patients with FCMS(8.6%of patients).Cholangitis and cholecystitis occurred in 11.4%and 5.7%of patients,respectively,in FCMS.Tumor growth occurred only in 10 cases among patients with UCMS after a median of 140 d,sludge occurred in nine,seven and one patients in HCMS,FCMS and UCMS,respectively(P=0.04).CONCLUSION Given the prolonged stent functioning time,the use of HCMS is preferred over the use of UCMS and FCMS for optimizing biliary drainage in patients with distal malignant biliary obstruction.Esam Elshimi Wesam Morad Omar Elshaarawy Ahmed Attia 2020World Journal of Gastrointestinal Endoscopy2020,12,9:5
5Wear and mechanical characterization of Mg-Gr self-lubricating composite fabricated by mechanical alloying显示文摘In this research,the wear and mechanical responses of pure magnesium-graphite(Mg-Gr)composite have been investigated aiming to get the optimum composition of reinforcement.The composite materials were fabricated by mechanical alloying.The percentage of graphite reinforcement was chosen as 3,5,7 and 10 wt.%to identify its potential for self-lubricating property under dry sliding conditions.The mechanical properties including hardness,tensile strength and flexural strength of the composites and the base material were tested.The wear tests were conducted by using a pin-on-disc tribometer.The results show that the mechanical properties decrease with increasing graphite content as compared to that of the base material.The wear rate and average coefficient of friction decrease with the addition of graphite and was found to be minimum at 5 wt.%graphite reinforcement.The addition of 5 wt.%graphite in the composite exhibits superior wear properties as compared to that of the matrix material and other compositions of the Mg-Gr composites.Azzat Esam Abdulqader Al-maamari AKMAsif Iqbal Dewan Muhammad Nuruzzaman 2019Journal of Magnesium and Alloys2019,7,2:2

10Effect of lead acetate toxicity on experimental male albino rat显示文摘Objective:To evaluate the effect of different doses of lead acetate(1/20,1/40 and 1/60 of LD_(50))on body weight gain,blood picture,plasma prolein profile and the function of liver,kidney and thyroid gland.Methods:Male albino rats were divided into four groups,the first group represented the health control animals,while the second,third and fourth groups were ingested orally with sub lethal doses of load acetate(1/20,1/40 and 1/60)of the oral LD_(50),respectively.One dose was ingested every two days during the experimental period(14 weeks)including the adaptation time.Blood was collected and used for all analysis.Results:The results showed that,the ingestion of Pb^(2+)induced significant stimulation in glutamic-pyruvic transaminase(ALT)and glutamic-oxalacetic transaminease(AST)activity.Also,total soluble protein and albumin contents of plasma were significantly decreased,while the content of globulin was changed by the Pb^(2+)treatments.The cholinesteiase activity was inhibited,but the activities of alkaline and acid phosphates and lactate dehydrogenase were stimulated,while plasma glucose level was elevated as a result of lead acetate intoxication.In case of blood picture,Pb^(2+)ingestion reduced the contents of hemoglobin and RBCs count of intoxicated rat's blood and the plasma levels of T3,T4 and blood WBCs count were decreased.Conclusions:It can be concluded that lead acetate has harmful effect on experimental male albino rats.Therefore,the present work advises people to prevent exposure to the lead compound to avoid injurious hazard risk.Nabil M Ibrahim Esam A Eweis Hossam S El-Beltagi Yasmin E Abdel-Mobdy 2012Asian Pacific Journal of Tropical Biomedicine2012,2,1:1

12Distinct editing functions of natural HLA-DM allotypes impact antigen presentation and CD4^(+) T cell activation显示文摘Classical human leukocyte antigen(HLA)molecules of the major histocompatibility classⅡ(MHCⅡ)complex present peptides for the development,surveillance and activation of CD4^(+) T cells.The nonclassical MHCⅡ-like protein HLA-DM(DM)catalyzes the exchange and loading of peptides onto MHCⅡmolecules,thereby shaping MHCⅡimmunopeptidomes.Natural variations of DM in both chains of the protein(DMA and DMB)have been hypothesized to impact peptide presentation,but no evidence for altered function has been reported.Here we define the presence of DM allotypes in human populations covered by the 1000 Genomes Project and probe their activity.The functional properties of several allotypes are investigated and show strong enhancement of peptide-induced T cell activation for a particular combination of DMA and DMB.Biochemical evidence suggests a broader pH activity profile for the new variant relative to that of the most commonly expressed DM allotype.Immunopeptidome analysis indicates that the compartmental activity of the new DM heterodimer extends beyond the late endosome and suggests that the natural variation of DM has profound effects on adaptive immunity when antigens bypass the canonical processing pathway.Miguel Álvaro-Benito Eliot Morrison Friederike Ebner Esam T.Abualrous Marie Urbicht Marek Wieczorek Christian Freund 2020Cellular & Molecular Immunology2020,17,2:1
