| 8 | Role of glycosylation in TGF-β signaling and epithelial-to-mesenchymal transition in cancer显示文摘Glycosylation is a common posttranslational modification on membrane-associated and secreted proteins that is of pivotal importance for regulating cell functions.Aberrant glycosylation can lead to uncontrolled cell proliferation,cell-matrix interactions,migration and differentiation,and has been shown to be involved in cancer and other diseases.The epithelial-to-mesenchymal transition is a key step in the metastatic process by which cancer cells gain the ability to invade tissues and extravasate into the bloodstream.This cellular transformation process,which is associated by morphological change,loss of epithelial traits and gain of mesenchymal markers,is triggered by the secreted cytokine transforming growth factor-β(TGF-β).TGF-βbioactivity is carefully regulated,and its effects on cells are mediated by its receptors on the cell surface.In this review,we first provide a brief overview of major types of glycans,namely,N-glycans,O-glycans,glycosphingolipids and glycosaminoglycans that are involved in cancer progression.Thereafter,we summarize studies on how the glycosylation of TGF-βsignaling components regulates TGF-βsecretion,bioavailability and TGF-βreceptor function.Then,we review glycosylation changes associated with TGF-β-induced epithelial-to-mesenchymal transition in cancer.Identifying and understanding the mechanisms by which glycosylation affects TGF-βsignaling and downstream biological responses will facilitate the identification of glycans as biomarkers and enable novel therapeutic approaches. | Jing Zhang Peter ten Dijke Manfred Wuhrer Tao Zhang | 2021 | Protein & Cell2021,12,2: | 4 |

| 11 | Bile leakage after loop closure vs clip closure of the cystic duct during laparoscopic cholecystectomy:A retrospective analysis of a prospective cohort显示文摘BACKGROUND Laparoscopic cholecystectomy(LC)is one of the most frequently performed surgical procedures.Cystic stump leakage is an underestimated,potentially life threatening complication that occurs in 1%-6%of the patients.With a secure cystic duct occlusion technique during LC,bile leakage becomes a preventable complication.AIM To investigate the effect of polydioxanone(PDS)loop closure of the cystic duct on bile leakage rate in LC patients.METHODS In this retrospective analysis of a prospective cohort,the effect of PDS loop closure of the cystic duct on bile leakage complication was compared to patients with conventional clip closure.Logistic regression analysis was used to develop a risk score to identify bile leakage risk.Leakage rate was assessed for categories of patients with increasing levels of bile leakage risk.RESULTS Of the 4359 patients who underwent LC,136(3%)underwent cystic duct closure by a PDS loop.Preoperatively,loop closure patients had significantly more complicated biliary disease compared to the clipped closure patients.In the loop closure cohort,zero(0%)bile leakage occurred compared to 59 of 4223(1.4%)clip closure patients.For patients at increased bile leakage risk(risk score≥1)rates were 1.6%and up to 13%(4/30)for clip closure patients with a risk score≥4.This risk increase paralleled a stepwise increase of actual bile leakage complication for clip closure patients,which was not observed for loop closure patients.CONCLUSION Cystic duct closure with a PDS loop during LC may reduce bile leakage in patients at increased risk for bile leakage. | Sandra C Donkervoort Lea M Dijksman Aafke H van Dijk Emile A Clous Marja A Boermeester Bert van Ramshorst Djamila Boerma | 2020 | World Journal of Gastrointestinal Surgery2020,12,1: | 4 |

| 13 | Systematic review of cystic duct closure techniques in relation to prevention of bile duct leakage after laparoscopic cholecystectomy显示文摘AIM To study the effect of different techniques of cystic duct closure on bile leakage after laparoscopic cholecystectomy(LC) for biliary disease.METHODS A systematic search of MEDLINE, Cochrane and EMBASE was performed. Rate of cystic duct leakage(CDL) was the primary outcome. Risk of bias was evaluated. Odds ratios were analyzed for comparison of techniques and pooled event rates for non-comparative analyses. Pooled event rates were compared for each of included techniques.RESULTS Out of 1491 articles, 38 studies were included. A total of 47491 patients were included, of which 38683(81.5%)underwent cystic duct closure with non-locking(metal)clips. All studies were of low-moderate methodological quality. Only two studies reported separate data on uncomplicated and complicated gallbladder disease. For overall CDL, an odds ratio of 0.4(95%CI: 0.06-2.48)was found for harmonic energy vs clip closure and an odds ratio of 0.17(95%CI: 0.03-0.93) for locking vs non-locking clips. Pooled CDL rate was around 1% for harmonic energy and metal clips, and 0% for locking clips and ligatures. CONCLUSION Based on available evidence it is not possible to either recommend or discourage any of the techniques for cystic duct closure during LC with respects to CDL,although data point out a slight preference for locking clips and ligatures vs other techniques. | 18 | Inventory and action plan for PFOS and related substances in Suriname as basis for Stockholm Convention implementation显示文摘Within the framework of the Stockholm Convention implementation in Suriname,the first inventory of perfluorooctane sulfonic acid(PFOS)has been developed.The major stocks of PFOS are firefighting foams with minor stocks in pesticides(ant baits)and possibly carpets.The inventory of firefighting foams assessed the import,current stocks and major use sites and recent releases.Considerable amount of PFOS and potentially PFOS containing firefighting foams are stored at the military base,the oil company and small firefighting departments throughout the country.The total amount of stored PFAS firefighting foam in Suriname is estimated to be at least 275.6 tonnes,where the major stockpile of 206 tonnes is at the oil storage facilities.For some of these foams PFOS content is confirmed and for others it is not clear if the foam contains PFOS or other per-or polyfluorinated alkylated substances(PFAS).The upper estimate of PFOS content is 4133 kg in these foams.Additionally,PFOS or other PFAS has also been discovered in fire extinguishers in institutions.The total amount of PFOS related substance EtFOSA imported with the pesticide sulfluramid was 75 kg.The imports were banned in 2014 and is controlled.A considerable amount of synthetic carpets are in stores even produced before 2003 where main use of PFOS from 3 M took place possibly containing PFOS related substances.A compilation of potentially PFOS contaminated sites has been made.Potentially contaminated sites result from firefighting foam use,dumpsites and sulfluramid use.The airport where firefighting foam was applied is above a major drinking water reservoir,where an urgent assessment is needed.Due to the lack of analytical capacity for PFOS and precursors,drinking water,firefighting foam or impacted carpets,could not been analysed.An action plan for the control of PFOS and other PFAS has been developed considering the synergy of the Stockholm Convention and the Strategic Approach to International Chemicals Management(SAICM)addressing this emerging pollution issue for developing countries. | Victorine Pinas Carmen Van Dijk Roland Weber | 2020 | Emerging Contaminants2020,6,1: | 3 |
