1Optimization of phenol degradation by Candida tropicalis Z-04 using Plackett-Burman design and response surface methodology显示文摘Statistical experimental designs were used to optimize the process of phenol degradation by Candida tropicalis Z-04,isolated from phenol-degrading aerobic granules.The most important factors influencing phenol degradation (p<0.05),as identified by a two-level Plackett-Burman design with 11 variables,were yeast extract,phenol,inoculum size,and temperature.Steepest ascent method was undertaken to determine the optimal regions of these four significant factors.Central composite design (CCD) and response surface analysis were adopted to further investigate the mutual interactions between these variables and to identify their optimal values that would generate maximum phenol degradation.The analysis results indicated that interactions between yeast extract and temperature,phenol and temperature,inoculum size and temperature affected the response variable (phenol degradation) significantly.The predicted results showed that the maximum removal efficiency of phenol (99.10%) could be obtained under the optimum conditions of yeast extract 0.41 g/L,phenol 1.03 g/L,inoculum size 1.43% (V/V) and temperature 30.04°C.These predicted values were further verified by validation experiments.The excellent correlation between predicted and experimental values confirmed the validity and practicability of this statistical optimum strategy.This study indicated the excellent ability of C.tropicalis Z-04 in degrading high-strength phenol.Optimal conditions obtained in this experiment laid a solid foundation for further use of this microorganism in the treatment of highstrength phenol effluents.Jiangya Zhou Xiaojuan Yu Cong Ding Zhiping Wang Qianqian Zhou Hao Pao Weimin Ca 2011Journal of Environmental Sciences2011,23,1:48
2CA19-9 antigen levels can distinguish between benign and malignant pancreaticobiliary disease显示文摘BACKGROUND:CA19-9 is a carbohydrate tumor-associated antigen which is frequently upregulated in pancreatobiliary neoplasia.However,it may also be elevated in patients with jaundice in the absence of a tumor due to biliary obstruction,and in other non-hepato-pancreatico-biliary conditions.This study aimed to evaluate whether CA19-9 levels could accurately differentiate between benign and malignant pancreatobiliary disease.METHODS:All patients referred to a single surgeon for investigation of pancreaticobiliary disease in 2003 in whom a firm diagnosis had been established were included.For malignant disease,a histological diagnosis was required but for benign disease a firm radiological diagnosis was deemed adequate.The patients were divided into 4 categories:pancreatic adenocarcinoma(PCa);cholangiocarcinoma(CCa);chronic pancreatitis(CP)and biliary calculous disease(Calc).Bilirubin and alkaline phosphatase levels corresponding to the point of assessment of CA19-9 were also noted.RESULTS:Final diagnoses were made of pancreatic adenocarcinoma(PCa,n=73),cholangiocarcinoma(CCa,n=19),ampullary carcinoma(Amp,n=7),neuroendocrine carcinoma(Neu,n=4),duodenal carcinoma(Duo,n=3),chronic pancreatitis(CP,n=115),and biliary calculous disease(Calc,n=27).Median CA19-9 levels(U/ml)were:PCa,653;CCa,408;Duo,403;Calc,27;CP,19;Neu,10.5;Amp,8(reference range:0-37).The CA19-9 levels were significantly greater for malignant than for benign disease,could differentiate PCa from CCa/Duo,and were significantly higher in unresectable than in resectable PCa.The sensitivity,specificity,positive predictive value(PPV)and negative predictive value(NPV)for CA19-9 were 84.9%,69.7%,67.7%and 86.1%,respectively.A ROC analysis provided an area under the curve for CA19-9 of 0.871(0.820-0.922),giving an optimal CA19-9 of 70.5 U/ml for differentiating benign from malignant pathology.Using this cut-off,the sensitivity was 82.1%,while specificity,PPV and NPV improved to 85.9%,81.3%and 86.5%,respectively.When standard radiology was included(US/ CT/MRCP)in the decision process,the results improved to 97.2%,88.7%,86.6%,and 97.7%.For benign disease,the CA19-9 correlated directly with the serum bilirubin,but for malignant disease,CA19-9 levels were elevated independent of the bilirubin level.CONCLUSIONS:CA19-9 is useful in the differentiation of pancreatobiliary disease and when using an optimized cut-off and combining with routine radiology,the diagnostic yield is improved significantly,thus stressing the importance of a multi-disciplinary approach to pancreatobiliary disease.Gareth Morris-Stiff Mary Teli Nicky Jardine Malcolm CA Puntis 2009Hepatobiliary & Pancreatic Diseases International2009,8,6:33
3WJD 5^(th) Anniversary Special Issues(2): Type 2 diabetes Type 2 diabetes and cardiovascular disease: Have all risk factors the same strength?显示文摘Diabetes mellitus is a chronic condition that occurs when the body cannot produce enough or effectively use of insulin.Compared with individuals without diabetes,patients with type 2 diabetes mellitus have a considerably higher risk of cardiovascular morbidity and mortality,and are disproportionately affected by cardiovascular disease.Most of this excess risk is it associated with an augmented prevalence of well-known risk factors such as hypertension,dyslipidaemia and obesity in these patients.However the improved cardiovascular disease in type 2 diabetes mellitus patients can not be attributed solely to the higher prevalence of traditional risk factors.Therefore other non-traditional risk factors may be important in people with type 2 diabetes mellitus.Cardiovascular disease is increased in type 2 diabetes mellitus subjects due to a complex combination of various traditional and non-traditional risk factors that have an important role to play in the beginning and the evolution of atherosclerosis over its long natural history from endothelial function to clinical events.Many of these risk factors could be common history for both di-abetes mellitus and cardiovascular disease,reinforcing the postulate that both disorders come independently from'common soil'.The objective of this review is to highlight the weight of traditional and non-traditional risk factors for cardiovascular disease in the setting of type 2 diabetes mellitus and discuss their position in the pathogenesis of the excess cardiovascular disease mortality and morbidity in these patients.Iciar Martín-Timón Cristina Sevillano-Collantes Amparo Segura-Galindo Francisco Javier del Caizo-Gómez 2014World Journal of Diabetes2014,5,4:32
4A Genome-wide Functional Characterization of Arabidopsis Regulatory Calcium Sensors in Pollen Tubes显示文摘Calcium, an ubiquitous second messenger, plays an essential and versatile role in cellular signaling. The diverse function of calcium signals is achieved by an excess of calcium sensors. Plants possess large numbers of calcium sensors, most of which have not been functionally characterized. To identify physiologically relevant calcium sensors in a specific cell type, we conducted a genome-wide functional survey in pollen tubes, for which spatiotemporal calcium signals are well-characterized and required for polarized tip growth. Pollen-specific members of calmodulin (CaM), CaM-like (CML), calcium-dependent protein kinase (CDPK) and calcineurin B-like protein (CBL) families were tagged with green fluorescence protein (GFP) and their localization patterns and overexpression phenotypes were characterized in tobacco pollen tubes. We found that several fusion proteins showed distinct overexpression phenotypes and subcellular localization patterns. CDPK24-GFP was localized to the vegetative nucleus and the generative cell/sperms. CDPK32-GFP caused severe growth depolarization. CBL2-GFP and CBL3-GFP exhibited dynamic patterns of subcellular localization, including several endomembrane compartments, the apical plasma membrane (PM), and cytoskeleton-like structures in pollen tubes. Their overexpression also inhibited pollen tube elongation and induced growth depolarization. 5On linking climate to Chinese dynastic change: Spatial and temporal variations of monsoonal rain显示文摘The causal correlation or linkage between the East Asian Monsoon (EAM) intensity and rise/fall of Chinese dynasties recently proposed by high-resolution paleoclimate reconstructions is examined in detail. Aside from many exceptions to the correlation, both instrumental and historical climate records show strong spatial variations of rainfall on annual-to-decadal scales over eastern China. The relationship between rainfall and EAM intensity also exhibits regional disparity. These observations suggest: (1) Tak-ing paleo-proxy records from a single locality in eastern China to imply changes in drought/wetness as to affect the cultural and political history of China is fraught with uncertainty. 7Endoscopic ultrasonography guided biliary drainage: Summary of consortium meeting, May 7^(th) , 2011, Chicago显示文摘Endoscopic retrograde cholangiopancreatography (ERCP) has become the preferred procedure for biliary or pancreatic drainage in various pancreatico-biliary disorders. With a success rate of more than 90%, ERCP may not achieve biliary or pancreatic drainage in cases with altered anatomy or with tumors obstructing access to the duodenum. In the past those failures were typically managed exclusively by percutaneous approaches by interventional radiologists or surgical intervention. The morbidity associated was significant especially in those patients with advanced malignancy, seeking minimally invasive interventions and improved quality of life. With the advent of biliary drainage via endoscopic ultrasound (EUS) guidance, EUS guided biliary drainage has been used more frequently within the last decade in different countries. As with any novel advanced endoscopic procedure that encompasses various approaches, advanced endoscopists all over the world have innovated and adopted diverse EUS guided biliary and pancreatic drainage techniques. This diversity has resulted in variations and improvements in EUS Guided biliary and pancreatic drainage; and over the years has led to an extensive nomenclature. The diversity of techniques, nomenclature and recent progress in our intrumentation has led to a dedicated meeting on May 7 th , 2011 during Digestive Disease Week 2011. More than 40 advanced endoscopists from United States, Brazil, Mexico, Venezuela, Colombia, Italy, France, Austria, Germany, Spain, Japan, China, South Korea and India attended this pivotal meeting. The meeting covered improved EUS guided biliary access and drainage procedures, terminology, nomenclature, training and credentialing; as well as emerging devices for EUS guided biliary drainage. 10Analysis on the decadal scale variation of the dust storm in North China显示文摘In this paper, the temporal variation characteristics of the dust storm in North China are investigated. Based on power spectrum analysis and wavelet analysis, 1956-1970 and 1985-1999 are chosen as the high-frequency and low-frequency dust storm decades respectively. Analysis results clearly show that the spring and wintertime anomalous atmospheric circulation between these two dec-ades are significantly different. Compared with the former decade, there are a strengthened polar vortex, enhanced westerlies near 50°N, and a weak East Asian major though in the winter of the latter decade. The north and center parts of the Siberian high and the Aleutian low become weak. The southerly and easterly wind anomalies appear over the north and east parts of China, which implies the weakening of East Asian winter monsoon. Meanwhile, northern China experiences warmer winters and wetter springs, which are in favor of the weakening of dust storm intensity in North China. There are significant out-of-phase relationships between dust frequency and wintertime westerly intensity, as well as between dust frequency and Arctic Oscillation. 13A novel mutation at the JK locus causing Jknull phenotype in a Chinese family显示文摘Urea transporters are a group of proteins that facilitate urea movement across biological membranes. Kidd blood group (JK antigen) and urea transporter of human erythrocytes are carried by the same protein UT-B. To investigate the molecular basis of the Jknull phenotype in the Chinese population, blood samples from Chinese individuals were screened using the 2 mol/L urea solution hemolysis test. Urea and water permeability of erythrocytes membrane was measured by stopped-flow light scattering. Genomic DNA was extracted from lymphocytes. UT-B gene of Jknull's family was analyzed using genomic PCR by primers designed to cover sequences of all exons and exon-intron boundaries in human UT-B gene. One Jknull subject was found from twenty thousand screened Chinese individuals, and it was confirmed that this individual did not express the erythrocyte urea transporter. Genomic sequence analysis of the Jknull individual showed that there were two point mutations, G→C, which is novel, and G→A, at the 3(-acceptor splice site (AG) of intron 5 of UT-B gene. Exon 6 is spliced out in the UT-B transcript due to either of these mutations. 15No apparent lock-in depth of the Laschamp geomagnetic excursion: Evidence from the Malan loess显示文摘The first-order chronological framework of the long-term Chinese loess/paleosol sequences was based mainly on magnetostratigraphy. However, there remain arguments on the lock-in ages of the major geomagnetic reversals. This study systematically compared the stratigraphic locations of the Laschamp excursion and Henrich events (H4 at~39 ka and H5 at~48 ka) recorded by the Luochuan loess profile in the central Chinese Loess Plateau. It may be the product of the dynamic processes involv-ing tumbling, grinding and compresso-shearing functions, so the spherical particulates with ul-tra-size of nm and mm are called grinding grain. The formation of the grinding grains is related with the ductile-brittle deformation, the lateral pressure of the slipping, and the very low grade of dynamic metamorphism of the rocks. Based on the systematic analysis, it is indicated that some elements like Nb and Ta are enriched in the grinding grains in the granitic rocks because of the chemical differentiation, and that the slipping lamellae bearing grinding grains in mudstone with smear efficiency prove a perfect seal for oil and gas reservoirs.
