| 2 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 5 | Association Between Diurnal Temperature Range and Respiratory Tract Infections显示文摘Objective This study aimed to assess the association between emergency-room visits for respiratory tract infection (RTI) with diurnal temperature range (DTR), a weather parameter closely associated with urbanization and global climate change. Methods We conducted a semiparametric time-series analysis to estimate the percentage increase in emergency-room visits for RTI associated with changes in DTR after adjustment for daily weather conditions (temperature and relative humidity) and outdoor air pollution. Results DTR was significantly associated with daily emergency-room visits for RTI. An increase of 1 °C in the current-day (L0) and in the 2-day moving average (L01) DTR corresponded to a 0.94% [95% confidence interval (CI), 0.34%-1.55%] and 2.08% (95% CI, 1.24%-2.93%) increase in emergency-room visits for RTI, respectively. Conclusion DTR was associated with increased risk of RTI. | 7 | Geometrical feature analysis and disaster assessment of the Xinmo landslide based on remote sensing data显示文摘At 5:39 am on June 24, 2017, a landslide occurred in the village of Xinmo in Maoxian County, Aba Tibet and Qiang Autonomous Prefecture(Sichuan Province, Southwest China). On June 25, aerial images were acquired from an unmanned aerial vehicle(UAV), and a digital elevation model(DEM) was processed. Landslide geometrical features were then analyzed. These are the front and rear edge elevation, accumulation area and horizontal sliding distance. Then, the volume and the spatial distribution of the thickness of the deposit were calculated from the difference between the DEM available before the landslide, and the UAV-derived DEM collected after the landslide. Also, the disaster was assessed using high-resolution satellite images acquired before the landslide. These include Quick Bird, Pleiades-1 and GF-2 images with spatial resolutions of 0.65 m, 0.70 m, and 0.80 m, respectively, and the aerial images acquired from the UAV after the landslide with a spatial resolution of 0.1 m. According to the analysis, the area of the landslide was 1.62 km2, and the volume of the landslide was 7.70 ± 1.46 million m3. The average thickness of the landslide accumulation was approximately 8 m. The landslide destroyed a total of 103 buildings. The area of destroyed farmlands was 2.53 ha, and the orchard area was reduced by 28.67 ha. A 2-km section of Songpinggou River was blocked and a 2.1-km section of township road No. 104 was buried. Constrained by the terrain conditions, densely populated and more economically developed areas in the upper reaches of the Minjiang River basin are mainly located in the bottom of the valleys. METHODS: Human AGS cells transfected with cagA and blank vector were treated with specific mitogen-activated protein kinase kinase (MEK) inhibitor. Total cell proteins were combined by strong anion exchange (SAX2) and weak cation exchange (CM10) ProteinChip arrays and analyzed using surface-enhanced laser desorption/ ionization time-of-flight mass spectrometry (SELDI-TOF-MS) proteomics technology. Protein expression profi les were compared with those of inhibitor-untreated cagA transfectants. SwissProt/TrEMBL database searching for differentially expressed proteins was carried out using the TagIdent tool with the pI and mass information. RESULTS: When a total of 16 proteins that showed expression differences in inhibitor-untreated cagA transfectants were compared with vector transfectants,three proteins with m/z 4229,8162 and 9084 were found to have no expression differences after treatment with MEK inhibitor,while the other 13 maintained the same expression differences after inhibitor treatment. Seven pieces of meaningful matching information for the three proteins were obtained from database searching. CONCLUSION: Biomarkers with m/z 4229,8162 and 9084 are ERK1/2 phosphorylation dependent,and therefore are the downstream molecules of ERK1/2 in the ERK/MAPK signaling pathway. | 12 | Synthesis of a Novel Core-shell Type Acrylic-polyurethane Hybrid Emulsion Containing Siloxane and Fluorine as well as Water and the Oil Resistances of Cured Film显示文摘Siliconated polyurethane (Si-PU) was synthesized using isophorone diisocyanate (IPDI), hydroxybutyl-terminated polydimethylsiloxane (PDMS), polytetramethylene ether glycol (PTMG), polypropylene glycol (PPG), 1,6-hexanediol (HDO), dimethylol propionic acid (DMPA) and triethylamine (TEA). Based on butyl acrylate (BA), 2, 2, 2-trifluoroethylmethacrylate (TFEMA) and Si-PU as a seed emulsion, a novel core-shell type acrylic-polyurethane hybrid emulsion, containing siloxane and fluorine (F-Si-PU), was prepared by seeded emulsion polymerization. The contents of siloxane and fluorine were determined according to the feed ratio. Fourier transform infrared spectroscopy (FTIR) was used to identify the chain structures of Si-PU and F-Si-PU. Investigation of transmission electron microscopy (TEM) confirmed the core-shell structure of F-Si-PU emulsion. | 14 | An experimental study on astrocytes promoting production of neural stem cells derived from mouse embryonic stem cells显示文摘Background The production of neural stem cells (NSCs) derived from embryonic stem (ES) cells was usually very low according to previous studies, which was a major obstacle for meeting the needs of clinical application. This study aimed at investigating whether astrocytes could promote production of NSCs derived from ES cells in vitro.Methods Mouse ES cells line-D3 was used to differentiate into NSCs with astrocytes as inducing stromal cells by means of three-stage differentiation procedure. Another group without astrocytes served as control. The totipotency of ES cells was identified by observation of cells’ morphology and formation of teratoma in severe combined immunodeficiency disease (SCID) mice. The quantity and purity of NSCs derived from ES cells were analyzed using clonogenic assay, immunohistochemical staining and flow cytometry assay. The plasticity of NSCs was detected by differentiating test. Octamer-binding transcription factor 4 (Oct-4) and nestin, the specific marker genes of ES cells and NSCs respectively, were detected continuously using reverse transcription-polymerase chain reaction (RT-PCR) method to monitor the process of cell differentiation.Results The ES cells of D3 line could maintain the ability of differentiating into cellular derivations of all three primary germ layers after continuous passage culture. At the end of two-stage of inducing process, 23.2±3.5 neurospheres per plate formed in astrocyte-induced group and only 0.8±0.3 per plate in the control group (clonogenic assay, P<0.01), and the ratio of nestin positive cells was (50.2±2.8)% in astrocyte-induced group and only (1.4±0.5)% in the control group (flow cytometry, P<0.01). With the induction undergoing, the expression of Oct-4 gradually decreased and then disappeared, while the expression of nestin was increased step by step, and the ratio of nestin positive cells was up to 91.4% by the three-stage differentiation. The nestin positive cells could be further induced into neurons, astrocytes, and oligodendrocytes in differentiating medium supplemented with fetal calf serum. | 15 | Massive cranium from Harbin in northeastern China establishes a new Middle Pleistocene human lineage显示文摘It has recently become clear that several human lineages coexisted with Homo sapiens during the late Middle and Late Pleistocene.Here,we report an archaic human fossil that throws new light on debates concerning the diversification of the Homo genus and the origin of H.sapiens.The fossil was recovered in Harbin city in northeastern China,with a minimum uranium-series age of 146 ka.This cranium is one of the best preserved Middle Pleistocene human fossils.Its massive size,with a large cranial capacity(1,420 mL)falling in the range of modern humans,is combined with a mosaic of primitive and derived characters.It differs from all the other named Homo species by presenting a combination of features,such as long and low cranial vault,a wide and low face,large and almost square orbits,gently curved but massively developed supraorbital torus,flat and low cheekbones with a shallow canine fossa,and a shallow palate with thick alveolar bone supporting very large molars.The excellent preservation of the Harbin cranium advances our understanding of several less-complete late Middle Pleistocene fossils from China,which have been interpreted as local evolutionary intermediates between the earlier species Homo erectus and later H.sapiens.Phylogenetic analyses based on parsimony criteria and Bayesian tip-dating suggest that the Harbin cranium and some other Middle Pleistocene human fossils from China,such as those from Dali and Xiahe,form a third East Asian lineage,which is a part of the sister group of the H.sapiens lineage.Our analyses of such morphologically distinctive archaic human lineages from Asia,Europe,and Africa suggest that the diversification of the Homo genus may have had a much deeper timescale than previously presumed.Sympatric isolation of small populations combined with stochastic long-distance dispersals is the best fitting biogeographical model for interpreting the evolution of the Homo genus. | Xijun Ni Qiang Ji Wensheng Wu Qingfeng Shao Yannan Ji Chi Zhang Lei Liang Junyi Ge Zhen Guo Jinhua Li Qiang Li Rainer Grun Chris Stringer | 2021 | The Innovation2021,2,3: | 6 |

| 20 | Alda-1 treatment promotes the therapeutic effect of mitochondrial transplantation for myocardial ischemia-reperfusion injury显示文摘Mitochondrial damage is a critical driver in myocardial ischemia-reperfusion(I/R)injury and can be alleviated via the mitochondrial transplantation.The efficiency of mitochondrial transplantation is determined by mitochondrial vitality.Because aldehyde dehydrogenase 2(ALDH2)has a key role in regulating mitochondrial homeostasis,we aimed to investigate its potential therapeutic effects on mitochondrial transplantation via the use of ALDH2 activator,Alda-1.Our present study demonstrated that time-dependent internalization of exogenous mitochondria by cardiomyocytes along with ATP production were significantly increased in response to mitochondrial transplantation.Furthermore,Alda-1 treatment remarkably promoted the oxygen consumption rate and baseline mechanical function of cardiomyocytes caused by mitochondrial transplantation.Mitochondrial transplantation inhibited cardiomyocyte apoptosis induced by the hypoxia-reoxygenation exposure,independent of Alda-1 treatment.However,promotion of the mechanical function of cardiomyocytes exposed to hypoxia-reoxygenation treatment was only observed after mitochondrial Alda-1 treatment and transplantation.By using a myocardial I/R mouse model,our results revealed that transplantation of Alda-1-treated mitochondria into mouse myocardial tissues limited the infarction size after I/R injury,which was at least in part due to increased mitochondrial potential-mediated fusion.In conclusion,ALDH2 activation in mitochondrial transplantation shows great potential for the treatment of myocardial I/R injury. | Xiaolei Sun Rifeng Gao Wenjia Li Yongchao Zhao Heng Yang Hang Chen Hao Jiang Zhen Dong Jingjing Hu Jin Liu Yunzeng Zou Aijun Sun Junbo Ge | 2021 | Bioactive Materials2021,6,7: | 5 |
