1Soil Microbial Characteristics Under Long-Term Heavy Metal Stress:A Case Study in Zhangshi Wastewater Irrigation Area,Shenyang显示文摘Soil samples were collected from Zhangshi Wastewater Irrigation Area in the suburb of Shenyang City,China,an area with a 30-year irrigation history with heavy metal-containing wastewater.The chemical properties and microbial characteristics of the soils were examined to evaluate the present situation of heavy metal pollution and to assess the soil microbial characteristics under long-term heavy metal stress.In light of the National Environmental Quality Standards of China,the soil in the test area was heavily polluted by Cd and to a lesser degree by Zn and Cu,even though wastewater irrigation ceased in 1993.Soil metabolic quotient (qCO_2) had a significant positive correlation,while soil microbial quotient (qM) had a negative correlation with content of soil heavy metals.Soil microbial biomass carbon (MBC) had significantly negative correlation with Cd,but soil substrate-induced respiration (SIR),dehydrogenase activity (DHA),cellulase activity, and culturable microbial populations had no persistent correlations with soil heavy metal content.Soil nutrients,except for phosphorous,showed positive effects on soil microbial characteristics,which to a certain degree obscured the adverse effects of soil heavy metals.Soil Cd contributed more to the soil microbial characteristics,but qM and qCO_2 were more sensitive and showed persistent responses to heavy metals stress.It could be concluded that qM and qCO_2 can be used as bioindicators of heavy metal pollution in soils.ZHANC Yan ZHANG Hui-Wen SU Zhen-Cheng ZHANG Cheng-Gang 2008Pedosphere2008,18,1:12
2Elevated IL-37 levels in the plasma of patients with severe coronary arterycalcification显示文摘Background Coronary artery calcification (CAC) is a predictor of cardiovascular events and plaque burden and is closely associatedwith chronic inflammation. Intedeukin (IL)-37 is a newly discovered member of the IL-1 family and is considered an anti-inflammatorycytokine. Our recent study on mice indicated that IL-37 could attenuate atherosclerosis and vascular calcification, which suggests that IL-37could be associated with the development of atherosclerosis and related diseases. The aim of this study was to investigate if IL-37 plays arole in the progression of CAC in patients. Methods Two hundred participants with suspected cardiovascular disease were recruited. Thelevels of plasma IL-37, osteoprotegerin (OPG), hypersensitive C-reactive protein (hsCRP) together with other biochemical parameters weremeasured, and a coronary calcium assessment was carried out by multi-detector row CT. A score of 〈 10 AU (Agatston units) denotes anabsence of CAC, a score of 11-100 AU denotes mild CAC, 101-400 denotes moderate CAC, and 〉 400 AU denotes severe CAC. ResultsOur initial data showed that there were no apparent differences in plasma IL-37 levels among patients with or without mild or moderate CAC.However, IL-37 levels were significantly increased in patients with severe CAC (P 〈 0.001). Similar results were observed for plasma OPGand hsCRP levels. When IL-37 levels in patients with severe calcification were compared with that in all of the other non-severe CAC groups,it became apparent that there was a significant positive correlation between IL-37 level and severe CAC (r = 0.360, P 〈 0.001; OR = 1.033)using Spearrnan's correlation and binary logistic regression analysis. 3Non-Fusion and Fusion Expression of β-Galactosidase from Lactobacillus bulgaricus in Lactococcus lactis显示文摘Objective To construct four recombinant Lactococcus lactis strains exhibiting high β-galactosidase activity in fusion or non-fusion ways,and to study the influence factors for their protein expression and secretion.Methods The gene fragments encoding β-galactosidase from two strains of Lactobacillus bulgaricus,wch9901 isolated from yogurt and 1.1480 purchased from the Chinese Academy of Sciences,were amplified and inserted into lactococcal expression vector pMG36e.For fusion expression,the open reading frame of the β-galactosidase gene was amplified,while for non-fusion expression,the open reading frame of the β-galactosidase gene was amplified with its native Shine-Dalgarno sequence upstream.The start codon of the β-galactosidase gene partially overlapped with the stop codon of vector origin open reading frame.Then,the recombinant plasmids were transformed into Escherichia coli DH5α and Lactococcus lactis subsp.lactis MG1363 and confirmed by determining β-galactosidase activities.Results The non-fusion expression plasmids showed a significantly higher β-galactosidase activity in transformed strains than the fusion expression plasmids.The highest enzyme activity was observed in Lactococcus lactis transformed with the non-fusion expression plasmids which were inserted into the β-galactosidase gene from Lactobacillus bulgaricus wch9901.The β-galactosidase activity was 2.75 times as high as that of the native counterpart.In addition,β-galactosidase expressed by recombinant plasmids in Lactococcus lactis could be secreted into the culture medium.The highest secretion rate(27.1%) was observed when the culture medium contained 20 g/L of lactose.Conclusion Different properties of the native bacteria may have some effects on the protein expression of recombinant plasmids.Non-fusion expression shows a higher enzyme activity in host bacteria.There may be a host-related weak secretion signal peptide gene within the structure gene of Lb.bulgaricus β-galactosidase,and its translation product may introduce the enzyme secretion out of cells in special hosts.CHUAN WANG CHAO-WU ZHANC HENG-CHUAN LIU QIAN YU AND XIAO-FANG PEI 2008Biomedical and Environmental Sciences2008,21,5:7
4Upregulation of MiR-126 Delays the Senescence of Human Glomerular Mesangial Cells Induced by High Glucose via Telomere-p53-p21-Rb Signaling Pathway显示文摘Diabetic kidney disease (DKD)is a microvascular complication of type 2 diabetes.The study of DKD mechanisms is the most important target for the prevention of DKD.Renal senescence is one of the important pathogeneses for DKD,but the mechanism of renal and cellular senescence is unclear.Decreased expression of circulating miR-126 is associated with the development of DKD and may be a promising blood-based biomarker for DKD.This study is to probe the effect and mechanism of miR-126 on the aging of human glomerular mesangial cells (HGMCs)induced by high glucose.HGMCs were cultured with Roswell Park Memorial Institute (RPMI-1640)in vitro.The effect of high glucose on morphology of HGMCs was observed 72h after intervention.The cell cycle was examined by flow cytometry.The telomere length was measured by Southern blotting.The expression levels of p53,p21 and Rb proteins in p53-p21-Rb signaling pathway and p-statl,p-stat3 in JAK/STAT signaling pathway were detected by Western blotting respectively.The expression of miR-126 was examined by qRT-PCR.MiR-126 mimics was transfected into HGMCs.The effects of miR-126 mimics transfection on cell morphology,cell cycle,telomere length,p53,p21,Rb,p-stat1 and p-stat3 were observed. The results showed that high glucose not only arrested the cell cycle in G1phase but also shortened the telomere length.High glucose led to high expression of p53,p21,Rb,p-statl and p-stat3 and premature senescence of HGMCs by activating the telomere-p53-p21-Rb and JAK/STAT signaling pathways.Moreover,the miR-126 was decreased in HGMCs induced by high glucose.It was suggested that the transfection of miR-126 mimics could inhibit the telomere-p53-p21-Rb and JAK/STAT signaling pathway activity in vitro and delay the senescence of HGMCs.The results may serve as a new strategy for the treatment of DKD.Dong-wei CAO Chun-ming JIANG Cheng WAN Miao ZHANC Qing-yan ZHANG Min ZHAO Bo YANG Da-long ZHU Xiao HAN 2018Current Medical Science2018,38,5:5
