2An Arabidopsis Class II Formin,AtFH19,Nucleates Actin Assembly,Binds to the Barbed End of Actin Filaments,and Antagonizes the Effect of AtFH1 on Actin Dynamics显示文摘Formin is a major protein responsible for regulating the nucleation of actin filaments,and as such,it permits the cell to control where and when to assemble actin arrays.It is encoded by a multigene family comprising 21 members in Arabidopsis thaliana.The Arabidopsis formins can be separated into two phylogenetically-distinct classes:there are 11 class I formins and 10 class II formins.Significant questions remain unanswered regarding the molecular mechanism of actin nucleation and elongation stimulated by each formin isovariant,and how the different isovariants coordinate to regulate actin dynamics in cells.Here,we characterize a class II formin,AtFH19,biochemically.We found that AtFH19 retains all general properties of the formin family,including nucleation and barbed end capping activity.It can also generate actin filaments from a pool of actin monomers bound to profilin.However,both the nucleation and barbed end capping activities of AtFH19 are less efficient compared to those of another well-characterized formin,AtFH1.Interestingly,AtFH19 FH1FH2 competes with AtFH1 FH1FH2 in binding actin filament barbed ends,and inhibits the effect of AtFH1 FH1FH2 on actin.We thus propose a mechanism in which two quantitatively different formins coordinate to regulate actin dynamics by competing for actin filament barbed ends.Yiyan Zheng Haibo Xin Jinxing Lin Chun-Ming Liu Shanjin Huang 2012Journal of Integrative Plant Biology2012,54,10:7
3Optimized pre-treatment of high strength food waste digestate by high content aluminum-nanocluster based magnetic coagulation显示文摘Coagulation-based pre-treatment efficiency of high strength digestate of food waste(HSDFW) anaerobic digestion is negated by organic ligand-catalyzed decomposition of coagulants. In this study, an efficient HSDFW pre-treatment method, magnetic seeds(MS) coagulation, was employed by using highly stable Keggin Al_(30) nanocluster(PAC_(30)), MS and polyacrylamide(PAM), and its operation was optimized by evaluating the performance of removing turbidity, total suspended solids(TSS), chemical oxygen demand(COD), and total phosphorous(TP) phosphate. Results showed that at the optimum dosage of 4.82 g/L, PAC_(30) demonstrated excellent removals as high as 98.93% ± 0.1% of turbidity, 98.04% ± 0.1% of TSS, 58.28% ± 0.3% of total COD, 99.98% ± 0.01% of TP and 99.50% ± 0.01% of dissolved phosphate, respectively. Apparent molecular weight(AMW) and three-dimensional excitation-emission matrix(3 D-EEM) fluorescence spectroscopy analyses demonstrated more efficient removal of dissolved organic matter(DOM), particularly non-biodegradable and hydrophobic components by PAC_(30) than commercial coagulant. The sedimentation was much improved from 40 min by coagulation/flocculation to about 5 min settling by MS coagulation. 16Directed evolution of a neutrophilic and mesophilic methanol dehydrogenase based on high-throughput and accurate measurement of formaldehyde显示文摘Methanol is a promising one-carbon feedstock for biomanufacturing,which can be sustainably produced from carbon dioxide and natural gas.However,the efficiency of methanol bioconversion is limited by the poor catalytic properties of nicotinamide adenine dinucleotide(NAD^(+))-dependent methanol dehydrogenase(Mdh)that oxidizes methanol to formaldehyde.Herein,the neutrophilic and mesophilic NAD^(+)-dependent Mdh from Bacillus stearothermophilus DSM 2334(Mdh_(Bs))was subjected to directed evolution for enhancing the catalytic activity.The combination of formaldehyde biosensor and Nash assay allowed high-throughput and accurate measurement of formaldehyde and facilitated efficient selection of desired variants.Mdh_(Bs)variants with up to 6.5-fold higher K_(cat)/K_(M)value for methanol were screened from random mutation libraries.The T153 residue that is spatially proximal to the substrate binding pocket has significant influence on enzyme activity.The beneficial T153P mutation changes the interaction network of this residue and breaks theα-helix important for substrate binding into two shortα-helices.Reconstructing the interaction network of T153 with surrounding residues may represent a promising strategy to further improve Mdh_(Bs),and this study provides an efficient strategy for directed evolution of Mdh.Jin Qian Liwen Fan Jinxing Yang Jinhui Feng Ning Gao Guimin Cheng Wei Pu Wenjuan Zhou Tao Cai Shuang Li Ping Zheng Jibin Sun Depei Wang Yu Wang 2023Synthetic and Systems Biotechnology2023,8,3:0
17Bubble-templated synthesis of nanocatalyst Co/C as NADH oxidase mimic显示文摘Designing highly active nanozymes for various enzymatic reactions remains a challenge in practical applications and fundamental research. In this work, by studying the catalytic functions of natural NADH oxidase(NOX), we devised and synthesized a porous carbon-supported cobalt catalyst(Co/C) to mimic NOX. The Co/C can catalyze dehydrogenation of NADH and transfers electrons to O_(2)to produce H_(2)O_(2).Density functional theory calculations reveal that the Co/C can catalyze O_(2)reduction to H_(2)O_(2)or H;O considerably. The Co/C can also mediate electron transfer from NADH to heme protein cytochrome c,thereby exhibiting cytochrome c reductase-like activity. The Co/C nanoparticles can deplete NADH in cancer cells, induce increase of the reactive oxygen species, lead to impairment of oxidative phosphorylation and decrease in mitochondrial membrane potential, and cause ATP production to be damaged. 18Disease X Testing:The results of an international external quality assessment exercise显示文摘The United Nations Secretary-General Mechanism(UNSGM)for investigation of the alleged use of chemical and biological weapons is the only established international mechanism of this type under the UN.The UNGSM may launch an international investigation,relying on a roster of expert consultants,qualified experts,and analytical laboratories nominated by the member states.Under the framework of the UNSGM,we organized an external quality assurance exercise for nominated laboratories,named the Disease X Test,to improve the ability to discover and identify new pathogens that may cause possible epidemics and to determine their animal origin.The“what-if”scenario was to identify the etiological agent responsible for an outbreak that has tested negative for many known pathogens,including viruses and bacteria.Three microbes were added to the samples,Dabie bandavirus,Mammarenavirus,and Gemella spp.,of which the last two have not been taxonomically named or published.The animal samples were from Rattus norvegicus,Marmota himalayana,New Zealand white rabbit,and the tick Haemaphysalis longicornis.Of the 11 international laboratories that participated in this activity,six accurately identified pathogen X as a new Mammarenavirus,and five correctly identified the animal origin as R.norvegicus.These results showed that many laboratories under the UNSGM have the capacity and ability to identify a new virus during a possible international investigation of a suspected biological event.The technical details are discussed in this report.Tian Qin Han Zheng Xuelian Luo Wen Zhang Jing Yang Yamin Sun Na Han Yuanhai You Liang Lu Xin Lu Di Xiao Shenshen Jiang Xuexin Hou Jinxing Lu Biao Kan Jianzhong Zhang Jianguo 2022Journal of Biosafety and Biosecurity2022,4,2:0
19From agricultural cellulosic waste to food delivery packaging: A mini-review显示文摘The growing food delivery service market has boosted the consumption of packaging materials, and this trend is projected to continue in the following years. The gap between industrial supply and consumer demand from a sustainable viewpoint leads to a need for agricultural cellulosic waste-based materials that bring the idea of trash-to-treasure to fruition. In this paper, we review up-to-date advancements surrounding the food delivery packaging that are derived from agricultural cellulosic waste. Two scenarios in which agricultural feedstock is used as a host or guest material are summarized, and sketch on the individual processing routine is depicted. We further evaluate how the chemical compositions and processing parameters influence the properties of the final products. 20Genome-wide identification and expression profiling of the YUCCA gene family in Brassica napus显示文摘Auxin is essential for plant growth and development,also closely related to plant architecture.YUCCA(YUC)proteins catalyse the rate-limiting step for endogenous auxin biosynthesis.In this study,we identified 57 Bn YUC genes in Brassica napus(rapeseed)genome,located on 17 chromosomes.Among them,A09 chromosome contained the most Bn YUCs as 6.Phylogenetic tree showed that all Bn YUC members were divided into 2 evolutionary clusters,including 7 sub-groups.Among the sub-groups,Clade I-C consisted 20 members,evolved independently from Arabidopsis thaliana,and motif 10 was unique.Genomic collinearity analysis showed that YUC collinear gene pairs widely existed in Brassica species,indicating that this family has a high retention rate in the evolution.Expression profile suggested that the Bn YUC family genes tended to express in specific growth/development stages as well as suffering stress/hormone treatments.It suggested that dominant expression of specific members existed with each development stage or treatment,indicating that each Bn YUC might have a clear division of work,and play a given biological function.This work was expected to build a basis for further study of the involvement of Bn YUC in growth,development and stress response.Ka Zhang Jinfang Zhang Cheng Cui Liang Chai Benchuan Zheng Jun Jiang Haojie Li Jinxing Tu 2022Oil Crop Science2022,7,3:0
