1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 2Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. 32018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 5Roux-en-Y choledochojejunostomy using novel magnetic compressive anastomats in canine model of obstructive jaundice显示文摘BACKGROUND:The traditional hand-sewn Roux-en-Y choledochojejunostomy is technically complicated,and the incidence of postoperative complications has remained high.A set of novel magnetic compressive anastomats was introduced to facilitate choledochojejunostomy and improve the prognosis of patients.METHODS:After ligating the common bile duct for 7 days,16 dogs were randomly divided into two groups (n=8 per group).Anastomats were used in the study group,and the traditional hand-sewn method was used in the control group for standard Roux-en-Y choledochojejunostomy.We compared the operation time,incidence of complications,gross appearance,and pathological disparity in stoma between the two groups in 1-month and 3-month follow-up examinations.RESULTS:The time spent on constructing the anastomosis for the study group was significantly shortened.Although no anastomotic stenosis occurred in the two groups,the narrowing rate of biliary-enteric anastomosis was much higher in the control group.There was one case of bile leakage in the control group,whereas no bile leakage occurred in the study group.A smoother surface,an improved layer apposition,and a lower local inflammatory response were identified in the anastomosis of the study group.CONCLUSION:The structures of the novel magnetic compressive anastomats are simple,and they are time-saving,safe and efficient for performing Roux-en-Y choledocho- jejunostomy procedures in a canine model of obstructive jaundice.Chao Fan,Xiao-Peng Yan,Shi-Qi Liu,Chun-Bao Wang,Jian-Hui Li,Liang Yu,Zheng Wu and Yi Lv Department of Hepatobiliary Surgery and Department of Pathology,First Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710061,China Department of Surgical Oncology,Third Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710068,China 2012Hepatobiliary & Pancreatic Diseases International2012,11,1:49
6FLOOD DISCHARGE AND ENERGY DISSIPATION BY JETS FROM OUTLETS IN HIGH ARCH DAM显示文摘In this thesis, the scale effect by an aerated water jet diffusing in the water upon the hydrodynamic pressure, the local riverbed scour by multiple layered jets and by those colliding in the vertical and the transverse directions, and the stability of the apron slab both in the inverted arch cushion pool and the flat bottom one by the large discharge and the high water head with 300m level are detailedly researched by means of model test and numerical simulation. A mathematic model simulating the destabilization of the flat bottomed slab is established to open out the mechanism of its stability. 8Multi-objective optimization of a mixed-flow pump impeller using modified NSGA-II algorithm显示文摘In order to maintain a uniform distribution of pareto-front solutions, a modified NSGA-II algorithm coupled with a dynamic crowding distance(DCD) method is proposed for the multi-objective optimization of a mixed-flow pump impeller. With the pump meridional section fixed, ten variables along the shroud and hub are selected to control the blade load by using a three-dimensional inverse design method. Hydraulic efficiency, along with impeller head, is applied as an optimization objective; and a radial basis neural network(RBNN) is adopted to approximate the objective function with 82 training samples. Local sensitivity analysis shows that decision variables have different impacts on the optimization objectives. Instead of randomly selecting one solution to implement, a technique for ordering preferences by similarity to ideal solution(TOPSIS) is introduced to select the best compromise solution(BCS) from pareto-front sets. The proposed method is applied to optimize the baseline model, i.e. a mixed- flow waterjet pump whose specific speed is 508 min?1?m3s?1?m. The performance of the waterjet pump was experimentally tested. Compared with the baseline model, the optimized impeller has a better hydraulic efficiency of 92% as well as a higher impeller head at the design operation point. Furthermore, the off-design performance is improved with a wider highefficiency operation range. After optimization, velocity gradients on the suction surface are smoother and flow separations are eliminated at the blade inlet part. 9Tree-ring precipitation records from Baiyinaobao, Inner Mongolia since A.D. 1838显示文摘Correlation function analysis shows that total precipitation from April to early July (to July 10) in growing season limits the ring width of Picea Koraiensis significantly in Baiyinaobao, Xilinhot, Inner Mongolia. Thus a transfer function is designed to reconstruct the total precipitation from April to early July over the region for the last 160 years. The explained variance of the reconstruction is 49.3% (and 45.7% when adjusted for loss of degrees of freedom, N = 31, r = 0.702, F = 13.608, p < 0.0001). There are three relatively wet periods in the reconstruction which are: 1869—1875, 18861921 and 19431968, and four relatively dry periods: 1851—1868, 1876—1885, 1922—1943 and 1969—1999. Among them, the wet periods of 1886—1921 and 1943—1968, and the dry period of 1922—1943 appeared almost at the same time with those in Mt. Qilian region. Meanwhile, the dry period of 1922—1943 corresponds to high temperature and low rainfall during the 1920s and thereafter, and wetness of 1943—1968 corresponds very well to low temperature and more precipitation after the 1940s in Northwest, Northern China and the Changjiang River drainage basin. Calculation shows that the reconstruction series is significantly correlated with local dry-wet index (r = -0.234, p < 0.007, N = 143). Wavelet analysis indicates that there is a 22-a cycle in the precipitation of April to early July during 18381920, and an 11-a cycle during 1920—1952. No more cycles that reach 95% confidence limits are identified after 1953. The precipitation from April to early July quickly dropped into low during the 1920s. 10Characteristics of one-year observation of VOCs,NOx, and O_3 at an urban site in Wuhan, China显示文摘A continuous online observation of ozone and its precursors(NOx, VOCs) was carried out in central urban Wuhan from September 2016 to August 2017. The concentration levels of ozone,NOx, VOCs and their variations in urban Wuhan were analyzed, as well as effects of VOCs on ozone photochemical generation and the main controlling factors for ozone production. During the observation period, the average concentrations of ozone and NOx in Wuhan was 22.63 and30.14 ppbv, respectively, and the average concentration of VOCs was 32.61 ppbv(42.3% alkanes,13.0% alkenes, 10.0% aromatics, 7.3% acetylene, 9.9% OVOCs, and 10.5% halohydrocarbons).Ozone concentration was higher in spring and summer as compared with autumn and winter,wheras VOCs and NOx concentratios were lower in spring and summer but higher in autumn and winter. Aromatics and alkenes, two of VOCs species, showed the highest contributions to ozone formation potential in Wuhan(35.7% alkenes, 35.4 aromatics, 17.5% alkanes, 8.6% OVOCs,1.6% halogenated hydrocarbons, and 1.4% acetylene). Among all VOCs species, those with the highest contribution were ethylene, m-xylene, toluene, propylene and o-xylene. 11Laparoscopic vs open D2 gastrectomy for locally advanced gastric cancer: A meta-analysis显示文摘AIM:To conduct a meta-analysis comparing laparoscopic(LGD2)and open D2 gastrectomies(OGD2)for the treatment of advanced gastric cancer(AGC).METHODS:Randomized controlled trials(RCTs)and non-RCTs comparing LGD2 with OGD2 for AGC treatment,published between 1 January 2000 and 12January 2013,were identified in the Pub Med,Embase,and Cochrane Library databases.Primary endpoints included operative outcomes(operative time,intraoperative blood loss,and conversion rate),postoperative outcomes(postoperative analgesic consumption,time to first ambulation,time to first flatus,time to first oralintake,postoperative hospital stay length,postoperative morbidity,incidence of reoperation,and postoperative mortality),and oncologic outcomes(the number of lymph nodes harvested,tumor recurrence and metastasis,disease-free rates,and overall survival rates).The Cochrane Collaboration tools and the modified Newcastle-Ottawa scale were used to assess the quality and risk of bias of RCTs and non-RCTs in the study.Subgroup analyses were conducted to explore the incidence rate of various postoperative morbidities as well as recurrence and metastasis patterns.A Begg’s test was used to evaluate the publication bias.RESULTS:One RCT and 13 non-RCTs totaling 2596patients were included in the meta-analysis.LGD2 in comparison to OGD2 showed lower intraoperative blood loss[weighted mean difference(WMD)=-137.87 m L,95%CI:-164.41--111.33;P<0.01],lower analgesic consumption(WMD=-1.94,95%CI:-2.50--1.38;P<0.01),shorter times to first ambulation(WMD=-1.03d,95%CI:-1.90--0.16;P<0.05),flatus(WMD=-0.98d,95%CI:-1.30--0.66;P<0.01),and oral intake(WMD=-0.85 d,95%CI:-1.67--0.03;P<0.05),shorter hospitalization(WMD=-3.08 d,95%CI:-4.38--1.78;P<0.01),and lower postoperative morbidity(odds ratio=0.78,95%CI:0.61-0.99;P<0.05).No significant differences were observed between LGD2 and OGD2 for the following criteria:reoperation incidence,postoperative mortality,number of harvested lymph nodes,tumor recurrence/metastasis,or three-or five-year diseasefree and overall survival rates.However,LGD2 had longer operative times(WMD=57.06 min,95%CI:41.87-72.25;P<0.01).CONCLUSION:Although a technically demanding and time-consuming procedure,LGD2 may be safe and effective,and offer some advantages over OGD2 for treatment of locally AGC.Zhen-Hong Zou Li-Ying Zhao Ting-Yu Mou Yan-Feng Hu Jiang Yu Hao Liu Hao Chen Jia-Ming Wu Sheng-Li An Guo-Xin Li 2014World Journal of Gastroenterology2014,20,44:25
12Studies of Chemical Constituents and Their Antioxidant Activities From Astragalus mongholicus Bunge显示文摘Objective To evaluate the antioxidant activities of different chemical constituents from Astragalus mongholicus Bunge and their protection against xanthine (XA)/xanthine oxidase (XO)-induced toxicity in PC12 cells. Methods The compounds of Astragalus mongholicus Bunge were isolated by chromatography and the structures were elucidated on the basis of spectral data interpretation. Their antioxidant activities were detected by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities in a cell-free system. Meanwhile, the effects against XA/XO-induced toxicity were assessed using MTT assay in PC12 cells. Results Ten principal constituents were isolated and identified as formononetin (I), ononin (II), calycosin (III), calycosin-7-O-β-D-glucoside (IV), 9,10-dimethoxypterocarpan-3-O-β-D-glucoside (V), adenosine (VI), pinitol (VII), daucosterol (VIII), β-sitoster (IX) and saccharose (X) from Astragalus mongholicus Bunge. The compounds I, III, and IV scavenged DPPH free radicals in vitro. Formononetin and calycosin were found to inhibit XA/XO-induced cell injury significantly, with an estimated EC50 of 50 ng/mL. Conclusion Compound Ⅱ, Ⅵ, and Ⅶ are first reported in this plant. 13Amplitudes, rates, periodicities and causes of temperature variations in the past 2485 years and future trends over the central-eastern Tibetan Plateau显示文摘Amplitudes, rates, periodicities, causes and future trends of temperature variations based on tree rings for the past 2485 years on the central-eastern Tibetan Plateau were analyzed. The results showed that extreme climatic events on the Plateau, such as the Medieval Warm Period, Little Ice Age and 20th Century Warming appeared synchronously with those in other places worldwide. The largest amplitude and rate of temperature change occurred during the Eastern Jin Event (343-425 AD), and not in the late 20th century. There were significant cycles of 1324 a, 800 a, 199 a, 110 a and 2-3 a in the 2485-year temperature series. The 1324 a, 800 a, 199 a and 110 a cycles are associated with solar activity, which greatly affects the Earth surface temperature. The long-term trends (>1000 a) of temperature were controlled by the millennium-scale cycle, and amplitudes were dominated by multi-century cycles. Moreover, cold intervals corresponded to sunspot minimums. 15Inhibitory effects of antisense RNA of HAb18G/CD147 on invasion of hepatocellular carcinoma cells in vitro显示文摘AIM: To study the inhibitory effects of antisense RNA of HAb18G/CD147 on invasion of hepatocellular carcinoma (HCC) cells in vitro.METHODS: Antisense RNA of HAb18G/CD147 vector PCIasHAb18G was constructed by reversely inserting HAb18G/CD147 cDNA to eukaryotic expression vector PCI-neo. The HCC cell line HHCC was transfected by PCI-asHAb18G via cation liposome. Expression of HAb18G/CD147 of transfected cells selected by G418 (geneticin) was observed by immunohistochemical SP staining and FACS (fluorescence activated cell sorting). Gelatin zymography was used to determine the effect of PCI-asHAb18G on reducing secretions of MMP2 and MMP-9 of the transfected cells. Boyden chamber was employed to test the invasion of HCC cells in vitro.RESULTS: The construction of antisense RNA vector PCIasHAb18G was verified correct by partial nucleotide sequencing and restricted endonuclease digestion. The expression of HAb18G/CD147 in transfected HHCC was inhibited by PCI-asHAb18G. Secretions of MMP-2 and MMP9 of transfected HHCC were reduced and the invasion of transfected HHCC was inhibited compared to HHCC,respectively.CONCLUSION: Invasion of HCC cells can be inhibited by antisense RNA of HAb18G/CD147. 17Three-dimensional morphometric analysis for hepatectomy of centrally located hepatocellular carcinoma:A pilot study显示文摘AIM: To describe a three-dimensional model(3DM) to accurately reconstruct anatomic relationships of centrally located hepatocellular carcinomas(HCCs).METHODS: From March 2013 to July 2014, reconstructions and visual simulations of centrally located HCCs were performed in 39 patients using a 3D subject-based computed tomography(CT) model with customdeveloped software. CT images were used for the 3D reconstruction of Couinaud's pedicles and hepatic veins, and the calculation of corresponding tumor territories and hepatic segments was performed using Yorktal DMIT software. The respective volume, surgical margin, and simulated virtual resection of tumors were also estimated by this model preoperatively. All patients were treated surgically and the results were retrospectively assessed. Clinical characteristics, imaging data, procedure variables, pathologic features, and postoperative data were recorded and compared to determine the reliability of the model.RESULTS: 3D reconstruction allowed stereoscopic identification of the spatial relationships between physiologic and pathologic structures, and offered quantifiable liver resection proposals based on individualized liver anatomy. The predicted values were consistent with the actual values for tumor mass volume(82.4 ± 109.1 m L vs 84.1 ± 108.9 m L, P = 0.910), surgical margin(10.1 ± 6.2 mm vs 9.1 ± 5.9 mm, P = 0.488), and maximum tumor diameter(4.61 ± 2.16 cm vs 4.53 ± 2.14 cm, P = 0.871). In addition,the number and extent of portal venous ramifications, as well as their relation to hepatic veins, were visualized. Preoperative planning based on simulated resection facilitated complete resection of large tumors located in the confluence of major vessels. 18Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. 19Chemical characterization of size-resolved aerosols in four seasons and hazy days in the megacity Beijing of China显示文摘Size-resolved aerosol samples were collected by MOUDI in four seasons in 2007 in Beijing. The PM10 and PM1.8mass concentrations were 166.0 ± 120.5 and 91.6 ± 69.7 μg/m^3, respectively,throughout the measurement, with seasonal variation: nearly two times higher in autumn than in summer and spring. Serious fine particle pollution occurred in winter with the PM1.8/PM10 ratio of 0.63, which was higher than other seasons. The size distribution of PM showed obvious seasonal and diurnal variation, with a smaller fine mode peak in spring and in the daytime. OM(organic matter = 1.6 × OC(organic carbon)) and SIA(secondary inorganic aerosol) were major components of fine particles, while OM, SIA and Ca_2+were major components in coarse particles. Moreover, secondary components, mainly SOA(secondary organic aerosol) and SIA,accounted for 46%–96% of each size bin in fine particles, which meant that secondary pollution existed all year. Sulfates and nitrates, primarily in the form of(NH_4)_2SO_4, NH_4NO_3, Ca SO_4, Na_2SO_4 and K_2SO_4, calculated by the model ISORROPIA II, were major components of the solid phase in fine particles. The PM concentration and size distribution were similar in the four seasons on non-haze days, while large differences occurred on haze days, which indicated seasonal variation of PM concentration and size distribution were dominated by haze days. 20Seasonal variations and size distributions of water-soluble ions in atmospheric aerosols in Beijing, 2012显示文摘The characteristics of water-soluble ions in airborne particulate matter in Beijing were investigated using ion chromatography. The results showed that the total concentrations of ions were 83.7 ± 48.9 μg/m3 in spring, 54.0 ± 17.0 μg/m3 in summer, 54.1 ± 42.9 μg/m3 in autumn, and 88.8 ± 47.7 μg/m3 in winter, respectively. Furthermore, out of all the ions, NO3-,SO42-and NH4+accounted for 81.2% in spring, 78.5% in summer, 74.6% in autumn, and 76.3%in winter. Mg2+and Ca2+were mainly associated with coarse particles, with a peak that ranged from 5.8 to 9.0 μm. Na+, NH4+and Cl-had a multi-mode distribution with peaks that ranged from 0.43 to 1.1 μm and 4.7 to 9.0 μm. K+, NO3-, and SO42-were mainly associated with fine particles, with a peak that ranged from 0.65 to 2.1 μm. The concentrations of Na+, K+,Mg2+, Ca2+, NH4+, Cl-, NO3-and SO42-were 2.69, 2.32, 1.01, 4.84, 16.9, 11.8, 42.0, and 44.1 μg/m3 in particulate matter(PM) on foggy days, respectively, which were 1.4 to 7.3 times higher than those on clear days.
