1The Msi1-mTOR pathway drives the pathogenesis of mammary and extramammary Paget’s disease显示文摘Mammary and extramammary Paget’s Diseases(PD)are a malignant skin cancer characterized by the appearance of Paget cells.Although easily diagnosed,its pathogenesis remains unknown.Here,single-cell RNA-sequencing identified distinct cellular states,novel biomarkers,and signaling pathways—including mTOR,associated with extramammary PD.Interestingly,we identified MSI1 ectopic overexpression in basal epithelial cells of human PD skin,and show that Msi1 overexpression in the epidermal basal layer of mice phenocopies human PD at histopathological,single-cell and molecular levels.Using this mouse model,we identified novel biomarkers of Paget-like cells that translated to human Paget cells.Furthermore,single-cell trajectory,RNA velocity and lineage-tracing analyses revealed a putative keratinocyte-to-Paget-like cell conversion,supporting the in situ transformation theory of disease pathogenesis.Mechanistically,the Msi1-mTOR pathway drives keratinocyte-Paget-like cell conversion,and suppression of mTOR signaling with Rapamycin significantly rescued the Paget-like phenotype in Msi1-overexpressing transgenic mice.Topical Rapamycin treatment improved extramammary PD-associated symptoms in humans,suggesting mTOR inhibition as a novel therapeutic treatment in PD.Yongli Song Christian F.Guerrero-Juarez Zhongjian Chen Yichen Tang Xianghui Ma Cong Lv Xueyun Bi Min Deng Lina Bu Yuhua Tian Ruiqi Liu Ran Zhao Jiuzhi Xu Xiaole Sheng Sujuan Du Yeqiang Liu Yunlu Zhu Shi-jun Shan Hong-duo Chen Yiqiang Zhao Guangbiao Zhou Jianwei Shuai Fazheng Ren Lixiang Xue Zhaoxia Ying Xing Dai Christopher J.Lengner Bogi Andersen Maksim V.Plikus Qing Nie Zhengquan Yu-Show 2020Cell Research2020,30,10:2
2Population Structure Analysis and Genome-Wide Association Study of Tea(Camellia sinensis(L.)Kuntze)Germplasm in Qiannan,China,Based on SLAF-Seq Technology显示文摘Duyun Maojian tea is a famous tea in China.In this study,the specific-locus amplified fragment(SLAF)sequencing method was used to analyze the population structure and conduct a genome-wide association study(GWAS)of 2 leaf traits of 123 tea plants in Qiannan,China.A total of 462,019 SLAF tags and 11,362,041 single-nucleotide polymorphism(SNP)loci were obtained.The results of phylogenetic tree analysis,cluster analysis,and principal component analysis showed that 123 tea germplasms were clustered into three groups,and the heterozygosity rates for Groups I,II,and II were 0.206,0.224,and 0.34,respectively.Generally,tea germplasms in a production area are clustered in a group,indicating that tea germplasms in different production areas have certain genetic diversity.The traditional Duyun Maojian tea core production areas,TS and DC-SJ,are clustered into Group I and Group II respectively,while the ZY production area is relatively independent in Group III.Furthermore,based on GWAS analysis,11 candidate genes related to leaf apex and 7 candidate genes related to leaf shape were obtained.This study clarified the genetic relationship among eight Duyun Maojian tea production areas and obtained candidate genes related to leaf apex and leaf shape development.The results showed that population structure and candidate genes are an effective basis for the breeding of Duyun Maojian tea germplasm.Fen Zhang Weili Tian Lu Cen Litang Lv Xiaofang Zeng Yulu Chen Yichen Zhao Yan Li 2022Phyton-International Journal of Experimental Botany2022,91,4:1
3Effect of SiO_(2) nano film deposited on copper and aluminium electrodes on the power frequency breakdown voltage of SF_(6) gas显示文摘In order to reveal the effect of insulating nano film on the insulation strength of SF_(6) gas gap,SiO_(2) nano film was deposited on the electrode surface by the plasma-enhanced chemical vapour deposition technique.The power frequency breakdown characteristics of SF_(6) gas under Cu and Al electrodes were obtained before and after coating.The results showed that when the Cu electrode was coated,the breakdown voltage was up to at most 11.5% higher than that of an uncoated Cu electrode in the pressure range of 0.2-0.5 MPa.When the Al electrode was coated,the breakdown voltage of SF_(6) gas was lower than that of an uncoated one in the pressure range of 0.2-0.5 MPa.The breakdown voltage of SF_(6) gas under the Cu electrode was slightly lower than that under the Al electrode,but the breakdown voltages under the coated Cu electrode were significantly higher than that under the coated Al electrode.Through SEM tests,it was found that the surface roughness of the electrode decreases after coating,but particles were formed on the surface of the Al electrode.The content of metal fluoride formed on the coated Al electrode under multiple breakdowns was significantly higher than that on the coated Cu electrode.The research results could provide references for improving insulation performance of GIS equipment.Yu Zheng Shengle Gong Lei Liu Zhihui Xiang Yichen Lv Jianhui Yu Wenjun Zhou 2023High Voltage2023,8,4:0
