| 1 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas. | Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. | 2 | Chemical weathering and CO_2 consumption of a high-erosion-rate karstic river: a case study of the Sanchahe River, southwest China显示文摘The Sanchahe River in southwest China is a tributary of the Wujiang River and experiences high erosion rates. Geochemical analysis was conducted on Sanchahe River basin samples collected in the wet and dry seasons of 2014 in order to better understand local chemical weathering processes, anthropogenic influences, and associated CO2 consumption. The samples' total dissolved solid concentrations were found to be significantly higher than that of the global river average. Ca2+was the dominant cation in the samples and accounted for 64 % and73 % of the total cations in the dry and wet seasons,respectively. HCO3-and SO42-were the dominant anions,accounting for 92 % of the total anions. Stoichiometry analyses of the river waters suggested that the water chemistry is controlled by carbonate dissolution by both carbonic and sulfuric acid. The chemical weathering rates of carbonate and silicate evaporites in the Sanchahe River basin were estimated to be approximately 109.2 and 11.0 t/(km2a), respectively, much higher than both the global mean values and the Wujiang River, a typical karstic river.The CO2 consumption by carbonate and silicate weathering are estimated to be 597.4 9 103 and 325.5 9 103mol/(km2a), which are much higher than corresponding values in the Wujiang River, indicating a high erosion rate in the Sanchahe River basin. | Yanling An Yiliang Hou Qixin Wu Lin Qing Longbo Li | 2015 | Chinese Journal Of Geochemistry2015,34,4: | 4 |
| 3 | Clinical application of traditional herbal medicine in five countries and regions: Japan;South Korea;China's Mainland;Hong Kong, China;Taiwan, China显示文摘Objective:To study the characteristics of the clinical application of traditional herbal medicine in five countries and regions where traditional medicine(TM)is widely used:Japan;South Korea;China's Mainland;Hong Kong,China;Taiwan,China.And to analyze,from multiple angles,the variations in the methods of TM used in each countries and regions.Methods:Fifty commonly used traditional herbs(TOP50)from classical formulae were selected,and descriptive statistics,correlation analyses,and Friedman test were used to compare methods of clinical application and dosages in the countries and regions of interest based on a retrospective review of randomly-selected medical records.Results:There were significant differences in sequencing TOP50 among the five countries and regions(P<.01).The adherence to the classical formulae,from strong to weak was Japan;China's Mainland;Taiwan,China;Hong Kong,China;South Korea.There were also significant differences in TOP50 dosing regimens(P<.01).Overall dosages of TOP50 from high to low were:China's Mainland;Hong Kong,China;Taiwan,China;South Korea;Japan.Finally,there were significant differences in the dosage intervals(P<.01),with dosage intervals from maximum to minimum as follows:China's Mainland;Taiwan,China;Hong Kong,China;South Korea;Japan.Conclusions:The clinical dosages of traditional herbal medicine used in the China's Mainland are relatively large compared with those prescribed in the other four countries and regions,and the range of dosages in China's Mainland is more flexible.On the other hand,clinical dosages of TOP50 are the lowest in Japan,where the dosage range is also the narrowest,supporting the idea that Doctors of Kampo medicine are highly cautious and are likely to adhere strictly to classical formulae,while Hong Kong,China and Taiwan,China are in the middle.The dosage of TOP50 in South Korea is significantly lower than that used in the China's Mainland;Hong Kong,China and Taiwan,China where the adherence to the classical formulae was the lowest,albeit slightly higher than that in Japan.The methods of clinical application of traditional herbal medicine in the five countries and regions were affected by multiple factors,including history,geography,climate,and culture. | Liming Chen Yanling Fu Lin Zhang Sijia Zhao Qing Feng Yihua Cheng Tosha Yanai Daji Xu Mingyu Luo Sang Woo An Woong Seok Lee Seong Hoon Cho Byung Wook Lee | 2015 | Journal of Traditional Chinese Medical Sciences2015,2,3: | 1 |
| 4 | The impact of sulfate reduction on the distribution of methylmercury in reservoirs in the city of Guiyang显示文摘In this paper, the relationship between sulfate reduction potential and mercury methylation potential was studied in the Aha, Baihua and Hongfeng reservoirs from Guiyang City. The methylmercury (MeHg) concentrations of lake water in the Aha Reservoir were greatly elevated as compared to those of the Hongfeng and Baihua reservoirs, which was correlated with its distinctly high SRB abundance, SO42-, and S2- concentrations. Among the three reservoirs, however, the highest MeHg was observed in in the top several centimenters of pore water profile in the Hongfeng Reservoir where the lowest S2- in pore water occurred. The distributions of MeHg in lake water and pore water showed the highest methylation potential occurred at water-sediment surface for the Aha Reservoir and the in the top several centimenters of sediments for the Hongfeng Reservoir. The system firstly uses variational level set active contour withgradients and phase information to complete automatic extraction of the boundaries of thyroid nodules images.Then according to thyroid ultrasound images and clinical diagnostic criteria, a new feature extraction methodbased on the fusion of shape, gray and texture is explored. Due to the imbalance of thyroid sample classes, thispaper introduces a weight factor to improve support vector machine, offering different classes of samples withdifferent weights.
