1Recent progress and challenges in screening and characterization of UGT1A1 inhibitors显示文摘Uridine-diphosphate glucuronosyltransferase 1 A1(UGT1 A1) is an important conjugative enzyme in mammals that is responsible for the conjugation and detoxification of both endogenous and xenobiotic compounds. Strong inhibition of UGT1 A1 may trigger adverse drug/herb–drug interactions, or result in metabolic disorders of endobiotic metabolism. Therefore, both the US Food and Drug Administration(FDA)and the European Medicines Agency(EMA) have recommended assaying the inhibitory potential of drugs under development on the human UGT1 A1 prior to approval. This review focuses on the significance,progress and challenges in discovery and characterization of UGT1 A1 inhibitors. Recent advances in the development of UGT1 A1 probes and their application for screening UGT1 A1 inhibitors are summarized and discussed in this review for the first time. Furthermore, a long list of UGT1 A1 inhibitors, including information on their inhibition potency, inhibition mode, and affinity, has been prepared and analyzed. Challenges and future directions in this field are highlighted in the final section. 2Reversing design methodology of investment casting die profile based on ProCAST显示文摘Turbine blade is one of the critical components of aircraft engine.The performance of the engine depends on the shape and dimensions of components,but superalloy blade material cannot be easily machined.Although investment casting is an ideal process for such net-shape components,it requires an accurate determination of the casting-die profile.In this paper,a reversing design methodology for investment casting die using ProCAST is proposed.By combining the methods of simplifying grid files and quick sorting,the efficiency of sorting and matching can be largely improved.Further,the mould/die cavity anti-deformation system can be easily built.With ProCAST,the optimized die profile for investment casting can be established.Dou Yangqing Bu Kun Dou Yangliu Dong Yiwei 2010China Foundry2010,7,2:10
3Effect of methylmercury on some neurotransmitters and oxidative damage of rats显示文摘In order to study the molecular mechanism of injury in rat organs induced by methylmercury, and the relationship between neurotransmitter and oxidative damage in the toxicity process of rat injury by methylmercury was studied. The control group was physiological saline of 0.9%, the concentration of exposure groups were 5 mg/(kg5d) and 10 mg/(kg5d) respectively. The content of AChE, ACh, NOS, NO, MDA, SOD, GSH-Px and GSH in different organs of rats were determined with conventional methods. The results showed that after exposure to methylmercury for 7 d, the mercury content in brain of exposure groups increased clearly and had significant difference compared with the control group(P<0.01). In rat’s brain, serum, liver and kidney, the content of ACh and AChE were all decreased; the content of NOS and NO were all increased; the content of MDA was increased compared with the control group, the exposure groups had significant difference (P<0.01); the content of SOD, GSH and GSH-Px was decreased compared with the control group, the exposure groups had significant difference(P<0.01). It could be concluded that methylmercury did effect the change of neurotransmitter and free radical. They participated in the toxicity process of injury by methylmercury. 5Kinetically Controlled Cynthesis of Cefaclor using Penicillin G Acylase显示文摘Enzymatic synthesis of cefaclor from 7-aminodesacetoxymethyl-3-chlorocephalosporainc acid(6-ACCA) and phenylglycine derivatives using penicillin G acylase was studied .Many factors that affect the conversion of 7-ACCA to cefaclor were examined.The immobilized enzyme from Bacillis megaterium gave a better catalytic properties and the higher conversion was obtained using phenylglycine methyl ester(PGME) as acyl donor.And the external mass transfer limitation could be eliminated when the stirring rate was more than 150r/min.Low temperature was beneficial for the synthesis and the results showed that the synthetase activity was hardly influenced by temperature while the amidase activity was affected greatly by temperature.The optimum reaction conditions were determined at pH 6.5 and 10℃,respectively.The best 7-ACCA conversion of 56% was achieved when the intial concentration of 7-ACCA and PGME was at 50 mM and 150mM,respectively.YANGLiu WEIDong-zhi XUEPing LUGuao-zhong 2003分子催化2003,17,2:7
6Cross-Reaction of SARS-CoV Antigen with Autoantibodies in Autoimmune Diseases显示文摘To investigate the significance of the SARS-associated coronavirus (SARS-CoV) antibody, detected by ELISA and indirect immunofluorescence assays (IFA) for the SARS-CoV Vero E6 cell lysates, in non-SARS subjects,114 serum samples from healthy controls and 104 serum specimens from autoimmune disease patients were collected. The results of ELISA showed that among 114 sera from healthy controls, 4 (3.5 %) were positive of SARS-CoV-IgG antibody and 114 (100%) were all negative of SARS-CoV-IgM antibody; the specificity of SARS-CoV-IgG antibody for SARS patients was 96.5%, but the specificity of both SARS-CoV-IgG and -IgM antibodies for SARS patients was 100%. In 58 cases with SLE, positive rates of SARS-CoV-IgG and -IgM antibodies were 32.8% (19/58) and 8.6% (5/58), respectively, in which 11 cases (19%) were positive of both SARS-CoV-IgG and -IgM antibodies; in 10 cases with SS, positive rate of both SARS-CoV-IgG and -IgM antibodies was 10% (1/10); in 16 cases with MCTD, positive rate of SARS-CoV-IgG was 37.5% (6/16), positive rate of both SARS-CoV-IgG and -IgM antibodies was 6.3% (1/16); in 20 cases with RA, one case was positive (5%) of SARS-CoV-IgG. However, of all samples with positive SARS-CoV-IgG and -IgM antibodies for autoimmune diseases and healthy controls, SARS-CoV RNA and antibodies were all negative by RT-PCR and IFA. All sera for negative or positive ELISA results were also negative or positive results using ELISA with Vero E6 cells lysates. 7Neurotransmitter level changes in domestic ducks(Shaoxing duck) growing up in typical mercury contaminated area in China显示文摘The neurotransmitter level changes of ducks exposed 8-month in a mercury-polluted site(Wanshan, China) and a reference site(Shanghai, China) were examined. Chemical analyses showed both higher mercury and selenium concentrations in the organ of Wanshan ducks. An increased content of acetylcholine(ACh) in brain and blood and a decreased activity of acetylcholinesterase(AChE) in blood were observed. Moreover, there was an increasing trend for nitric oxide synthase(NOS) activity and nitric oxide(NO) production in duck brain, but a reduction of NOS activity in duck serum. The possible explanations were due to the interactive effect of selenium accumulation and the sublethal exposure level of mercury in Wanshan area. 8Algebraic Conditions for Classifying the Positional Relationships Between Two Conics and Their Applications显示文摘In many fields of computer science such as computer animation, computer graphics, computer aided geometric design and robotics, it is a common problem to detect the positional relationships of several entities.Based on generalized characteristic polynomials and projective transformations, algebraic conditions are derived for detecting the various positional relationships between two planar conics, namely, outer separation, exterior contact, intersection, interior contact and inclusion. Then the results are applied to detecting the positional relationships between a cylinder (or a cone) and a quadric. 9Ultra-small nanodots coated with oligopeptides providing highly negative charges to enhance osteogenic differentiation of hBMSCs better than osteogenic induction medium显示文摘For bone regenerative engineering,it is a promising method to form skeletal tissues differentiating from human bone morrow mesenchyme stem cells(hBMSCs).However,it is still a critical challenge to efficiently control ostogenesis and clearly reveal the influence factor.To this end,the fluorescent gold nanodots(Au NDs) with highly negative charges as osteogenic induction reagent are successfully synthesized,which display better than commercial osteogenic induction medium through the investigations of ALP activity(2.5 folds) and cytoskeleton staining(1.5 folds).Two kinds of oligopeptides with different bio-structures(cysteine,Cys and glutathione,GSH) are selected for providing surficial charges on Au NDs.It is revealed that Au-Cys with more negative charges(-51 mV) play better role than Au-GSH(-19 mV) in osteogenic differentiation,when both of them have same size(~2 nm),sphere shape and show similar cell uptake amount.To explore deeply,osteogenesis related signaling pathways are monitored,revealing that the enhancement of osteogenic differentiation was through autophagy signaling pathway triggered by Au-Cys.And the promotion of highly negative charges in osteogenic diffe rentiation was further proved via sliver nanodots(Ag NDs,Ag-Cys and Ag-GSH) and carbon nanodots(CDs,Cys-CDs and GSH-CDs).This work indicates part of insights during hBMSCs differentiation and provides a novel strategy in osteogenic differentiation process.Yueqi Zhao Lin Meng Kai Zhang Yuanqing Sun Yue Zhao Zhe Yang Yangliu Lin Xinrui Liu Hongchen Sun Bai Yang Quan Lin 2021Chinese Chemical Letters2021,32,1:1
